Abstract
Die Erfindung betrifft Mäuse, in denen das endogene Immunsystem defekt ist und in die sich besonders gut und ohne Konditionierung selbst geringe Mengen von humanen blutbildenden Stammzellen transferieren lassen die dann ihrerseits lebenslang reife humane Blutzellen bilden (Differentierung) und sich selbst regenerieren (Selbsterneuerung). Vorteilhaft und überraschend weisen die erfindungsgemäßen Mäuse mit dem NSG(NOD/SCID/IL2rγnull)-Hintergrund nach dem Transfer prozentual mehr humane Leukozyten im Blut auf als die zuvor von den Erfindern generierten BALB/c Rag2–/– Il2rg–/– KitWv/Wv Mäusen, und den NSG Mäusen ohne eine Mutation im Kit Gen (s. a. 1B), was darauf hinweist, dass die Einnistung und/oder Expansion durch Selbsterneuerung humaner Zellen in den erfindungsgemäßen Mäusen erhöht ist.
Description
- Die Erfindung betrifft Mäuse, in denen das endogene Immunsystem defekt ist und in die sich besonders gut und ohne Konditionierung selbst geringe Mengen von humanen blutbildenden Stammzellen transferieren lassen die dann ihrerseits lebenslang reife humane Blutzellen bilden (Differentierung) und sich selbst regenerieren (Selbsterneuerung).
- Stand der Technik
- Mäuse, in die humane Zellen oder auch Gewebe transferiert sind, sind äußerst wichtige Tiermodelle für die biomedizinische translationale Forschung, für die Impfstoff- und Antikörperentwicklung sowie für die Herstellung von humanen Zellen und Geweben, und sind die einzige Möglichkeit Mechanismen der Entwicklung und Funktion komplexer Systeme (z. B. das hämatopoetische System) in vivo zu untersuchen und zu modulieren.
- Damit die Mäuse die humanen Zellen oder auch Gewebe nicht abstoßen, werden nach dem Stand der Technik hoch immunodefiziente Mäuse als Empfänger gewählt. In den letzten Jahrzehnten wurden dazu eine Vielzahl von genetisch modifizierten Mäusen entwickelt (für eine Übersicht siehe Schultz et al. 2007. Nature Reviews Immunology 7, 118–130). So ist z. B. bekannt, dass in NSG(NOD/SCID/IL2rγnull)-Mäusen durch Transfer von humanen Zellen ein funktionales humanes Immunsystem etabliert werden kann. Damit die humanen Zellen in diesen Mäusen anwachsen ist aber eine vorhergehende Gamma-Bestrahlung (sogenannte Konditionierung) der Mäuse notwendig, um restliche endogene Immun- und Stammzellen zu zerstören (s. Ishikawa F et al. 2005. Blood 106 (5): 1565–73). Nachteilig hat diese Konditionierung nicht unerhebliche Nebenwirkungen, vergleichbar mit einer Strahlenkrankheit (u. a. Hautreizungen, neurologische Störungen, Diarrhoe, Appetitlosigkeit).
- In einer jüngeren Arbeit der Erfinder (C. Waskow et al. 2009 Nature Methods 6: 267–26) wird der Transfer von allogenen Maus-Knochenmarkzellen in Rag2–/– Il2rg–/– KitW/Wv mutante Tiere ohne vorherige Bestrahlung oder chemotherapeutischer Behandlung beschrieben.
- Aufgabe der Erfindung
- Aufgabe der Erfindung ist es Mäuse anzugeben, in die sich besonders gut und ohne Konditionierung humane Zellen, insbesondere blutbildende Stammzellen, transferieren lassen.
