本发明属于药食同源类配方与食品技术领域,具体是将 “药食同源之中药”的传统中医精髓理论和“多重益生菌”有机结合起来,添加以蛋白短肽、益生元、氨基酸、碳水化合物、复合维生素、复合矿物质为辅料制成的催乳全营养配方食品及其制备方法。
背景技术
产后缺乳指产妇分娩后哺乳期内泌乳量少或无泌乳,不能满足哺育婴儿需要, 中医称之为“乳汁不行”或“乳汁不足”;多发生于产后第2-15日内甚或整个哺乳期,占所有产妇22.2%左右;催乳是一种中医治法,又称通乳、下乳,是治疗产后缺乳的方法,通过科学的方法,达到促进母乳乳汁分泌的目的;中医药治疗产后缺乳有着悠久的历史,积累了丰富的经验,如宋代陈自明著《妇人大全良方》引《灵苑方》有“下乳汁立效方”;中药催乳是中医传统的催乳方法,也是对于真性缺乳的主要疗法,已经形成了比较完整的理论体系,临床实践中取得较好的疗效;母乳含有免疫物质300余种,如多种分泌性免疫球蛋白、促生长因子、肠道激素、活性酶、吞噬细胞及淋巴细胞等,能增强婴儿的免疫力,营养丰富,适合婴儿消化吸收,最有利于婴儿的生长发育;母乳喂养能增进母婴情感的交流,促进婴儿的智能发育,还有利于母亲健康,通过婴儿吸吮乳汁可刺激产妇子宫收缩,减少产后出血,抑制排卵,推迟月经复潮,减少乳腺癌和卵巢癌的发生;防治产后缺乳对于提高母乳喂养率,具有重要意义;
《诸病源候论》云:“产后乳无汁候,即产则血水俱下,津液暴竭,经血不足";《经效产宝》则认为“气血虚弱,经络不调”为缺乳的病因;陈无择在《三因极一病证方论》中分虚实论缺乳:“产妇有二种乳脉不行,有气血盛而壅闭不行者,有血少气弱涩而不行者;虚当补之,盛当疏之”;因此选用既能养血滋阴,又可行气活血之药治疗产后缺乳;
由于现代社会生活节奏紧张、食品质量、环境污染、怀孕年龄逐步增大、人们生活习惯改变等各种因素的影响,近年来产后缺乳在我国发病率有逐步上升趋势;对产后缺乳的患者进行及时有效的治疗, 改善缺乳症状, 满足婴儿喂养的需要, 对于提高人口质量有重要意义;现代医学对产后缺乳缺乏有效的治疗措施和治疗药物, 中医药对产妇产后缺乳治疗有较多的文献报道;根据祖国医学理论, 女子妊娠时气血下注以养胎, 分娩后气血上行以化乳;乳汁为血所化生, 而赖气的运行;缺乳者多由气血虚弱、乳络不通所致;女子平素气血虚弱, 复因分娩过程耗气太过, 失血过多, 或脾胃功能虚弱者, 气血生化无源, 冲任气血一时补充不及,乳汁化生乏源, 而见乳汁甚少, 或无乳可下;《傅青主女科》: “妇人产后绝无点滴之乳, 人以为乳管之闭者也, 谁知是气与血之两涸乎;夫乳乃气血之所化而成也, 无血固不能生乳汁, 无气亦不能生乳汁”;女子以肝为先天之本,肝主藏血,主疏泄,乳房、胁肋为肝经所布, 故乳汁分泌跟肝气的疏泄有关;催乳方扶助改善产妇机体气血虚弱,补气生血, 通络行乳; 津液血乳同源, 由后天脾胃摄人的水谷精华所化生, 处方药物同时补脾健胃, 活血通络催乳;
本研究结果显示,催乳全营养配方食品使产妇泌乳量有显著增加;方中当归、阿胶、人参、黄芪补血养血行血;通草、漏芦疏肝解郁理气,通络行乳;甘草通行十二经,祛邪热,坚筋骨,健脾胃,得中和之性,有调补之功,协和诸药,使之不争,调和诸药寒凉温热,为全方之使药;全方药物又互有佐使,例如阿胶补血,止血,滋阴润燥,黄芪补气,并鼓舞气血运行,补益中土,温养脾胃,用黄芪大补脾胃之气,资生血之源,益气而生乳;阿胶与当归、黄芪等补益气血药同用,气血双补,医家认为气无形,血则有形;有形不能速生,必得无形之气以生之;黄芪用之于当归之中,自能助之以生血也,配伍人参,治血虚神怯,虚倦;方中人参能大补元气,当归活血化疲,二者配伍更具补血活血之功,参、归气血双补;蒲公英化热毒,解食毒,漏芦味苦降泻,通草人胃经,使胃气上达而下乳汁,与黄芪相佐,有通经下乳之功;全方诸药合用生化有源,补气生血,共起通络行乳、利水通乳、健脾通乳、理血产乳之效;
