Abstract
The invention provides a traditional Chinese medicine for treating herpes zoster and postherpetic pain and a preparation method thereof, and relates to the field of traditional Chinese medicines. The traditional Chinese medicine for treating herpes zoster and postherpetic pain comprises the following traditional Chinese medicine raw materials: earthworm, snakeskin, common cnidium fruit, ground beetle, liquorice, divaricate saposhnikovia root, common jasminorange, weeping forsythia, snow chrysanthemum, fineleaf schizonepeta herb, dahurian patrinia herb and sabia japonica. The traditional Chinese medicine composition prepared by the invention can quickly relieve pain and quickly converge the wound surface aiming at the herpes zoster, further controls local spreading, promotes scabbing and healing of the wound, has obvious antiviral effect, fully plays a role in treating both symptoms and root causes of the traditional Chinese medicine formula, namely comprehensive effects of resisting virus, relieving pain, resisting inflammation and relieving itching, and obviously shortens the course of disease treatment; aiming at the postherpetic pain of herpes zoster, the medicine can clear and activate the channels and collaterals, detoxify and detoxify, regulate qi and blood and relieve the pain of patients together.

At present, no specific medicine is available for treating herpes zoster, and the specific medicine is generally used as a treatment principle of resisting virus, nourishing nerves, relieving pain and inflammation, preventing secondary infection and shortening the course of disease.
The cure cycle of the herpes zoster is long, a large number of blisters are generated on the body, stabbing pain can be generated, great pain is brought to people, antibiotics are mostly adopted for treating the herpes zoster when the herpes zoster is treated, the drug resistance of people is enhanced by using the antibiotics, and the health of people is endangered.

