Verfahren zur Herstellung von Norpatchoulenol
Norpatchoulenol ist ein ungesättigter tricyclischer,
in Patchouliöl vorkommender und dessen Geruch in charakteristischer
Weise bestimmender Alkohol der Formel
XIII
Für die Gewinnung dieses Alkohols, der auch als ITordehydropatchoulol
bezeichnet werden kann, standen bisher, ausser der Isolierung aus natürlichein Patchouliöl .■;·..-in j MethD

zur Verfugung. Es wurde nun eine synthetische Methode zu dessen
Gewinnung gefunden. Diese ist dadurch gekennzeichnet, dass man ein Glvkol der Formel

XII
dehydratisiert.
Die erfindungsgemässe Dehydratisierung kann auf an sich
"bekannte Art und V/eise durchgeführt werden. Die anzuwendende Methode richtet sich unter anderra nach der absoluten Konfiguration
des die sekundäre 0H~Gruppe tragenden Kohlenstoffs (trans- bzw. cis-Elimination).
Das Glykol der Formel XII kann seinerseits aus einem
Monoäther des Glykols XII, und zwar einem solchem, in welchem
die sekundäre OH-Gruppe mit einer leicht abspaltbaren Gruppe, wie der Benzylgr-uppe, veräthert ist, durch Aetherspaltung,
insbesondere mittels Hydrogenolyse, erhalten werden, Bin geeigneter Vorläufer für die Gewinnung des Glykols XII ist demgemäss
z.B. der Benzyläther der Formel

der nach an sich bekannten Methoden hydrogenolysiert werden kann.
Der Benzyläther XI b und der entsprechende Methyläther, d.h. die Verbindungen der allgemeinen Formel

worin R Methyl oder Benzyl bedeutet, sind neu und bilden ebenfalls Gegenstand der Erfindung.
Anhand des nachfolgenden Iteaktionsschemas soll gezeigt
werden, wie die neuen Monoäther der allgemeinen Formel XI erhalten werden können.

Die Synthese nimmt ihren Ausgang vom ungesättigten
Mcyclischen Aldehyd I, der nach J.Org.Chem. 27 (1972), 2871
erhalten werden kann. Behandlung dieses Aldehyds mit Bromessigsäureäthylester
in-Gegenwart von Zink gemäss Reformatsky liefert
den Ester II (Diastereomerengemisch), dessen Estergruppe mittels LiAlH. zur primären· Alkoholgruppe reduziert werden kann.
Durch Umkristallisation aus Petroläther lässt sich aus dem erhaltenen Gemisch der zwei diastereomeren Glykole III eines
in Form von Kristallen vom Smp, 113-1140C gewinnen. Die im
folgenden beschriebenen Reaktionen basieren auf .diesem Isomeren.
Zunächst wird die primäre OH-Gruppe des Glykole III mittels Triphenylchlormethan veräthert, worauf die sekundäre
OH-Gruppe des erhaltenen Tritylathers IY veräthert wird. Durch
Methylierung erhält man so den Methyl-trityl-äther Y a und
mittels Benzylierung den Benzyl-trityl-äther Yb.
Durch Hydroborierung und Oxydation lassen sich die Verbindungen der Formel V in die entsprechenden Ketone der Formel
VI und letztere durch Methylierung in die Verbindungen der
Formel VII überführen. Aus diesen erhält man nach hydrogenolytischer
oder hydrolytischer Abspaltung der Tritylgruppe, die
freien primären Alkohole der Formel VIII. Ueber die Tosylate
der Formel IX gelangt man sodann zu den Halogeniden X (worin "HaI'1 Chlor, Bron oder Iod bedeutet) die nach Cyclisierung
schliesalicii die tricycliachen I-Ionoäther der Forsiel SI liefern.

a) In einen 100 ml-Kolben gibt man 3,3 g (15 mMol) des
Diols III, 6 g Tritylchlorid (21 mMol), 50 ml wasserfreies Benzol und 3 ml Pyridin. Das Gemisch wird 2
Stunden unter Rückfluss gehalten. Nach Abkühlung wird das ausgefällte Pyridiumchlorid abfiltriert, das Lösungsmittel
abdestilliert und der Rückstand in einem Petroläther-Aether-Gemisch
(8:2) aufgenommen. Nach Filtration über 100 g neutrales AIpO., der Aktivität 1 erhält man
. 4 g des Tritylmonoäthers IV

