技术领域

背景技术
为了从空气中去除造成环境污染的有害物质,经常使用这样一种滤器,该滤器含有:蜂房;多孔的陶瓷;由玻璃纤维、金属纤维、或碳纤维支承的再生纤维素;由木质纸浆、锯屑、椰子壳、煤、苯酚、合成纤维或其它原料制成的活性碳颗粒、粉末或纤维材料;或沸石。已经知道这些材料在某种程度上是有用的。作为另一种用于除去有害物质的技术,日本未审专利申请公开号为No.2002-355299已经公开了使用一种光催化剂的空气清洁过滤器,其中起光催化作用的功能性颗粒(氧化钛,氧化锌,氧化铁等)由活性碳纤维支承。此外,日本未审专利申请公开号为No.2001-95552已经公开了一种烟草烟雾过滤器,其包括含有至少CaO,B2O3,SiO2,和Al2O3的多孔玻璃。
近来逐渐关注由二噁类化合物引起的空气污染和烟雾引起的健康损害,因此,日益希望得到一种比以往更有效地除去空气中的二噁类化合物和由烟雾产生的有害物质的方法。在使用空调,鼓风加热器,空气清洁器,电动真空清洁器,干燥器和其它电动或电子设备时,上面所述的过滤器能有效地去除灰尘,但是不利的是,不能有效地除去二噁类化合物。
由于上述情况,本发明的目的是提供一种除去对人体有害的化学物质的有效方法,该化学物质例如是空气中的二噁类化合物和烟草烟雾产生的芘;一种除去有害物质的滤器;以及安装滤器的烟雾器具、面具、电动和电子设备。

实施本发明的最佳方式
在本发明中使用的DNA包含两条螺旋形的多核苷酸链(双螺旋)。优选地,它从鱼的睾丸(testes)中得到,例如鲑鱼,青鱼或鳕鱼,或者来自哺乳动物或鸟类的胸腺,例如母牛,猪或鸡。在DNA的双链中,结构互补的平面碱基对垂直于螺旋轴,从两条多核苷酸链的糖-磷酸酯骨架延伸至螺旋中心,每一对碱基是氢键结合的。这种双链DNA在碱基对之间有空间存在,在B-形式的DNA中,空间测量大约宽度为1.1nm,高度为0.34nm。平面小分子能捕获入所述空间中。这里称之为插入。小分子的电荷或疏水性促进了这种现象。很多对人体有害的物质,例如二噁类化合物,包括了多个苯环,因此具有一种平面结构。因此本发明的发明者发现环境空气中的二噁类化合物或吸用烟草产生的有害物质能利用DNA有效地除去。
本发明能除去有害物质的方法涉及上述DNA特性的使用。在这种方法中,使含有有害物质例如二噁类化合物的气体与DNA接触从而被DNA捕获,因此被除去。同样,本发明涉及一种用于除去有害物质的滤器,包括DNA和能支承DNA的纤维状滤器材料。在本发明中使用的纤维状滤器材料指的是纤维的集合体。然而,这不必局限于纤维,任何材料都可以被使用,只要它和纤维在状态上相似,并且能够支承DNA和允许含有有害物质的气体渗透。优选地,它可以是纤维素纤维,例如纸;玻璃纤维;天然纤维;合成纤维,例如聚乙烯,聚丙烯,或者尼龙树脂;或者蜂房。
本发明中用于除去有害物质的滤器能产生有效除去有害物质的效果,只要DNA以能够在纤维之间自由移动的游离状态支承于纤维状滤器材料的至少某些区域的纤维之间,其中含有有害物质的气体将渗透入所述区域中。然而DNA也能被整个纤维状滤器材料支承。在本发明中,DNA被纤维状滤器材料支承意味着DNA以不会从纤维状滤器材料脱落的状态存在。DNA的形式没有特别限制,但是DNA可以是粉末,纤维或膜。
以游离状态在滤器物质的纤维中支承DNA的方式没有特别地限制,可以采用多种方式。例如,如图1中显示,通过从动物组织中提取,纯化和冷冻干燥成粉末、纤维或膜形式而制备的预定数量的DNA1被直接和均衡地分散或撒到纤维状滤器材料2从而相互接触,从而被支承在纤维状滤器材料2的纤维之间。这样,被纤维状滤器材料支承的DNA1被支持在纤维状滤器材料2的纤维2’中,不会从纤维中脱落,如图2中显示的,它是图1的片断放大图。
