Die
vorliegende Erfindung betriftt ein Verfahren zur Herstellung optisch
aktiver Alkohole entsprechend der Ansprüche. Genauer betrifft die vorliegende
Erfindung ein neues, hoch zweckmäßiges Verfahren
zur Herstellung optisch aktiver Alkohole, die für verschiedene Anwendungen,
wie als Zwischenprodukte zur Synthese pharmazeutischer Chemikalien,
Flüssigkristall-Materialien
und Mittel zur Racemattrennung brauchbar sind.

Verschiedene
Verfahren zur Herstellung optisch aktiver Verbindungen sind bislang
bekannt. Als Verfahren zur asymmetrischen Synthese optisch aktiver
Alkoholverbindungen sind beispielsweise die folgenden Verfahren
bekannt:
- (1) ein Verfahren, das Enzyme wie
Bäckerhefe
verwendet; und
- (2) ein Verfahren zur asymmetrischen Hydrierung von Carbonylverbindungen
unter Verwendung von Metall-Komplex-Katalysatoren. Besonders für das Verfahren
(2) gibt es eine große
Zahl von Beispielen asymmetrischer Katalysereaktionen, wie beispielsweise:
(1) Die asymmetrische Hydrierung von Carbonylverbindungen mit funktionellen
Gruppen durch optisch aktive Rutheniumkatalysatoren, wie detailliert
in Asymmetric Catalysis in Organic Synthesis, Hrsg. R. Noyori.,
S. 56-82 (1994), beschrieben; (2) ein Verfahren über eine Wasserstofftransfer-artige
Reduktion mittels chiraler Komplexkatalysatoren von Ruthenium, Rhodium oder
Iridium, wie in Chem. Rev., Vol. 92, S. 1051-1069 (1992), beschrieben;
(3) ein Verfahren zur asymmetrischen Hydrierung mit Weinsäure mittels
eines veränderten
Nickelkatalysators mit Weinsäure,
wie in Oil Chemistry, S. 882-831 (1980) und Advances in Catalysis,
Vol. 32, S. 215 (1983), Hrsg. Y. Izumi, beschrieben; (4) ein asymmetrisches
Hydrosilylierungs-Verfahren, wie in Asymmetric Synthesis, Vol. 5,
Kap. 4 (1985), Hrsg. J. D. Morrison und J. Organomet. Chem. Vol.
346, S. 413-424 (1988), beschrieben; und (5) ein Boranreduktions-Verfahren
in Gegenwart eines chiralen Liganden, wie in J. Chem. Soc., Perkin
Trans. 1, S. 2039-2044 (1985) und J. Am. Chem. Soc., Vol. 109, S.
5551-5553 (1987), beschrieben.

Mit
dem herkömmlichen
Verfahren mittels Enzymen können
Alkohole mit einer relativ hohen optischen Reinheit isoliert werden,
aber die hierfür
geeigneten Reaktionssubstrate sind begrenzt, und die absolute Konfiguration
der resultierenden Alkohole ist auf eine spezifische festgelegt.
Durch das asymmetrische Hydrierungs-Verfahren mittels eines Übergangsmetall-Komplex-Katalysators
können
optisch aktive Alkohole mit hoher Selektivität hergestellt werden, jedoch
muss ein druckresistenter Reaktor hierfür verwendet werden, da Wasserstoffgas
als Quelle für
Wasserstoff verwendet wird, was bezüglich der Betriebsschwierigkeit
und -sicherheit unvorteilhaft ist. Des Weiteren unterliegt das Verfahren über die
asymmetrische Wasserstofftransfer-artige Reduktion unter Verwendung
von herkömmlichen
Metallkomplex-Katalysatoren Beschränkungen, da dieses Verfahren
Reaktionsbedingungen unter Erhitzen benötigt und die Selektivität der Reaktion
ungenügend ist,
was aus praktischen Gesichtspunkten von Nachteil ist.

Demzufolge
besteht bisher ein Bedürfnis
nach einem neuen, allgemein anwendbaren Verfahren zur Synthese optisch
aktiver Alkohole unter Verwendung eines hoch aktiven und hoch selektiven
Katalysators und ohne die Verwendung von gasförmigem Wasserstoff.

Doch
bislang hat sich kein hoch effizientes und hoch selektives Verfahren
zur Herstellung solcher sekundären
Alkohole über
eine asymmetrische Synthesereaktion mittels Katalysatoren, die den
oben beschriebenen ähnlich
sind, etabliert.
-
Bezüglich der
optisch aktiven sekundären
Alkohole ist ein Verfahren zur Herstellung optisch aktiver sekundärer Alkohole
durch Racemattrennung von racemischen sekundären Alkoholen für einige
Reaktionssubstrate bekannt, die nur schwer reduziert werden können, obwohl
eine exzellente optische Reinheit kaum erreicht wird (Asymmetric
Catalysis in Organic Synthesis, Hrsg. R. Noyori). Da die Wasserstofftransfer-artige
Reduktion nach dem Verfahren eine reversible Reaktion ist, wird
die Dehydrierungs-Oxidation als ihre Gegenreaktion gemäß dem Verfahren
verwendet. Daher wird dieses Verfahren als ein kinetisches Verfahren
zur Racemattrennung bezeichnet. Gemäß dieser Methode wurde jedoch
noch kein Verfahren zur Herstellung optisch aktiver sekundärer Alkohole
mit Katalysatoren mit hoher Effizienz beschrieben.

Zassinovich

beschreiben asymmetrische Wasserstofftransfer-Reaktionen, die durch
homogene Übergangsmetall-Katalysatoren
vorangetrieben werden. Ruthenium(II)-Katalysatoren werden beschrieben.

Krasik
und Alper, Tetrahedron, Vol. 50, Nr. 15, 11. April 1994, S. 4347-4354
beschreiben Schiff'sche Basen
als zugegebenen chiralen Liganden für die von [Ru(η6-C6H6)Cl2]2-katalysierte Wasserstofftransfer-Reduktion
von Ketonen mit 2-Propanol.

