Abstract
本发明涉及药食两用发酵技术领域,公开了一种桑椹灵芝乳酸菌共发酵产物及其提取物的制备方法及应用,桑椹、灵芝、乳酸菌均为“药食两用”或“新资源目录”名单中的材料,具有食品安全保证,同时还各自具有较高的生理活性,对于促进机体免疫力提升具有显著效果,三种材料不是简单搭配,桑椹是时令水果,富含花色苷和多酚类物质;灵芝并非采用子实体,而是通过对灵芝菌深层液体发酵获得高生物量和多糖含量的灵芝菌丝球,提升免疫力效果更佳;乳酸菌有益于肠道健康,可同时促进机体免疫能力。三种材料的搭配可以互相增强各自的效果,具有显著的创新性。

技术领域
本发明涉及药食两用发酵技术,特别涉及灵芝液体深层发酵和乳酸菌发酵双重发酵工艺,具体属于一种可增强免疫力的桑椹灵芝乳酸菌共发酵产物及其制备方法和应用。
背景技术
根据中华预防医学会新型冠状病毒肺炎防控专家组发表的一项新冠肺炎流行病学研究报告,分析发现,感染者中接近80%的新冠肺炎为普通型或轻症,超过86%的感染者年龄在30-79岁,死亡病例中80%为60岁以上老年人,由此可见,免疫力水平较低的感染者发病情况较为急促和严重。而由于当前新冠肺炎尚无特效药物和疫苗可用,目前基本是对症治疗,提高人体的免疫功能显得尤为重要。人体若免疫力低下则容易患各种疾病,而且更易转化成为新冠肺炎重症患者,而轻症患者基本在加强营养与提高免疫和对症护理之后得以治愈。由上可见,除做好应当的防护措施外,人民群众应努力提高自身免疫力,除了加强锻炼和保障休息外,通过摄食具有增强机体免疫力的食品,是提升个人免疫水平的重要方式。
发明内容
发明目的:针对现有技术中存在的问题,本发明提供一种可增强免疫力的桑椹灵芝乳酸菌共发酵产物及其制备方法和应用,能够有效增强机体免疫能力,工艺简单、绿色安全。
技术方案:本发明提供的一种桑椹灵芝乳酸菌共发酵产物的制备方法,包括以下步骤:1)称取葡萄糖、玉米粉、磷酸氢二钾、酵母膏装入摇瓶中,加入蒸馏水摇匀,用纱布及牛皮纸封口,于蒸汽灭菌器中126℃灭菌25min,即得灵芝种子液培养基;2)将所述灵芝种子液培养基中接入灵芝菌丝,用纱布及牛皮纸封口,放入摇床中,于23~31℃,150rpm条件下,摇瓶避光振荡培养2~6天,即得灵芝种子液;3)取桑椹,清洗榨汁,量取桑椹汁加入摇瓶中,加入蒸馏水,混匀;用纱布及牛皮纸封口,在126℃条件下灭菌25min后,即得桑椹培养基;所述桑椹培养基中桑椹汁与蒸馏水的质量比为(0.6~1.6)︰(2.5~7.5);所述桑椹培养基的PH值为5~7;4)将步骤2)制得的所述灵芝种子液按照4~12%的接种量转接于所述桑椹培养基中,得接种灵芝的桑椹培养基;5)分别对保加利亚乳杆菌、嗜热链球菌和两歧双歧杆菌进行乳酸菌菌种活化培养,转接于步骤4)所得的接种灵芝的桑椹培养基中,将所得培养基放入摇床中,于培养温度23~31℃,摇床转速150rpm,避光发酵2~7天,得乳酸菌发酵桑椹灵芝产物;所述乳酸菌发酵灵芝和桑椹溶液的配比浓度为2:1、1:1、1:2;6)将步骤5)所得乳酸菌发酵桑椹灵芝产物,冷冻干燥,粉碎过筛,即得桑椹灵芝乳酸菌共发酵产物。
优选地,步骤1)中所述灵芝种子液培养中,葡萄糖、玉米粉、磷酸氢二钾、酵母浸膏、蒸馏水的质量比为(0.2~1.5)︰(0.3~0.8):(0.13~0.38)︰(0.15~0.35)︰(24~76)。
优选地,步骤6)中所述冷冻干燥条件为:冻结温度为-10~-35℃,冷阱温度为-28~-32℃,真空度为8~18Pa,冻干最终温度为30~75℃。
优选地,在所述步骤6)之后,还将所述桑椹灵芝乳酸菌共发酵产物粉碎后过30目~100目筛。
本发明还提供了一种上述桑椹灵芝乳酸菌共发酵产物的制备方法制备的桑椹灵芝乳酸菌共发酵产物的提取物的制备方法,包括以下步骤:将桑椹灵芝乳酸菌共发酵产物置于40~60℃烘箱内干燥,干燥产物过50~90目筛,得桑椹灵芝乳酸菌共发酵产物粉末;取桑椹灵芝乳酸菌共发酵产物粉末,置于圆底烧瓶中,加5~9倍量蒸馏水,浸泡20~30min,加热回流0.5~1.5h,用5~10层纱布过滤,滤渣再加5~9倍量蒸馏水回流0.5~1.5h,滤过,合并滤液;将滤液浓缩至1.135 g/ml生药浓度,得桑椹灵芝乳酸菌共发酵产物提取液;将所述桑椹灵芝乳酸菌共发酵产物提取液浓缩至干,即得桑椹灵芝乳酸菌共发酵产物提取物。
