Abstract
Description
Die Erfindung betrifft neue Steroid-Styrylfarbstoff-Konjugate, die sich überraschend physiologisch wie Steroid-Hormone verhalten und die sowohl in vivo als auch in vitro in Zellen und Geweben steroidspezifisch mit Proteinen Wechselwirken oder an diese binden. Dies dient einerseits in der funktionellen Analytik dem Nachweis der Interaktion von Steroid-Hormonen in Zellen und Geweben, z. B. mittels mikroskopischer Methoden, andererseits ermöglicht es auch quantitative Bestimmungen von Steroid-Hormonen und von deren Bindungsproteinen, beispielsweise mittels spektroskopischer Verfahren.The invention relates to novel steroid-styryl dye conjugates which behave surprisingly physiologically like steroid hormones and which interact steroid-specifically with proteins or bind to them both in vivo and in vitro in cells and tissues. Dabei können nur potentielle Bindungsstellen und Proteine detektiert werden, die bereits bekannt sind und für die es entsprechende Antikörper gibt. Da Steroidhormone generell in sehr komplexer Weise eine Vielzahl genomischer und nichtgenomischer Effekte induzieren, die über Steroid-Protein-Wechselwirkungen vermittelt werden, ist davon auszugehen, dass viele (wenn nicht gar die meisten) dieser Steroid-Targets unbekannt sind. Ein direkter Nachweis derartiger Steriodinteraktionen in lebenden Zellen ist in der Fachwelt bislang nicht bekannt geworden und wäre von fundamentaler Bedeutung, weil erstens durch die Veränderung der nativen Steroidstruktur, z. B. durch den Einbau von Farbstoffen zu Steroid-Farbstoff-Konjugaten, die Hormonwirkung verändert wird bzw. völlig verloren geht, weil zweitens die Konstrukte von lebenden Zellen nicht aufgenommen (Membranbarriere) werden bzw. weil sie drittens zytotoxisch sind.So far, it is necessary to detect the corresponding binding proteins by immunohistochemistry, so as to indirectly draw conclusions about potential binding sites of said steroid hormones. Direct detection of such steriod interactions in living cells has not previously been reported in the art and would be of fundamental importance because, first, by altering the native steroid structure, e.g. B. by the incorporation of dyes to steroid-dye conjugates, the hormone action is changed or completely lost, because secondly, the constructs of living cells are not taken up (membrane barrier) or because they are thirdly cytotoxic.
Steroidhormone gehören zu den wichtigsten Botenstoffen im menschlichen Körper. Sie werden dabei vor allem von spezifischen Bindungsproteinen transportiert. Auf Grund ihrer Fettlöslichkeit sind sie in der Lage, Zellmembranen zu überwinden und in das Innere von Zellen zu gelangen. Sie binden beispielsweise an Rezeptoren im Zellkern, die als Transkriptionsfaktoren wirken. Steroide beeinflussen daher direkt die Eiweißsynthese, das Zellwachstum und die Zellteilung. Steroidhormone spielen in der Klinik eine große Rolle. Sie werden u. a. als Medikamente verwendet (Antibabypille, Cortisonsalbe, Vitamin D). Steroide spielen aber auch bei der Entstehung bestimmter Krebsarten eine bedeutende Rolle. Krebserkrankungen der Brustdrüse, der Gebärmutter, der Eierstöcke, der Prostata und des Hodens werden unmittelbar mit Steroiden in Zusammenhang gebracht. Für die klinische Diagnostik und nachfolgende Therapie dieser Erkrankungen ist es daher höchst bedeutsam, festzustellen, in wie weit Tumorgewebe auf Steroidhormone reagieren oder nicht. In der pathologischen Routinediagnostik werden zum Beispiel Biopsien von Brustkrebsgewebe, die intraoperativ gewonnen werden, auf das Vorhandensein von Estrogenrezeptoren untersucht. Dazu wird üblicherweise eine Gewebeprobe im histologischen Schnittpräparat mit einem Antikörper gegen den Estrogenrezeptor immungefärbt. Je nachdem, ob das Gewebe Estrogenrezeptor positiv oder negativ ist, wird dann eine Antiestrogentherapie verordnet.Steroid hormones are among the most important messengers in the human body. For the clinical diagnosis and subsequent treatment of these diseases it is therefore highly significant to determine to what extent tumor tissues react to steroid hormones or not. In routine pathological diagnostics, for example, biopsies of breast cancer tissue obtained intraoperatively are examined for the presence of estrogen receptors. For this purpose, usually a tissue sample in the histological section preparation is immunostained with an antibody against the estrogen receptor. Depending on whether the tissue estrogen receptor is positive or negative, then an anti-tumoral therapy is prescribed.