在用于除去有害物质的滤器中,优选地,DNA以和另一种大分子化合物的混合物的形式,至少在含有有害物质的气体能够渗透的纤维状滤器材料的区域被支承。通过和另一种大分子化合物的组合,与单独使用相比,DNA在除去水之后能更加坚固地被纤维状滤器材料支承。结果,减少了DNA从纤维状滤器材料脱落的可能性,因而,用于除去有害物质的滤器即使在长期使用中也表现了稳定的能除去有害物质的效果。任何大分子化合物都可以被使用,只要它不影响除去有害物质的效果并且能与DNA混合和能通过物理力(例如分子间力或静电力)和化学键(例如共价键,离子键或配位键)之一或两者吸附到纤维状滤器材料上。可举例的大分子化合物包括:丙烯酸盐,例如聚丙烯酸钠和聚甲基丙烯酸钠;和纤维素,例如甲基纤维素,羧甲基纤维素,和他们的盐。
也优选地是,本发明的用于除去有害物质的滤器中,DNA被固定到纤维状滤器材料上,从而至少在含有有害物质的气体能够渗透的纤维状滤器材料的区域被支承。在这种结构中既然DNA更加坚固地固定到纤维状滤器材料,DNA就不易从纤维状滤器材料上脱落。结果,即使滤器使用很长的时间,本发明的用于除去有害物质的滤器也几乎不会由于DNA的脱落而降低除去有害物质的效果,。
在本发明用于除去有害物质的滤器的一个变体中(其中DNA固定到纤维状滤器材料上),DNA通过紫外线暴露固定到纤维状滤器材料。在这一变化中,优选地,辐射到DNA的紫外线的波长范围在250到270nm。
可选择的,用于除去有害物质的滤器中,DNA通过席夫碱形成产生的化学键固定到纤维状滤器材料上。例如,DNA固定到纤维素纤维(即纤维状滤器材料)得到用于除去有害物质的滤器,正如图3所显示的。特别地是,(a)允许NaIO4在25℃作用到纤维素纤维(I)一个小时以产生纤维素纤维(II),在纤维素纤维(II)中,纤维素分子的一部分具有醛基;接着(b)加入5mg/mlDNA,在80℃反应8小时。因而得到用于除去有害物质的滤器(III),其中DNA固定到纤维素纤维上。在用于除去有害物质的滤器的另一个变体中(其中DNA通过席夫碱形成产生的化学键被固定),DNA可以先通过席夫碱形成产生的化学键固定到玻璃珠的表面,玻璃珠支承于纤维状滤器材料。固定DNA到玻璃珠的方法中,(a)允许NaIO4在25℃作用到DNA上3个小时以产生具有醛基的DNA;接着,(b)允许具有醛基的DNA和胺化的玻璃珠在25℃反应3个小时;(c)NaBH3CN被加入到产生的中间体中反应6个小时,正如图4中显示的。产生的固定有DNA的玻璃珠在纤维状滤器材料的纤维间被支承,正如图5中显示的,例如,以游离的状态被支承。因此,构造了本发明的用于除去有害物质的滤器。
对于用于除去有害物质的滤器的另一种变体(其中DNA固定到纤维状滤器材料上),DNA通过凝胶状态或以从凝胶中除去水分得到的多孔形式固定到纤维状滤器材料上。为了制备滤器,纤维状滤器材料浸入到DNA的水溶液中,一种分散剂例如聚丙烯酸钠加入到溶液中形成凝胶。接着,取出(draw up)纤维状滤器材料,或者取出后,将水分从粘附到纤维状滤器材料的凝胶中除去以得到多孔形式。可选择的,DNA可以与水和凝胶剂混合,例如丙烯酰胺或硅胶,以形成溶胶,纤维状滤器材料浸入溶胶,或用溶胶包裹或喷洒。所述溶胶干燥或冷冻干燥得到一种凝胶,或者进一步,除去产生的凝胶中的水得到一种多孔形式。明胶,琼脂,和此类物质可以作为凝胶剂使用。
