Abstract

本发明涉及一种肿瘤病尿样检测试剂,由分别存放的A、B、C、D四个试剂组成,试剂A是含硫酸汞0.5—15%的硫酸溶解液,试剂B为含亚硝酸0.001—1%的无机酸或水溶解液,试剂C为含对氨基苯磺酸0.01—0.5%的水或有机酸溶解液;试剂D是含萘胺衍生物0.05—10%无机酸或有机酸溶解液。将试剂A、B、C、D依次滴入尿样中进行反应,健康人尿样反应变成玫瑰红色,肿瘤病人因尿中含大量单羟酚,反应后尿样不变色,因而可用于对肿瘤病患疾的快速筛查。

Description

肿瘤病尿样检测试剂
本发明涉及一种肿瘤病尿样检测试剂,用于检测尿样中单羟酚的含量,以对肿瘤病患疾进行快速筛查。
癌症是严重危害人类健康的主要疾病之一,早期发现是治愈的关键,但至今尚未有简便快速且经济有效的检测手段,待发现症状时往往已是中晚期了,这不仅要花费昂贵的治疗费,而且通过治疗延长的存活期相当有限。目前国际上根据肿瘤有丝分裂时会释放端粒酶的机理已采用先进的端粒酶PCR检测技术,但PCR仪器昂贵,在我国推广应用尚有困难。
本发明的目的是提供一种可快速、简便地筛查肿瘤患疾的尿样检测试剂,它可对端粒酶的代谢物单羟酚在尿液中的含量进行快速检测,使肿瘤病得以早期发现和早期治疗。
本发明试剂由分别存放在各自容器内的A、B、C、D四个试剂组成,试剂A是硫酸汞的硫酸溶解液,其中硫酸汞含量为0.5-15%,硫酸为0.15-15%,稳定剂2-3%,其余为水;试剂B为亚硝酸的无机酸或水溶解液,其中含亚硝酸0.001-1%,其余为无机酸或水;试剂C为对氨基苯磺酸的水或有机酸溶解液,其中对氨基苯磺酸的含量为0.01-0.5%,稳定剂1%,其余为水或有机酸;试剂D是萘胺衍生物的无机酸或有机酸溶解液,其中萘胺衍生物的含量为0.05-10%,稳定剂1%,其余为无机酸或有机酸。以上均为重量百分数。
上述试剂C的对氨基苯磺酸可与试剂D的萘胺衍生物反应产生成红色生成物。由于肿瘤病人尿液中有较大量的单羟酚,滴入试剂A后,单羟酚在酸性环境下与汞盐反应生成金属络合物,再滴入试剂B和C,金属络合物便在亚硝酸盐存在下,与对氨基苯磺酸发生偶氮反应,使试剂C中的对氨基苯磺酸在反应  中被消耗或大部分被消耗,再滴入试剂D时,无反应发生,因此肿瘤病人尿样用本试剂检测后不变色或基本不变色。而健康人的尿液中单羟酚含量极微,滴入试剂A后无金属络合物生成,因而滴入试剂B和C后也无偶氮反应产生,致使试剂C中的对氨基苯磺酸无消耗,因此再滴入试剂D时,对氨基苯磺酸就会与萘胺衍生物发生反应使尿样变成玫瑰红色。
综上所述,采用本发明试剂检测尿样时,只需观察尿样颜色的的变化即可初步判别是否患有肿瘤病,即尿样变成鲜艳的玫瑰红色的是阴性,尿样不变色的有是阳性,二者之间色差明显,容易识别。因此,本发明试剂可快捷简便地对人群进行肿瘤病筛查,对于尿检呈阳性者可通过进一步检查确珍,有利于肿瘤病的早期发现和早期治疗。
下面说明本发明的实施例。
按下列配比配制试剂A、B、C、D并分别装入四个试剂盒内:
试剂A:将0.5克的硫酸汞溶解在3mol/l的浓硫酸100ml中,加入3号稳定剂3克;
试剂B:将0.001克的亚硝酸钠溶解在蒸馏水或无机酸100ml中。
试剂C:将领克的对氨基苯磺酸溶解在蒸馏水或有机酸100ml中,并加入2号稳定剂1克;
试剂D:将0.5克萘胺衍生物溶解在无机酸或有机酸100ml部中,并加入3号稳定剂。
以上试剂检测尿样的过程如下:
取尿样1ml,滴入试剂A 60微升,紧接着滴入试剂B 60微升,试剂C 60微升,摇匀后静置3-5分钟,待充分反应后滴入试剂D 60微升摇匀后静置备分钟,若尿样呈现鲜艳的玫瑰红色为阴性,表示被检者无肿瘤病;反之,若尿样变成鲜艳的玫瑰红色为阳性,可初步确定被检者有肿瘤患疾,应对病人进一步检查确珍。

Claims (1)

  1. 肿瘤病尿样检测试剂,其特征是由分别存放在各自容器内的A、B、C、D四个试剂组成,试剂A是硫酸汞的硫酸溶解液,其中硫酸汞含量为0.5-15%,硫酸为0.15-15%稳定剂2-3%,其余为水;试剂B为亚硝酸的无机酸或水溶解液,其中含亚硝酸0.001-1%其余为无机酸或水;试剂C为对氨基苯磺酸的水或有机酸溶解液,其中对氨基苯磺酸的含量为0.01-0.5%,稳定剂1%,其余为水或有机酸;试剂D是萘胺衍生物的无机酸或有机酸溶解液,其中萘胺衍生物的含量为0.05-10%,稳定剂1%其余为无机酸或有机酸,以上均为重量百分数。
