Beispiel 1: Schachbrettitration von drei Seren auf Antikörper gegen Tollwutvirus Antigen.Example 1: Checkerboard titration of three sera Antibodies against rabies virus antigen.
Fig. 1a ist eine Photokopie-Darstellung des Testergeb nisses mit einem Serum mit Antikörpern gegen Tollwutvi rus dargestellt. Die Fig. 1 zeigt 5 Streifen Nitrozel lulosepapier, die jeweils in 6 Felder aufgeteilt sind. In diese Felder wurden 2 µl eines konzentrierten und gereinigten inaktivierten Tollwutantigens mit 680IU/ml in Verdünnungen von 1 : 150 bis 1 : 1200 aufgetropft. Die Antigenmenge einer Verdünnung von 1 : 200 enthielt somit 0,007 IU/2 µl. Nach dem Trocknen der Streifen für 15 Minuten bei +37°C wurden 4 ml der Serumverdünnungen von 1 : 50 bis 1 : 600 jeweils mit einem Streifen für 10 Minuten bei +37°C unter leichtem Schütteln inkubiert. Danach wurde die Flüssigkeit abgesaugt und die Streifen gewaschen, indem diese in ca. 5 ml Waschpuffer für 5 Minuten bei +37°C unter leichtem Bewegen eingelegt wurden. Dieser Vorgang wurde 3 Mal wiederholt. Der letzte Waschpuffer wird gründlich abgesaugt und ansch ließend 4 ml eines an Peroxydase gekoppeltes Anti-Human IgG (Konjugat) in einer Verdünnung von 1 : 900 (abhängig von Charge) für 10 Minuten bei +37°C unter leichtem Schütteln inkubiert. Der Vorgang wird zweimal mit Waschpuffer und zweimal mit Phosphatgepufferter Salzlö sung wiederholt. Während des letzten Waschvorgangs wird das Substrat (4 Chloronaphtol + H2O2) hergestellt und kurz bei +37°C vorgewärmt. Nach Beendigung des Wasch vorgangs werden 3 ml Substrat auf die Streifen gegeben und 5 Minuten unter leichtem Schütteln bei +37°C inkubiert. Anschließend an die Farbentwicklung werden die Streifen unter fließendem Wasser abgespült und sofort beurteilt. Das hier gezeigte Ergebnis (Fig. 1a) zeigt eine starke Farbreaktion der Serumverdünnungen bis 1 : 600 bei Verwendung der Antigenverdünnungen 1 : 50 bis 1 : 200. Die Serumverdünnung 1 : 50 hat noch bei einer Antigenverdünnung von 1 : 1200 eine deutliche Reaktion. Dies ist auch bei den Serumverdünnungen 1 : 100 und 1 : 200 der Fall. Die Serumverdünnung 1 : 600 war bei dieser Antigenverdünnung negativ, aber noch bei einer Antigen verdünnung von 1 : 400 positiv. Aus diesem Grunde wurden in den nachfolgenden Untersuchungen mit diesem Antigen eine Verdünnung von 1 : 200 eingesetzt. Fig. 2 zeigt das Testergebnis, das mit dem beschriebe nen Blood Drop ELISA durchgeführt wurde. Aus einer geronnenen nicht zentrifugierten Blutprobe wurde eine Probe eines Probanden, von dem der positive Tollwutvirus-Antikörperstatus bekannt war, in Verdün nungspuffer 1 : 75 verdünnt und dann in diese Verdünnung ein Träger mit Tollwutvirusantigen getaucht. Nach weiterem Ablauf der Reaktion wie in Beispiel 1 darge stellt, wurde eine positive Reaktion nachgewiesen. Weiterhin wurde ein Tropfen Kapillarblut von ca. 20 µl aus der Fingerbeere eines Probanden ohne Tollwutvirus-Antikörper direkt in ein für die Aufnahme des Antigenträgers vorgesehenes Gefäß getropft, welches 1500 µl Verdünnungspuffer enthielt. Nach Ablauf des in Beispiel 1 beschriebenen Ablaufes der Reaktion war das Ergebnis negativ. Fig. 2 shows the test result, which was carried out with the described Blood Drop ELISA. Die Kennzeichnung eines Befundes durch "+" bedeutet, daß Antikörper gegen das HIV I vorhanden sind. Die Befundsangabe "-" bedeu tet, daß Antikörper gegen das HIV I nicht vorhanden sind. Ferner erfüllen sie den Zweck, daß sie, nach der vorläufigen Reaktion, die Testmischung wieder aus der Ausnehmung 12 absaugen können, einfach indem sie mit der offenen Spitze in die Ausnehmung 72 gehalten werden, zusammengedrückt und dann losgelassen werden, so daß die Flüssigkeit aus der Ausnehmung 12 wieder in diese Behälter 86 bzw. 88 hineingesaugt wird.