Alternativ können diese Antikörper auch direkt mit Fluoreszenzfarbstoffen oder mit radioaktiven Isotopen gekoppelt (gelabelt) werden. Andere steroidbindene Proteine bleiben unerkannt. Außerdem sind diese Verfahren nicht auf hochauflösende bildgebende mikroskopische Techniken übertragbar und auf Antikörpern basierende Labels sind nur bedingt in lebenden Geweben applizierbar. Die Divergenz der auftretenden Streustrahlung verhindert hier die Anbindung optisch hochauflösender bildgebender Verfahren. Solche Methoden lassen sich außerdem naturgemäß für eine klinische Diagnostik nur sehr eingeschränkt anwenden. Patienten würden dabei radioaktiver Strahlung unmittelbar ausgesetzt. Molecular Imaging ist ein schnell wachsendes interdisziplinäres Forschungsgebiet. Bildgebende Verfahren ermöglichen oftmals eine frühzeitige Diagnose und bessere Klassifizierung von Tumorerkrankungen sowie das Therapiemonitoring. Neben optischen Verfahren spielen Computertomographie, Ultraschall, Magnetresonanzspektroskopie und -tomographie immer noch trotz ihrer ebenfalls immensen Nachteile (Sicherheitsbestimmungen, schwierige Handhabung, Kosten, Spezialgeräte) ebenso wie nuklearmedizinische Techniken wie PET und SPECT eine wichtige Rolle. So kann beispielsweise der Estrogenrezeptorstatus beim Mammakarzinom mit radioaktiv markiertem 16β-[18F1-17β-estradiol mittels PET ermittelt werden. Mit Ausnahme der optischen Verfahren haben alle hier genannten bildgebenden Verfahren den gravierenden Nachteil der oben beschriebenen nur sehr begrenzten optischen Auflösung.Due to their excellent detection sensitivity, fluorescence markers play an outstanding role in the detection of specific biomolecules in cells and tissues. In der sehr umfangreichen Steroid-Literatur wurden bislang auch in anderem Zusammenhang sowohl Steroid-Farbstoff-Konjugate als auch Steroid-Verbindungen mit Styrylstrukturfragmenten beschrieben. So werden beispielsweise Steroid-Fluorochrom-Konjugate beschrieben, die Styryl-Fragmente enthalten. Diese stammen aus dem Bereich der physiko-chemischen Grundlagenforschung bzw. den Materialwissenschaften und verwenden zumeist Cholesterol bzw. dessen Abkömmlinge als spezifisches Strukturfragment zur Erzeugung von Chiralität bzw. Amphiphilie zum Triggern molekularer Selbstorganisationsprozesse oder anderer Materialeigenschaften. Auf diesen Konjugaten beruhende bildgebende Verfahren zur Erkennung von Proteininteraktionen spielen in diesem Zusammenhang keine Rolle. Außerdem haben Cholesterin und seine Derivate in Biologie und Materialwissenschaft zwar eine herausragende Bedeutung als strukturbestimmende und strukturstabilisierende Gruppen, zeigen aber keinerlei Hormonwirkung und gehören damit nicht zur Klassse der Steroidhormone. Rückschlüsse oder gar die Detektion der in situ-Bindung bzw. Wechselwirkung von gonalen z. Vorschrift 1 – allgemeine Arbeitsvorschrift für die Knoevenagel-Kondensation: 2,0 mmol der C-H-aciden Verbindung und 2,1 mmol der entsprechenden Carbonylverbindung werden in Gegenwart von 0,2 ml Piperidin in 12 ml absolutem Ethanol 60 min unter Rückfluss erhitzt. Weniger reaktionsfähige Recktanten erfordern ggf. längere Reaktionszeiten (Dünnschichtkontrolle). Man lässt über Nacht im Tiefkühlschrank (bei ca. –18°C) stehen, saugt ab, wascht mit wenig kaltem Ethanol. In der Regel sind die so erhaltenen Rohprodukte von ausreichend hoher Reinheit (> 95%, NMR- und Dünnschicht-Kontrolle). Bleibt die Fällung des Rohproduktes aus, kann dies durch langsamen Zusatz von wenig Wasser erreicht werden. In Alkoholen oder Wasser leicht lösliche quartäre Ammoniumsalze können als Tetraphenylfluoroborat durch Zusatz einer äuqimolaren Menge von Natriumtetrafluoroborat (30%-ige Lösung in Methanol oder Ethanol) gefällt werden. Nicht kristallisierende oder schwer fällbare oder zur Zersetzung neigende Reaktionsprodukte werden nach dem Entfernen des Lösungsmittels am Rotationsverdampfer säulenchromatographisch gereinigt (Kieselgel, Essigester/Hexan, bei salzartigen Verbindungen unter Zusatz von 1 bis 5% Methanol).Protocol 1 - general procedure for the Knoevenagel condensation: 2.0 mmol of the C-H-acidic compound and 2.1 mmol of the corresponding carbonyl compound are refluxed in the presence of 0.2 ml of piperidine in 12 ml of absolute ethanol for 60 min. Vorschrift 2 – Kondensation aromatischer hydroxysubstituierter Aldehyde mit C-H-aciden Verbindungen in Acetanhydrid und anschließende Spaltung des gebildeten Acetates – allgemeine Vorschrift am Beispiel der Herstellung von 2-(4-Hydroxystyryl)-8-hydroxychinolin:
