The extraction method of the cordyceps sinensis comprises the following steps:
step 1: selecting a cordyceps sinensis raw material, crushing the cordyceps sinensis raw material, adding a 6% 30% BG aqueous solution, heating to 70 ℃, carrying out water bath for 4 hours, and filtering by using a 1-micron filter paper plate to obtain a cordyceps sinensis stock solution;
step 2: taking the stock solution in the step 1, adding 6 percent BG, and diluting with water to be colorless and transparent
And step 3: and (3) filtering the liquid obtained in the step (2) by using a 0.22-micron filter paper plate to obtain the cordyceps sinensis extract.
The invention uses the high-temperature water bath technology to strengthen the extraction and separation process, can effectively improve the extraction and separation rate, shorten the extraction time, save the cost, and even can improve the quality and the yield of products, and when the cosmetics contain a small amount of cordyceps sinensis extract, the cosmetics have obvious whitening and anti-aging effects.
Example two of the inhibition of tyrosinase activity by Cordyceps sinensis mask fluid:
the purpose is as follows: and (5) measuring the half inhibition concentration of the cordyceps sinensis mask on the tyrosinase activity.
Materials: cordyceps sinensis mask
Sample preparation:
buffer solution: k2HPO4(8.71g/500mL)+KH2PO4(6.805g/500mL)→pH=6.80 (±0.02)
Substrate: about 0.3mg/mL of L-tyrosine
A spectrophotometer: uv, 480nm
The method comprises the following steps:
(1) enzyme concentration selection:
taking a small amount of enzyme, dissolving the enzyme by using a buffer solution, wherein the color is brown; adding 500. mu.L of (L-tyrosine (0.3 mg/mL)), 500. mu.L of buffer solution, 450. mu.L of pure water and 50. mu.L of enzyme into a 1.5mL centrifuge tube; before adding enzyme, water bath is carried out for 20min at 37 ℃; after the enzyme was added, the absorbance was measured at a wavelength of 480nm in a spectrophotometer at 37 ℃ in a water bath (enzyme addition-test) for 10 min. The enzyme concentration was adjusted to give an Absorbance (ABS) of between 0.45 and 0.55. The enzyme solution at this concentration was used as an experimental enzyme solution.
(2) Experimental addition
(3) Measuring absorbance at 480nm, making table, and plotting to obtain IC50。
Calculating the formula: inhibition (%) = [1- (S-SB)/(C-CB) ] + 100
Note that: before adding enzyme, water bath is carried out at 37 ℃ for 20 min; after adding enzyme, water bath at 37 ℃ (adding enzyme-testing time) for 10min for light splitting
The absorbance was measured at a wavelength of 480 nm. Water bath at 37 deg.C for 20min without enzyme.
Results
The results of the cordyceps sinensis mask on inhibiting the tyrosinase activity are shown in the table:
S:Sample SB:Sample Blank C:Control CB:Control Blank
and (4) conclusion: the Cordyceps facial mask has tyrosinase activity inhibiting effect, and IC50 value is about 25.5 μ L/mL.
Thus: the examples show that the cordyceps sinensis facial mask prepared according to the formula and the process of the examples has a remarkable whitening effect. The lowest IC50 value is 24 mu L/mL, namely the product contains 2.4% of cordyceps sinensis extract and has obvious whitening effect
