Biomateriál na bázi nanoviákenných vrstev a způsob jeho přípravyBiomaterial based on nanofiber layers and method of its preparation
Oblast technikyTechnical field
The present invention relates to a biomaterial based on nanofibrous layers and to a process for its preparation.
Dosavadní stav technikyBACKGROUND OF THE INVENTION
The preparation of various forms of solids characterized by the presence of pores or specific surface areas corresponding to the structural units of the system in the order of several nanometers to hundreds of nanometers is currently in the forefront of material engineering. Virtually all important features of such systems are derived from the extremely large specific surface area. While porous polymer-based materials and nanomaterials prepared through organized supermolecular structures have been extensively studied for a long time, fiber tens in the order of tens to hundreds of nanometers has received increased attention only in the last five years. These fibers can be used to form fibrous layers of good mechanical properties directly during the spinning process. The mechanical properties and morphology are favorably influenced by the anisotropic nature of the fibrous layer. The pores in such layers have a specific geometry which makes the fiber surfaces easily accessible.
2004 Mar-Apr; 25(7-8): 1 289-97].Nanofibers are generally described as fibers whose diameter ranges in the submicron region, that is in the range up to 1000 nm. They have a number of exceptional properties, such as large fiber specific surface area, high fiber porosity and small pore diameter. In terms of cell cultures, the structure of nanofibrous layers approximates that of the extracellular matrix. This is consistent with repeated observations of higher cell adhesion to nanofibres than to microfibers or layers of identical polymers [Schindler, Ahmedl, Kamal, NurEKA, Grafe TH, Young Chung H, et al. Synthetic nanofibrillar matrix promotes in vivo-like organization and morpho30 genesis for cell in culture. Využití mikrovláken k vytvoření porézních trojrozměrných tkáňových náhrad obsahujících buňky je známo již od roku 1993; tato technologie sloužila nejprve k experimentální přípravě náhrad kloubních chrupavek [Robinson D, Efrat M, Mendes DG, Halperin N, Něvo Z. Implants compo40 sed of carbon fiber mesh and bone-marrow-derived, chondrocyte-enriched cultures for jo int surface reconstruction. Bull Hosp Jt Dis. 1993 Spring; 53(1): 75-82.], Toto využití mikrovláken je chráněno patentem [US 5 759 830, US 5 770 417]. The use of microfibers to create porous three-dimensional tissue replacements containing cells has been known since 1993; This technology was first used for experimental preparation of articular cartilage replacements [Robinson D, Efrat M, Mendes DG, Halperin N, Neva Z. Implants compo40 sediment of carbon fiber mesh and bone-marrow-derived, chondrocyte-enriched cultures for jo int surface reconstruction. Bull Hosp Jt Dis. 1993 Spring; 53 (1): 75-82.], This use of microfibers is protected by a patent [US 5,759,830, US 5,770,417]. The spatial network of microfibers ensures the mechanical strength of the material, maintains its three-dimensional shape and the spatial arrangement of the cells. However, this method is not applicable to nonwoven nanofiber fabrics, since the material would collapse under its own weight when it reaches the porosity required to interpose cells between the fibers (pores in the order of units to tens of micrometers).
1).The standard procedure for maintaining both tissue and stem cell cultures is monolayer cultivation; for this cultivation, specially coated materials adhere to the cells (tissue plastic, laminin-coated glass, polylysine, tissue plastic covered with inactivated fibroblasts, etc.). Alternative cultivation methods include suspension cultivation or thin gel-based materials such as MatriGel®. All the cultivation methods described have disadvantages. The growth of cells in a monolayer culture is conditioned by their adhesion to the material on which they are cultured; the medium only has access to the capical (away from the culture surface) layer; the properties of the cells change upon reaching a continuous (conCZ 300805 B6 tluent) layer; the substances produced by the cells are released into the medium and do not remain around the cells. The advantage of monolayer cultivation is - besides being a very well standardized procedure - the ability to monitor individual cells in a microscope (Fig. 1).
2).When cultivating cells on fibers or in macroporous materials whose pore size is in the order of tens of micrometers, the cells behave similarly as in monolayer - the flat surfaces of the material grow (Fig. 2).
Vrstvy z netkané nanovlákenné textilie připravené metodou elektrospinningu je možné využít ke ío kultivaci buněk v monolayeru podobně jako například tkáňový plastik; první experimenty s buněčnými kulturami na nanovlákenných textiliích byly publikovány v roce 2002 [Li WJ,The electrospinning nonwoven nanofiber layers can be used to cultivate cells in a monolayer similar to a tissue plastic; the first experiments with cell cultures on nanofibrous textiles were published in 2002 [Li WJ,
Laurencin CT, Caterson EJ, Tuan RS, Ko FK. Electrospun nanofibrous structure: a novel scaffold for tissue engineering. J Biomed Mater Res. 2002 Jun 15; 60(4): 613-21.]. Laurencin CT; Caterson EJ; Tuan RS; Ko FK. Electrospun nanofibrous structure: a novel scaffold for tissue engineering. J Biomed Mater Res. 2002 Jun 15; 60 (4): 613-21.]. The system for the cultivation of cells on nanofibrous layers is protected by patent [US 6,790,455]. The expected beneficial use of nanofibrous layers in biomedicine has led to the filing of the patent application [WO 2005/025630], in which the use of nanofibres for tissue replacement construction is not mentioned.