parching Lumbricus, periostracum Serpentis, fructus Cnidii and Eupolyphaga Seu Steleophaga with wine, extracting Lumbricus, periostracum Serpentis, fructus Cnidii and Eupolyphaga Seu Steleophaga with water, and filtering to obtain filtrate 1;
processing fructus forsythiae and herba Patriniae with honey to obtain fructus forsythiae and herba Patriniae with honey respectively;
extracting fructus forsythiae, herba Patriniae, Glycyrrhrizae radix, radix Saposhnikoviae, folium Et cacumen Murrayae, fructus forsythiae, herba Centellae, herba Schizonepetae, and caulis Sinomenii with water, and filtering to obtain filtrate 2;
and mixing the filtrate 1 and the filtrate 2, adding 1-2% by mass of white sugar, and uniformly mixing to obtain a traditional Chinese medicine extracting solution.
Earthworm: it is cold in nature and salty in taste, enters liver, spleen and bladder channels, has effects of clearing heat and calming endogenous wind, relieving asthma, dredging collaterals and promoting urination, and can be used for treating high fever and infantile convulsion, tetany, lung heat cough and asthma, arthralgia, hemiplegia and pyretic anuresis.
Snakeskin: salty and sweet in flavor, mild in nature. It enters liver meridian. Has effects in expelling pathogenic wind, arresting convulsion, removing nebula, and removing toxic materials. Can be used for treating infantile convulsion, spasm, nebula, pharyngitis, swelling, and skin pruritus.
Fructus cnidii: pungent and bitter with warm nature. It enters kidney meridian. Has effects of eliminating dampness, dispelling pathogenic wind, killing parasites, relieving itching, warming kidney and tonifying yang. Can be used for treating pudendal pruritus, leukorrhagia, eczema, pruritus, lumbago due to damp arthralgia, sexual impotence due to kidney deficiency, and infertility due to cold womb.
Ground beetle: cold in nature, salty in taste and slight toxic in property. It enters liver meridian. Has effects in removing blood stasis and nourishing bones and muscles. Can be used for treating fracture of tendons and bones, amenorrhea due to blood stasis, and abdominal mass.
Wind prevention: pungent and sweet in flavor, slightly warm in nature. It enters bladder, spleen and liver meridians. Has effects of dispelling pathogenic wind, relieving exterior syndrome, eliminating dampness, relieving pain and relieving spasm. Can be used for treating exterior syndrome due to exogenous pathogenic factor, rubella pruritus, rheumatalgia, tetanus, and spleen deficiency with excessive dampness.
Murraya paniculata (thunb.) repertoire: pungent, slightly bitter, warm in nature and slightly toxic. It enters heart, liver and lung meridians. Has effects in promoting qi and blood circulations, dispelling blood stasis, relieving pain, and removing toxic materials. Can be used for treating traumatic injury, rheumatalgia, gastralgia, toothache, tetanus, epidemic encephalitis B, insect bite, snake bite, and local anesthesia. Can be used for treating gastralgia, rheumatalgia, toothache, traumatic injury, and insect bite.
Fructus forsythiae: slightly cold in nature and bitter in taste. It enters lung meridian, heart meridian and small intestine meridian. Has effects in clearing away heat and toxic materials, relieving swelling, dispersing pathogen accumulation, inhibiting microorganisms, inhibiting phosphodiesterase and lipoxygenase, relieving emesis, resisting liver injury, relieving inflammation, inhibiting elastase activity, and reducing spontaneous hypertension.
Snow chrysanthemum: sweet in flavor and neutral in nature. Has the effects of clearing away heat and toxic materials and eliminating dampness, and can be used for treating chronic dysentery, conjunctival congestion, swelling and pain, damp-heat dysentery, etc.
Herba Patriniae: cool in nature, pungent and bitter in flavor. It enters stomach meridian, large intestine meridian and liver meridian. Clear heat and remove toxicity, dispel stasis and expel pus.
Caulis Sinomenii: bitter and pungent in flavor and warm in nature. It enters liver meridian. Dispel wind, remove dampness, activate blood and remove toxicity. Can be used for treating rheumatic arthralgia, arthroncus of knee, edema, tinea pedis, swelling and pain due to traumatic injury, fracture, deep abscess, osteomyelitis, suppurative arthritis, spondylitis, pyocutaneous disease, and skin pruritus.
Herba schizonepetae: pungent in flavor and slightly warm in nature. It enters lung and liver meridians. Relieving exterior syndrome, dispelling pathogenic wind, promoting eruption and eliminating sore. Can be used for treating common cold, headache, measles, rubella, and pyocutaneous disease.
Licorice root: sweet in flavor and neutral in nature. It enters heart, lung, spleen and stomach meridians. Invigorating spleen and replenishing qi, clearing away heat and toxic materials, eliminating phlegm and relieving cough, relieving spasm and pain, and harmonizing the above drugs. Can be used for treating weakness of spleen and stomach, asthenia, palpitation, short breath, cough, excessive phlegm, abdominal distention, limb spasm, pain, carbuncle, suppurative sore, and relieving drug toxicity and strong toxicity.

Aiming at the residual pain after herpes zoster and the imbalance of qi, blood, yin and yang as the basis of pain, the reignition of lingering toxin is the pain factor, and the key to pain is that the damage of the channels and collaterals is not repeated. The traditional Chinese medicinal materials are sealed and soaked with the wine, so that the wine can be fully absorbed by the traditional Chinese medicinal materials, the loss of the wine is reduced in the frying process, and the wine frying effect of the traditional Chinese medicinal materials can be enhanced.
In some embodiments of the invention, the mass ratio of the raw materials to the wine in the wine frying step is (5-8): 1, the wine is 52-60 degrees, and the wine frying step adopts high-alcohol wine which can enhance the penetration of the Chinese medicinal materials, and preferably selects white wine with 50-60 degrees. The mass ratio of the traditional Chinese medicinal materials to the white spirit is (2-3): 1, the waste of white spirit can be fully reduced on the basis of ensuring the permeation effect.
In some embodiments of the present invention, the honey-roasting specifically comprises the following steps: heating honey to 113-118 ℃, adding boiled water for diluting when the water content is 10-15%, adding the crushed fructus forsythiae and herba patriniae after heating and boiling, continuously stir-frying uniformly until the honey becomes dark yellow, loose and non-sticky, and the coolness in the fructus forsythiae and the herba patriniae can be inhibited by the honey-frying operation.

