本发明公开了一种具有口服降糖活性的鱼明胶水解产物及其应用,所述鱼明胶水解产物主要是由如下方法制得:1)使鱼明胶与木瓜蛋白酶和中性蛋白酶混合,同时加入桑黄酮和α‑萘黄酮,酶解,以形成鱼明胶产物A;2)使鱼明胶产物A与碱性蛋白酶和胃蛋白酶混合,同时加入泽兰黄酮和黄芩黄酮I,继续酶解,以形成鱼明胶终产物,即得。与现有技术相比,本发明提供了一种新型的鱼明胶水解产物,其具有显著的口服降糖效果,可以用于保健食品的制备。此外,制备方法简单易操作,利用黄酮显著促进了水解酶活性,显著降低了食品级水解酶的使用量和水解反应时间,大大降低的生产成本。

技术领域
本发明属于鱼明胶水解产物技术领域,具体涉及一种具有口服降糖活性的鱼明胶水解产物及其应用。
背景技术
水解明胶亦称胶原多肽,是胶原蛋白或明胶经过一定程度的水解而得到的。水解明胶与明胶相比具有较强的水溶性、易被人体吸收等特性。另外,水解明胶还具有较多的生理功能,例如抗高血压、预防与治疗骨质疏松、治疗胃溃疡等,还具有抗衰老、抗氧化等功能。近年来,水解明胶的生产也越来越引起人们的重视。鱼类加工业的快速发展,产生了大量的副产物,其中主要包括鱼皮明胶,占鱼总重量10%左右。鱼皮中的主要成分就是胶原蛋白,是巨大的潜在可利用蛋白资源,可用来制备胶原蛋白、明胶等产品,而鱼明胶在医药、化妆品、保健品等附加值产品领域已经得到广泛的利用,因此,有效利用鱼皮废弃物提取胶原蛋白,不仅可以变废为宝,而且可作为一种新型安全的胶原蛋白资源,用于弥补猪,牛等哺乳动物胶原蛋白资源的不足,以及对进一步促进水产养殖业的可持续性发展,提高水产养殖加工业的附加值具有重大的现实意义。
国内外很多的报道对鱼皮的处理采用强酸碱的方法,利用酸或者碱在一定外界作用条件下提取胶原蛋白,此种处理方法不仅对环境污染大,并且操作复杂,耗时较长且危险。
发明内容
发明目的:为了解决现有技术中存在的问题,本发明提供了一种具有口服降糖活性的鱼明胶水解产物及其应用,所述鱼明胶水解产物安全、口服降糖活性高,并且制备方法简单易操作。
技术方案:为达到上述发明目的,本发明采用以下技术方案:
一种具有口服降糖活性的鱼明胶水解产物,所述鱼明胶水解产物主要是由如下方法制得:
1)使鱼明胶与木瓜蛋白酶和中性蛋白酶混合,同时加入桑黄酮和α-萘黄酮,酶解以形成鱼明胶产物A;
2)使鱼明胶产物A与碱性蛋白酶和胃蛋白酶混合,同时加入泽兰黄酮和黄芩黄酮I,继续酶解,以形成鱼明胶终产物,即得。
优选的,步骤1)中,所述木瓜蛋白酶和中性蛋白酶的用量为0.01%~0.02%(w/w),其中,木瓜蛋白酶和中性蛋白酶的质量比为(1-3):(3-1)。
优选的,步骤1)中,所述桑黄酮和α-萘黄酮的使用量为0.1%~0.2%(w/w),其中,桑黄酮和α-萘黄酮的质量比为(1-3):(3-1)。
优选的,步骤1)中,所述酶解的时间为0.5-1小时,pH 5~7,温度40~65℃。
优选的,步骤2)中,所述碱性蛋白酶和胃蛋白酶的用量为0.01%~0.02%(w/w),其中,碱性蛋白酶和胃蛋白酶的质量比为(1-3):(3-1)。
优选的,步骤2)中,所述泽兰黄酮和黄芩黄酮I的使用量为0.15%~0.25%(w/w),其中,泽兰黄酮和黄芩黄酮I的质量比为(1-3):(3-1)。
优选的,步骤2)中,所述继续酶解的时间为0.5-1小时,pH 5~7,温度40~65℃。
本发明最后提供了所述的鱼明胶水解产物在制备口服降糖药物或降糖食品中的应用。
本发明不采用传统的酸碱处理方法,而是直接采用环保安全的酶解法,利用各种不同的蛋白酶在一定的外界环境条件下对胶原进行限制性降解,将末端肽切割下来,由于胶原肽链间的共价交联键是由分子末端的赖氨酸或者羟赖氨酸相互作用形成的,末端肽被切下后,含有三螺旋结构的主体部分仍然紧密联结,但可溶于低浓度有机酸或者中性溶液中,从而被提取出来的方式来处理鱼明胶并利用酶促进剂显著提高酶解效率、降低酶使用量,得到具有口服降糖活性的鱼明胶水解产物,并且减少环境污染。本发明鱼明胶水解产物安全、口服降糖活性高,制备方法简单易操作,明显提高鱼明胶水解产物附加值。
有益效果:与现有技术相比,本发明提供了一种新型的鱼明胶水解产物,其具有显著的口服降糖效果,可以用于保健食品的制备。此外,制备方法简单易操作,利用黄酮显著促进了水解酶活性,显著降低了食品级水解酶的使用量和水解反应时间,大大降低的生产成本。