Andererseits
wird eine große
Zahl von Übergangsmetall-Komplexen
als Katalysatoren für
Organometall-Reaktionen verwendet; insbesondere aufgrund der hohen
Aktivität
und Stabilitäts
und einer daraus resultierenden einfachen Handhabbarkeit von Seltenerdmetallkomplexen
wurden trotz der hohen Kosten synthetische Reaktionen entwickelt,
die sich dieser Komplexe bedienen. Der Fortschritt dieser asymmetrischen
Synthesereaktionen, die chirale Komplexkatalysatoren verwenden,
ist bahnbrechend; es sind viele Berichte erschienen, die die Verwirklichung
hoch effizienter organischer Synthesereaktionen beschreiben.

Darunter
sind bereits viele asymmetrische Reaktionen, die chirale Komplex-Katalysatoren mit
optisch aktiven Phosphin-Liganden als Katalysator verwenden, entwickelt
worden, und einige davon werden industriell angewendet (Asymmetric
Catalysis in Organic Synthesis, Hrsg. R. Noyori).

Bei
den Komplexen von optisch aktiven Stickstoffverbindungen, die an Übergangsmetalle
wie Ruthenium, Rhodium und Iridium koordiniert sind, sind viele
solche Komplexe bekannt, die zusätzlich
exzellente Eigenschaften als Katalysatoren für eine asymmetrische Synthese-Wirkung haben. Um
die Eigenschaften dieser Katalysatoren zu verbessern, gibt es viele
Vorschläge,
die die Verwendung optisch aktiver Stickstoffverbindungen mit spezifischen
Strukturen betreffen (Chem. Rev., Vol. 92, S. 1051-1069 (1992)).

Beispielsweise
erschienen Berichte über
(1) optisch aktive 1,2-Diphenylethylendiamine und Rhodium-Diamin-Komplexe
mit Cyclohexandiamin-Liganden, wie in Tetrahedron Asymmetry, Vol.
6, S. 705-718 (1995) beschrieben; (2) Rhuthenium-Imid-Komplexe mit
Bisaryliminocyclohexan-Liganden, wie in Tetrahedron Vol. 50, S.
43474354 (1994) beschrieben; (3) Iridium- Pyridin-Komplexe mit Pyridinliganden,
wie in Japanese Patent Offenlegung Nr. 62-281861 und 63-119465;
(4) optisch aktive 1,2-Diphenylethylendiamine oder Iridium-Diamin-Komplexe
mit Cyclohexandiaminliganden, wie im japanischen Patent mit der
Veröffentlichungs-Nr.
JP 62273990 beschrieben;
(5) Ruthenium-Diamin-Komplexe von RuCl[p-TsNCH(C
6H
5)CH(C
6H
5)-NH
2](aren)(chlor-(N-p-toluolsulfonyl-1,2-diphenylethylendiamin)(aren)ruthenium)(Aren
steht hier für
Benzol, das sowohl unsubstituiert wie auch substituiert sein kann),
hergestellt durch Koordination von Ruthenium mit optisch aktivem
N-p-Toluolsulfonyl-1,2-diphenylethylendiaminen
[hier als „p-TsNHCH(C
6H
5)CH(C
6H
5)NH
2" bezeichnet wird],
wie in J. Am. Chem. Soc., Vol. 117, S. 7562-7563 (1995); J. Am.
Chem. Soc., Vol. 118, S. 2521-2522 (1996) und in J. Am. Chem. Soc.,
Vol. 118, S. 4916-4917 (1996), beschrieben.

Auch
wenn diese Komplexe verwendet werden, bleiben gegenwärtig dennoch
Probleme, die zur praktischen Anwendung überwunden werden müssen, wie
ungenügende
Katalysator-Aktivität, Nachhaltigkeit
und optische Reinheit, abhängig
von den jeweiligen Reaktionen und Reaktions-Substraten.

Um
die zuvor genannten Probleme zu überwinden,
stellt die vorliegende Erfindung ein Verfahren zur Herstellung optisch
aktiver sekundärer
Alkohole bereit, wobei man racemische sekundäre Alkohole oder meso-artige
Diole einer Wasserstofftransfer-Reaktion in Gegenwart optisch aktiver
Ruthenium-Diamin-Komplex-Katalysatoren, die durch folgende allgemeine
Formel (VII) dargestellt werden, unterwirft
wobei * für ein asymmetrisches Kohlenstoffatom
steht, R
01 und R
02 gleich
oder verschieden sind, und unabhängig
voneinander für
eine Alkylgruppe oder eine Phenylgruppe oder eine Cycloalkylgruppe,
die unsubstituiert oder mit einer Alkylgruppe substituiert sein
kann, stehen; oder R
01 und R
02 gemeinsam
einen alicyclischen Ring bilden, der unsubstituiert oder mit einer
Alkylgruppe substituiert ist; R
03 für eine Methansulfonylgruppe,
Trifluormethansulfonylgruppe, Naphthylsulfonylgruppe, Kampfersulfonylgruppe
oder Benzolsulfonylgruppe, die unsubstituiert oder mit einer Alkylgruppe,
einer Alkoxylgruppe oder einem Halogenatom substituiert ist, oder
eine Benzoylgruppe, die unsubstituiert oder mit einer Alkoxycarbonylgruppe
oder Alkylgruppe substituiert ist, steht; R
04 für ein Wasserstoffatom
oder eine Alkylgruppe steht; X für
eine aromatische Verbindung die unsubstituiert oder mit einer Alkylgruppe
substituiert ist, steht; und m und n jeweils für 0 oder 1 stehen.

Gemäß der vorliegenden
Erfindung werden die oben beschriebenen charakteristischen Verfahren
zur Herstellung optisch aktiver Verbindungen bereitgestellt. Die
Einzelheiten sind nachstehend beschrieben.

Das
für die
erfindungsgemäße Wasserstofftransfer-artige
asymmetrische Reduktion zu verwendende Katalysator-System ist sehr
charakteristisch und bislang unbekannt.
-
Der
optisch aktive Ruthenium-Diamin-Komplex, der wie oben beschrieben
durch die folgende Formel (VII) dargestellt ist, als ein aus einem Übergangsmetall
und einer optisch aktiven Stickstoff-haltigen Verbindung als Ligand
bestehender Metallkomplex, ist als Katalysator zur Herstellung optisch
aktiver sekundärer
Alkoholverbindungen geeignet, wobei man racemische sekundäre Alkohole
oder meso-Typ-Diole einer Wasserstoff-Transferreaktion unterwirft;
daher kommt diesem Komplex größere Bedeutung
zu.