NMR: 1,10, 1,70, ~3-4, 5,36.
b) In einen mit Rührer versehenen Kolben gibt man 5,9 g (13 mMol) Tritylmonoäther IV, 60 ml Glym, 2 g (14 mMol)
Methyljodid und fügt dann in kleinen Portionen 0,8 g
Natriumhydrid (50$, 15 mMol) zu. Hierauf erhitzt man
während 2 Stunden zum Rückfluss, fügt succesive weitere 2 g Methyljodid und 0,5 g Natriumhydrid zu, worauf die
Reaktion nach weitern 2 Stunden beendigt ist. Das Reaktionsgemisch
wird mit 100 ml Aether und dann mit 5 ml Wasser versetzt. Die ätherische lösung wird dann mit
gesättigter NaCl-Lösung neutral gewaschen. Man trocknet über Natriumsulfat und destilliert das Lösungsmittel.
Man erhält 7,4 g rohen Diäther V a, der mittels Chromato-
. graphie über 100 g neutrales Aluminiumoxyd gereinigt wird. Die Ausbeute an dünnschichtchromatographisch
homogenem Diäther V a beträgt 6,65 g.

c) Ig des Diäthers V a, gelöst in 15 ml Tetrahydrofuran
wird bei O0C mit einem Strom Diboran (BpH,-) behandelt.
Die Reaktion wird dünnschichtchromatographisch verfolgt. Nach Verschwinden des Ausgangsmaterials fügt man einige
Tropfen Wasser zu und giesst dann das Gemisch in 20 ml Wasser. Man extrahiert mit 4 mal 10 ml Benzol, wäscht
mit 2 mal 10 ml gesättigter NaCl-Lösung, trocknet über
Natriumsulfat und destilliert das Lösungsmittel. Man erhält 1,1 g rohes Boran.
Zur Oxydation des erhaltenen Borans wird dieses in 15 ml Pyridin gelöst und diese Lösung zu einer Lösung von
1,6 g CrO^ in 16 ml Pyridin gegeben. Man rührt über
Nacht bei Raumtemperatur, giesst in 60 ml Aether und filtriert den Niederschlag, den man mit Aether wäscht.
Die organische Phase wird mit 1Obiger, kalter Salzsäure
neutral gewaschen. Nach Trocknen und Destillation des Aethers, erhält man 1,1 g rohes Keton VI a. Nach Chromatographie
über 25 g neutrales Aluminiumoxyd (Elution mit Petroläther-Aether 8:2) werden.200 mg Keton VI a mit
folgenden Spektraldaten erhalten:

d) Herstellung von Tritylkalium: 100 mg in feine Lamellen zerschnittenes Kalium werden zu einer Lösung von 600 mg
Triphenylmethan in 5 ml Glym gegeben. Das Gemisch wird
bei Raumtemperatur unter Inertatmosphäre 16 Stunden
gerührt.
Alkylierung: In einen mit Rührer versehenen 25 ml-Kolben
gibt man eine Lösung von 200 mg des Ketons VI a in 5 ml Glym und fügt eine genügende Menge Tritylkalium zu.
Wenn die rote Färbung bleibt, rührt man 20 Minuten und

versetzt dann mit 0,5 ml Methyljodid. Nach. 6 Stunden bei
Raumtemperatur giesst man das Reaktionsgemisch in 25 ml Wasser und extrahiert dann mit 3 mal 20 ml Aether. Die
Extrakte werden mit Wasser und- dann mit gesättigter NaCl-Lösung gewaschen. Nach Trocknen über Natriumsulfat
verdampft man das Lösungsmittel. Nach Chromatographie des Rohprodukts über 50 g neutrales Aluminiumoxyd erhält
man 25 mg methyliertes Keton VII a neben 100 mg Ausgangsmaterial
.

e) Eine Lösung von 1,45 g von rohem Keton VII a in 100 ml Aethanol wird in Gegenwart von 0,25 g.Palladium auf
Kohle (55έ) mit Wasserstoff behandelt. Dauer der Hydrogenolyse:
17 Stunden. Nach Filtration des Katalysators destilliert man das Lösungsmittel und ehromatographiert
den Rückstand über eine Kolonne von 40 g Silicagel. Elution mit einem Petroläther-Aether-Gemisch (3:7) ergibt
125 mg des Alkohols VIII a.

