the test method comprises the following steps: 1mL of sodium phosphate buffer (1/15mol/L, pH 6.8), 0.5mL of test sample and 1mL of tyrosinase solution (tyrosinase was prepared into a 100U/mL solution using a phosphate buffer solution) were sequentially added to the test tube, water bath was performed at 30 ℃ for 10min, 2.5mL of L-tyrosinase solution (mass fraction 0.05%) as a substrate was added, and then immediately after the time was counted, the absorbance B at 475nm was measured at 20min of the reaction, and the absorbance A was measured in the same manner as in the case where 0.5mL of test sample was not added.
Wherein A is the absorbance of the test sample when the test sample is not added, and B is the absorbance of the test sample.
Three replicates were run for each group and the results are tabulated in table 1.
TABLE 1
Examples of the invention | Inhibition ratio (%) |
Example 1 | 97.24 |
Example 2 | 98.00 |
Example 3 | 99.67 |
Example 4 | 98.52 |
Example 5 | 98.63 |
Comparative example 1 | 72.35 |
Comparative example 2 | 85.31 |
Comparative example 3 | 90.23 |
Comparative example 4 | 92.20 |
Comparative example 5 | 93.01 |
Comparative example 6 | 95.44 |
As can be seen from Table 1, the samples of examples 1-5 of the present invention have good inhibitory effect on tyrosinase, and the inhibitory rate thereof is 97.24-99.67%, wherein the inhibitory rate of the sample of example 3 on tyrosinase is up to 99.67%. The results of the experiments of comparative examples 1 to 6 are all inferior to those of example 3, and it can be seen that the key effects of the composition and the content of the raw materials on the inhibition of tyrosinase by the freckle removing liquid in the invention are that the freckle removing liquid with good inhibition effect on tyrosinase can be obtained only by preparing the freckle removing liquid by adopting the scheme within the protection scope of the invention.
Second, clinical test of speckle removing effect
The test population: 110 volunteers (40 male and 70 female) aged 30-50 were randomly divided into 11 groups of 10 individuals.
A sample to be tested: the products prepared in examples 1 to 5 and comparative examples 1 to 6 (all samples were subjected to sensitization test, and the test results were all safe and harmless).
The test method comprises the following steps: the melanin content, whiteness and moisture content of the face of a tested group before the sample to be tested is smeared are measured, the tested person is smeared with the sample to be tested corresponding to each group after the face is cleaned in the morning and evening respectively, the sample is naturally absorbed when the person is patted and pressed by finger abdomen, any other skin care products related to moisturizing, freckle removing and whitening are not used in the test period, the face change condition of the tested person is detected after the tested person is used for 15 days, and the test result is counted to be shown in a table 2.
TABLE 2
Examples of the invention | Melanin change ratio (%) | Percent change in whiteness (%) | Moisture content change rate (%) |
Example 1 | -18.00 | 2.76 | 21.86 |
Example 2 | -18.53 | 2.82 | 22.06 |
Example 3 | -19.24 | 3.23 | 23.15 |

Comparative example 1 | -10.05 | 1.06 | 12.36 |
Comparative example 2 | -12.09 | 1.45 | 19.24 |
Comparative example 3 | -13.37 | 1.53 | 20.53 |
Comparative example 4 | -14.20 | 1.89 | 15.14 |
Comparative example 5 | -15.32 | 2.06 | 16.23 |
Comparative example 6 | -17.06 | 2.43 | 20.26 |
As can be seen from table 2, the anti-freckle liquid containing carnosine according to examples 1 to 5 of the present invention can reduce melanin in the skin and improve the whiteness and moisture content of the face, wherein the anti-freckle liquid according to example 3 is most preferable, and has a melanin reduction rate of 19.24%, a whiteness improvement rate of 3.23%, and a moisture content change rate of 23.15%. The freckle removing liquid in the comparative examples 1 to 6 is relatively inferior to that in the example 3, it is understood that the important significance of the synergistic combination of the components and the content of the raw materials of the freckle removing liquid is that when a part of single or multiple components are replaced, the synergy can generate certain imbalance, and finally the function of the product is weakened.
Finally, it should be noted that the above-mentioned contents are only used for illustrating the technical solutions of the present invention, and not for limiting the protection scope of the present invention, and that the simple modifications or equivalent substitutions of the technical solutions of the present invention by those of ordinary skill in the art can be made without departing from the spirit and scope of the technical solutions of the present invention.