Example 1
The traditional Chinese medicine for treating herpes zoster and postherpetic pain comprises the following raw materials: 25g of earthworm, 13g of snakeskin, 26g of common cnidium fruit, 10g of ground beetle, 10g of liquorice, 13g of divaricate saposhnikovia root, 8g of weeping forsythia capsule, 8g of common jasminorange herb, 5g of snow chrysanthemum, 6g of fineleaf schizonepeta herb, 5g of dahurian patrinia herb and 8g of sabia japonica.
The preparation method comprises the following steps: putting the earthworm, the snakeskin, the common cnidium fruit and the ground beetle into a sealable container, adding 30g of white spirit with the alcohol content of 52 degrees, uniformly mixing, sealing the container, taking out after 2 days, putting into an iron pan, keeping the temperature at 80-90 ℃, turning over and frying for 10min, then changing the color of the raw materials, pouring out, and cooling for later use; taking another pot, pouring honey into the pot, heating the honey to 113-118 ℃, adding boiling water for diluting when the water content is 10% -15%, adding fructus forsythiae and herba patriniae which are crushed into 30 meshes after the honey is boiled, continuously stir-frying the mixture evenly until the honey is dried to be dark yellow, loose and not sticky, pouring the mixture out, and cooling the mixture for later use;
adding water into Lumbricus, periostracum Serpentis, fructus Cnidii and Eupolyphaga Seu Steleophaga parched with wine, decocting with strong fire to boil, turning to slow fire, decocting for 1.5 hr, pouring out filtrate, decocting the residue with water for 2 hr, pouring out filtrate, and mixing the two filtrates to obtain filtrate 1; adding water into honey-fried fructus forsythiae and herba Patriniae, Glycyrrhrizae radix, radix Saposhnikoviae, folium Et cacumen Murrayae, fructus forsythiae, herba Centellae, herba Schizonepetae and caulis Sinomenii, boiling with strong fire, turning to slow fire, decocting for 2 hr, pouring out filtrate, decocting the residue with water for 1 hr, pouring out filtrate, and mixing the filtrates to obtain filtrate 2; and mixing the filtrate 1 and the filtrate 2, adding white sugar accounting for 2% of the mass of the filtrate, uniformly mixing, concentrating until the relative density is 1.1 at 50-60 ℃, and preparing the external ointment for treating herpes zoster.
The topical ointment is applied to herpes, and is applied once a day.
Example 2
The traditional Chinese medicine for treating herpes zoster and postherpetic pain comprises the following raw materials: 26g of earthworm, 12g of snakeskin, 25g of common cnidium fruit, 13g of ground beetle, 13g of liquorice, 12g of divaricate saposhnikovia root, 10g of weeping forsythia capsule, 7g of common jasminorange herb, 7g of snow chrysanthemum, 5g of fineleaf schizonepeta herb, 6g of dahurian patrinia herb and 9g of sabia japonica.
The preparation method comprises the following steps: putting the earthworm, the snakeskin, the common cnidium fruit and the ground beetle into a sealable container, adding 35g of white spirit with the alcohol content of 60 degrees, uniformly mixing, sealing the container, taking out after 1 day, putting into an iron pan, keeping the temperature at 80-90 ℃, turning over and frying for 12min, then changing the color of the raw materials, pouring out, and cooling for later use; taking another pot, pouring honey into the pot, heating the honey to 113-118 ℃, adding boiling water for diluting when the water content is 10% -15%, adding fructus forsythiae and herba patriniae which are crushed into 20 meshes after the honey is boiled, continuously stir-frying the mixture evenly until the honey is dried to be dark yellow, loose and not sticky, pouring the mixture out, and cooling the mixture for later use;

