Abstract
本发明属于口腔保健用品领域,具体涉及含辅酶Q10的牙膏及其制备方法。所述含辅酶Q10的牙膏包含透明质酸‑还原型辅酶Q10接枝复合物、芳香酯酶LGG_02908和弱碱性牙膏基料。本发明人已令人惊讶地发现,将还原型辅酶Q10接枝到透明质酸上,可以在牙膏的加工和保存期间,很好地防止还原型辅酶Q10被氧化成氧化型辅酶Q10。通过搭配在常温和弱碱性pH下具有较低活性而在弱酸性和口腔温度下具有较高活性的芳香酯酶LGG_02908,可以在使用本发明的牙膏进行口腔护理时,将还原型辅酶Q10从透明质酸上释放出来,从而在口腔中发挥其功效。

技术领域
本发明属于口腔保健用品领域,具体涉及含辅酶Q10的牙膏及其制备方法。
背景技术
还原型辅酶Q10是人体新陈代谢不可缺少的物质,存在于人体细胞“线粒体”中,参与能量ATP合成,具有抗氧化及细胞膜修复作用,随着人体年龄的增长,20岁以后人体辅酶Q10合成能力下降,浓度低于一定量时,会引起代谢异常,诱发牙周炎、牙龈炎、牙龈萎缩等牙疾。
牙膏是人们日常用品之一,用它可以清洁口腔牙齿和防牙菌斑作用。目前市场上的牙膏种类繁多,各具特色,以其气味和功效独特为广大消费者所喜爱与接受。随着人们对医疗保健的认识逐渐提高,现有牙膏已远远不能满足消费者的不同需求,特别是针对口腔炎症导致牙龈出血的大多数人,刷牙时出血量之多使人体抵抗力下降,易滋生其他疾病,功能性医疗牙膏对提高人类健康具有不可替代作用。
现已开发出多种含辅酶Q10的牙膏,以期通过日常口腔护理来发挥辅酶Q10的功效。然而,还原型辅酶Q10容易被分子氧氧化成氧化型辅酶Q10,在将还原型辅酶Q10加工成牙膏时,或者在加工后的保存过程中,还原型辅酶Q10的稳定化仍然是重要的课题。在上述的加工或保存期间,很难将氧完全除去或遮蔽,导致残存或者混入的氧会产生较大的恶劣影响。
发明内容
为了解决上述问题,本发明提供了一种含辅酶Q10的牙膏,其包含透明质酸-还原型辅酶Q10接枝复合物、芳香酯酶LGG_02908和弱碱性牙膏基料。
进一步地,所述透明质酸-还原型辅酶Q10接枝复合物如下制备:
(1)将透明质酸溶解在溶于反应溶剂中,加入1-乙基-(3-二甲基氨基丙基)碳二亚胺和1-羟基苯并三唑,在氮气保护下,将接头上的羧基活化;
(2)将还原型辅酶Q10溶于适当的溶剂,缓慢滴加到经活化的透明质酸溶液中,进行羟基与羧基的酯化反应,待反应结束,透析除杂,冷冻干燥,即得所述透明质酸-还原型辅酶Q10接枝复合物。
进一步地,所述透明质酸-还原型辅酶Q10接枝复合物的结构如下式所示:
进一步地,所述反应溶剂选自甲酰胺或二氯甲烷。
进一步地,所述适当的溶剂选自二氯甲烷、四氢呋喃、二甲亚砜或二氯甲烷。
进一步地,所述透明质酸选自高分子量透明质酸、中分子量透明质酸或低分子量透明质酸。
进一步地,所述弱碱性牙膏基料的pH为8-9。
进一步地,所述弱碱性牙膏基料包括摩擦剂、润湿剂、发泡剂、粘合剂、防腐剂、甜味剂、去离子水。
进一步地,所述摩擦剂为碳酸钙、磷酸氢钙、氢氧化铝、三水合氧化铝中的一种或者两种以上的混合物。
进一步地,所述润湿剂为甘油、山梨醇、丙二醇、聚乙二醇中的一种或两种以上的混合物。
进一步地,所述发泡剂为月桂醇硫酸钠、月桂酰肌氨酸钠、月桂酰谷氨酸钠、椰油酰胺丙基甜菜碱中的一种或多种混合物。
进一步地,所述粘合剂为羧甲基纤维素钠、羟乙基纤维素、卡拉胶、瓜尔胶、黄原胶中的一种或两种以上的混合物。