“药食同源”是中华原创医学之中对人类最有价值的贡献之一;中医历来强调“药疗不如食疗”,食疗成为现代人追求返璞归真、健康饮食的最佳选择;让人们在享受美食的过程中祛除病痛,避免了打针、吃药,甚至手术之苦;对于特殊营养食品,在美国、欧盟等发达国家都有相应的规定和开发应用;而我国GB29922-2013《特殊医学用途配方食品通则》食品安全国家标准中全营养食品明确规定:2.1.1全营养配方食品:可作为单一营养来源满足目标人群营养需求的特殊医学用途配方食品;2.1.2特定全营养配方食品:可作为单一营养来源满足目标人群在特定疾病或医学状况下营养需求的特殊医学用途配方食品;
研究证明中医脾与胃肠道正常菌群之间存在着十分密切的关系,催乳与肠道菌群的失调密切相关,脾虚证是反映机体脾胃生理机能不足的综合征,当机体肠胃功能异常时,肠道的微生态系统会受到严重的破坏,大肠杆菌等条件性病原菌大量增殖,双歧杆菌、乳杆菌等益生菌数量显著下降;同时菌群失调又反过来影响机体对营养物质的吸收,降低机体免疫力,削弱肠道的屏障功能,进一步加重疾病,中医传统医学认为:“脾(即消化系统)为后天之本”,肠胃健康是健康之本,人体营养的99%需要靠肠道吸收,肠内疾病也是万病之源;但随着年龄越大,肠道中有益菌群(益生菌)比例逐渐减少;如双岐杆菌在婴幼儿时期占98%,到中年时降至10%,65岁以后仅占5%;人越老,益生菌越少,肠道功能也随之逐渐下降,越来越多的研究显示,正常菌群在人体消化、免疫和抗病等方面有诸多不可替代的作用;肠道菌群组成的改变与多种疾病,特别是肠道疾病的发生发展密切相关,而从已有临床研究发现:益生菌能起到调节肠道菌群,控制肠道炎症作用,是一种安全有效的干预调节方法;
直接口服益生菌虽有一定效果,但是由于益生菌一在肠道内无法定植,且停留、占位很少,易流失,停服后有害菌易复发,相关患者需要长期服用,治疗周期长,往往使用后复发率非常高;为缩短相关患者的治疗周期,减少停药后复发率,本团队通过近叁年来对益生菌在肠道内定植机理的关健技术研究,努力寻求改进益生菌在肠道内定植的“药食同源之中药”复方,考虑到益生菌在肠道内定植的几个关键要素:益生元的存在能起到定植作用、肠道pH环境的降低和提高益生菌细胞的肠道粘附能力等,基于上述要素,本课题组从数十种“药食同源之中药”中筛选合适的食材,首次发现了添加当归、人参、芡实、茯苓、乌药叶和甘草等药食同源之中药成分的提取物及水苏糖、低聚异麦芽糖、酵母、L-谷氨酰胺等后,对外源益生菌有很强的定植作用,为肠道微生态制剂中益生菌的组方选择提供实验依据;
目前市场上有利用以食源性短肽为主要原料,添加药食两用的生姜、肉桂、芡实、茯苓、甘草等的提取物、复合维生素和矿物质组成复合配方,还有利用益生菌为主要原料的产品,但还没有发现结合催乳患者的体质特征,依据传统中医的精髓理论,因人而异,黄金配伍以多种药食两用的中药提取物精华、多种益生菌、短肽为主要原料,以益生元、碳水化合物、氨基酸、多种维生素、多种矿物质为辅料制成的特定全营养配方食品或产后催乳特殊膳食产品;

现代科学的发展已经证明:中草药能改变肠道菌群组成及恢复肠道菌群失调,肠道菌群能提高中草药的生物利用率;但是中药的黄酮类及生物碱都具有非常强的杀菌功能,也就是说在肠道内中药的黄酮类及生物碱都具有非常强的杀菌(包括益生菌)作用,解决中药杀菌功能+益生菌共同作用这个世界技术难题.
