Durch Liebrecht und Röhmann ist bekannt, dafs neutrale Case'inalkalilösungen durch Silbersalze und dergl. bei vorsichtigem Zusatz derselben nicht gefällt werden. Dem gegenüber zeigen die neutralen Pflanzenglobulinalkalilösungen ein abweichendes Verhalten; es gelingt nicht, sie mit Schwermetallsalzen zu versetzen, ohne dafs ein auch beim Erwärmen unlöslicher Niederschlag entsteht. Wohl aber kann man das Entstehen dieses Niederschlags vermeiden, wenn man statt einer neutralen Globulinlösung eine alkalische (gegen Phenolphtale'in) verwendet. Hierbei hat der Erfinder die überraschende Beobachtung gemacht, dafs die ursprünglich alkalische Globulinlösung nach Zusatz des Schwermetallsalzes und erfolgter Reaction gegen Phenolphtalei'n sauer oder neutral reagirt, vorausgesetzt, dafs der verwendete Alkaliüberschufs nicht zu grofs war. Auf diese Beobachtung gründet der Erfinder sein Verfahren. Zu diesem Behuf versetzt man zunächst eine neutrale Pflanzenglobulinalkalilösung mit einem bestimmten gröfseren Ueberschufs von Alkali und giebt hierauf die berechnete Menge Schwermetallsalz (etwa Quecksilberchlorid oder Silbernitrat) zu; diese alkalische Lösung titrirt man zurück mit Y10 Normalsäure, entweder bis sich eben ein bleibender Niederschlag zu zeigen beginnt oder mit Hülfe von Phenolphtalem bis zum Farbenumschlag. Nunmehr löst man die Hauptmenge des Globulinalkalis in Wasser, versetzt mit dem aus dem Vorversuch sich ergebenden Alkaliüberschufs und sodann mit der Lösung der berechneten Menge Schwermetallsalz.