进一步地,所述甜味剂为糖精钠、安赛蜜、纽甜中的一种或两种以上的混合物。
进一步地,所述牙膏还包含中草药提取物。
进一步地,所述中草药提取物可以选自以下的一种或几种:金银花水提取物、白芍水提取物、茯苓醇提取物、五味子醇提取物。
如本文所用,金银花水提取物、白芍水提取物、茯苓醇提取物和五味子醇提取物为本领域的通用原材料,均可通过自制或购买获得。
金银花水提取物的一种自制方法可以为:把金银花粉碎后,加去离子水,金银花与去离子水的比例为1:5-1:20(g:ml),浸泡6-24小时,加热煮沸1-3小时,分离出药液,向药渣中加入去离子水,金银花与去离子水的比例为1:5-1:20(g:ml),加热煮沸1-3小时,将两次煮沸后的药液合并,过滤,离心后,弃去沉淀将上清液浓缩,烘干或冻干,制得金银花水提取物。
白芍水提取物的一种自制方法可以为:把白芍粉碎后,加去离子水,白芍与去离子水的比例为1:5-1:20(g:ml),浸泡6-24小时,加热煮沸1-3小时,分离出药液,向药渣中加入去离子水,白芍与去离子水的比例为1:5-1:20(g:ml),加热煮沸1-3小时,将两次煮沸后的药液合并,过滤,离心后,弃去沉淀将上清液浓缩,烘干或冻干,制得白芍水提取物。
茯苓醇提取物的一种自制方法可以为:把茯苓粉碎后,加无水乙醇,茯苓与无水乙醇的比例为1:5-1:20(g:ml),浸泡6-24小时,水浴回流1-3小时,过滤,离心后,弃去沉淀,将上清液浓缩,烘干或冻干,制得茯苓醇提取物。
五味子醇提取物的一种自制方法可以为:把五味子粉碎后,加无水乙醇,五味子与无水乙醇的比例为1:5-1:20(g:ml),浸泡6-24小时,水浴回流1-3小时,过滤,离心后,弃去沉淀,将上清液浓缩,烘干或冻干,制得五味子醇提取物。
进一步地,所述牙膏按重量份数包含5-10份透明质酸-还原型辅酶Q10接枝复合物、0.1-0.3份芳香酯酶LGG_02908、25-30份摩擦剂、5-10份润湿剂、1-2份发泡剂、1.5-2.5份粘合剂、0.1-0.3份甜味剂、1-2份中草药提取物、20-30份去离子水。
本发明还提供了如本文所述的含辅酶Q10的牙膏的制备方法,其包括将透明质酸-还原型辅酶Q10接枝复合物、芳香酯酶LGG_02908和弱碱性牙膏基料混合、搅拌、研磨并真空脱气,获得牙膏膏体。
进一步地,所述制备方法包括将透明质酸-还原型辅酶Q10接枝复合物与中草药提取物混合,加入润湿剂和去离子水搅拌、制成去离子水混悬液,再与摩擦剂、粘合剂、发泡剂、甜味剂混合、搅拌、研磨并真空脱气,最后加入芳香酯酶LGG_02908搅拌、研磨并真空脱气,获得牙膏膏体。
本发明的有益效果
本发明人已令人惊讶地发现,将还原型辅酶Q10接枝到透明质酸上,可以在牙膏的加工和保存期间,很好地防止还原型辅酶Q10被氧化成氧化型辅酶Q10。通过搭配在常温和弱碱性pH下具有较低活性而在弱酸性和口腔温度下具有较高活性的芳香酯酶LGG_02908(关于芳香酯酶LGG_02908的制备以及性能描述,参考Bin-Chun Li等人,3Biotech(2021)11:496,其通过引用整体并入本文),可以在使用本发明的牙膏进行口腔护理时,将还原型辅酶Q10从透明质酸上释放出来,从而在口腔中发挥其功效。
具体实施方式
以下结合具体实施例来进一步说明本发明,但实施例并不对本发明做任何形式的限定。除非特别说明,本发明采用的试剂、方法和设备为本技术领域常规试剂、方法和设备。