Experimental example 1
Selecting 100 patients with herpes zoster inquiry as study objects between 2018 and 2020, wherein 43 men and 57 women are aged 18-67 years, the affected parts are waist 39, breast 18, back 22, hip 10 and head, neck and shoulder 11; 100 patients were divided into 5 groups at random, wherein the experimental groups 1 to 3 used the external ointments prepared in examples 1 to 3, respectively, the experimental group 4 used acyclovir, the application method referred to the specification, the experimental group 5 was a blank group, no external application product was used, and the herpes blistering position was retested after 7 days, wherein the clinical symptoms were significantly improved to significant effect, the clinical symptoms were slightly improved to effective, the clinical symptoms were not changed to any effect, and the total effective rate was (number of effects + number of effects)/total number of people. As can be seen from table 1, compared with the control group (acyclovir), the ointments prepared in embodiments 1 to 3 of the present invention still have a higher effective rate, which indicates that the embodiments of the present invention have a very good therapeutic effect on treatment of herpes zoster.
Experimental example 2
100 postherpetic pain were selected as subjects. The inclusion criteria were that the patients were postherpetic neuralgia by electrocardiographic examination. 40 male and 60 female patients aged 22-69 years were randomly and averagely divided into 4 groups, wherein the experimental groups 1-3 were administered with the oral liquid prepared in examples 4-6 of the present invention, the experimental group 4 was a control group, vitamin C was administered at a prescribed dose every day, and the treatment time was two weeks. The degree of neuralgia before and after the treatment of the patients was evaluated by the pain Visual Analogue Scale (VAS). VAS evaluation criteria: 0 part indicates no pain, 1 to 3 parts indicates mild pain, 4 to 6 parts indicates moderate pain, and 7 to 10 parts indicates severe pain. The experimental results are shown in table 2.

As can be seen from Table 2, the oral liquid prepared in the examples 4-6 has a significant pain relief effect after two weeks, which shows that the oral liquid prepared in the examples 4-6 of the present invention has a good therapeutic effect on the pain after herpes zoster.
In conclusion, the traditional Chinese medicine for treating herpes zoster and herpes zoster residual pain and the preparation method thereof are disclosed by the embodiment of the invention. The traditional Chinese medicine composition prepared by adopting the earthworm, the snake skin, the common cnidium fruit, the ground beetle, the liquorice, the divaricate saposhnikovia root, the common jasminorange, the weeping forsythia capsule, the snow chrysanthemum, the fineleaf schizonepeta herb, the dahurian patrinia herb and the caulis sinomenii as raw materials can quickly relieve pain and quickly converge a wound surface so as to control local spreading and promote scabbing and healing of a sore opening aiming at the herpes zoster, has an obvious antiviral effect, fully plays a role in treating both symptoms and root causes of a traditional Chinese medicine formula, namely has comprehensive effects of resisting viruses, relieving pain, resisting inflammation and relieving itching, and obviously shortens the course of treatment period; aiming at the postherpetic pain of herpes zoster, the medicine can clear and activate the channels and collaterals, detoxify and detoxify, regulate qi and blood and relieve the pain of patients together.

The traditional Chinese medicine for treating herpes zoster and post-herpetic pain according to claim 1, which is prepared from the following raw materials in parts by weight: 20-30 parts of earthworm, 10-15 parts of snakeskin, 20-30 parts of fructus cnidii, 8-15 parts of ground beetle, 8-15 parts of liquorice, 10-15 parts of divaricate saposhnikovia root, 5-12 parts of fructus forsythiae, 5-10 parts of murraya paniculata, 3-8 parts of coreopsis tinctoria, 3-8 parts of schizonepeta, 3-8 parts of herba patriniae and 5-10 parts of caulis sinomenii.

parching Lumbricus, periostracum Serpentis, fructus Cnidii and Eupolyphaga Seu Steleophaga with wine, extracting Lumbricus, periostracum Serpentis, fructus Cnidii and Eupolyphaga Seu Steleophaga with water, and filtering to obtain filtrate 1;
processing fructus forsythiae and herba Patriniae with honey to obtain fructus forsythiae and herba Patriniae with honey respectively;
extracting fructus forsythiae, herba Patriniae, Glycyrrhrizae radix, radix Saposhnikoviae, folium Et cacumen Murrayae, fructus forsythiae, herba Centellae, herba Schizonepetae, and caulis Sinomenii with water, and filtering to obtain filtrate 2;
and (3) mixing the filtrate 1 and the filtrate 2, adding white sugar accounting for 1-2% of the mixed filtrate, and uniformly mixing to obtain the traditional Chinese medicine extracting solution.