Zur
genaueren Beschreibung der optisch aktiven Ruthenium-Diamin-Komplexe
der Formel (VII) kann die durch X dargestellte aromatische Verbindung,
die unsubstituiert oder mit einer Alkylgruppe, beispielsweise C1-C4-Alkylgruppen,
substituiert ist, beispielsweise Benzol, Toluol, Xylol, Mesitylen,
Hexamethylbenzol, Ethylbenzol, tert-Butylbenzol, p-Cymol und Cumol
bedeuten und umfasst bevorzugt Benzol, Mesitylen und p-Cymol.
-
R01 und R02 können lineare
oder verzweigte Alkylgruppen, beispielsweise C1-C4-Alkylgruppen
sein. Genauer können
die Alkylgruppen Methyl, Ethyl, n-Propyl, Isopropyl, n-, iso-, sec-
und tert-Butyl, sein. Besonders bevorzugt ist die Gruppe Methyl,
Ethyl, n-Propyl oder iso-Propyl.

Wenn
R01 und R02 miteinander
verbunden sind und eine alicyclische Gruppe bilden, kann diese Gruppe geeigneterweise
ein C5-C7-gliedriger
Ring sein. Die Alkylgruppe, die gegebenenfalls ein Substituent hiervon sein
kann, ist beispielsweise eine C1-C4-Alkyl-Substituenten-Gruppe, wozu die Methylgruppe, Ethylgruppe, n-Propylgruppe,
Isopropylgruppe und n-, iso-, sec- und tert-Butylgruppe zählen. Vorzugsweise
ist die Alkylgruppe Methyl.

R01 und R02 in Form
einer Phenylgruppe, wobei R01 und R02 eine Alkylgruppe, beispielsweise eine
Methylgruppe, tragen können,
umfassen insbesondere Phenyl, o-, m- und p-Tolylgruppen.

R01 und R02, die für eine Cycloalkylgruppe
stehen, enthalten Kohlenstoffatome in einem 5 bis 6-gliedrigen Ring,
wozu bevorzugt Cyclopentyl oder Cyclohexyl zählen.

In
stärker
bevorzugten Beispielen stehen R01 und R02 unabhängig
voneinander für
Phenyl oder R01 und R02 stehen
gemeinsam für
Tetramethylen (-(CH2)4-).

R03 steht für eine Methansulfonylgruppe,
eine Trifluormethansulfonylgruppe, eine Naphthylsulfonylgruppe,
eine Kampfersulfonylgruppe oder eine Benzolsulfonylgruppe, die unsubstituiert
oder mit einer Alkylgruppe, z. B. einer C1-C3-Alkylgruppe, einer Alkoxygruppe, z. B.
einer C1-C3-Alkoxygruppe,
oder einem Halogenatom substituiert ist, oder eine Benzoylgruppe,
die mit einer Alkoxycarbonylgruppe, beispielsweise einer C1-C4-Alkoxycarbonylgruppe,
oder einer Alkylgruppe, beispielsweise C1-C4-Alkylgruppe, substituiert sein kann.

Genauer
steht R03 für eine Benzylsulfonylgruppe,
die unsubstituiert oder mit einer C1-C3-Alkylgruppe, einer
C1-C3-Alkoxylgruppe
oder einem Halogenatom substituiert sein kann, und umfasst Benzylsulfonyl,
o-, m- und p-Toluolsulfonyl, o-, m- und p-Ethylbenzylsulfonyl, o-,
m- und p-Methoxybenzylsulfonyl, o-, m- und p-Ethoxybenzylsulfonyl,
o-, m- und p-Chlorbenzylsulfonyl, 2,4,6-Trimethylbenzylsulfonyl,
2,4,6-Triisopropylbenzylsulfonyl, p-Fluorbenzylsulfonyl und Pentafluorbenzylsulfonyl,
und umfasst besonders bevorzugt Benzylsulfonyl oder p-Toluylsulfonyl.
Genauer umfasst R03 das für C1-C4-Alkoxycarbonyl
steht, Methoxycarbonyl, Ethoxycarbonyl, Isopropyloxicarbonyl, und
tert-Butoxycarbonyl, bevorzugt beinhaltet es Methoxycarbonyl oder
tert-Butoxycarbonyl. Genauer beinhaltet R03,
das für
eine Benzoylgruppe steht, die unsubstituiert oder mit einer C1-C4-Alkylgruppe
substituiert sein kann, Benzoyl, o-, m- und p-Methylbenzoyl, o-,
m- und p-Ethylbenzoyl, o-, m- und p-Isopropylbenzoyl, und o-, m-
und p-tert-Butylbenzoyl,
bevorzugt umfasst es Benzoyl oder p-Methylbenzoyl.

In
den besonders bevorzugten Beispielen steht R03 für Methansulfonyl,
Trifluormethansulfonyl, Benzylsulfonyl oder p-Toluolsulfonyl. R04, das für
ein Wasserstoffatom oder eine Alkylgruppe, beispielsweise C1-C4-Alkylgruppe
steht, umfasst besonders z. B. Wasserstoff, Methyl, Ethyl, n-Propyl,
Isopropyl, n-, iso-, sec- und tert-Butyl, und besonders bevorzugt
umfasst es ein Wasserstoffatom oder eine Methylgruppe.

Bevorzugt
werden die racemischen sekundären
Alkohole, die erfindungsgemäß als Rohstoff-Verbindungen
verwendet werden, durch die folgende Formel (VIII) dargestellt.
Die racemischen Alkohole sind selbstverständlich nicht auf die von der
Formel dargestellten beschränkt.