2 mmol 8-Hydroxychinaldin werden mit 2,1 mmol 4-Hydroxybenzaldehyd und 3 ml Acetanhydrid versetzt und 6 Stunden unter Rückflussbedingungen erhitzt. Man engt am Rotationsverdampfer ein und rührt das erhaltene braune Öl 30 min mit 10 ml 10%-iger Natronlauge bei Raumtemperatur. Man saugt oder dekandiert ab, wascht mit Wasser und hydrolyiert das erhaltene ockerfarbene Acetat durch einstündiges kochen mit 10 ml 10%-iger Natronlauge. Man filtriert heiß (G4-Fritte) und säuert nach dem Abkühlen auf Raumtemperatur mit Salzsäure an und saugt das ausgefallene ockerfarbene Produkt ab. Nach dem Waschen mit Wasser, Natriumhydrogenkarbonat und zuletzt nochmals mit Wasser wird aus Ethanol umkristallisiert: 68% leuchtend dottergelbes Produkt (F 234–236°C). Mit Vorteil (höhere Reinheit und Ausbeute) kann auch das Hydrochlorid durch Ausfällen aus einer ethanolischen Lösung des Rohproduktes mit 1–2 ml konz. Salzsäure gefällt und bei Bedarf aus Ethanol/Wasser mit Zusatz einer geringen Menge an konzentrierter Salzsäure umkristallisiert werden: 70% gelbes amorphes Pulver (Rohprodukt: F = 215–217°C, > 185°C Zers.).Protocol 2 - Condensation of aromatic hydroxy-substituted aldehydes with CH-acidic compounds in acetic anhydride and subsequent cleavage of the acetate formed - general procedure using the example of the preparation of 2- (4-hydroxystyryl) -8-hydroxyquinoline

1,36 g ß-Estradiol (5 mmol), 2,76 g wasserfreies und vorgetrocknetes Kaliumcarbonat (20 mmol), 100 mg 18-Krone-6 (0,38 mmol) und 8,64 g 1,4-Dibrombutan (40 mmol) werden in 80 ml trocknem Toluol bei 110°C in einer Argon-Atmosphäre intensiv gerührt. Nach 20 Stunden (Dünnschichtkontrolle, Laufmittel Essigester/Hexan = 2:1, Kieselgel Silufol-Platten) wird durch Zugabe von 40 ml Wasser und 30 ml Essigester aufgearbeitet. Das Gemisch wird noch ca. 10 min intensiv gerührt und die Phasen werden getrennt. Man extrahiert jeweils noch einmal mit 30 ml Lösungsmittel nach, trocknet die vereinten organischen Phasen über Natriumsulfat und engt am Rotationsverdampfer ein. Das erhaltene farblose Öl wird säulenchromatografisch gereinigt (200 g Kieselgel 60, Elutionsmittel: Essigester/Hexan, Gradient 1:10 bis 2:1). Nach Umkristallisation aus Methanol und Trocknung im Exsikkator (P2O5) werden 1,3 g (Ausbeute 66,1%, M = 393,36 g/mol) 3-(4'-Brombut-1'-yloxy)-17β-hydroxy-estra-1,3,5(10)-trien erhalten: F = 101,5–103,0°C), Rf = 0,58 (Essigester/Hexan = 2:1, Silufol).Rule 3 - General procedure for the alkylation of 3-hydroxyestra-1,3,5 (10) -trienes using the example of 17-β-estradiol (

Picolin werden unter Argon 3 Stunden auf 100°C erhitzt. Anschließend wird das hellbräunlich-heterogene Reaktionsgemisch auf Raumtemperatur abgekühlt und mit Ether sorgfältig 3 × gewaschen. Das amorphe beige Rohprodukt (1,25 g, 77,9% Ausbeute, M = 486,49 g/mol; Rf = 0,45 Methanol/Wasser/Essigsäure = 4:2:0,5; Silufol) kann direkt weiterverarbeitet werden. 140 mg N-1'[(17β-hydroxy-estra-1,3,5(10)-trien-3-yloxy)-but-4'-yl]-4-methylpyridinium Perchlorat (0,277 mol) und 100 mg 4-Dimethylaminobenzaldehyd (0,670 mmol) werden in 20 ml absolutem Ethanol 3 Stunden unter Rühren und Rückflußbedingungen gekocht. Man lässt über Nacht bei 4°C stehen, saugt ab (G4-Fritte) und wäscht gründlich mehrmals mit Ether. Nach Umkristallisation aus Ethanol/Aceton (4:1), waschen mit Ether und Trocknung erhält man 153 mg (86,7% Ausbeute, M = 637,21 g/mol) eines leuchtend weinrotes Produktes: F = 140–145°C.