发明目的
本发明的目的是在继承中医精髓理论治病方法上,开创性将多种提高产后缺乳患者的免疫力的益生菌与“药食同源之中药”复方的提取物有机结合起来, 添加以短肽、氨基酸、碳水化合物、多种维生素、多种矿物质为辅料,不仅能够补充催乳患者所必需营养素,改善机体营养状况,还能利用药食两用中草药中的功能因子调节机体功能,益生菌能起到调节肠道菌群,毒副作用小、安全有效,但是中药的黄酮类及生物碱都具有非常强的杀菌功能,也就是说在肠道内中药的黄酮类及生物碱都具有非常强的杀菌(包括益生菌)作用,我们通过了20多年的技术攻关,取得了中药中药杀菌功能+益生菌共同作用技术,解决了这个世界技术难题;并且研发了人们容易接受的适合催乳患者食用的一种产后催乳特殊膳食。
本发明的集成创新制备方法为:第1步半仿生加超声波提取,该方法是利用超声波的特性来实现药材物质分子的加速运动,进而提高药液的浸透力,从而提高提取的效率,超声波提取是一个物理提取的过程,一般不会改变药材中有效成分的化学性质和结构,第2步提取液真空浓缩后,醇沉或超滤法去除杂质及农药残留来提纯提取液,第3步喷雾干燥的集成创新方法制成本草精华微囊化保护技术,有效解决了口服过程中高温以及强酸胃液对益生菌的灭活;采用三层蛋白质深埋微胶囊技术,再与本发明的目的集成创新的制备方法并微囊化处理后的中药提取物混合均匀的制剂工艺,以解决中药杀菌功能+益生菌共同作用这个世界技术难题。
发明内容
本研发团队根据《特殊医学用途配方食品通则》的要求,结合产后缺乳患者的体质特征,综合当今美国哈佛大学的先进生理学、医药、营养学及临床应用等研究成果,依据“消中寓补”、“均衡调养”的传统中医的精髓理论(肝、脾、肾三脏同调为主原则),采用2种以上药食两用的中药提取物精华、多种益生菌、短肽为主要原料,以益生元、碳水化合物、氨基酸、多种维生素及矿物质辅料,不仅可作为单一营养来源满足产后缺乳患者的营养需求,还有补气活血、补脾健胃、通络行乳、利水通乳、理血产乳及提高免疫力的全营养配方食品、特殊膳食、特定全营养配方食品或非全营养配方食品(组合成的复合配方);

本发明所述的中药提取物是指黄芪、炮穿山甲、漏芦、王不留行、桑寄生、穿山甲、路路通、党参、木通、熟地黄、川芎、杜仲、通草、益母草、阿胶、人参、当归、丝瓜仁、丝瓜络、木瓜、大枣、玉米须、桔梗、赤小豆、砂仁、陈皮、肉桂、覆盆子、山药、小茴香、金银花、甘草、蒲公英、葛根、黄花菜、枸杞子其中的6种以上或全部;
2.益生元
以促益生菌肠道定植、肠道收敛、降低肠道pH环境,也不被大多数的肠道腐败菌利用,但可以促进人体内有益细菌——乳杆菌、双歧杆菌的生长繁殖,从而可以抑制腐败菌生长,有助于改善和维持肠道正常功能;长期食用可以延缓衰老、通便、提高免疫力、减轻肝脏负担、提高营养素的吸收率,特别是能够改善机体对钙、铁、锌等金属离子的吸收;可选择性地促进有益菌(双歧、乳酸)的生长繁殖外,还可代谢为酸性产物如乳酸等有机酸,使结肠内pH值降低,抑制肠道有害细菌的生长;使小肠内水电解质滞留在肠腔,产生高渗效果,刺激肠蠕动引起缓泻,从而促进肠道内毒素及其他毒性物质的排出,乳果糖还可直接灭活肠道中内毒素、降低肠道pH值;