R6 steht für
eine aromatische monocyclische oder polycyclische Kohlenwasserstoffgruppe,
die unsubstituiert oder substituiert sein kann, oder für eine heteromonocyclische
oder polycyclische Gruppe, die Heteroatome, wie Stickstoff, Sauerstoff,
Schwefelatome und dergleichen enthält, und steht besonders für aromatische
monocyclische oder polycyclische Gruppen, wie Phenylgruppen, 2-Methylphenyl,
2-Ethylphenyl, 2-Isopropylphenyl, 2-tert-Butylphenyl, 2-Methoxyphenyl, 2-Chlorphenyl,
2-Vinylphenyl, 3-Methylphenyl, 3-Ethylphenyl, 3-Isopropylphenyl,
3-Methoxyphenyl, 3-Chlorphenyl, 3-Vinylphenyl, 4-Methylphenyl, 4-Ethylphenyl,
4-Isopropylphenyl,
4-tert-Butylphenyl, 4-Vinyphenyl, Cumenyl, Mesityl, Xylyl, 1-Naphthyl,
2-Naphthyl, Anthryl, Phenanthryl,
und Indenyl; hetero-monocyclische oder polycyclische Gruppen wie
Thienyl, Furyl, Pyranyl, Xanthenyl, Pyridyl, Pyrrolyl, Imidazolyl,
Indolyl, Carbazoyl, und Phenthronylyl; und Ferrocenyl. Des Weiteren
steht R7 für ein Wasserstoffatom, eine
gesättigte
oder ungesättigte
Kohlenwasserstoff-Gruppe, oder eine funktionelle Gruppe, die Heteroatome
enthält,
wozu beispielsweise Alkylgruppen wie Methyl, Ethyl, Propyl, Isopropyl, Butyl,
Pentyl, Hexyl, und Heptyl; Cycloalkylgruppen, wie beispielsweise
Cyclopropyl, Cyclobutyl, Cyclopentyl und Cyclohexyl; und ungesättigte Kohlenwasserstoffe
wie Benzyl, Vinyl, und Allyl zählen.
R6 und R7 können unter
Ausbildung eines Rings miteinander verbunden sein, und in diesem
Fall umfasst R7 beispielsweise eine gesättigte oder
ungesättigte
alicyclische Gruppe, die ein cyclisches Keton, wie beispielsweise
Cyclopentanon, Cyclohexanon, Cycloheptan, Cyclopentenon, Cyclohexenon,
und Cycloheptenon bildet; oder eine gesättigte und ungesättigte alicyclische
Gruppe mit einer Substituentengruppe, die eine Alkylgruppe, eine
Arylgruppe, eine ungesättigte
Alkylgruppe oder eine lineare oder cyclische Kohlenwasserstoffgruppe
an jedem der einzelnen Kohlenstoffatome aufweist.

Zusätzlich werden
die meso-artigen Diole beispielsweise durch die folgende Formel
(IX) dargestellt.

Die
meso-Diole sind selbtverständlich
nicht auf die durch die Formel dargestellten beschränkt.

In
diesem Fall sind R8 und R9 gleich
und stehen für
eine gesättigte
oder ungesättigte
Kohlenwasserstoffgruppe, die unsubstituiert oder substituiert ist,
oder R8 und R9 können miteinander
verbunden sein, um eine gesättigte
oder ungesättigte
alicyclische Gruppe zu bilden, die unsubstituiert oder substituiert
sein kann.

Genauer
kann der Ruthenium-Diamin-Komplex in der vorliegenden Erfindung
beispielsweise so sein, dass m und n in der Formel (VII) gleichzeitig
Null sind. Vorliegend steht η für die Anzahl
an Kohlenstoffatomen, die bei ungesättigten Liganden an das Metall
gebunden sind, und daher wird Hexahapto (6 Kohlenstoffatome an das
Metall gebunden) durch η6 dargestellt; p-Ts steht für p-Toluolsulfonylgruppen;
Ms steht für
eine Methansulfonylgruppe; und Tf steht für eine Trifluormethansulfonyl-Gruppe.

Unter
den Verbindungen, die durch die allgemeine Formel (VII) dargestellt
werden, können
Komplexe der Formel (VII), bei denen m und n gleichzeitig 0 sind,
wie im Folgenden beschrieben, hergestellt werden. Genauer kann Ru[(S,S)-,(R,R)-TsNCH(R01)CH(R02)NH](η6-p-cymol)(((S,S)
und (R,R)-N-toluolsulfonyl-1,2-disubstituiertes Ethylendiamin)(η6-p-cymol)ruthenium
(wobei R01 und R02 gleich
sind, wie oben beschrieben und Ts eine p-Toluolsulfonylgruppe ist), durch Umsetzung
eines Rohmaterials [RuC12(η6-p-cymol)]2(tetrachlorbis(η6-p-cymol)diruthenium),
das nach dem in J. Chem. Soc., Dalton Trans., S. 233-241 (1974)
beschriebenen Verfahren hergestellt ist, mit (S,S)-, (R,R)-TsNHCH(R01)CH(R02)NH2((S,S) und
(R,R)-N-p-toluolsulfonyl-1,2-disubstituiertem Ethylendiamin) in
Gegenwart eines Alkalimetallhydroxids oder eines Alkalimetallalkoholats
in einem Lösungsmittel,
hergestellt werden.
-
Die
Umsetzung verläuft
im Allgemeinen quantitativ, wenn man ein Rohmaterial [RuCl2(η6-p-cymol)]2(tetrachlorbis(η6-p-cymol)diruthenium (1 Mol) und (S,S)-,
(R,R)-TsNHCH (R01)CN(R02)NH2(((S,S) und (R,R)-N-p-toluolsulfonyl-1,2-disubstituiertes
Ethylendiamin) (2 Mol) mit Alkalimetallhydroxid oder Alkalimetallalkoholate
in einem Schutzgasstrom, wie Stickstoff, Helium oder Argon, in einem
inerten Lösungsmittel,
bei Temperaturen von –10
bis 50°C,
für 30
Minuten bis zu 3 Stunden, umsetzt und das Reaktionsprodukt stehen lässt, bevor
man durch ein Verfahren zur Flüssigkeitsabtrennung
die wässrige
Phase entfernt und anschließend
das Lösungsmittel
unter vermindertem Druck entfernt.