f) 101 mg des Alkohols VIII a werden in 9 ml Pyridin gelöst. Man kühlt auf -120C ab und fügt 300 mg Tosylchlorid zu.
Man lässt 14 Stunden bei ca. -140C stehen, giesst dann das Reaktionsgemisch in 50 ml lO^ige Salzsäure und
extrahiert mit Aether (3 mal 20 ml). Die organische Phase wird mit lO^iger-Salzsäure und dann mit Bicarbonatlösung
gewaschen. Das erhaltene Tosylat IX a (100 mg) braucht für die nachfolgende Reaktion nicht gereinigt

zu werden.
g) 100 mg des erhaltenen Tosylats IX a, gelöst in 5 ml
Aceton, werden 20 Stunden bei Raumtemperatur mit 500 mg NaJ behandelt. Das Reaktionsgemisch wird in 30 ml Wasser.
gegossen und dann mit 3 mal 15 ml Aether extrahiert. " Nach Waschen mit Wasser und Trocknen über Sulfat wird
das Lösungsmittel abgedampft. Man erhält 87 mg rohes Jodid X a, das über 6 g neutralem Aluminiumoxyd chromatographiert
wird. Elution mit einem Petroläther-Aether-Gemisch
8:2 ergibt 68 mg Jodid_X a.
h) In einen 10 ml-Kolben gibt man 68 mg Jodid X a, gelöst
in 5 Eil Tetrahydrofuran, und dann 100 mg fein zerschnittenes
Natrium. Das Gemisch wird 4 Stunden unter Rückfluss gerührt, dann auf Raumtemperatur abgekühlt,
das Lösungsmittel entfernt und das Natrium mit Tetrahydrofuran gewaschen. Man fügt 1 ml Wasser zu und säuert
mit 10?S-iger Salzsäure an. Nach Zugabe von 25 ml Aether
wird dekantiert, mit Bicarbonatlösung gewaschen und über Sulfat getrocknet. Nach Abdampfen des Lösungsmittels
erhält man 44 mg rohes Cyclisierungsprodukt XI a. Chromatographie über eine Kolonne von 5 g Silicagel und
Elution mit einem Petroläther-Aether-Gemisch 8:2 liefert
17 mg Cyclisierungsprodukt XI a.

In Analogie zu Beispiel Ib) erhält man aus dem Tritylmonoäther
IV bei der Umsetzung mit Benzylchlorid (vgl. Canad. J. of Chem. 44 (1966), 1591) den Diäther V b mit folgenden
Spektraldaten:

• Die Oxydation des erhaltenen Diäthers V b gemäss Beispiel
1 c führt zum Keton VI b, das in Analogie zum Beispiel Id) zur Verbindung VII b methyliert werden kann. Nach selektiver
Abspaltung der Tritylgruppe, z.B. mit Essigsäure in homogener
ätherischer Phase bei Raumtemperatur (vgl. z.B. J. Chem. Soc. 1°,56, 3459)» wird der erhaltene Monoäther VIII b in Analogie
zu Beispiel 1 f) in das Tosylat IX b, dieses geraäss Beispiel
Ig) in das Jodid X b übergeführt, worauf schliesslich die Cyclisation gemäss Beispiel lh) zum tricyclischen Benzyläther
XI b führt. -Dessen Hydrogenolyse zum freien Diol XII kann
nach an sich bekannten Methoden der hydrogenoly tischen Benzylätherspaltung
durchgeführt werden. Die' Dehydratisierung des Diols XII zum Norpatchoulenol XIII kann bei Vorliegen eines
Diols XII mit S-Konfiguration des die sekundäre OH-G-ruppe tragenden
Kohlenstoffs wie nachstehend beschrieben vorgenommen werden:
Man mischt 400 mg des genannten Diols in 1 ml Pyridin mit einer Lösung von 500 mg (2,06 mMol) p-Brombenzolsulfonylchlorid
in 1 ml Pyridin. Das Gemisch wird 1,5 Stunden bei O0C
gerührt, dann auf Eis gegossen und mit Aether extrahiert. Nach Extraktion, Neutralwaschen, Trocknen und Destillieren des
Aethers erhält man 300 mg eines leicht gelblichen kristallisierten Produkts, das über eine Kolonne von 10 g Silicagel chromato-

graphiert wird. Elution mit einem Petroläther-Aether-Gemisch
(9:1) liefert zunächst 110 mg eines kristallinen Gemisches von 2 Produkten, mit Norpatchoulenol XIII als Hauptprodukt.
Weitere Elution mit Petroläther-Aether-G-emisch 7:3 liefert
160 mg von nicht umgesetztem Diol XII. Schmelzpunkt von so erhaltenem
reinem Norpatchoulenol: 180-1830C.