4.多种益生菌
使益生菌能大部分通过胃的强酸环境到达肠道,以促进益生菌的增殖,调节肠内微生态平衡,促进对微量元素的吸收,增强脾胃运动功能,加速药效的吸收,改善黏膜表面的微生物群或酶的平衡,或刺激机体特异性或非特异性免疫机制,提高机体定植抗力或免疫力;Xing等用双歧杆菌和乳酸杆菌预处理肝缺血再灌注大鼠,可以明显降低肠源性内毒素血症的水平并预防肝损伤的发生;选择性肠道脱污染也是调整肠道微生态的重要措施,有研究应用肠道不吸收抗生素利福昔明(rifaximin),通过抑制肠道中产尿素酶及产氨细菌的生长,来增强抵抗力;

本发明所述的氨基酸是指L-谷氨酰胺、蛋氨酸、异亮氨酸、酪氨酸、苯丙氨酸、赖氨酸、精氨酸、组氨酸、色氨酸、瓜氨酸、鸟氨酸、牛磺酸其中的一种、几种或全部混合;
7.脂肪粉
脂肪,俗称油脂,由碳、氢和氧元素组成;它既是人体组织的重要构成部分,又是提供热量的主要物质之一;食物中的脂肪在肠胃中消化,吸收后大部分又再度转变为脂肪;它主要分布在人体皮下组织、大网膜、肠系膜和肾脏周围等处;脂肪是由甘油和脂肪酸组成的三酰甘油酯,其中甘油的分子比较简单,而脂肪酸的种类和长短却不相同;脂肪酸分三大类:饱和脂肪酸、单不饱和脂肪酸、多不饱和脂肪酸;脂肪的来源可分下述二种:动物性来源:动物体内贮存的脂肪:如猪油、牛油、羊油、鱼油、骨髓、肥肉、鱼肝油等;动物乳中的脂肪:如奶油等;植物性来源:植物性脂肪来源主要是从植物中的果实内提取,如芝麻、葵花子、茶生、核桃、松籽、黄豆等;通过多种脂肪酸复合,调整饱和脂肪酸、单不饱和脂肪酸、多不饱和脂肪酸的含量比为1:1:1,ω-6和ω-3的比例为10:1~2:1,优选5:1~2:1,首选2:1;特别添加了2-8份的ω-3脂肪酸,来源包括鱼油、藻油中的一种或两种组合后加入包埋物料,经均质得到乳化液,然后将乳化液通过喷雾干燥设备中进行干燥,再经过筛分得到脂肪粉;

维生素和矿物质是人体必需的营养素,本发明中添加的维生素为维生素和矿物质是人体必需的营养素;
本发明中添加的维生素为维生素A、维生素D、维生素E、维生素K、维生素K1、维生素C、维生素B1、维生素B2、维生素B6、维生素B12、烟酸、叶酸、泛酸、胆碱、生物素其中的一种、几种或全部混合;
本发明中添加的矿物质为钙、磷、钾、钠、镁、氯、铁、锌、硒、铜、锰、碘、氟、铬、钼其中的一种、几种或全部混合,本发明所述的矿物质添加为该矿物质的有机或无机化合物;
本发明中添加的维生素矿物质混合物,是严格按照等量递增法原则,特定混合工艺进行混合得到的混合物,混合速率为12-15r/min,混合时间为20-40min;