一种能除去有害物质的滤器的更优选的变体中(其中DNA固定到纤维状滤器材料上),DNA通过在DNA双链之间插入结合到纤维状滤器材料上的补骨脂素从而固定到纤维状滤器材料上。例如,正如图6中显示的,补骨脂素(以X指定的部分)结合到聚亚烷基(Y指定的部分,n表示1或大于1的整数),聚亚烷基进一步结合到一种至少部分被磷酸化的纤维状滤器材料上。有这种结构的补骨脂素插入到待捕获的DNA中。在这一变体中,由于补骨脂素具有一种平面结构,补骨脂素通过插入到双链DNA的一个碱基对a和另一个碱基对b之间的空间而在DNA中被捕获,正如图7中显示的。结果,DNA固定到纤维状滤器材料上,其中聚亚烷基作为间隔基。在这个例子中,通过紫外线辐射,DNA被坚固地固定到补骨脂素上。可见光也可以取代紫外线将DNA固定到补骨脂素上。使用可见光与使用紫外光的情况不同,其在于DNA可以被坚固地固定到补骨脂素,即便DNA没有被干燥,例如,甚至在水中。因此,与紫外线的情况相比,固定条件相对不受限制。在本发明使用补骨脂素的滤器中,有害物质,例如二噁类化合物在DNA碱基对之间的一些空间,而不是在补骨脂素插入的空间被捕获,正如图7所显示的。在这一变体中使用的间隔基可以是聚乙二醇基而不是聚亚烷基。
在本发明用于除去有害物质的滤器中,DNA可以通过偶联剂固定到纤维状滤器材料上。对于这种用于除去有害物质的滤器,DNA和硅胶(一种硅烷偶联剂)和,如果需要,另一种添加剂混合来制备一种水溶液或一种悬浮液。一种载体浸入到水溶液中或悬浮液中,或者纤维状滤器材料由水溶液或悬浮液涂布或喷雾以至于载体充满水溶液或悬浮液,接着除去水分。这种形式的变体对以下情况特别有利,即其中DNA(作为一种有机物质)更加坚固地固定到一种无机的纤维状滤器材料。优选的偶联剂是,例如,硅烷偶联剂。
用于除去有害物质的滤器可以通过将DNA的水溶液充满纤维状滤器材料,接着从纤维状滤器材料中除去水分而制备。这样DNA以一种游离的状态在纤维状滤器材料的纤维中支承。例如,将纤维状滤器材料用刷子或类似物涂渍DNA水溶液,接着在热空气中或用加热器干燥,优选的在60℃或更低,从而得到能除去有害物质的滤器。可选择的,如图8所显示,纤维状滤器材料4浸入到含有合适浓度DNA的水溶液中,接着从DNA水溶液3中取出来的纤维状滤器材料4在60℃或更低温度下干燥,该温度不会导致DNA的热变性。
另外,对于用于除去有害物质的滤器,在充满DNA水溶液后,或者充满含有DNA的溶液以及DNA和另一种大分子化合物的混合物后,纤维状滤器材料暴露到紫外线下,因此DNA被固定到纤维状滤器材料上。
很多类型的有害物质能被包含在本发明中。然而,既然本发明采用了在DNA中的插入,本发明对处理具有平面结构的有机化合物特别有利,例如具有芳环的那些物质。特别的,本发明对处理二噁类化合物非常有利,例如二苯并对二氧芑(DD),氧芴(DF),联苯(BP),多氯化二苯并对二噁类化合物(PCDDs),多氯化氧芴类(PCDFS),和共平面PCB。
本发明的用于除去有害物质的滤器能适宜地被并入或者配置于多种产品,包括:吸烟器具,例如香烟和香烟过滤器;面具;气体面具;焚化炉中用于收集或除去灰尘的滤器设备;烟囱;汽车消声器;和负离子发生器。另外,它优选地用于安装在使用温度或温度和湿度可控制气体的电动或电子设备的流动通道的气体过滤器中。使用温度或温度和湿度可控制的气体的电动或电子设备的例子包括空调,鼓风加热器和湿度调节器。在这样的电动或电子设备中气体过滤器安装在控制进入设备的气体的温度或温度和湿度的位置和排出气体的位置之间。例如,对于空调,气体过滤器安置于温度在空调内部设定于预定值的气体排放处。气体过滤器包括滤器,因此包含在经过气体过滤器的气体中的灰尘能被除去,从而,净化气体从设备中排放到外面。