Das
Alkalimetallhydroxid oder Alkalimetallalkoholat umfasst insbesondere
NaOH, NaOCH3, NaOC2H5, KOH, KOCH3, KOC2H5, LiOH, LiOCH3, und LiOC2H5, besonders bevorzugt umfasst es NaOH oder
KOH. Die Menge an Alkalimetallhydroxid oder Alkalimetallalkoholat
beträgt
das 5 bis 10 fache der Menge an Ruthenium. Das inerte Lösungsmittel
umfasst geeigneterweise beispielsweise Kohlenwasserstoffe wie Benzol,
Toluol, Xylol, Cyclohexan, und Methylcyclohexan; Ether, wie Dimethylether,
Diethylether, Isopropylether, Methyl-tert-butylether, Tetrahydrofuran,
1,3-Dioxolan, und 1,4-Dioxan; halogenierte Kohlenwasserstoffe, wie
Chloroform, Methylenchlorid und Chlorbenzol.

Der
Komplex kann durch ein weiteres Verfahren hergestellt werden. Genauer
kann Ru[(S,S)-, (R,R)-TsNHCH(R01)CH(R=2)NH](η6-p-cymol)(((S,S) und (R,R)-N-p-toluolsulfonyl-1,2-disubstituiertes
Ethylendiamin)(η6-p-cymol)ruthenium (wobei R01 und
R02 gleich sind wie oben beschrieben und
Ts für
p-Toluolsulfonylgruppe steht) durch Umsetzung eines Rohprodukts

Ethylendiamin) mit einem tertiären
Amin (beispielsweise Triethylamin) hergestellt ist, wie beispielsweise
in J. Am. Chem. Soc., Vol. 117, S. 7562-7563 (1995), J. Am. Chem.
Soc., Vol. 118, S. 2521-2522 (1996) und J. Am. Chem. Soc., Vol.
118, S. 4916-4917 (1996) beschrieben, in Gegenwart von Alkalimetall-Hydroxiden oder Alkalimetall-Alkoholaten
in einem Lösungsmittel
leicht hergestellt werden.

Die
Umsetzung verläuft
m Allgemeinen quantitativ, wenn man RuCl[(S,S)-,(R,R)-TsNCH(R01)CH(R02)NH2](η6-p-cymol)(Chlor-((S,S) und (R,R)-N-p-toluolsulfonyl-1,2-disubstituiertes
Ethylendiamin)(η6-p-cymol)ruthenium (1 Mol) als Rohmaterial
mit Alkalimetallhydroxiden oder Alkalimetallalkoholaten in einen
Schutzgasstrom, wie Stickstoff, Helium oder Argon, in einem inerten
Lösemittel,
bei Temperaturen von –10
bis 50°C
für 30
Minuten bis zu 3 Stunden umsetzt und das Reaktionsprodukt stehen lässt, bevor
man die wässrige
Phase durch ein Flüssigkeits-Trennungs-Verfahren
entfernt und anschließend das
Lösungsmittel
unter vermindertem Druck entfernt.
-
Das
Alkalimetall-Hydroxid oder Alkalimetall-Alkoholat umfasst insbesondere
NaOH, NaOCH3, NaOC2H5, KOH, KOCH3, KOC2H5, LiOH, LiOCH3, und LiOC2H5, bevorzugt umfasst es NaOH oder KOH. Die Menge
an Alkalimetall-Hydroxid oder Alkalimetall-Alkoholat beträgt 1 bis
2 Mal die Menge in Mol an Ruthenium. Das inerte Lösungsmittel
umfasst angebrachter Weise beispielsweise Kohlenwasserstoffe wie
Benzol, Toluol, Xylol, Cyclohexan und Methylcyclohexan; Ether wie
Dimethylether, Diethylether, Diisopropylether, Methyl-tert-Butylether,
Tetrahydro furan, 1,3-Dioxolan und 1,4-Dioxan; und halogenierte Kohlenwasserstoffe
wie Chloroform, Methylenchlorid und Chlorbenzol.

Der
Komplex, der durch die allgemeine Formel (V) dargestellt wird, wobei
m und n gleichzeitig 1 sind, kann wie im Folgenden beschrieben,
hergestellt werden. Genauer kann RuH[(S,S)-,(R,R)-TsNCH(R01)CH(R02)NH2](η6-p-cymol)(Hydrid-((S,S) und (R,R)-N-toluolsulfonyl-1,2-disubstituiertes
Ethylendiamin)(η6-p-cymol)ruthenium) (wobei R01 und
R02 gleich sind wie oben beschrieben und
Ts eine p-Toluolsulfonylgruppe ist), auf einfache Weise hergestellt
werden, indem man ein Rohmaterials in Form von Ru[(S,S)-, (R,R)-TsNHCH(R01)CH(R02) NH](η6-p-cymol)(((S,S,) und (R,R)-N-Toluolsulfonyl-1,2-disubstituiertes
Ethylendiamin)(η6-p-cymol)ruthenium)
(wobei R01 und R02 gleich
sind wie oben definiert; und Ts für eine p-Toluolsulfonylgruppe
steht) in einem Alkohol-Lösungsmittel
umsetzt. Die Reaktion wird gewöhnlich
quantitativ durchgeführt,
indem man Ru[(S,S)-,(R,R)-TsNCH(R01)CH(R02)NH](η6-p-cymol) (((S,S,) und (R,R)-N-toluolsulfonyl-1,2-disubstituiertes
Ethylendiamin)(η6-p-cymol)ruthenium) (wobei R01 und
R02 gleich sind wie oben definiert; und
Ts für
eine p-Toluolsulfonylgruppe steht) als Rohprodukt in einem Schutzgasstrom
in einem Alkohol-Lösungsmittel
bei Temperaturen von 0 bis 100°C
für eine
Zeit von 3 Minuten bis zu 1 Stunde für die Wasserstofftransfer-Reaktion
umsetzt, und anschließend
das Lösungsmittel
unter vermindertem Druck entfernt. Geeignete Alkohol-Lösungsmittel
umfassen beispielsweise Methanol, Ethanol, n-Propanol, Isopropanol,
n-Butanol, iso-Butanol
und sec-Butanol.
-
Der
Komplex kann durch ein weiteres Verfahren hergestellt werden. Genauer
kann RuH[(S,S)-,(R,R)-TsNCH(R01)CH(R02)NH2](η6-p-cymol)(Hydrid-((S,S) und (R,R)-N-toluolsulfonyl-1,2-disubstituiertes
Ethylendiamin)(η6-p-cymol)ruthenium) (wobei R01 und
R02 wie oben definiert sind und Ts für eine p-Toluolsulfonylgruppe
steht), auf einfache Weise hergestellt werden, indem man beispielsweise Ru[(S,S)-,(R,R)-TsNCH(R01)CH(R02)NH](η6-p-cymol)(((S,S) und (R,R)-N-toluolsulfonyl-1,2-disubstituiertes Ethylendiamin)(η6-p-cymol)ruthenium) (wobei R01 und
R02 gleich sind wie oben definiert; und
Ts für
eine p-Toluolsulfonylgruppe steht) als Rohmaterial in einem Lösungsmittel
mit Wasserstoff unter Druck umsetzt.

