Abstract
The invention provides an essential oil composition with pain relieving effect and application thereof, wherein the essential oil composition comprises the following components in parts by weight: 1.5 to 5 parts of lavender essential oil, 1 to 5 parts of yam essential oil, 0.5 to 3 parts of wood essential oil, 1 to 5 parts of radix angelicae pubescentis essential oil, 1 to 5 parts of notopterygium root essential oil, 1 to 5 parts of fructus cnidii essential oil, 1 to 2 parts of ailanthus altissima root essential oil and 20 to 50 parts of base oil. The invention reasonably selects the components of lavender essential oil, mount-Chinese yam essential oil, wood essential oil, pubescent angelica essential oil, notopterygium essential oil, cnidium essential oil, chinese toon root essential oil, base oil and the like to cooperate with each other, and the components are carefully modulated according to the theory of traditional Chinese medicine, so that the invention has good effect of relieving bone and muscle pain, also has certain effect on migraine and nervous headache, is safe, has no toxic or side effect, and is suitable for any adult population to use.

Preferably, the essential oil composition further comprises: 0.1 to 1 part of borneol essential oil and/or 0.1 to 1 part of peppermint essential oil.

The invention reasonably selects the components of lavender essential oil, mount-Chinese yam essential oil, wood essential oil, pubescent angelica essential oil, notopterygium essential oil, cnidium essential oil, chinese toon root essential oil, base oil and the like to cooperate with each other, and the components are carefully modulated according to the theory of traditional Chinese medicine, so that the invention has good effect of relieving bone and muscle pain, also has certain effect on migraine and nervous headache, is safe, has no toxic or side effect, and is suitable for any adult population to use.

