DEUTSCHES PATENTAMTGERMAN PATENT OFFICE
The invention relates to the recovery of glutamic acid from protein hydrolysates, in particular from the hydrolysates of gluten.
The known processes for producing and recovering glutamic acid from proteinaceous materials such as gluten include hydrolysis with acid under suitable conditions. The resulting hydrolyzate, which contains the individual amino acids in acidic solution, is treated in various ways in order to obtain the individual amino acids or their mixtures. For example, tyrosine, and leucine can thus be that the pH value of the hydrolyzate and applied to approximately 4 to 7, after removal of insoluble impurities, the solution is concentrated above the saturation point of tyrosine and leucine also removed. After removal of the p H value is the remaining solution to a value of about 3.2 which is the isoelectric point of Glutammsäure set, in which direct crystallization of glutamic acid is possible.
Ferner bestehen Korro-However, none of the known methods lead to ashless glutamic acid without assistance long, complicated and costly treatment. When hydrochloric acid as a hydrolyzing agent is used, sodium hydroxide is generally used as a neutralizing agent. That I The resulting sodium chloride is not a deleterious contaminant of glutamic acid, but it can pass into the mother liquor, from which glutamic acid crystallizes. Furthermore, there are corrosive

Sulfuric acid is cheaper and less corrosive than hydrochloric acid, but the by-products resulting from its use, namely sodium sulfate and calcium sulfate, are harmful impurities of glutamic acid, and in addition, the former contaminates the mother liquor.
Ziel der Erfindung ist ein neues Verfahren zurThe aim of the invention is a new method for
ίο Gewinnung von Glutaminsäure in guter Ausbeute und in einem hohen Reinheitsgrad aus Schwefelsäurehydrolysaten von Glutenen, welche im wesentlichen aschefrei und leicht und in hohen Ausbeuten in Mononatriumglutamat überführbarίο Obtaining glutamic acid in good yield and in a high degree of purity from sulfuric acid hydrolysates of gluten, which are essentially ash-free and easily convertible into monosodium glutamate in high yields

It has been found that glutamic acid is obtained in good yield and with a high degree of purity A sulfuric acid hydrolyzate can be obtained from gluten when a new ash removal stage is switched on under critically controlled conditions.

N to 7 is used for the purpose of removing leucine and tyrosine, the greater part of the resulting calcium sulfate can easily, e.g. B. by filtration, but the remaining calcium sulfate, as well as the calcium bound to the amino acids, can only be removed with great difficulty, if at all. In general, it later crystallizes with the glutamic acid. It has been found that the concentrate contained in the last glutamic acid back 'residual calcium sulphate can be brought into a state which enables easy removal when the first pH value of the concentrate to 2.8 to 3.5, preferably, 3 , 2, adjusted with sulfuric acid and then the liquid is quickly heated to 65 to 100 °, preferably to about 80 °, and held at this temperature for 5 to 30 minutes, preferably about 15 minutes at 80 °. A possible and useful measure in the process is the addition of decolorizing carbon after adjusting the p H -value before the heat treatment. After the heat treatment, the hot liquid should be filtered immediately, the filtrate should be cooled and the glutamic acid should be crystallized out. The heat treatment described converts the calcium sulfate into an easily filterable form and prevents premature crystallization of the glutamic acid, which could otherwise be lost at this point. By adding decolorizing charcoal at this point, the subsequent crystallization of glutamic acid is favored and dyes are removed. The crystals of glutamic acid are larger, more uniform, and easier to filter than if the charcoal treatment was omitted.
According to the invention it is possible in a single Crystallization of glutamic acid in a purity of 94 to 98% and above and with a ash as low as 0.04 per cent. This is an advantage over the known methods, which ', usually, to \. Ash content of 4 to 7% lead. Accordingly, the invention represents a great advance in the art because it for the first time the advantages of sulfuric acid as a hydrolyzing agent for proteins without the disadvantages the difficult removal of insoluble ash from glutamic acid.