本发明中能除去有害物质的滤器也被作为空气过滤器安装在电动或电子设备的空气或加热的空气流入、流出或流过的空气出口、入口或流动通道。冷却器,空气冷却器,和类似的电动或电子设备包括空气清洁器,电子真空清洁器,干燥器,和用于汽车发动机的冷却器。安装在这些设备中的空气过滤器包括滤器。吸入设备中的空气或加热空气在经过空气过滤器时被过滤除去灰尘和外来颗粒。例如,在电子真空清洁器中,空气过滤器安装在空气和灰尘和外来颗粒被吸入的位置和空气被排出的位置之间。因此,包含在空气中的灰尘和外来颗粒在通过空气过滤器时被除去。
正如上面所描述的,本发明的用于除去有害物质的滤器包括由纤维状滤器材料支承的DNA。支承的DNA的数量大致根据存在的有害物质的浓度决定。例如,环境空气中的二噁类化合物存在的级别是10-12g/m3。因此,为了有效除去二噁类化合物,由纤维状滤器材料支承的DNA的数量级至少在10-9克/m3,其中所述体积是含有有害物质的空气所渗透的区域的体积。
本发明用于除去有害物质的滤器,除了含有DNA和纤维状滤器材料之外,如果需要,还可以含有多种添加剂,例如着色剂和除臭剂。

实施例1
图9(a)和9(b)显示了使用本发明用于除去有害物质的滤器的香烟。图9(a)是香烟5的透视图,图9(b)是(a)的沿线A-A的断面图。香烟5包括一个含有化学或天然纤维层7的嘴端6,以及邻接嘴端6的用于除去有害物质的滤器9。在用于除去有害物质的滤器9中,DNA8以游离的状态由化学或天然纤维层7支承。由于用于除去有害物质滤器9的存在,抽吸烟草产生的有害物质,例如萘,有效地被DNA 8捕获。因此,在被人体吸收之前减少了有害物质。这对保持健康非常有利。用于除去有害物质的滤器9不会太多减少烟草的独特味道,因此,香烟的味觉质量不会受到不利影响。

实施例5
图13是一种气体面具24的透视图,它是使用本发明能除去有害物质的滤器的气体面具中的一种。气体面具24包括面具脸部25,头带26,滤罩框架27,和在滤罩框架27中的能除去有害物质的滤器28。除去有害物质的滤器28包括通过紫外线暴露而被HEPA(高效颗粒空气(high-efficiency particulate air))滤器支承的DNA。通过HEPA滤器(它能高效地捕获颗粒)支承DNA,除去有害物质的效果被进一步增强。例如,在有毒气体容易弥漫的有火的位置,带上气体面具24能有效地阻止有毒气体被人体吸收。
为了检查能除去有害物质的滤器多么有效地除去有害物质,进行了下面的测试。
测试实施例1
对具有本发明的能除去有害物质的滤器和具有已知滤器的香烟进行了实验来检查它们能除去吸烟产生的有害物质的效果。
(i)香烟的制备
制备1:已知香烟的纤维状滤器层浸入到DNA和聚丙烯酸钠的重量比10%的溶液中10秒钟,拿出来后,滤器层在40℃干燥。产生的能除去有害物质的滤器作为香烟的过滤器以产生Research Cigarette2R4F(商品名,Kentucky Unv.,U.S)香烟A。
制备2:已知香烟的纤维状滤器层浸入到DNA和聚丙烯酸钠的重量比10%的溶液中10秒钟,拿出来后,滤器层在40℃干燥。产生的能除去有害物质的滤器与活性炭结合作为香烟的过滤器以产生Research Cigarette 2R4F(商品名,Kentucky Unv.,U.S)香烟B。
制备3:仅仅使用已知香烟的纤维状滤器层作为香烟的滤器以产生Research Cigarette 2R4F(商品名,Kentucky Unv.,U.S)C。

测试实施例2
在这一测试实施例中,对本发明的能除去有害物质的滤器进行试验来检查除去环境空气中二噁类化合物的效果,特别是多氯化的二苯并对二氧芑(PCDDs),多氯化的氧芴(PCDF5),和共平面的PCB。