- Erfindungsbeschreibung:
- Erfindungsgemäß wird die Aufgabe gelöst durch eine Maus, welche folgende Mutationen aufweist:
- i. eine Mutation in beiden Allelen des Prkdc-Gen, bevorzugt Prkdcscid,
- ii. eine Mutation in beiden Allelen des Gen kodierend für die IL-2-Rezeptor-gamma-Kette, bevorzugt Il2rgtm1Wjl/SzJ,
- iii. eine Mutation im Gen kodierend für den Kit Rezeptor
- Vorteilhaft und überraschend weisen die erfindungsgemäßen Mäuse (s. a.
1A ) nach dem Transfer prozentual mehr humane Leukozyten im Blut auf als die zuvor von den Erfindern generierten BALB/c Rag2–/– Il2rg–/– KitWv/Wv Mäusen, und den NSG Mäusen ohne eine Mutation im Kit Gen (s. a.1B ), was darauf hinweist, dass die Einnistung und/oder Expansion durch Selbsterneuerung humaner Zellen in den erfindungsgemäßen Mäusen erhöht ist. - Die oben genannten Mutationen i. bis iii. sind sogenannte loss of function Mutationen, d. h. das Genprodukt ist durch die Mutation im Gen funktionslos (Nullmutationen i. und ii.) oder in seiner Funktion eingeschränkt (Mutation iii.). Die Mutationen im Prkdc-Gen und im Gen kodierend für die IL-2-Rezeptor-gamma-Kette sind sogenannte Nullmutationen, d. h. es wird kein oder ein funktionsloses Genprodukt gebildet. Die Mutation im Gen kodierend für den Kit Rezeptor resultiert bevorzugt nur in einer eingeschränkten Funktion des Rezeptors oder ist eine konditionale Nullmutation.
- Die erfindungsgemäßen Mäuse werden nachfolgend auch NSG Kitmut genannt. Dabei steht die Abkürzung NSG für „non-obese diabetic“ (NOD), „severe-combined immune deficient“ (Scid), „gamma“ (IL2 receptor gamma chain), und Kitmut für die Mutationen im Gen kodierend für den Kit-Rezeptor (CD117 oder auch v-kit – NCBI UniGene Mm.247073).
- Die bevorzugten Mutationen im Gen kodierend für den Kit-Rezeptor sind KitW, KitWv und/oder KitW41. Dabei steht KitW für die Mutation der Base Guanin zu Adenin in der Splice-donor Stelle in Intron 10, KitWv für die Mutation der Base von Cytosin zu Thymin in Position 2007 und KitW41 für die Mutation der Base Guanin zu Adenin in Position 2519. Mindestens eines der beiden Allele des Kit-Rezeptor-Gens trägt eine dieser Mutationen – bevorzugt tragen beide Allele dieselbe oder auch unterschiedliche Kit-Mutationen. Bevorzugte Allel-Kombinatioen sind ausgewählt aus KitW/Wv, KitWv/Wv, KitW41/W41, KitW/+, KitWv/+ und KitW41/+, besonders bevorzugt KitWv/Wv. Dabei steht „+“ für ein nicht mutiertes Allel (Wildtyp).
- Die erfindungsgemäßen Mäuse mit Mutationen in beiden Allelen des Kit Rezeptors zeigen nach Transfer von humanen Zellen eine erhöhte Anzahl an humanen hämatopoetischen Spenderzellen im Blut im Vergleich zu Mäusen mit einer Kit Mutation in einem Allel oder zu Kit Wildtyp Tieren (
1A ). - Im Knochenmark und in der Milz ist in den erfindungsgemäßen Mäusen nach dem Transfer ebenfalls eine erhöhte Anzahl humaner Leukozyten messbar in den erfindungsgemäßen NSG KitWv/Wv und NSG KitWv/+ Tieren im Vergleich zu NSG Tieren ohne eine Mutation im Kit Gen, wobei die Anzahl an humanen Zellen in Tieren mit Mutationen in beiden Kit-Allelen höher ist als in Tieren mit Mutationen in nur einem Kit-Allel (s. a.
