Claims (17)
1. Gær- eller filamentøs svampe-værtscelle transformeret med et nukleinsy-rekonstrukt omfattende en nukleotidsekvens, der koder xylose isomerase omfattende en aminosyresekvens, som har mindst 70 % sekvensidentitet med aminosyresekvensen SEQ ID NO: 1, bestemt med anvendelse af Need-leman og Wunsch-algoritme, BLOSSUM 62-sammenligningsmatrix, en Gap penalty på 12 og en Gap-længde-penalty på 4, idet nukleinsyrekonstruktet, efter transformation af værtscellen, giver værtscellen evnen til at isomerise xylose til xylulose, hvilket giver værtscellen evnen til at vokse på xylose som carbonkilde.
2. Transformeret værtscelle ifølge krav 1, hvor værtscellen er gær, som tilhører en af slægterne: Saccharomyces, Kluyveromyces, Candida, Pichia, Schi-zosaccharomyces, Hansenula, Kloeckera, Schwanniomyces og Yarrowia.
3. Transformeret værtscelle ifølge krav 2, hvor gæret tilhører en af følgende arter: S. cerevisiae, S. bulderi, S. barnetti, S. exiguus, S. uvarum, S. diastati-cus, K. lactis, K. marxianus og K. fragilis.
4. Transformeret værtscelle ifølge krav 1, hvor værtscellen er en trådagtig svamp, som tilhører en af følgende slægter: Aspergillus, Trichoderma, Humi-cola, Acremonium, Fusarium og Penicillium.
5. Transformeret værtscelle ifølge et hvilket som helst af de foregående krav, hvor nukleotidsekvensen, der koder en xylose-isomerase, er operabelt koblet til en promoter, som forårsager tilstrækkelig ekspression af xylose-isomerasen i værtscellen til at give værtscellen evnen til at isomerisere xylose til xylulose.
6. Transformeret værtscelle ifølge krav 5, hvor promoteren ikke er følsom over for katabolisk repression i værtscellen.
7. Transformeret værtscelle ifølge et hvilket som helst af de foregående krav, hvor værtscellen omfatter en genetisk modifikation, som resulterer i et kendetegn valgt fra gruppen bestående af: (a) forøget transport af xylose ind i værtscellen; (b) forøget xylulose-kinaseaktivitet; (c) forøget flux af pentose phosphat-pathway; (d) nedsat følsomhed over for katabolisk repression; (e) forøget tolerance over for ethanol, osmolaritet eller organiske syrer; og (f) reduceret produktion af biprodukter.
8. Transformeret værtscelle ifølge krav 7, hvor den genetiske modifikation består af overekspression af et endogent gen, ekspression af et heterologt gen, eller en kombination deraf, og hvor det endogene, eller heterologe gen er valgt fra gruppen bestående af et gen, som koder: en hexose- eller pento-se-transporter, en xylulose-kinase; et enzym fra pentose-phosphat-pathway, et glycolytisk enzym og et ethanologent enzym.
9. Transformeret værtscelle ifølge krav 7, hvor den genetiske modifikation består i inaktivering af et endogent gen, hvor genet er valgt fra gruppen bestående af et hexose-kinase-gen, Saccharomyces MIG1 og MIG2 gener og hybridiserende homologer deraf.
10. Transformeret værtscelle ifølge et hvilket som helst af de foregående krav, hvor værtscellen udtrykker et eller flere enzymer, som giver værtscellen evnen til at producere mælkesyre, eddikesyre, raysyre, aminosyrer, 1,3-propan-diol, ethylen, glycerol, β-lactam-antibiotika og cephalosporiner.
11. Transformeret værtscelle ifølge krav 10, hvor værtscellen indeholder en genetisk modifikation, som resulterer i nedsat alkohol-dehydrogenaseaktivitet.
12. Fremgangsmåde til fremstilling af ethanol, hvor fremgangsmåden omfatter trinnene: (a) at fermentere et medium indeholdende en kilde af xylose med en transformeret værtscelle som defineret i et hvilket som helst af kravene 1 - 9, hvorved værtscellen fermenterer xylose til ethanol, og eventuelt (b) genvinde ethanolen.
13. Fremgangsmåde ifølge krav 12, hvor mediet også indeholder en glucose-kilde.
14. Fremgangsmåde ifølge krav 12 eller 13, hvor den volumetriske ethanol-produktivitet er mindst 0,5 g ethanol pr. liter pr. time.
15. Fremgangsmåde ifølge et hvilket som helst af kravene 12 - 14, hvor ethanoludbyttet er mindst 50 % af det teoretiske udbytte på 0,51 g ethanol pr. g glucose eller xylose.
16. Fremgangsmåde til fremstilling af et fermenteringsprodukt valgt fra gruppen bestående af mælkesyre, eddikesyre, ravsyre, aminosyrer, 1,3-propan-diol, ethylen, glycerol, β-lactam-antibiotika og cephalosporiner, hvor fremgangsmåden omfatter trinnene: (a) at fermentere et medium indeholdende en kilde af xylose med en transformeret værtscelle som defineret i krav 10 eller 11, hvorved værtscellen fermenterer xylose til fermenteringsproduktet, og eventuelt (b) genvinde fermenteringsproduktet.
17. Fremgangsmåde ifølge krav 16, hvor mediet også indeholder en glucose-kilde.