Die
Reaktion verläuft
im Allgemeinen quantitativ, indem man RuH[(S,S)-,(R,R)-TsNCH(R01)CH(R02)NH2](η6-p-cymol)(Hydrid-((S,S) und (R,R)-N-toluolsulfonyl-1,2-disubstituiertes
Ethylendiamin)(η6-p-cymol)ruthenium) (worin R01 und
R02 wie oben definiert sind; und Ts für eine p-Toluolsulfonylgruppe
steht) als Rohmaterial in einem inerten Lösungsmittel bei einer Temperatur
von 0 bis 50°C für 30 Minuten
bis zu 24 Stunden (bevorzugt 1 bis 10 Stunden) unter Wasserstoff-Druck
hydriert und anschließend
das Lösungsmittel
unter vermindertem Druck entfernt. Der Wasserstoff-Druck ist dabei
in einem Bereich von 1 bis 150 atm, bevorzugt 20 bis 100 atm.

Geeignete
inerte Lösungsmittel
umfassen beispielsweise Kohlenwasserstoffe, wie Benzol, Toluol,
Xylol, Hexan, Heptan, Cyclohexan, und Methylcyclohexan; und Ether,
wie Dimethyl ether, Diethylether, Diisopropylether, Methyl-tert-butylether,
Tetrahydrofuran, 1,3-Dioxolan und 1,4-Dioxan.

Ein
optisch aktives Diamin der Formel (S,S)-, (R,R)-R03NHCH(R01)CH(R02)NH2((S,S) und (R,R)-N-substituiertes-1,2-disubstituiertes
Ethylendiamin) (wobei R01, R02 und
R03 wie oben definiert sind) wird unter
Verwendung von (S,S)-,(R,R)-NH2CH(R01)CH(R02)NH2((S,S) und (R,R)-1,2-disubstituierter Ethylendiamine
als Rohprodukt in herkömmlicher
Weise hergestellt [Protective Groups in Organic Synthesis, Vol.
2, S. 309-405 (1991)].

Genauer
werden (S,S)-,(R,R)-TsNHCH(R01)CH(R02)NH2((S,S) und
(R,R)-N-p-Toluolsulfonyl-1,2-disubstituierte Ethylendiamine) (wobei
R01 und R02 wie
oben definiert sind; und Ts für
p-Toluolsulfonylgruppe steht) leicht hergestellt, indem beispielsweise
((S,S)-,(R,R)-NH2CH(R01)CH(R02)NH2((S,S) und
(R,R)-1,2-disubstituierte Ethylendiamine) als Rohmaterialien mit
TsCl(p-Toluolsulfonylchlorid) in Gegenwart einer Base (beispielsweise
eines tertiären
Amins, eines Alkalimetallsalzes oder dergleichen) in einem Lösungsmittel
umgesetzt werden.

Die
Reaktion verläuft
im Allgemeinen quantitativ, indem man ((S,S)-,(R,R)-NH2CH(R01) CH(R02)NH2((S,S) und (R,R)-1,2-disubstituierte Ethylendiamine)
(1 Mol) und TsCl (p-Toluolsulfonylchlorid) (1 Mol) mit einer Base
(z. B. Triethylamin) in einem inerten Lösungsmittel (z. B. Toluol,
Tetrahydrofuran und Methylenchlorid) in einem Schutzgas-Strom wie
Stickstoff, Helium oder Argon oder dergleichen bei Temperaturen von
0 bis 50°C
für 30
Minuten bis zu 3 Stunden umsetzt, anschließend die erhaltene Mischung
mit Wasser versetzt, und das Reaktionsprodukt stehen lässt, bevor
die flüssige
Phase durch ein Verfahren zur Flüssigkeitsabtrennung
entfernt und das Lösungsmittel
unter vermindertem Druck abgedampft wird.

Das
optisch aktive Diamin (S,S)-,(R,R)-NH2CH(R01)CH(R02)NH2((S,S) und (R,R)-1,2-disubstituierte Ethylendiamine) (wobei
R01 und R02 wie
oben definiert sind) ist bekannt und ist bisweilen kommerziell erhältlich oder
kann durch herkömmliche
Verfahren oder durch herkömmliche
Verfahren zur Racematspaltung hergestellt werden (Tetrahedron Lett.,
Vol. 32, S. 999-1002)
(1991), Tetrahedron Lett., Vol. 34, S. 1905-1908 (1993)].

(S,S)
und (R,R)-1,2-Diphenylethylendiamin und (S,S) und (R,R)-1,2-Cyclohexandiamin
sind kommerziell erhältlich.
-
Beispielsweise
kann das optisch aktive Diamin der allgemeinen Formel (e) durch
folgendes Verfahren hergestellt werden [Tetrahedron Lett., Vol.
32, S. 999-1002 (1991)].
-
Das
optisch aktive Diamin der allgemeinen Formel (e) ((S,S)-,(R,R)-1,2-disubstituiertes
Ethylendiamin) kann einfach und mit hoher Ausbeute hergestellt werden,
indem man Cyclophosphate aus optisch aktiven 1,2-disubstituierten
Ethylendiolen als Ausgangsmaterial darstellt, diese mit Amidin zum
Imidazolin umsetzt, und anschließend den Imidazolin-Ring unter
Verwendung einer Säure-Katalyse öffnet.
-
Der
Ruthenium-Diamin-Komplex kann isoliert und verwendet werden, aber
wenn man diesen Komplex in einer Reaktionslösung herstellt, wird der erhaltene
Komplex als Katalysator für
asymmetrische Synthese und dergleichen verwendet.
-
Das
Verfahren zur Herstellung optisch aktiver sekundärer Alkohole unter Verwendung
eines Wasserstofftransfer-artigen Oxidations-Katalysators wird nun
im Folgenden beschrieben.

meso-2-Cyclooctan-1,4-diol, 5,8-Dihydroxy-1,4,4a,5,8,8a-Hexahydro-endo-1,4-methanonaphtharene
und dergleichen.