2A ). - Das heißt, im Organ der Hämatopoese, dem Knochenmark, und in einem Organ in dem Immunabwehr stattfindet, der Milz, ist vorteilhaft eine hohe Anzahl an humanen Spenderzellen vorhanden. Im Knochenmark der erfindungsgemäßen Mäuse sind nach dem Transfer die Frequenzen von humanen myeloiden Zelltypen weit höher im Vergleich zu konventionellen NSG Tieren (s. a.
2B ), was einen deutlichen positiven Einfluss auf die Entwicklung von Immunreaktionen haben kann. Immunreaktionen initiiert auf der Basis von humanen Zellen in Mäusen sind in der biomedizinischen translationalen Forschung von besonders hohem Interesse weil diese Vorgänge so komplex sind, dass nur in vivo Studien aussagekräftig sind. - Das heißt, die erfindungsgemäßen Tiere sind wesentlich sensitiver für die Aufnahme selbst kleinster Mengen humaner hämatopoetischer Stammzellen (HSCs) im Vergleich zu den aus dem Stand der Technik bekannten Balb/c Rag2–/– Il2rg–/– KitWv/Wv Mäusen, und erlauben eine zeitlich fast unbegrenzte Einnistung humaner HSCs.
- Die Erfindung umfasst auch eine erfindungsgemäße Maus in die humane Zellen transferiert sind.
- Zur Ausbildung eines humanen Immunsystems in der Maus werden HSCs transferiert. Diese können beispielsweise aus dem Knochenmark, Nabelschnurblut oder auch der fetalen Leber stammen. Bevorzugt sind CD34-angereicherte humane Blutstammzellen aus dem Nabelschnurblut, dem Knochenmark, oder dem Blut von G-CSF behandelten Spendern.
- Bevorzugt werden pro Maus 1 × 104 bis 5 × 105 humane Zellen transferiert. Die Zellen werden dazu bevorzugt intravenös oder direkt in den Knochen injiziert.
- Alternativ oder zusätzlich können andere humane Zellpopulationen, wie andere pluripotente Zellen, Thymuszellen, Hepatozyten, neuronale Zellen, und Derivate aller drei Keimblätter, Krebsstammzellen, sowie solide Organe in die erfindungsgemäßen Mäuse transferiert werden.
- Die Erfindung betrifft auch die Verwendung der erfindungsgemäßen Mäuse für die biomedizinische Forschung, für die Impfstoff- und Antikörperentwicklung sowie für die Herstellung von humanen Zellen und Geweben.
- Die erfindungsgemäßen Mäuse eignen sich insbesondere als Tiermodelle zur Untersuchung der humanen Hämatopoese, Immunität, Zell- und Gewebeerneuerung, Stammzellfunktionen- und differenzierung, Krebs und human-spezifischen Infektionen, und Transplantatabstoßung (Graft versus host Disease).
- Erfindungsgemäße Mäuse, die wie oben beschrieben ein humanes Immunsystem ausgebildet haben, eignen sich sich insbesondere für die Impfstoff- und Antikörperentwicklung.
- Ausführungsbeispiele:
- Die Erfindung wird durch nachfolgende Abbildungen und Ausführungsbeispiele erläutert.

1 zeigt, dass die erfindungsgemäßen Mäuse nach Transfer humaner Zellen eine höhere Anzahl humaner Leukozyten aufweisen.1A zeigt die erfindungsgemäßen Mäusen (NSG KitWv/Wv und NSG KitWv/+) und Vergleichsbeispiele NSG Kit+/+.1B zeigt die Vergleichsbeispiele Rag2–/– Il2rg–/– KitWv/Wv, Rag2–/– Il2rg–/– sublethal bestrahlt („irradiated“) und Rag2–/– Il2rg–/–. Den Tieren wurden jeweils 3 × 105 CD34-angereicherte humane Nabelschnurblutzellen injiziert und die Beteiligung humaner Leukozyten wurde über einen Zeitraum von 16 Wochen gemessen. -
2 zeigt, dass die erfindungsgemäßen Mäuse nach Transfer humaner Zellen eine höhere Anzahl humaner Leukozyten im Knochenmark und in der Milz und eine erhöhte Frequenz von reifen humanen Granulozyten im Knochenmark aufweisen.2A zeigt den Prozentsatz humaner Leukozyten im Knochenmark und in der Milz von erfindungsgemäßen Mäusen (NSG KitWv/Wv und NSG KitWv/+) 18 Wochen nach Transplantation von 3 × 105 CD34-angereicherten humanen Nabelschnurblutzellen. NSG Kit+/+ ist ein Vergleichsbeispiel. - Ausführungsbeispiel 1:Generation von NSG KitWv/+ und NSG KitWv/Wv Tieren.