The essential oil is volatile aromatic substance extracted from flowers, leaves, stems, roots or fruits of plants by steam distillation, supercritical extraction, extrusion, cold soaking or solvent extraction, and most of the essential oil has single direct use, limited efficacy and is easy to cause irritation, allergy and other phenomena. The invention is prepared by mutually and cooperatively combining the components of lavender essential oil, mount-Chinese yam essential oil, wood essential oil, radix angelicae pubescentis essential oil, notopterygium essential oil, fructus cnidii essential oil, chinese toon root essential oil, basic oil and the like according to the theory of traditional Chinese medicine, has good effect of relieving bone and muscle pain, also has certain effect on migraine and nerve headache, and is safe and free from toxic and side effects.
To achieve the object, a first aspect of the present invention provides an essential oil composition having pain relieving effect, comprising, in parts by weight: 1.5 to 5 parts of lavender essential oil, 1 to 5 parts of yam essential oil, 0.5 to 3 parts of wood essential oil, 1 to 5 parts of radix angelicae pubescentis essential oil, 1 to 5 parts of notopterygium root essential oil, 1 to 5 parts of fructus cnidii essential oil, 1 to 2 parts of ailanthus altissima root essential oil and 20 to 50 parts of base oil.
According to the invention, the lavender essential oil has the effects of relieving pain, resisting depression, disinfecting, sterilizing, relieving congestion and swelling, reducing blood pressure, calming and recovering the essence, the lavender essential oil is matched with the yam essential oil with the effects of removing blood stasis and relieving swelling, the radix angelicae pubescentis essential oil with the effects of resisting inflammation and relieving pain can have good effects of relieving bone and muscle pain, the addition of the notopterygium root essential oil and the fructus cnidii essential oil has good effects of dispelling wind and removing dampness, sterilizing and diminishing inflammation, and the effects of relieving pain of the compound essential oils are further enhanced by the aid of the toona sinensis root essential oil and the wood essential oil. Wherein, the Chinese toon root is a medicine for clearing heat and drying dampness, and has the effects of astringing and inducing astringency. The costustoot has the effects of strengthening the spleen and promoting digestion, and can help the human body to dredge the whole body qi movement, but simultaneously contains a plurality of volatile oil, ether alcohols and other components, and the components have the effects of sterilizing and diminishing inflammation. In the invention, the use of the ailanthus altissima root essential oil and the wood essential oil can consolidate the efficacies of the above essential oils.
In order to further enhance the pain relief effect, in the present invention, the essential oil composition further comprises: 0.1 to 1 part of suberect spatholobus stem essential oil, 1 to 3 parts of pseudo-ginseng essential oil and 0.1 to 1 part of ginseng essential oil.
In the invention, the spatholobus stem and the pseudo-ginseng have the effects of promoting blood circulation and removing obstruction in collaterals, and have good effects of promoting blood circulation and relieving pain when being compounded with the climbing fern for bones and muscles. And for nervous headache and migraine, proper amount of ginseng is better.
Preferably, the essential oil composition of the present invention further comprises: 0.1 to 1 part of borneol essential oil and/or 0.1 to 1 part of peppermint essential oil. Wherein, the borneol essential oil has excellent transdermal function, is a good permeation promoter, and has the function of promoting blood circulation. The penetration promoting function of the mint essential oil is inferior to that of borneol essential oil, but the mint essential oil has good efficacy on headache, nerve disorder and spasm, so the mint essential oil is compounded for migraine and nervous headache.
In the present invention, in order to better relieve the low back pain, the essential oil composition further comprises: 1 to 5 parts of lemon essential oil, 1 to 5 parts of mugwort essential oil and 0.5 to 3 parts of patchouli essential oil.
And in order to better relieve back, neck and shoulder pain, the essential oil composition further comprises: 1-5 parts of kuh-seng essential oil and 3-5 parts of rhizoma cibotii essential oil.
And in order to relieve migraine and nervous headache, the essential oil composition further comprises: 1 to 5 parts of kuh-seng essential oil, 1 to 5 parts of bitter apricot seed essential oil and 1 to 5 parts of grapefruit oil.
In the present invention, the base oil is castor oil. The castor oil has good convergence efficacy, can relieve pain and itching, and can better play the effect of relieving pain by being compounded with the essential oil component. However, the base oil is not limited to castor oil, and any of grape seed oil, wheat germ oil, olive oil, jojoba oil and the like may be used as the base oil of the present invention.
In the invention, the essential oil composition further comprises 0.1-5 parts of auxiliary agent according to parts by weight.
The auxiliary agent is selected from one or more of the following groups: bisabolol, tocopherol, glycerol, sodium hyaluronate, and squalene.
Preferably, the auxiliary agent is bisabolol and squalene which are compounded according to the mass ratio of 2:1. Bisabolol is an active ingredient extracted from spring Huang Juli, has good solubilizing property, is used as a solubilizer of a mixed essential oil component in the invention, has the effects of sterilizing, diminishing inflammation, resisting irritation, repairing and the like, and can enhance the pain relieving effect of an essential oil composition when being matched with squalene.