Der
optisch aktive Diamin-Ligand der allgemeinen Formel (VII), d. h.
die (R,R)-Form oder die (S,S)-Form, kann als Rutheniumdiaminkomplex
für die
erfindungsgemäße Wasserstofftransfer-artige
Oxidation zufriedenstellend verwendet werden. Abhängig von
der Auswahl kann eine Zielverbindung der gewünschten absoluten Konfiguration
hergestellt werden. Solche Ru thenium-Diamin-Komplexe können in
einem Molverhältnis
von 1:10000 bis 1:10, bevorzugt 1:2000 bis 1:200 bezogen auf das
Substrat verwendet werden.
-
Zur
Durchführung
der Reaktion werden das Substrat und der Ruthenium-Diamin-Komplex
zum reinen Keton oder zu einer geeigneten Mischung von Keton mit
einem inerten Lösungsmittel
gegeben, um eine homogene Lösung
für die
Reaktion bei einer Reaktionstemperatur von 0 bis 100°C, bevorzugt
10 bis 50°C,
für 1 bis
100 Stunden, bevorzugt 3 bis 50 Stunden, herzustellen.
-
Ketone
umfassen beispielsweise Aceton, Methylethylketon, Diethylketon,
Diisopropylketon, Methyl-tert-Butylketon, Cyclopentanon, und Cyclohexanon.
Besonders bevorzugt ist Aceton. Diese Ketone können geeigneterweise einzeln
oder in einer Mischung mit einem inerten Lösungsmittel verwendet werden.
Die Ketone können
in einer 0,1- bis 30fachen Menge (Volumen/Gewicht) in Abhängigkeit
der Art des Substrats, aber bevorzugt in einer 2- bis 5fachen Menge
(Volumen/Gewicht) verwendet werden.
-
Geeignete
inerte Lösungsmittel
umfassen beispielsweise Kohlenwasserstoffe, wie Benzol, Toluol,
Xylol, Hexan, Heptan, Cyclohexan und Methylcyclohexan; und Ether,
wie Dimethylether, Diethylether, Diisopropylether, Methyl-tert-butylether,
Tetrahydrofuran, 1,3-Dioxolan, und 1,4-Dioxan.

Erfindungsgemäß kann die
Umsetzung in einem Batch- oder kontinuierlichen Verfahren durchgeführt werden.

Herstellung
von optisch aktiven Alkoholen

Nachstehend
sind Herstellungsbeispiele für
optisch aktive Alkohole angegeben und außerdem wird das erfindungsgemäße Verfahren
im Detail beschrieben. Tabelle 1, 2 und 3 zeigen alle Reaktionssubstrate, Übergangsmetall-Komplexe
und optisch aktive Amin-Verbindungen als chirale Liganden, die als
typische Beispiele verwendet werden.

Beispiele 1 bis 19 – Vergleichsbeispiele

Zu
trockenem 2-Propanol (5,0 ml) wurden verschiedene Aminoalkohol-Verbindungen
(0,05 mmol) als chirale Liganden optisch aktiver Aminoverbindungen,
wie in Tabelle 3 gezeigt, und der in Tabelle 2 angegebene Ruthenium-Aren-Komplex
(0,0125 mmol) gegeben; man rührte
die Mischung unter Argon- oder Stickstoff-Atmosphäre für 20 Minuten
bei 80°C
und kühlte
die resultierende Mischung auf Raumtemperatur ab, worauf man gefrorenes
und entgastes trockenes 2-Propanol (45,0 ml), verschiedene entgaste
und destillierte Carbonylverbindungen (5 mmol), wie in Tabelle 1
gezeigt, und eine Lösung
von 0,05 molarer KOH in 2-Propanol (2,5 ml, 0,125 mmol) in dieser
Reihenfolge zugab; anschließend
rührte
man bei Raumtemperatur. Nach beendeter Reaktion wurde die Reaktionsmischung
mit verdünnter
Salzsäure
sauer gestellt, das 2-Propanol unter vermindertem Druck entfernt
und schließlich
gesättigte
NaCl-Lösung
zugegeben. Das resultierende Produkt wurde mit Ethylacetat extrahiert,
mit gesättigter
Natriumchloridlösung
gewaschen und über
trockenem Natriumsulfat getrocknet. Das Lösungsmittel wurde aus dem Produkt
destillativ entfernt. Um den Umsatz zu errechnen, wurde das Endprodukt
durch 1HNMR (CDCl3)
analysiert. Dann wurde das Produkt durch Dünnschicht-Kieselgel-Chromatographie
gereinigt und die isolierte Alkoholfraktion wurde verwendet, um
die optische Reinheit und die absolute Konfiguration mittels HPLC
und GLC zu bestimmen. Die Ergebnisse sind in Tabelle 4 zusammengefasst. Außerdem können der
Umsatz und die optische Reinheit der erprobten Reaktionslösungen gleichzeitig
durch GLC errechnet werden.

Beispiele 20 bis 23 – Vergleichsbeispiele

Unter
Verwendung desselben Verfahrens wie im Beispiel 1, wurden Aminophosphin-Verbindungen als optisch
aktive Aminverbindung der Reaktion verwendet. Die Ergebnisse sind
in Tabelle 4 zusammengefasst.

