Description
According to the previously known processes for the production of pure L- and D L-cysteine hydrochloride from keratins, hair is usually hydrolyzed with hydrochloric acid and, after the acid has been blunted or distilled off, a weakly acidic pH value is set, as a result of which sparingly soluble cystine precipitates. For cleaning, recrystallization is carried out in various ways using a lot of activated carbon. The yields of pure cystine are about 6 0 J 0 , based on uncleaned hair. In the subsequent reduction to cysteine or cysteine hydrochloride, further losses in yield occur, so that these processes work unfavorably.
A number of analytical methods for the quantitative determination of cystine are known (Houben - Weyl, methods of organic chemistry, 4th edition, 11/2, p. 430), in which the cystine by conversion into the poorly soluble cysteine copper (I) mercaptide quantitatively from the hydrolysates is isolated. By decomposing the mercaptide with hydrogen sulfide or rhodanides, the cystine becomes as cysteine hydrochloride or after oxidation as Preserved cystine in substance. These procedures are because of their awkwardness and the quantities involved expensive chemicals are only of analytical interest.
. . .U.S. Patent 2,376,186 discloses a Method for the production of cysteine is described in which the mentioned deficiencies are partially eliminated. Here the acidic hydrolyzate is blunted by lye and the cystine by adding copper (I) oxide precipitated as an insoluble cysteine copper (I) mercaptide complex! This complex is used in the usual Way worked up by decomposition with hydrogen sulfide, an aqueous solution of Cysteine accumulates. Crystallized cysteine can be separated from it by concentration. The achievable yields are inadequate in this procedure. In addition, the cysteine obtained is not pure L-cysteine, but, depending on the hydrolysis conditions, a more or less strongly racemized product. Further Disadvantages are the not inconsiderable consumption of copper (I) oxide and working with the poisonous Hydrogen sulfide and the problematic separation of large amounts of the finest copper sulfide sludge of the oxygen-sensitive cystic solution. . . .
The invention relates to a process for the production of l- and DL-cysteine hydrochloride or DL-cystine hydrochloride by acid hydrolysis of keratins and isolation of the cysteine or cystine contained in the hydrolysates, which is characterized in that the hydrolysis is carried out with sulfuric acid, the sulfuric acid hydrolyzate stirred with excess, catalytic amounts of copper oxide or copper sulfate containing copper powder at 40 to 100 0 C and then brought by dilution with water at about 40 to 80 ° C to a content of below 10 ° / 0 free acid, the precipitated precipitate of the Cysteine copper (I) mercaptides suctioned off and washed with water, then hydrochloric acid is added to the mercaptide, whereby it is completely or partially dissolved, and the resulting solution is reduced electrolytically using a diaphragm, the resulting cysteine hydrochloride solution is suctioned off from the copper powder formed, evaporated and for the separation of J. cysteine hydr chloride is recrystallized from water.
■■'''■'··' "; ■'-: ? Die Menge des einzusetzenden Kupferpulvers liegtThe keratin is hydrolyzed with sulfuric acid at a concentration of 40 to 80 ° / 0. By concentrating the mother liquor, crystallized cysteine hydrochloride racemate can furthermore be separated off or correspondingly racemized cystine can be obtained by oxidation. The copper powder obtained in the electrolytic reduction can be used again directly for the production of the cysteine copper (l) mercaptide. ■■ '' ■ '··'';■' -: The amount of the copper powder is

The degree of dilution is not critical here.
According to the previously known processes, the formation of the cysteine copper (I) mercaptide was achieved by adding copper (I) oxide or chloride to the hydrolyzate previously blunted by the addition of base. In the process according to the invention, on the other hand, the considerable amounts of alkali which would be required to blunt the acid in the hydrolyzate are saved. Of the present process at 40 to 8O 0 C obtained according mercaptide precipitate can be vacuumed in contrast to the situation with the known methods without any difficulties, because it fails in coarse form. In addition, that in the strongly acidic region (/ o strength 10 ° H 2 SO 4) and precipitated at elevated temperature Mercaptidniederschlag extremely pure is obtained in the present process, so that other amino acids are not included therein. The purity is also shown by the fact that practically no activated carbon is required during the entire processing, which also has a considerable cost-reducing effect. It is surprising that the precipitation is quantitative under these conditions, although Lucas and Beveridge (Biochem. Journal, 34, p. 1359) found the mercaptide precipitate to be very soluble in cold 0.5 η-sulfuric acid. This surprisingly low solubility is due to the absence of chlorine ions. Another decisive advantage of the method according to the invention is the extremely low consumption of copper. When using copper (I) oxide, for stoichiometric reasons, twice the amount is needed than when using copper powder, since half of the amount is converted into divalent copper when the cystine is reduced to cysteine, which is discarded with the mother liquor:

Addition of 15 ml of concentrated HCl partly in. All metals can be used as cathode materials. Solution: With magnetic stirring, cathodes are more noble than iron; expediently, copper is electrolyzed. A cylinder is used as the diaphragm. A suitable diaphragm material is the 40 drical clay cell, which is filled with about 100 ml of dilute HCl, the usual acid-resistant, clay-based. The anode consists of a graphite rod, cylinder with fine porosity. Graphite in dilute hydrochloric acid copper sheet can advantageously be used as the anode material for the cathode made of a sheet of copper bent around the clay cell. Use as anolyte after 12 to 14 amp hours. If you choose the amount of hydrochloric acid consumed, the reduction is complete and the concentration in the cathode compartment is not too high, the sample filtrate is free of copper ions, as can be seen from the copper produced as a powder during the electrolysis, addition of hydrogen sulfide water Soil is sinking. Sucking off the copper can. During the electrolysis, the mercaptide powder poses no problems because it has completely dissolved and the equivalent amount is roughly enough. It can be used again directly, ie copper powder has been deposited for it. Without having to rewash and dry it, it is vacuumed up to 50%, washed with water and dried. Use mercaptid formation in the next batch. About 22 g of copper powder are recovered. If, in the course of the entire cysteine production, one only wants to use the amount of oxidized catalyst added as a catalyst to re-form mercaptides, then the copper can no longer be washed and dried. omitted.