Řada autorů se pokoušela o vytvoření trojrozměrné tkáňové náhrady s využitím nanovláken. Many authors attempted to create three-dimensional tissue replacement using nanofibers.
It has been patented to use a nanofibrous layer as the basis on which smooth muscle cells are cultured and thereafter other cell layers separated by biomaterial (e.g., polymer-based) are formed [US 6,428,802]. The possibility of tissue replacement by cell cultivation on parallel layers from a network of nanofibres and the underlying layer (carriers, in the original substrate) was also published. Furthermore, these layers can be separated from each other by any biomaterial [WO 2005/047493], but even here the formation of a homogeneous tissue by interconnecting the layers containing the cells is not described.
Part 4: Growth of rat bone marrow stromal cells in three-dimensional hydrogels with positive and negative surface charges and in polyelectrolyte complexes. Journal of materials science. 2006 Sep; 17(9): 82933.].Materials based on 2-hydroxyethyl methacrylate (HEMA) copolymers have been prepared repeatedly. These copolymers are highly biocompatible and the adhesion of cells to their surfaces depends on their electrical charge [Lesny P, PradnyM, JendelovaP, Michalek J, Vacik, Sykova E. Macroporous hydrogels based on 2-hydroxyethyl methacrylate. Part 4: Growth of rat bone marrow stromal cells in three-dimensional hydrogels with positive and negative surface charges and in polyelectrolyte complexes. Journal of Materials Science. 2006 Sep; 17 (9): 82933.].
Podstata vynálezuSUMMARY OF THE INVENTION
The object of the present invention is a biomaterial based on nanofibrous layers, which consists of at least two nanofibrous layers covered on both sides by continuously living cells, which layers are interconnected by the growth of cells, the nanofibrous layers are non-woven and consist of synthetic polymers or copolymers of monomers selected from the group consisting of methacrylic acid esters, methacrylic acid amides, urethanes, vinyl alcohol and lactic acid monomers and derivatives thereof.
Preferably, the monomers are selected from the group consisting of 2-hydroxyethyl methacrylate, 2-ethoxyethyl methacrylate and 2-hydroxypropyl methacrylamide.
It is a feature of the invention that the cells are selected from the group consisting of connective tissue cells, epithelial cells, parenchymatous organ cells, and mesenchymal stem cells derived from bone marrow or adipose tissue. Preferably, the cells are selected from the group consisting of chondrocytes, fibroblasts, hepatocytes and mesenchymal stem cells derived from bone marrow or adipose tissue.
The ability of a polymer to form into fibers, i.e., fiberize, is influenced by a variety of process and material parameters such as electric field strength, electrical conductivity, viscosity, molar mass, surface tension, polymer concentration, solvent, dielectric properties of polymer solution, hydrophilicity / hydrophobicity , the polymerization degree and the degree of polymer branching, or the experiment set-up. The properties of the spinning material are influenced not only by the chemical composition of the polymer, but also by the spinning parameters. By changing these parameters it is possible to influence not only the spinning process, but also the structure of the resulting layer and the fineness of the fibers within certain limits. However, optimum process and system parameters need to be sought separately for each electrospinned polymer solution, since the parameters found are transferable to a very limited extent.
The method of preparation of nanofibres of the synthetic polymer or copolymer according to the present invention can be the electrospinning method "Nanospider", in which the fibers are formed by the electrostatic field from a thin layer of polymer solution carried by a wading roller which is also a positive electrode. which is also a counter electrode [CZ 294274 (B6), WO 2005/024101],
Contact with and adhesion to extracellular matrix molecules are very important factors for most cells that determine their adhesion and growth in the body. Cell culture in two-dimensional culture systems does not reflect the natural environment in the body. The structure of the nanofibrous layer is compared to the structure of the basement membrane.
The cells adhere strongly to the nanofibrous layers of the present invention formed of polymers with a nanofibrous structure, even when they do not show affinity for the polymers without the nanofiber structure. After the cells are inoculated on the nanofiber layer, the nanofiber layer grows densely in the days to weeks (depending on the cell concentration) and their protuberances also grow between the fibers. If the layer of nanofibres is sufficiently thin (units up to tens of micrometers), it exceeds the cell growth to the opposite side of the layer; from a layer thickness of about 100 microns, the cells on the other side of the layer no longer exceed.