(i)空气过滤器的制备
在商业上可获得的空气过滤器的整个区域(长20cm宽30cm)上,均匀喷洒1000mg的DNA粉末。接着空气过滤器被暴露到波长为260nm的紫外线下从而使空气过滤器支承DNA。这样制备了本发明的能除去有害物质的滤器。
(ii)测量方法

1、一种除去有害物质的方法,其中有害物质与DNA接触而被DNA捕获,从而被除去。

8、根据权利要求6的用于除去有害物质的滤器,其中DNA通过席夫碱形成产生的化学键固定于纤维状滤器材料或包含在纤维状滤器材料中的玻璃珠。
9、根据权利要求6的用于除去有害物质的滤器,其中DNA以凝胶状态或以从凝胶中除去水分得到的多孔形式固定到纤维状滤器材料上。
10、根据权利要求9的用于除去有害物质的滤器,其中凝胶是丙烯酰胺凝胶。
11、根据权利要求6的用于除去有害物质的滤器,其中通过在DNA的双链间插入补骨脂素使DNA固定到纤维状滤器材料,所述补骨脂素通过间隔基结合于纤维状滤器材料。
12、根据权利要求6的用于除去有害物质的滤器,其中DNA通过偶联剂固定到纤维状滤器材料。
13、一种制备用于除去有害物质的滤器的方法, 包括以下步骤:用DNA的水溶液充满纤维状滤器材料;接着除去纤维状滤器材料中的水分,从而DNA以一种游离的状态支承于纤维状滤器材料的纤维中。
14、一种制备用于除去有害物质的滤器的方法,包括以下步骤:用一种含有DNA的水溶液以及DNA和另一种大分子化合物的混合物充满纤维状滤器材料;接着除去纤维状滤器材料中的水分,借此DNA被纤维状滤器材料支承。
15、一种制备用于除去有害物质的滤器的方法,包括以下步骤:用一种DNA的水溶液充满纤维状滤器材料;接着将纤维状滤器材料暴露于紫外线下,借此DNA被固定到纤维状滤器材料上。
16、一种制备用于除去有害物质的滤器的方法,包括以下步骤:用一种含有DNA的水溶液以及DNA和另一种大分子化合物的混合物充满纤维状滤器材料;接着将纤维状滤器材料暴露到紫外线下,借此DNA被固定到纤维状滤器材料上。
17、一种制备用于除去有害物质的滤器的方法,包括通过席夫碱形成产生的化学键将DNA固定到纤维状滤器材料或包含在纤维状滤器材料中的玻璃珠上的步骤。
18、一种制备用于除去有害物质的滤器的方法,包括以下步骤:将通过间隔基结合到纤维状滤器材料上的补骨脂素插入DNA的双链之间,借此固定DNA到纤维状滤器材料上。
19、一种制备用于除去有害物质的滤器的方法,包括以下步骤:用溶胶状态的DNA和水以及凝胶剂充满纤维状滤器材料,接着干燥或冷冻干燥纤维状滤器材料以除去水分,借此DNA以凝胶状态或者从凝胶中除去水分得到的多孔形式被固定到纤维状滤器材料上。
20、根据权利要求1的用于除去有害物质的方法,其中有害物质是二噁类化合物。
21、一种包括权利要求2到12中任意一项权利要求所述的用于除去有害物质的滤器的吸烟器具。
22、一种包括权利要求2至12中任意一项权利要求所述的用于除去有害物质的滤器的面具。
23、一种包括权利要求2至12中任意一项权利要求所述的用于除去有害物质的滤器的气体面具。
24、一种安装在使用温度或温度和湿度受控的气体的电动或电子设备的气体流动通道中的气体过滤器,该气体过滤器包括权利要求2至12中任一项所述的用于除去有害物质的滤器。
25、安装在空气或热空气借以流出或流入的电子或电动设备的空气出口、入口或流动通道的空气过滤器,该空气过滤器包括权利要求2至12中任一项所述的用于除去有害物质的滤器。
26、一种用于收集或除去焚化炉中的灰尘的滤器设备,该滤器设备包括权利要求2至12中任一项所述的用于除去有害物质的滤器。