下述实施例中所述实验方法,如无特殊说明,均为常规方法;所述试剂和材料,如无特殊说明,均可从商业途径获得。
实施例1
一种如上所述的具有口服降糖活性的鱼明胶水解产物的制备方法,包括如下步骤:
1)使用鱼明胶与0.01%(w/w)木瓜蛋白酶和中性蛋白酶(1:1)在pH 5.5和55℃的温度环境下混合,同时加入0.1%(w/w)的桑黄酮和α-萘黄酮(1:1),酶解,反应0.5小时形成鱼明胶产物A;
2)使鱼明胶产物A与0.02%(w/w)碱性蛋白酶和胃蛋白酶(1:1)在pH 6.5和60℃的温度环境下混合,同时加入0.25%(w/w)泽兰黄酮和黄芩黄酮I(1:1),继续酶解,反应1.0小时形成鱼明胶终产物,制得成品;
实施例2
一种如上所述的具有口服降糖活性的鱼明胶水解产物的制备方法,包括如下步骤:
1)使用鱼明胶与0.02%(w/w)木瓜蛋白酶和中性蛋白酶(2:1)在pH 6.0和60℃的温度环境下混合,同时加入0.2%(w/w)的桑黄酮和α-萘黄酮(2:1),酶解,反应1.0小时形成鱼明胶产物A;
2)使鱼明胶产物A与0.01%(w/w)碱性蛋白酶和胃蛋白酶(1:2)在pH 6.0和65℃的温度环境下混合,同时加入0.2%(w/w)泽兰黄酮和黄芩黄酮I(2:1),继续酶解,反应0.5小时形成鱼明胶终产物,制得成品;
实施例3
利用高脂高糖饲料喂养的C57小鼠构建高血糖模型,小鼠自由进食普通正常饲料为对照组,小鼠自由进食高脂高糖饲料16周为模型组,小鼠自由进食高脂高糖饲料16周同时自由饮用鱼明胶水解产物水溶液(5%,v/v)为干预组,小鼠自由进食高脂高糖饲料16周同时自由饮用西地那汀(2.5%,v/v)为阳性药物组。16周后进食24小时后,采集血液,检测空腹血糖水平和空腹糖化血红蛋白水平。实验结果如图1所示,发现鱼明胶水解产物显著降低小鼠血糖水平,降糖效果与阳性药物西地那汀相当。

1.一种具有口服降糖活性的鱼明胶水解产物,其特征在于,所述鱼明胶水解产物主要是由如下方法制得:
1)使鱼明胶与木瓜蛋白酶和中性蛋白酶混合,同时加入桑黄酮和α-萘黄酮,酶解,以形成鱼明胶产物A;
2)使鱼明胶产物A与碱性蛋白酶和胃蛋白酶混合,同时加入泽兰黄酮和黄芩黄酮I,继续酶解,以形成鱼明胶终产物,即得。
2.根据权利要求1所述的具有口服降糖活性的鱼明胶水解产物,其特征在于,步骤1)中,所述木瓜蛋白酶和中性蛋白酶的用量为0.01%~0.02%(w/w),其中,木瓜蛋白酶和中性蛋白酶的质量比为(1-3):(3-1)。
3.根据权利要求1所述的具有口服降糖活性的鱼明胶水解产物,其特征在于,步骤1)中,所述桑黄酮和α-萘黄酮的使用量为0.1%~0.2%(w/w),其中,桑黄酮和α-萘黄酮的质量比为(1-3):(3-1)。
4.根据权利要求1所述的具有口服降糖活性的鱼明胶水解产物,其特征在于,步骤1)中,所述酶解的时间为0.5-1小时,pH 5~7,温度40~65℃。
5.根据权利要求1所述的具有口服降糖活性的鱼明胶水解产物,其特征在于,步骤2)中,所述碱性蛋白酶和胃蛋白酶的用量为0.01%~0.02%(w/w),其中,碱性蛋白酶和胃蛋白酶的质量比为(1-3):(3-1)。
6.根据权利要求1所述的具有口服降糖活性的鱼明胶水解产物,其特征在于,步骤2)中,所述泽兰黄酮和黄芩黄酮I的使用量为0.15%~0.25%(w/w),其中,泽兰黄酮和黄芩黄酮I的质量比为(1-3):(3-1)。
7.根据权利要求1所述的具有口服降糖活性的鱼明胶水解产物,其特征在于,步骤2)中,所述继续酶解的时间为0.5-1小时,pH 5~7,温度40~65℃。
8.权利要求1-7任一项所述的鱼明胶水解产物在制备口服降糖药物或降糖食品中的应用。
