Die kommerzielle Markierung eines Oligonukleotid-Primers mit einem gewöhnlichen Flureszenzfarbstoff (FAM, HEX, TET oder ROX) kostet in der Regel zwischen 160 und 200 DM. Eine Selbstmarkierung ist für viele kleinere Laboratorien nicht praktikabel, da der zusätzliche instrumentelle und manuelle Mehraufwand (DNA-Synthesizer, HPLC-Reinigung) die Kostenersparnis nicht rechtfertigt. Im Rahmen von Genfindungs-Projekten muß man in der Regel durch Kopplungsanalysen an Familienstammbäumen gewisse Kandidatenloci mittels der Analyse polymorpher Mikrosatelliten-Marker ein- beziehungsweise ausschließen. Dabei gibt es gegenwärtig die Regel, daß zur Erreichung einer hohen Sicherheit und zum Ausschluß möglicher Rekombinationen mindestens drei polymorphe Mikrosatelliten-Marker sowohl auf der zentromeren als auch auf der telomeren Seite des Kandidatengens untersucht werden müssen. Die Kosten allein für die Fluoreszenzmarkierung der erforderlichen Oligonukleotid-Primer belaufen sich damit auf 960-1200 DM pro Genlocus.