Beispiele 24 bis 41

Die
in Tabelle 2 gezeigten chiralen Ruthenium-Komplexe wurden unter
Verwendung des in Beispiel 1 beschriebenen Verfahrens und unter
Verwendung optisch aktiver Aminoverbin dungen hergestellt. Die Komplex-Katalysatoren
und die Carbonylverbindungen wurden zu einer Mischung von Ameisensäure und
Triethylamin (5:2) für
eine Reaktion bei Raumtemperatur und für eine bestimmte Zeit gegeben.
Nach beendeter Reaktion wurde die Reaktionsmischung mit Wasser verdünnt, um
das Produkt mit Ethylacetat zu extrahieren. Nach Trocknen der organischen
Phase über
trockenen Natriumsulfat und Entfernen des Lösungsmittels durch Abdampfen,
wurde das 1HNMR (CDCl3)
ausgewertet, um die Umsetzung zu bestimmen. Die optische Reinheit und
die absolute Konfiguration wurden durch HPLC und GLC bestimmt. Die
Ergebnisse sind in Tabelle 5 zusammengefasst. Der Umsatz und die
optische Reinheit von jeder erprobten Reaktionslösung können gleichzeitig durch GLC
berechnet werden.

Gemäß der vorliegenden
Erfindung können
optisch aktive Alkohole mit einer hohen optischen Reinheit und einer
hohen synthetischen Ausbeute hergestellt werden.

Herstellung von optisch
aktiven sekundären
Alkoholen durch kinetische Racemattrennung von Alkoholen

Im
Folgenden werden Herstellungsbeispiele für optisch aktive sekundäre Alkohole
gezeigt, und Einzelheiten des erfindungsgemäßen Verfahrens werden beschrieben.
Jedoch ist die Erfindung nicht auf diese Beispiele begrenzt. Tabelle
9 zeigt zusammenfassend racemische sekundäre Alkohole oder meso-artige
Diole, die als typische Beispiele verwendet werden und Tabelle 10
zeigt Ruthenium-Diamin-Komplexe.

Die Reaktionslösung
wurde bei Raumtemperatur für
5 Minuten gerührt,
und durch Zugabe von Wasser (7 ml) verfärbte sich die Reaktionslösung von

Umkristallisieren aus Methanol, wurde der Katalysator Nr. 12 aus
Tabelle 10 als orange Kristalle erhalten. Die Ausbeute betrug 85
%.

(7 ml) und Ru[(S,S)-p-TsNCH(C6H5)CH(C6H5)NH](η6-p-cymol)((S,S)-N-p-Toluolsulfonyl-1,2-diphenylethylendiamin)(η6-p-cymol)ruthenium) (306,2 g; 0,5 mmol),
das in Beispiel 72 hergestellt wurde, eingefüllt und bei Raumtemperatur
und einem Wasserstoff-Druck von 20 atm umgesetzt. Nach Entfernen
des Lösungsmittels,
Waschen mit kaltem Pentan und anschließendem Umkristallisieren aus
Methanol, wurden orange Kristalle erhalten (420 mg). Die Ausbeute
betrug 70 %.

Anstelle
von Ru[(S,S)-p-TsNCN(C6N5)CH(C6H5)NH](η6-p-cymol)(((S,S)-N-p-Toluolsulfonyl-1,2-diphenylethylendiamin)(η6-p-cymol)ruthenium) wurde Ru[(S,S)-N-p-TsNCH(C6H5) CH(C6H5)NH](η6-mesitylen) (((S,S)-N-p-Toluolsulfonyl-1,2-diphenylethylendiamin)(η6-mesitylen)ruthenium), das in Beispiel 72
hergestellt wurde, verwendet, und unter Anwendung des Verfahrens
aus Beispiel 75-a wurde, der zuvor genannte Katalysator Nr. 13 aus
Tabelle 10 erhalten. Die Ausbeute betrug 60 %.

Synthese von (R)-1-Indanol
-
Ru[(S,S)-p-TsNCH(C6H5)CH(C6H5)NH](η6-p-cymol)((S,S)-N-p-Toluolsulfonyl-1,2-diphenylethylendiamin)(ruthenium-η6-p-cymolmesitylen) (6,0 mg; 10 μmmol), das
in Beispiel 71 hergestellt wurde, und 1-Indanol (671 mg; 5 mmol)
wurden in einem zuvor im Vakuum getrockneten und mit Argon befüllten Schlenk-Kolben
vorgelegt, Aceton (2,5 ml) wurde zugegeben und die Reaktionsmischung
wurde für
6 Stunden bei 28°C
gerührt.
Das Lösungsmittel
wurde bei vermindertem Druck destillativ entfernt, der Rückstand
wurde durch Kieselgel-Chromatographie (Eluent; Ethylacetat:Hexan
= 1:3) aufgereinigt, um (R)-Indanol (286 mg) als farblose Kristalle
zu erhalten. Die Ausbeute betrug 84 %.

erhaltene (R)-1-Indanol wurde durch HPLC untersucht (high performance
liquid chromatography) und das erhaltene (R)-1-Indanol hatte eine
optische Reinheit von 97 % ee.

Gemäß dem in
Beispiel 79 beschriebenen Verfahren wurden die in Tabelle 9 gezeigten
racemischen sekundären
Alkohole und meso-artigen Diole als Reaktionssubstrate mit den optisch
aktiven Ruthenium-Diamin-Komplexen unter geeigneten Reaktionsbedingungen
und Re aktionszeiten umgesetzt, um die jeweiligen optisch aktiven
sekundären
Alkohole in hohen Ausbeuten zu erhalte. Die Ergebnisse sind in Tabelle

Gemäß der vorliegenden
Erfindung werden optisch aktive Alkohole bereitgestellt, die in
vielen Bereichen pharmazeutischer Produkte, Synthesezwischenstufen
hiervon, Lebensmitteln, Geschmacksstoffen, Kosmetika und Flüssigkristall-Materialien
nützlich
sind.

Der
in der vorliegenden Erfindung verwendete Ruthenium-Diamin-Komplex
ist als chiraler Katalysator, der höhere Selektivität und Aktivität gewährleistet,
von industriellem Nutzen, da dieser Komplex für organische Synthesen, wie
asymmetrische synthetische Reaktionen, verwendet werden kann. Wird
der Komplex als Katalysator für
Wasserstofftransfer-artige asymmetrische Reduktionen von racemischen
sekundären
Alkoholen oder meso-artigen Diolen verwen det, können optisch aktive sekundäre Alkohole,
die als Zwischenprodukte von Wirkstoffen nützlich sind, mit hoher Effizienz
hergestellt werden.