实施例1:透明质酸-还原型辅酶Q10接枝复合物的制备
称取5g透明质酸(分子量500K道尔顿)溶于200ml二氯甲烷中,通氮气保护,室温水浴磁力搅拌,缓慢加入3mmol 1-乙基-(3-二甲基氨基丙基)碳二亚胺(EDC)和3mmol 1-羟基苯并三唑,活化30min,将溶于二氯甲烷的3mmol还原型辅酶Q10在搅拌下滴加到透明质酸溶液中,室温反应24h,在蒸馏水中透析3天(透析袋截留分子量3000),冷冻干燥得到透明质酸-还原型辅酶Q10接枝复合物。核磁共振氢谱1H-NMR表征(1H NMR:δ5.53(1H),5.37(5H),5.15(1H),5.00(1H),4.49(1H),4.22(1H),4.04(1H),3.95(1H),3.84(6H),3.80(2H),3.50(1H),3.41(3H),3.39(3H),3.36(1H),3.22(2H),2.13(3H),2.02(3H),1.96(2H),1.92(1H),1.91(6H),1.86(1H),1.66(2H),0.85(3H))证实获得了如下式所示的透明质酸-还原型辅酶Q10接枝复合物(接枝率62%):
实施例2
本实施例提供了一种含辅酶Q10的牙膏,其按重量份数包含5份透明质酸-还原型辅酶Q10接枝复合物、0.2份芳香酯酶LGG_02908、30份碳酸钙、8份丙二醇、2份月桂酰肌氨酸钠、2份黄原胶、0.2份糖精钠、2份中草药提取物(其由下述组分按重量份组成:白芍水提取物20份、茯苓醇提取物10份、五味子醇提取物10份)和25份去离子水。
所述含辅酶Q10的牙膏的制备过程包括:将透明质酸-还原型辅酶Q10接枝复合物与中草药提取物混合,加入丙二醇和去离子水搅拌、制成去离子水混悬液,再与碳酸钙、黄原胶、月桂酰肌氨酸钠、糖精钠混合、搅拌、研磨并真空脱气,最后加入芳香酯酶LGG_02908搅拌、研磨并真空脱气,获得牙膏膏体,通过无菌罐装得到牙膏成品。
实施例3
本实施例提供了一种含辅酶Q10的牙膏,其按重量份数包含10份透明质酸-还原型辅酶Q10接枝复合物、0.2份芳香酯酶LGG_02908、25份氢氧化铝、6份聚乙二醇、1份椰油酰胺丙基甜菜碱、2份瓜尔胶、0.2份安赛蜜、2份中草药提取物(其由下述组分按重量份组成:金银花水提取物20份、茯苓醇提取物10份、五味子醇提取物10份)和30份去离子水。
所述含辅酶Q10的牙膏的制备过程包括:将透明质酸-还原型辅酶Q10接枝复合物与中草药提取物混合,加入聚乙二醇和去离子水搅拌、制成去离子水混悬液,再与氢氧化铝、瓜尔胶、椰油酰胺丙基甜菜碱、安赛蜜混合、搅拌、研磨并真空脱气,最后加入芳香酯酶LGG_02908搅拌、研磨并真空脱气,获得牙膏膏体,通过无菌罐装得到牙膏产品。
对比例1
本对比例提供了一种含辅酶Q10的牙膏,其与实施例2的区别在于其使用透明质酸和还原型辅酶Q10的1:1质量比的混合物替代透明质酸-还原型辅酶Q10接枝复合物,其余成分、其含量以及制备方法与实施例2相同。
对比例2
本对比例提供了一种含辅酶Q10的牙膏,其与实施例2的区别在于不含芳香酯酶LGG_02908,其余成分、其含量以及制备方法与实施例2相同。
性能测试:
将实施例2-3和对比例1-2制备的20g牙膏膏体保存在25℃暴露于空气中的条件下。经过24小时后,将牙膏膏体分散在二氯甲烷中,搅拌30min,测pH后取一半溶液过滤收集上清液,再经超滤膜过滤取滤液备用,将剩余的一半溶液用盐酸调节pH至6.8,继续搅拌5min,过滤弃去沉淀物,再经超滤膜过滤取滤液,通过HPLC测定滤液中还原型辅酶Q10和氧化型辅酶Q10的重量比,其结果显示在表1中。

色谱柱:SYMMETRY C18(Waters制造)250mm(长)4.6mm(内径);
流动相:C2H5OH:CH3OH=4:3(v:v);
检测波长:210nm;
流速:1ml/min;
还原型辅酶Q10的保留时间:9.1min,氧化型辅酶Q10的保留时间:13.3min。

从表1的结果可知,本发明实施例2和3制备的牙膏膏体在空气中暴露24h后仍检测不到氧化型辅酶Q10的产生,说明透明质酸-还原型辅酶Q10接枝复合物可以保护还原型辅酶Q10避免被空气氧化;而在调节pH之前和之后(分别模拟牙膏的保存和使用场景),可以明显看到还原型辅酶Q10从透明质酸上的释放,表明本发明采用的芳香酯酶LGG_02908可以用来调控透明质酸-还原型辅酶Q10接枝复合物在牙膏保存与使用过程中的还原型辅酶Q10的释放。
而相比之下,未进行接枝而是直接混合时(对比例1),在暴露于空气24h后,可以检测到显著量的氧化型辅酶Q10的产生。而未添加芳香酯酶LGG_02908时(对比例2),在调节pH至6.8之后,仍检测不到大量还原型辅酶Q10的释放。

1.一种含辅酶Q10的牙膏,其特征在于,包含透明质酸-还原型辅酶Q10接枝复合物、芳香酯酶LGG_02908和弱碱性牙膏基料;
所述透明质酸-还原型辅酶Q10接枝复合物如下制备:
(1)将透明质酸溶解在溶于反应溶剂中,加入1-乙基-(3-二甲基氨基丙基)碳二亚胺和1-羟基苯并三唑,在氮气保护下,将接头上的羧基活化;
(2)将还原型辅酶Q10溶于适当的溶剂,缓慢滴加到经活化的透明质酸溶液中,进行羟基与羧基的酯化反应,待反应结束,透析除杂,冷冻干燥,即得所述透明质酸-还原型辅酶Q10接枝复合物;
所述反应溶剂为二氯甲烷,
所述适当的溶剂为二氯甲烷;
所述透明质酸的分子量500K道尔顿;
所述弱碱性牙膏基料的pH为8-9。
2.根据权利要求1所述的牙膏,其特征在于,所述弱碱性牙膏基料包括摩擦剂、润湿剂、发泡剂、粘合剂、防腐剂、甜味剂和去离子水。
3.根据权利要求2所述的牙膏,其特征在于,所述摩擦剂为碳酸钙、磷酸氢钙、氢氧化铝、三水合氧化铝中的一种或者两种以上的混合物;
所述润湿剂为甘油、山梨醇、丙二醇、聚乙二醇中的一种或两种以上的混合物;
所述发泡剂为月桂醇硫酸钠、月桂酰肌氨酸钠、月桂酰谷氨酸钠、椰油酰胺丙基甜菜碱中的一种或多种混合物;
所述粘合剂为羧甲基纤维素钠、羟乙基纤维素、卡拉胶、瓜尔胶、黄原胶中的一种或两种以上的混合物;
所述防腐剂为苯甲酸钠、尼泊金甲酯、尼泊金丙酯、脱氢乙酸钠、山梨酸钾中的一种或两种以上的混合物;
所述甜味剂为糖精钠、安赛蜜、纽甜中的一种或两种以上混合物。
4.根据权利要求3所述的牙膏,其特征在于,所述牙膏还包含中草药提取物。
5.根据权利要求4所述的牙膏,其特征在于,所述牙膏按重量份数包含5-10份透明质酸-还原型辅酶Q10接枝复合物、0.1-0.3份芳香酯酶LGG_02908、25-30份摩擦剂、5-10份润湿剂、1-2份发泡剂、1.5-2.5份粘合剂、0.1-0.3份甜味剂、1-2份中草药提取物、20-30份去离子水。