The advantage of cell cultures on nanofibrous layers is the possibility of bilateral growth of cells on nanofibrous layer. At the same time, cells growing on the nanofibrous layer retain the ability to grow in space, which is manifested by the fact that when two nanofibrous layers containing cells come closer, these layers grow together.
Another object of the present invention is a process for the preparation of a biomaterial according to the present invention, comprising at least two nanofiber layers interconnected by cell growth, comprising preparing a synthetic polymer or copolymer of monomers selected from the group consisting of methacrylic acid esters, meth40 acrylic acid amides, urethane, vinyl alcohol and monomers derived from lactic acid and its derivatives, prepared synthetic polymer or copolymer with a special electrostatic spinning method, followed by the electrostatic spinning prepared spinning layer of synthetic polymer or copolymer and sowing cells and culturing under standard conditions for tissue culture cultivation achieving a continuous (confluent) coating of the nanofibrous layer, and then at least two layers of a spun synthetic polymer or copolymer, poros The cells are stacked manually or automatically, and the stack is loaded at 0.5 to 2 kPa and cultured under standard tissue culture conditions for 1 to 4 weeks.
Standard conditions for culturing tissue cultures include the use of a combination of media suitable for the cell type, fetal calf serum or its substitute, antibiotics or growth factors, and cultured in incubators at 37 ° C and 5% CO 2; 3 days.
Preferably, the monomers are selected from the group consisting of 2-hydroxyethyl methacrylate, 2-ethoxyethyl methacrylate and 2-hydroxypropyl methacrylamide.
It is a feature of the method of the invention that the cells are selected from the group consisting of connective tissue cells, epithelial cells, parenchymatous organ cells and mesenchymal stem cells derived from bone marrow or adipose tissue. Preferably, the cells are selected from the group consisting of chondrocytes, fibroblasts, hepatocytes and mesenchymal stem cells derived from bone marrow or adipose tissue.
io Celý postup podle vynálezu tedy spočívá v neoddělitelných procesech zahrnujících přípravu a charakterizaci polymeru, proces zvláknění, osídlení nanovláken vhodnou buněčnou kulturou, kultivaci buněk na vrstvě nanovláken a zformování výsledného materiálu do podoby vhodné k vybrané aplikaci v tkáňovém inženýrství nebo konvenčním léčebném procesu. Thus, the whole process according to the invention consists in inseparable processes including preparation and characterization of the polymer, spinning process, nanofibers settlement by suitable cell culture, cultivation of cells on the nanofibers layer and forming the resulting material into a form suitable for selected tissue engineering or conventional treatment process.
In the field of biomedical applications, it is necessary to take into account the quality of the material used from several points of view. The first aspects are the resulting useful lysico-chemical properties such as mechanical (strength, hardness, toughness, elasticity), swelling (equilibrium water content), trans20 port (permeability), optical (refractive index) and surface (roughness, wettability, contact angle) ) Properties. Other important aspects are the physical and chemical structure of the material, which significantly affects the biotolerance of the material in applications in the environment. It is a required interaction of the material with living tissue, ie cell culture, its chemical stability, or controlled biodegradation. Requirements for the properties of biomaterials are often anti-permissive and a compromise must be sought between them. Therefore, for many applications, there is only a narrow selection of suitable chemical structures.
Another problem of bio-applications is the need for long-term tests of the effects themselves and in particular the possible adverse effects of the substances used. For all these reasons, there is a natural effort to test in new applications already established and therefore long-term used tested materials, which are well-suited to satisfy the toxicity or tolerance tests. In the field of hydrogels, such a material is cross-linked poly (2-hydroxyethyl methacrylate) and some of its copolymers or poly (2-hydroxypropyl methacrylamide),
Vhodnou kombinací parametrů zatížení, tloušťka vrstvy, procento plochy porostlé buňkami, počet vrstev a typy buněk v jednotlivých vrstvách - můžeme dosáhnout i vzniku relativně komplexních tkání. Příklad: kombinací vrstev porostlých hepato40 cyty a vrstev s buňkami endotelu lze vytvořit tkáňovou náhradu s podobným zastoupením buněk, jaké je v jatemí tkáni.The biomaterial according to the present invention can be used as a three-dimensional organ replacement: by assembling nanofibrous layers overgrown with cells and their uniform loading, three-dimensional implants can be formed. By a suitable combination of load parameters, layer thickness, percentage of cell area, number of layers and cell types in each layer - relatively complex tissues can also be produced. Example: by combining layers overgrown with hepato40 cells and layers with endothelial cells, a tissue replacement with a similar proportion of cells to that in liver tissue can be formed.

12B).After this time, a part of the material, which was subjected to a pressure higher than 0.5 kPa, developed sparse mesenchymal tissue overgrown with all layers of nanofibres and extracellular matrix deposition (Fig. 12A); In the unloaded part of the material there was no interconnection of layers (Fig. 12B).