- NSG KitWv/W Mäuse wurden durch das 15-fache Rückkreuzen von Mäusen mit Kit-Wv Mutationen mit NSG Mäusen generiert. Zu diesem Zweck wurden C57Bl/6 KitWv/+ Mäuse (Japan-SLC, Shizuoka, Japan) mit NSG Mäusen (Jackson Laboratory, USA) gekreuzt und deren Nachkommen für Sirpa des NOD genetischen Hintergrunds, Prkdc, Il2rg, KitWv und die Fellfarbe typisiert. Sobald die erforderlichen Genloci homozygot vorlagen (SirpaNOD/NOD Prkdc–/– Il2rg–/–) wurde nicht mehr typisiert. Nach 15 Rückkreuzungen (kongene Tiere) wurden NSG KitWv/+ Tiere miteinander verpaart oder direkt für Experimente genutzt.
- Ausführungsbeispiel 2: Transplantation humaner hämatopoetischer Stammzellen:
- Nabelschnurblutzellen von mindestens zwei Spendern wurden gemischt und über eine Dichtegradientenzentrifugation für Leukozyten angereichert, und anschließend wurden CD34+ Zellen über einen kommerziell erhältlichen Kit (Miltenyi Biotech) angereichert. Die Zellen wurden danach aliquotiert und pro Maus 3 × 105 Zellen in einem Volumen von 150 Mikroliter intravenös injiziert.
- Tabelle 1 zeigt die Zusammensetzung der humanen Zellen im Knochenmark von den erfindungsgemäßen Mäusen (NSG KitWv/Wv und NSG KitWv/+,) zum gleichen Zeitpunkt nach dem Transfer wie zu
1A beschrieben. Die zelluläre Zusammensetzung von humanen Zellen im Knochenmark von NSG Kit+/+, Blut von BALB/c Mäusen und Knochenmark von Menschen ist zum Vergleich gezeigt. N ≥ 6 für alle Spalten. Claims (4)
- Maus, welche folgende Mutationen: i. eine Mutation in beiden Allelen des Prkdc Gens, ii. eine Mutation in beiden Allelen des Gen kodierend für die IL-2-Rezeptor-gamma-Kette, iii. eine Mutation in mindestens einem Allele des Gens kodierend für den Kit Rezeptor sowie den genetischen Hintergrund bzw. Mutationen der NOD-Maus aufweist, wobei die Mutationen i. bis iii. loss of function Mutationen sind.
- Maus nach Anspruch 1, dadurch gekennzeichnet, dass humane Zellen in die Maus transferiert sind.
- Maus nach Anspruch 2, dadurch gekennzeichnet, dass die transferierten humanen Zellen hämatopoetische Stammzellen oder Stammzellen aus dem Nabelschnurblut sind und in der Maus ein humanes Immunsystem ausbilden, oder dass die transferierte Zellen Krebsstammzellen sind, oder Stammzellen einer der drei Keimblätter sind.
- Verwendung einer Maus nach einem der Ansprüche 1 bis 3 für die biomedizinische Forschung, für die Impfstoffentwicklung, Antikörperentwicklung oder die Herstellung von humanen Zellen oder Geweben.
