Example 1
The example provides a natto composition for regulating intestines and stomach and reducing blood fat, which comprises the following components in parts by weight (10 g for every 1 part by weight): 25 parts of natto, 8 parts of rhizoma polygonati extract and 5 parts of radix puerariae extract.
As shown in fig. 1, the preparation method of the natto composition for conditioning intestines and stomach and reducing blood fat comprises the following steps:
step (1), obtaining natto: soaking semen glycines in water for 24 hr; then pressurizing the soaked soybeans (under 2-10 atmospheric pressure) and steaming for 2 hours, cooling the steamed soybeans to room temperature, and uniformly spraying bacillus natto liquid; controlling the temperature of natto at 40 ℃ and the relative humidity at 80-90%, and fermenting for 14h; refrigerating fermented natto at 4 ℃, taking out, freeze-drying, crushing and sieving with a 50-60-mesh sieve to obtain the fermented natto;
step (2), obtaining a rhizoma polygonati extract: reflux-extracting pulverized rhizoma Polygonati in 80% ethanol 10 times of the total weight at 85deg.C for 2 hr, collecting extractive solution, and concentrating under reduced pressure to obtain extract; loading the extract on a polyamide column, eluting with ethanol with the volume fraction of 18% and 4 times of the column volume; eluting with 42% ethanol with 10 times of column volume, collecting 42% ethanol eluate, concentrating under reduced pressure, and vacuum drying to constant weight to obtain rhizoma Polygonati extract;
step (3), obtaining a kudzuvine root extract: adding the crushed kudzuvine root with a sieve of 30-50 meshes into water at the temperature of 70 ℃ and under the pressure of 53Kpa for first reduced pressure distillation extraction, and stopping reduced pressure distillation when the volume of distillate is half of the volume of the initial water adding amount; then adding half of the volume of the initial water adding amount, carrying out second reduced pressure distillation extraction under the same parameter condition, and stopping reduced pressure distillation when the volume of the distillate is half of the volume of the initial water adding amount; collecting and mixing the first vacuum distillation extractive solution and the second vacuum distillation extractive solution, concentrating under reduced pressure, and vacuum drying to constant weight to obtain radix Puerariae extract;
step (4): mixing the natto obtained in the step (1), the rhizoma polygonati extract obtained in the step (2) and the radix puerariae extract obtained in the step (3), grinding, and sieving with a 50-60-mesh sieve to obtain the natto composition for regulating intestines and stomach and reducing blood fat.

This example provides a natto composition for regulating intestines and stomach and reducing blood lipid, which is different from example 1 only in that: the natto composition for conditioning intestines and stomach and reducing blood fat comprises the following components in parts by weight (10 g for every 1 part by weight): 45 parts of natto, 15 parts of rhizoma polygonati extract and 10 parts of radix puerariae extract; the rest steps and parameters are the same.

This example provides a natto composition for regulating intestines and stomach and reducing blood lipid, which is different from example 1 only in that: the natto composition for conditioning intestines and stomach and reducing blood fat comprises the following components in parts by weight (10 g for every 1 part by weight): 37 parts of natto, 13 parts of rhizoma polygonati extract and 6 parts of radix puerariae extract; the rest steps and parameters are the same.
Example 4
The example provides a natto composition for regulating intestines and stomach and reducing blood fat, which comprises the following components in parts by weight (10 g for every 1 part by weight): 37 parts of natto, 13 parts of rhizoma polygonati extract, 6 parts of radix puerariae extract and 2 parts of liquorice extract.
The preparation method of the natto composition for regulating intestines and stomach and reducing blood fat comprises the following steps:

step (3), obtaining a kudzuvine root extract: adding crushed radix Puerariae into water at 70deg.C and 53Kpa pressure, performing first reduced pressure distillation extraction, and stopping reduced pressure distillation when the volume of distillate is half of the volume of the initial water addition amount; then adding half of the volume of the initial water adding amount, carrying out second reduced pressure distillation extraction under the same parameter condition, and stopping reduced pressure distillation when the volume of the distillate is half of the volume of the initial water adding amount; collecting and mixing the first vacuum distillation extractive solution and the second vacuum distillation extractive solution, concentrating under reduced pressure, and vacuum drying to constant weight to obtain radix Puerariae extract;
step (4), obtaining licorice extract: reflux extracting pulverized Glycyrrhrizae radix with 10 times of water at 100deg.C for 2 hr, collecting extractive solution, concentrating under reduced pressure, and vacuum drying to constant weight to obtain Glycyrrhrizae radix water extract;
step (5): mixing the natto obtained in the step (1), the rhizoma polygonati extract obtained in the step (2) and the radix puerariae extract obtained in the step (3) with the licorice extract obtained in the step (4), grinding, and sieving with a 50-60-mesh sieve to obtain the natto composition for regulating intestines and stomach and reducing blood fat.

This example provides a natto composition for regulating intestines and stomach and reducing blood lipid, which is different from example 1 only in that: regulating natto into red yeast rice; the rest steps and parameters are the same.

This example provides a natto composition for regulating intestines and stomach and reducing blood lipid, which is different from example 1 only in that: the Polygonatum sibiricum extract is obtained in the preparation method step (2): reflux-extracting pulverized rhizoma Polygonati skin with 10 times of 80% ethanol at 85deg.C for 2 hr, collecting extractive solution, and concentrating under reduced pressure to obtain extract; loading the extract on a polyamide column, eluting with ethanol with the volume fraction of 18% and 4 times of the column volume; eluting with 65% ethanol with a volume fraction of 10 times of column volume, collecting 65% ethanol eluate, concentrating under reduced pressure, and vacuum drying to constant weight to obtain rhizoma Polygonati extract; the rest steps and parameters are the same.
Comparative example 5
This example provides a natto composition for regulating intestines and stomach and reducing blood lipid, which is different from example 1 only in that: in the preparation method, in the kudzu root extract obtained in the step (3): pulverizing radix Puerariae, sieving with 10 mesh sieve, adding 8 times of 60% ethanol, reflux extracting under heating for 4 times each for 1 hr, filtering with 100 mesh filter cloth, and mixing filtrates; placing the filtrate in HPD-100 macroporous adsorbent resin column chromatography, sequentially eluting with 3 times of column water and 5 times of column volume of 85% ethanol, collecting 85% ethanol eluate, recovering ethanol, concentrating under reduced pressure at 50deg.C and 0.08MPa until no alcohol is present, steaming in water bath at 80deg.C to obtain fluid extract, and drying in vacuum drying oven at 70deg.C and 0.08MPa to obtain radix Puerariae extract; the rest steps and parameters are the same.
Test example 1
The present example examined the hypolipidemic efficacy of the natto compositions for conditioning the intestines and stomach and reducing blood lipid provided in examples 1 to 4 and comparative examples 1 to 5.
The testing method comprises the following steps: the method comprises the steps of equally dividing 110 cleaning-grade Wistar rats with male and female halves into 11 groups of 10 rats each; the test groups are normal groups, high-fat control groups and experimental groups 1 to 9 respectively. The normal group was fed with only ordinary basal feed every day, the high fat control group was fed with only high fat feed every day (high fat feed: 10.0% sucrose, egg yolk powder 10%, 15% lard, 1.2% cholesterol, 0.2% sodium cholate and 63.6% ordinary feed), the experimental groups 1 to 9 were fed with high fat feed each day with the natto composition for regulating gastrointestinal hypolipidemia provided in examples 1 to 4 and comparative examples 1 to 5, respectively, and SD rats of each group were free to drink water. SD rats were sacrificed after 30 days of continuous feeding, and the total serum cholesterol TC and triglyceride TG content of each group of SD rats were determined after intraperitoneal venous blood sampling. Wherein, the natto composition in experiment group 1 to 9 is added in an amount of 5mg/kg to the daily fed high fat feed, experiment group 1 corresponds to the natto composition for regulating gastrointestinal blood lipid obtained in example 1, experiment group 2 corresponds to the natto composition for regulating gastrointestinal blood lipid obtained in example 2, experiment group 3 corresponds to the natto composition for regulating gastrointestinal blood lipid obtained in example 3, experiment group 4 corresponds to the natto composition for regulating gastrointestinal blood lipid obtained in example 4, experiment group 5 corresponds to the natto composition for regulating gastrointestinal blood lipid obtained in comparative example 1, experiment group 6 corresponds to the natto composition for regulating gastrointestinal blood lipid obtained in comparative example 2, experiment group 7 corresponds to the natto composition for regulating gastrointestinal blood lipid obtained in comparative example 3, experiment group 8 corresponds to the natto composition for regulating gastrointestinal blood lipid obtained in comparative example 4, and experiment group 9 corresponds to the natto composition for regulating gastrointestinal blood lipid obtained in comparative example 5.
The test results are shown in Table 1.