1.一种产妇产后催乳特殊膳食,其特征在于其原料成分含量包括以重量计的下列组分:按下列原料选其中的6种以上药食两用的提取物:黄芪5份、炮穿山甲5份、漏芦5份、王不留行5份、桑寄生5份、穿山甲5份、路路通5份、党参5份、木通5份、熟地黄5份、川芎5份、杜仲5份、通草5份、益母草5份、阿胶3份、当归3份、人参3份、丝瓜仁3份、丝瓜络3份、木瓜3份、大枣3份、玉米须3份、桔梗3份、赤小豆3份、砂仁3份、陈皮3份、肉桂3份、覆盆子3份、山药3份、小茴香1份、金银花1份、甘草1份、蒲公英1份、葛根1份、黄花菜1份、枸杞子1份;微胶囊化后的双歧杆菌80份、乳杆菌80份、嗜热链球菌440份、L-谷氨酰胺100份、γ氨基丁酸50份、短肽130份、益生元100份、复合氨基酸50份、松谷膳食纤维80份、脂肪粉130份、复合维生素5份、复合矿物质50份;制作时将按下列原料选其中的6种以上药食:黄芪、炮穿山甲、漏芦、王不留行、桑寄生、穿山甲、路路通、党参、木通、熟地黄、川芎、杜仲、通草、益母草、阿胶、人参、当归、丝瓜仁、丝瓜络、木瓜、大枣、玉米须、桔梗、赤小豆、砂仁、陈皮、肉桂、覆盆子、山药、小茴香、金银花、甘草、蒲公英、葛根、黄花菜、枸杞子先采用半仿生或生物酶解加超声波提取,真空浓缩,醇沉或超滤法去除杂质及农药残留,喷雾干燥的集成创新方法制成本草精华;再进行与生物酶解提取的短肽、益生元、氨基酸、水溶性膳食纤维、MCT粉、α-亚麻酸、亚油酸、双歧杆菌(青春双歧杆菌、乳双歧杆菌、两歧双歧杆菌、短双歧杆菌、婴儿双歧杆菌、长双歧杆菌)、乳杆菌(嗜酸乳杆菌粉、干酪乳杆菌、保加利亚乳杆菌、德氏乳杆菌、乳亚种格氏乳杆菌、发酵乳杆菌、瑞士乳杆菌、唾液乳杆菌、鼠李糖乳杆菌、罗伊氏乳杆菌、植物乳杆菌、副干酪乳杆菌、约氏乳杆菌嗜热链球菌)、复合维生素及矿物质混合,混合时间为20min,混合速率为45r/min,得到混合均匀的粉状的制剂产品。
2.根据权利要求1所述的产妇产后催乳特殊膳食,还包括以重量计的下列组分:全脂乳粉15份,乳清蛋白粉20份,可食用植物水提浓缩粉13份(木瓜10、覆盆子10、大枣10、小茴香8、桂圆8、玉米须8、陈皮8、山药6、甘草6、蒲公英5、葛根5、枸杞子5),复配益生菌15份(麦芽糊精、乳双歧杆菌、长双岐杆菌、两岐双岐杆菌、嗜酸乳杆菌、鼠李糖乳杆菌、植物乳杆菌),乳糖10,鲈鱼粉10,果糖2、黑糖2,海洋鱼低聚肽粉(3%),L-谷氨酰胺(3%),抗性糊精,菊粉(2%),粉末油脂2(亚麻籽油、葵花籽油、辛烯基琥珀酸淀粉钠、麦芽糊精、二氧化硅),海藻糖,醋酸维生素A,胆钙化醇,dl-α-醋酸生育酚,植物甲萘醌,盐酸硫胺素,核黄素,盐酸吡哆醇,氰钴胺,L-抗坏血酸,烟酸,叶酸,泛酸钙,生物素,酒石酸氢胆碱,氧化镁,碳酸钙,焦磷酸铁,葡萄糖酸锌,亚硒酸钠,(DHA)二十二碳六烯酸。
