Auf Grund ihrer hohen Spezifität zu Nucleinsäuren, der leichten Aufnahme in biologische Systeme sowie ihrer photochemischen Eigenschaften, die das "Einfrieren" doppelsträngiger Nucleinsäurebereiche ermöglichen, eignen sich Psoralene als ideales Werkzeug zur Untersuchung sekundärer DNA- und RNA-Strukturen (vgl. C.-K. J. Shen et al., Proc. Natl. Acad. Sci. USA 73 [1976], 2649-53). Neben diesem biochemischen Anwendungsbereich werden Psoralenderivate schon seit längerer Zeit mit großem Nutzen im klinischen Bereich zur Behandlung von Psoriasis und ähnlichen Hauterkrankungen verwendet (vgl. J. H. Parrish et al., N. Engl. J. Med. 291 [1974], 1207-1211), sowie in neuester Zeit auch erfolgreich in der Photochemotherapie von cutanen T Zell Lymphoma, einer als bislang nahezu unheilbaren Hautkrebserkrankung (R. Edelson, N. 1,6 g (2,54 mMol) des modifizierten 2′-Desoxyadenosinderivates
(Verbindung IV nach Beispiel 4) werden durch zweimaliges
Einengen von 10 ml Pyridin getrocknet und anschließend
in 20 ml trockenem Pyridin gelöst. Zu dieser Mischung
gibt man unter Argonatmosphäre 936 mg (2,79 mMol) 1,3-Dichlor-
1,1,3,3-tetraisopropyl-disiloxan und rührt 2 Stunden
bei Raumtemperatur. Hiernach fügt man 892 mg (6,33 mMol)
Benzoylchlorid zu und läßt die Mischung über Nacht rühren.
Nach Beendigung der Reaktion wird die Reaktionsmischung bis
zur Trockne eingeengt, zur Beseitigung anhaftender Pyridinreste
ein weiteres Mal aus Toluol eingeengt und anschließend
in 50 ml Dichlormethan aufgenommen. Es wird einmal mit 30 ml
5%iger Natriumhydrogencarbonat-Lösung ausgeschüttelt, die
organische Phase über Natriumsulfat getrocknet und nach
Filtrieren bis zur Trockne eingeengt. Das Produkt wird dann
an 30 g Kieselgel chromatographiert. Als Elutionsmittel
dient 3% Methanol/Dichlormethan.
Ausbeute: 2,4 g (91,4%); Rf: (3% Methanol/Dichlormethan)

2,4 g (2,27 mMol) des vollständig geschützten Adenosinderivats
der Formel V nach Beispiel 5 werden in 10 ml trockenem
Tetrahydrofuran gelöst und mit 5,7 ml (2,5 Äquivalente)
einer 1M-Lösung von Tetrabutylammoniumfluorid versetzt. Die
Lösung wird 15 Minuten bei Raumtemperatur gerührt und nach
Abschluß der Reaktion bis zur Trockne eingeengt, in 20 ml
Dichlormethan aufgenommen und zweimal mit 10 ml 5%iger
Natriumhydrogencarbonat-Lösung extrahiert. Nach Trocknen
über Natriumsulfat und Einengen des Filtrats bis zur Trockne
nimmt man den Rückstand in 20 ml Pyridin auf, fügt 10 ml
konzentrierte Ammoniaklösung zu und rührt 30 Minuten bei Raumtemperatur.
Die Reaktionsmischung wird bis zur Trockne eingeengt
und anschließend zur Entfernung anhaftender Pyridinreste
noch zweimal aus je 10 ml Toluol eingeengt. Der Rückstand
wird in 50 ml Dichlormethan aufgenommen, mit 10 ml
5%iger Natriumhydrogencarbonat-Lösung extrahiert, die organische
Phase über Natriumsulfat getrocknet und bis zur Trockne
eingeengt. Die Aufreinigung des Produktes erfolgt an 20 g
Kieselgel mit 3% Methanol/Dichlormethan als Elutionsmittel.

800 mg (1,10 mMol) des Adenosinderivates (VI) nach Beispiel 6
und 495 mg (1,4 mMol) 4,4′-Dimethoxytritylchlorid werden
unter Argonatmosphäre in 10 ml trockenem Pyridin gelöst und
1 Stunde lang bei Raumtemperatur gerührt. Nach Abschluß der
Reaktion fügt man 5 ml Methanol zu und beläßt 15 Minuten bei
Raumtemperatur. Nach Einengen bis zur Trockne wird das Produkt
ohne weitere Aufarbeitung direkt an 10 g Kieselgel gereinigt.
Als Elutionsmittel dient 3% Methanol/Dichlormethan.
Ausbeute: 500 mg (42%); Rf: (3% Methanol/Dichlormethan)

500 mg (0,46 mMol) des Adenosinderivates der Formel VII
nach Beispiel 7 werden in 5 ml trockenem 1,2-Dichlorethan
unter Argonatmosphäre gelöst, mit 100 µl (2,5 Äquivalente)
Diisopropylethylamin versetzt. Jetzt gibt man 100 µl (1,1
Äquivalente) Chlor-(β-cyanoethoxy)-N,N-diisopropylaminophosphin
zu und rührt 1,5 Stunden bei Raumtemperatur. Die
Reaktion wird durch Zugabe von 5 ml 5%iger Natriumhydrogencarbonat-
Lösung gestoppt und dann 15 Minuten lang bei Raumtemperatur
belassen. Nun verdünnt man die Reaktionsmischung
durch Zugabe von 10 ml Dichlormethan, extrahiert die organische
Phase mit 15 ml 5%iger Natriumhydrogencarbonat-
Lösung, trennt die Phasen und trocknet anschließend über
Natriumsulfat. Das Produkt wird an 15 g Kieselgel gereinigt.
Als Elutionsmittel und zum Packen der Säule wird folgendes
Gemisch verwandt: Lösung A: Dichlormethan/Triethylamin =
10/1; Lösung B: n-Hexan/Triethylamin = 10/1. Lösung A und
Lösung B werden zur Elution zu gleichen Teilen gemischt.
Ausbeute: 400 mg (72,5%); Rf: (Ethylacetat/Dichlormethan/
Triethylamin = 45/45/10); beide Isomere bei 0,77 und 0,85

TA-Positionen erkennen, wobei jeweils das Adenosin Psoralen-modifiziert ist (Ausbeute der Photoaddition mehr als 90%). In Spur 4a wurde die reaktive AT-Position um ein Basenpaar verschoben; die schwach ausgeprägte Photoreaktion dokumentiert eindeutig, daß eine Spacerlänge (L) von 5 Kohlenstoffatomen das Optimum darstellen. In Spur 5a ist die Möglichkeit eines Thymidinmonoadduktes bzw. eines ebenfalls möglichen Crosslinks zwischen Thymidin und Cytidin dargestellt. Die schwach ausgeprägte Photoaddition zeigt, daß diese Reaktionen möglich sind.