As can be seen from Table 1, the natto compositions for regulating intestines and stomach and reducing blood lipid provided by examples 1 to 4 of the present application are superior to the natto compositions for regulating intestines and stomach and reducing blood lipid provided by comparative examples 1 to 5 in terms of blood lipid reduction. Therefore, the natto composition provided by the embodiment of the application has excellent blood lipid reducing effect, can provide more drug choices for patients with hyperlipidemia, and widens the application range of rhizoma polygonati, radix puerariae and the like.
Test example 2
50 volunteers suffering from dyspepsia and constipation are selected, the natto composition for regulating intestines and stomach and reducing blood fat provided in the embodiment 4 is taken, a proper amount of pharmaceutical auxiliary materials are added to prepare tablets, the daily dosage is 5g, after the oral administration is carried out for 2 weeks, the symptoms of dyspepsia and constipation of 40 volunteers disappear, the symptoms of dyspepsia and constipation of 5 volunteers are improved, the symptoms of dyspepsia and constipation of 5 volunteers are not obviously improved, the total effective rate is 90%, and the tablet has excellent regulating effect on intestines and stomach.

25-45 parts of natto, 8-15 parts of rhizoma polygonati extract and 5-10 parts of radix puerariae extract.
3. The gastrointestinal hypolipidemic natto composition of claim 2, further comprising, in parts by weight: 1-3 parts of licorice extract.
4. The gastrointestinal hypolipidemic natto composition of claim 2, wherein the gastrointestinal hypolipidemic natto composition comprises the following components in parts by weight:
37 parts of natto, 13 parts of rhizoma polygonati extract and 6 parts of radix puerariae extract.
5. The natto composition for regulating gastrointestinal blood lipid lowering according to any one of claims 1 to 4, wherein the preparation method of the rhizoma polygonati extract comprises the following steps:
adding the crushed rhizoma polygonati skin into ethanol with the volume fraction of 75-85% and the weight of 8-15 times of the crushed rhizoma polygonati skin at the temperature of 80-90 ℃ for reflux extraction for 1-3 hours, collecting an extracting solution, and concentrating under reduced pressure to obtain an extract;
loading the extract on a polyamide column, eluting with ethanol with the volume fraction of 15-20% which is 3-5 times of the column volume; eluting with ethanol with the volume fraction of 40-45% and the volume fraction of 6-10 times of the column volume, collecting ethanol eluate with the volume fraction of 40-45%, concentrating and drying to obtain the rhizoma polygonati extract.
6. The natto composition for regulating gastrointestinal blood lipid lowering according to any one of claims 1 to 4, wherein the preparation method of the kudzuvine root extract comprises the following steps:
adding crushed radix Puerariae into water, performing reduced pressure distillation, stopping reduced pressure distillation when the volume of distillate is half of the volume of the initial water adding amount, collecting reduced pressure distillation extractive solution, concentrating and drying to obtain radix Puerariae extract;
wherein, the working parameters of the reduced pressure distillation extraction comprise: the temperature is 68-72 ℃, the pressure is 50-55 Kpa, and the extraction times are 1-3 times.
7. The gastrointestinal hypolipidemic natto composition of any one of claims 1 to 4, wherein the licorice extract is an aqueous licorice extract.
