Vorrichtung zur dielektrophoretischen Sammlung suspendierter TeilchenDevice for the dielectrophoretic collection of suspended particles
The invention relates to a device for the dielectrophoretic collection of suspended particles, in particular biological cells and / or vesical, in the high-frequency alternating electric field. This process is used in particular in hybrid cell technology, medical-biological research and biotechnology «
Membrane Biol. 67, 165 - 182 (1982) /.The collection of suspended biological cells and / or vesicles such as liposomes or cell shadows to linear aggregates for the purpose of their subsequent Fuaionierung done in inhomogeneous high-frequency electric field. In known arrangements, the inhomogeneity of the high-frequency radio-frequency field is achieved by a corresponding electrode geometry, for example by parallel cylinder electrodes or a tip / plate electrode arrangement / U. Zimmermann, 3, Vienken; α. Membrane Biol. 67 , 165-182 (1982).
Cells and / or vesicles is only with sufficiently large field strength of the electr. High frequency field possible. If this field strength in a suspension vessel with the o.g. Realized electrodes for the narrowest part of the electrode gap, then the cells
Dies begrenzt die Ausbeute der dielektrophoretisch gesammelten biologischen Zellen und/oder Vesikel erheblich.and / or vesicles outside this space are no longer aggregated by the electric field. This significantly limits the yield of the dielectrophoretically collected biological cells and / or vesicles.
The object of the invention is to avoid the disadvantages of the known processes and to increase the proportion aen dielectrophoretically collected biological cells and / or vesicles essential.
The object of the invention is to treat a particle suspension as a whole dielectrophoretically homogeneous, d. H. to allow almost the same field strength at each location of the suspension volume to form aggregates. The object is achieved with a device for the dielectrophoretic collection of suspended particles according to the invention in that two parallel plate electrodes are arranged in a vessel in which the suspended particles, in particular biological cells and / or vesicles, are located. This produces a homogeneous high frequency electric field of large spatial extent. Also suitable as electrodes are two coaxially arranged cylinders of sufficiently large diameter, between which an annular gap for receiving the particle suspension is present. The inhomogeneous electric field required for the dielectrophoresis is generated by the suspended biological cells and / or vesicles themselves, if the electrical conductivity of the suspension medium is chosen to be smaller than that of the cell interior and the frequency of the applied alternating field is so high that the AC resistance of the cell and vesicle membranes become negligible. The electric field lines then preferably pass through the cell or vesicle interior and cause a gradient of the electric field strength in the vicinity of the particle, which leads to the formation of linear particle
aggregaten kommt.aggregates comes.
The advantage of the device according to the invention is that the yield of particle aggregates is substantially increased compared to the conventional arrangements. Furthermore, the transfer of aggregates, z. B "into an incubation vessel, easier to carry out.
Two planar electrodes made of cell-compatible material (2. 8 stainless steel) with a round or rectangular cross-section are arranged parallel and opposite each other at a small distance (eg 1 mm) so that a free volume for the particle suspension results between the electrodes. The two plate electrodes are connected to an electrical high frequency generator (eg 1 MHz). The dielectrophoretically to be collected biological cells and / or vesicles are suspended in an isotoraic medium of low electrical conductivity (eg., 5.4% glucose solution) and placed in the electrodes · space. After the application of the electrical high-frequency voltage (ζ B. 100 V), linear cell and / or vesicle aggregates form in the suspension, which are preferably arranged in the direction of the electrode normal.
