Description
Verfahren zur Gewinnung von kristallinem Dihydrostreptomycin-Hydrochlorid Gegenstand der Erfindung ist ein Verfahren zur Herstellung von reinem kristallinen Dihydrostrel)tomycin-Hydrochlorid. Die kristalline Verbindung wird erfindungsgemäß dadurch gewonnen, (laß eine Lösung von 1)iliydrostreiitoinycin-Hydrochlorid in wasserfreiem Methanol oder im wesentlichen wasserfreien Methanol hergestellt wird, der relative Betrag von Methanol derart vermindert wird, daß eine übersättigte Lösung entsteht, die übersättigte Lösung mit Spuren von Kristallen von 1)ihydrostreptomycin-Hydrochlorid lwiinlift wird und die Lösung längere Zeit in Bew-egung gehalten wird, wobei Kristallisation des 1)ilrydrostrel>tomycili-Hydroc'hlorids stattfinden. Dies ist wahrscheinlich darauf zurückzuführen, daß sehr feine Kriställchen von Dihydrostreptomycin-Hydrochlorid ;in der Apparatur oder der Atmosphäre vorhanden sind. Hierbei ist die Entfernung von Calciumchlori,d aus dem Reaktionsgemisch im Anschluß an die Reduktion von primärer Bedeutung. Dieser Schritt umfaß@t auch dass Niederschlagen von Dihydrostreptomycin-Hydrochloriid aus der methanolischen Calciumchlorid enthaltenden Reduktionsmischung mit Isopropanol. Hierbei wird die Methanollösung vorzugsweise dem Isopropanol zugefügt. Der so entstehendie amorphe Niederschlag von Dihydrostreptomycin-Hydrochlorild kann leicht durch 'Filtration gewonnen werden.