In the present invention, the individual essential oils may be obtained by a commercially available method.
In a preferred preparation method of the invention, the base oil and the auxiliary agent are mixed uniformly, and then each single essential oil is added for further uniform mixing. The mixing means may be stirring, rolling, turning, shaking, etc.
The essential oil composition provided by the invention is simple in use method, and can be directly smeared on painful parts of a body, and the operation effects of pressing, massaging, acupoint pressing and the like are better.
In a third aspect, the present invention provides a medicament having pain relieving effect, comprising the above-mentioned essential oil composition.
In the invention, the medicine also comprises pharmaceutically acceptable auxiliary materials. Depending on the different pharmaceutical dosage forms: different auxiliary materials are adopted for the ointment, the emulsion, the gel, the liniment or the patch. The common auxiliary materials of the ointment are as follows: vaseline, liquid paraffin, lanolin, beeswax, animal oil, vegetable oil, glyceryl monostearate, stearyl alcohol, etc. and their mixture. The common auxiliary materials of the emulsion are emulsifying agents. The common auxiliary materials of the emulsion are as follows: lipophilic emulsifying agent such as cetyl alcohol and cholesterol, nonionic emulsifying agent such as polysorbate and sorbitan fatty acid, and sodium oleate. First stage clinical evaluation
1) Investigation of relief efficacy against Low back pain
35 male subjects and 35 female subjects with ages of 30-65 were randomly selected, and the male subjects and female subjects were randomly divided into 7 groups, respectively using the essential oil compositions of examples 1-5 (corresponding to test groups 1-5) and comparative examples 1-2 (corresponding to control groups 1-2), the male and female proportions of each group were also 1:1, and these patients had pain symptoms caused by different degrees of lumbar injuries, and the subjects repeatedly applied the essential oil composition 3 times a day and a night to lumbar pain parts, each time for 3-5 mL, and were used for 4 weeks in combination with 10min massage or pressing to promote absorption. And (3) judging the effect: and (3) curing: the symptoms of the lumbar pain completely disappear; the effect is shown: at least 80% resolution of the symptoms of lumbar pain; the method is effective: the symptoms of the lumbar pain subside by 40 to 60 percent; invalidation: there was no change or exacerbation of the symptoms of lumbar pain. The results are shown in Table 1:

The total effective rate of the test group 8 reaches 100%, and the cure rate can also reach 40%. The control group 3 removes the ailanthus altissima essential oil and the wood essential oil, the total effective rate is only 50 percent, the cure rate is 0, and the number of significant effects is also reduced. After the castor oil is replaced by the control group 4, the cure rate is 0 and the total effective rate is also reduced although the number of significant persons is increased.
3) Investigation of relief efficacy against headache
35 male subjects and 35 female subjects were randomly selected, the ages were 30-65 years, the male subjects and female subjects were randomly divided into 7 groups, the essential oil compositions of examples 11-15 (corresponding to test groups 11-15) and comparative examples 5-6 (corresponding to control groups 5-6) were used respectively, the male and female proportions of each group were also 1:1, these patients had migraine or nervous headache, the subjects repeatedly applied the essential oil composition 2 times per night at the headache site, each time was 0.5-1 mL, and 5min massage or pressing was used in combination to promote absorption for 2 weeks. And (3) judging the effect: and (3) curing: headache symptoms completely disappeared; the effect is shown: at least 80% regression of headache symptoms; the method is effective: headache symptom is resolved by 40 to 60 percent; invalidation: the headache symptoms are unchanged or aggravated. The results are shown in Table 3:

As can be seen primarily from the data in table 3, the essential oil composition of test group 15 has the best soothing effect on head pain, the total effective rate can reach 100%, and the cure rate is the highest, reaching 80%. Secondly, the test group 14 has the total effective rate of 100 percent and the cure rate of 60 percent. The total effective rate of the test groups 11 to 13 can reach more than 80 percent. The control group 5 removes the ailanthus altissima essential oil and the wood essential oil, the total effective rate can reach 70 percent, and the number of significant effects is slightly reduced. After the castor oil replacement, control group 4 had not changed from test group 11.
Second phase clinical evaluation
(1) Investigation of relief efficacy against Low back pain
50 male subjects and 50 female subjects with ages between 30 and 65 were randomly selected, and were randomly divided into 2 groups, and the essential oil compositions of example 5 (corresponding to test group I) and example 10 (corresponding to control group I) in CN201910005254.5 were respectively used, wherein the ratio of male and female in each group was 1:1, and the patients had pain caused by waist injuries of different degrees, and the subjects repeatedly applied the essential oil compositions 3 times a day and 3-5 mL each time at the waist pain part, and the subjects were continuously used for 4 weeks in combination with 10min massage or pressing to promote absorption. And (3) judging the effect: and (3) curing: the symptoms of the lumbar pain completely disappear; the effect is shown: at least 80% resolution of the symptoms of lumbar pain; the method is effective: the symptoms of the lumbar pain subside by 40 to 60 percent; invalidation: there was no change or exacerbation of the symptoms of lumbar pain. The results are shown in Table 4:

As can be seen from the data in table 5, the essential oil composition of test group II has remarkable effect of relieving back and neck and shoulder pain, and the total effective rate reaches 100%, which is better than that of control group II.
(3) Investigation of relief efficacy against headache
50 male subjects and 50 female subjects with ages between 30 and 65 were randomly selected, and the male subjects and female subjects were randomly divided into 2 groups, and the essential oil compositions of example 15 (corresponding to test group III) and CN201911276697.4 (corresponding to control group III) were used respectively, the ratio of male and female in each group was 1:1, and these patients had migraine or nervous headache, and the subjects repeatedly applied the essential oil composition 2 times per night at the headache part, each time by 0.5 to 1mL, and the subjects were continuously used for 2 weeks in combination with 5min massage or pressing to promote absorption. And (3) judging the effect: and (3) curing: headache symptoms completely disappeared; the effect is shown: at least 80% regression of headache symptoms; the method is effective: headache symptom is resolved by 40 to 60 percent; invalidation: the headache symptoms are unchanged or aggravated. Note that: among the null population in test group III, 1 was allergic to essential oil odor and was discarded after 1 day of use.
As can be seen from the data in table 6, the essential oil composition of test group III has a remarkable soothing effect on head pain, and the total effective rate reaches 90%. Although the total effective rate of the control group III is higher than that of the test group III, the cure rate and the obvious efficiency of the test group III are far higher than those of the control group III.
Typical case

Cases five and Cao Mou, women, chefs, had nervous headache for up to 5 years, and the headache symptoms disappeared about 1 month after using the essential oil of example 15 of the present invention.

Claims (8)
1. An essential oil composition having pain relief effect, characterized in that: the components are as follows by weight: 1.5-5 parts of lavender essential oil, 1-5 parts of yam essential oil, 0.5-3 parts of wood essential oil, 1-5 parts of radix angelicae pubescentis essential oil, 1-5 parts of notopterygium root essential oil, 1-5 parts of fructus cnidii essential oil, 1-2 parts of ailanthus altissima root essential oil and 20-50 parts of castor bean oil.
2. An essential oil composition having pain relief efficacy as claimed in claim 1, wherein: the essential oil composition further comprises: 0.1-1 part of suberect spatholobus stem essential oil, 1-3 parts of pseudo-ginseng essential oil and 0.1-1 part of ginseng essential oil.
3. An essential oil composition having pain relieving effect according to claim 1 or 2, characterized in that: the essential oil composition further comprises: 0.1-1 part of borneol essential oil and/or 0.1-1 part of peppermint essential oil.
4. An essential oil composition having pain relief efficacy according to claim 3, wherein: the essential oil composition further comprises: 1-5 parts of lemon essential oil, 1-5 parts of mugwort essential oil and 0.5-3 parts of wrinkled gianthyssop essential oil.
5. An essential oil composition having pain relief efficacy according to claim 3, wherein: the essential oil composition further comprises: 1-5 parts of kuh-seng essential oil and 3-5 parts of rhizoma cibotii essential oil.
6. An essential oil composition having pain relief efficacy according to claim 3, wherein: the essential oil composition further comprises: 1-5 parts of kuh-seng essential oil, 1-5 parts of bitter almond essential oil and 1-5 parts of grapefruit oil.
7. The method for preparing the essential oil composition with pain relieving effect according to any one of claims 1 to 6, which is characterized in that: the method comprises the following steps: preparing essential oil by steam distillation, weighing the components according to the formula, and mixing uniformly.
8. A medicament having pain relief efficacy, characterized in that: an essential oil composition according to any one of claims 1 to 6.
