Abstract

New aq. nutrient medium for microorganisms is based on usual carbon and nitrogen sources; usual drug-inactivating ingredients; and opt. usual solidifying agents, buffer substances and trace elements. The drug-inactivating ingredients are (per litre of water) 0.2-0.5 g N-acetyl-L-cysteine, 3.5-15 mg Ca2+ 50-150 mg Mg2+, 40-75 mg p-aminobenzoic acid and 7.5-15 mg thymine or thymidine. The medium opt. also contains "Tween" (RTM) 80, lecithin, Mn2+, Fe2+ or Fe3+, and sodium, carbonate and phosphate or hydrogen phosphate ions. The medium is used for determn. of microbe counts in specimens contg. antagonists, antibiotics or residues of other chemotherapeutic agents, esp. on water, milk (prods) and meat prods. It is also useful in determn. of microbe counts in specimens from patients undergoing antibacterial therapy, helping to avoid continuation of treatment with ineffective drugs but avoiding the need to cease drug administration prior to taking the specimen.

Description

Vorrichtung zur KeimzahlermittlungDevice for determining the number of germs

The invention relates to a device for determining the number of germs; she consists of a flat support, which is attached to each of the two main surfaces with a new one Nutrient medium is coated.

Devices of this type have long been in use.

Such devices are used to determine the germ content in test samples, e.g. in urine.

The devices commonly used up to now consist of a slide, one side with C.L.E.D. (BROLACIN agar) or nutrient agar, the other side is coated with MacConkey agar or eosin methylene blue agar (EMB agar). the one side serves as a breeding ground for all types of germs, the other only for gram-negative ones Bacteria.

The disadvantage of the known device is that it is not an accurate one Provides picture of the germ count, if in the examination sample inhibitors or antibiotics or it contains other chemotherapy residues.

C.L.E.D. bietet anspruchsvollen Keimen keine ausreichende Wachstumsbasls, ebenso MaeConkey-Agar.These residues can damage the germs or be bacteriostatic influence. The nutrient medium usually used are then not able to to enable a sufficient growth of the germs or the damaged germs to animate. C.L.E.D. does not provide sufficient growth basls for demanding germs, also MaeConkey agar.

The second most important - especially in urinary tract infections - grows with nutrient agar Pathogen Proteus, which also occurs frequently as a life custom 1-continant, uncontrolled (raves); this makes it impossible to determine the number of germs.

Darüberhinaus kann z.B.The device according to the invention avoids the disadvantages mentioned. The support, preferably a slide, is coated with two new nutrient media. With one of them becomes through a useful new combination of the Components in suitable proportions - even with demanding germs achieved rapid and abundant growth. Reversibly damaged and therefore incapable of growth Germs are revitalized, as the residues of the above-mentioned antimicrobial effects Substances are neutralized or inactivated. That way it becomes a wrong one negative result of the bacterial count that simulates a successful therapy, prevented. Continuation of treatment with ineffective therapeutic agents is avoided. This can significantly reduce patient relapses. Furthermore can e.g.

Erweist sich das Medikament als unwirksam, so wird es sofort abgesetzt.the microbiological urine test already 2 to 3 days after the start chemotherapy treatment can be carried out without stopping the drug, to check the patient's responsiveness to the chemotherapy agent. Proves If the drug is found to be ineffective, it will be discontinued immediately.

The sensitivity test is repeated for more appropriate chemotherapy drugs to be identified and deployed early enough.

The same applies to the second nutrient medium, but instead of Contains D-lactose meso-inositol.

The combination of the two culture media also makes it possible to determine whether the infection is caused by E. coli.

After incubation on nutrient medium I they are yellow, on nutrient medium II no yellow colonies, the indole test is carried out (red color reaction with Kovacs reagent). About 90% E. coli strains (i.e. the Lactose-cleaving strains). This is used, for example, for urine tests an important preliminary clarification has already been achieved, as E. coli is one of the most common pathogens of urinary tract disease is.

Also in the hygiene sector, e.g. bacterial count determinations in waters, Milk and dairy products, as well as meat products, can be ailing or damaged Microorganisms can be recorded more precisely with the help of the new nutrient media.