本发明还提供了一种上述桑椹灵芝乳酸菌共发酵产物提取物在制备提高免疫力保健食品中的应用。
本发明还提供了一种上述桑椹灵芝乳酸菌共发酵产物提取物在制备提高免疫力功能型食品中的应用。
本发明还提供了一种上述桑椹灵芝乳酸菌共发酵产物提取物在制备提高免疫力药物中的应用。
有益效果:本发明制备的桑椹灵芝乳酸菌共发酵产物具有如下优点和效果:
灵芝是我国已有两千年食用历史的著名真菌,别名芝草、瑞草,是担子菌纲、多孔菌科、灵芝属真菌。灵芝是中华医学宝库中的一宝,自古人们把灵芝奉为仙草。现代研究发现,灵芝菌丝体和孢子中含有的活性成分多达400余种,主要有灵芝多糖、麦角甾醇、低级脂肪酸、多种氨基酸等营养成分。通过液体深层发酵可以是灵芝菌丝球富集更多的多糖类物质,并发挥其生理功效。灵芝能消除呼吸道系统的炎症,特别是对治疗慢性支气管炎、支气管哮喘等效果显著,对于缓解肺炎患者的“炎症风暴”有一定效果;灵芝还具有显著的抗肿瘤、抗氧化、促进肠道健康和免疫调节功能,可以提高机体的免疫力,有效抵抗外来病毒侵袭;灵芝还对辅助治疗神经衰弱失眠有显著疗效,促进机体休息和恢复,对于机体康复有较好的效果。
桑椹作为桑科植物桑的复果,具有极高的食物营养和医用价值,在我国具有悠久的栽植和食用历史,早在两千多年前,桑椹已是中国皇帝御用补品,现已被列为“药食两用”目录。桑椹具有免疫促进作用,对脾脏有增重作用,对溶血性反应有增强作用,可防止人体动脉硬化、骨骼关节硬化,促进新陈代谢。桑椹中含有大量花色苷物质,具有诸多生理活性,如提高免疫力、维持肠道菌群多样性、明目、抗氧化等。
乳酸菌是一类能利用可发酵糖生产大量乳酸的细菌通称,乳酸菌与人类的生活关系密切,无论是食品发酵,还是工业乳酸发酵以及医疗保健领域,乳酸菌的菌株都被广泛应用,具有较高的安全性和维持肠道菌群的功能,已被列入“新资源食品”名单。常用的乳酸菌种有保加利亚乳杆菌、干酪乳杆菌、瑞士乳杆菌等。乳杆菌是人肠道中优势菌群之一,它们也常和双歧杆菌、嗜热链球菌共同用于微生态制剂中,嗜酸乳酸菌的抗肿瘤、降低胆固醇和肠道致病菌的拮抗作用显著,通过维持肠道乳酸菌群的定殖,促进人体肠道健康,进而促进机体免疫力提升和身体健康。

本发明选取在我国分布广泛,具有丰富野生植物资源的桑椹为研究对象,利用现代发酵技术对其深入开发,丰富了桑椹的应用范围,实现了药用植物资源的综合利用。
本发明选取中医临床常用药用真菌灵芝为发酵工程菌,利用液体深层发酵技术,通过响应曲面发优化了乳酸菌-桑椹-灵芝的发酵工艺,相较于灵芝传统固体培养方法,极大地减少了灵芝培养时间,在原有灵芝药用功效基础上拓展了灵芝在中药生物工程领域的用途。
本发明发酵工艺操作简便,发酵周期短、不易污染、能耗成本较低,安全可靠,绿色环保,适合于大规模工业化生产。
本发明所述的桑椹灵芝乳酸菌共发酵产物可以被制成各种剂型产品,如口服液、保健饮品、胶囊剂、片剂、颗粒剂、滴丸、软胶囊剂、糖衣片剂、薄膜衣片剂、分散片、袋茶、功能性食品等。
具体实施方式
下面结合具体实施方式对本发明进行详细的介绍。
实施方式1:
称取葡萄糖1.5g、玉米粉0.6g、磷酸氢二钾0.37g、酵母膏0.3g,装入250ml摇瓶中,加入50ml蒸馏水摇匀,纱布及牛皮纸封口,于蒸汽灭菌器中126℃灭菌25min,即得灵芝种子液培养基。在制作好的灵芝种子液培养基中接入灵芝菌丝,纱布及牛皮纸封口,放入摇床中,25℃,150rpm摇瓶振荡培养,5天后即得灵芝种子液。取桑椹,清洗榨汁,量取桑椹汁加入摇瓶中,加入蒸馏水,混匀;桑椹汁与蒸馏水质量比为1:5,混匀,调整Ph值为7。瓶口用纱布及报纸封口,在126℃条件下灭菌25min后,即得桑椹培养基。将灵芝种子液按照6%的接种量转接于桑椹培养基中,得到接种灵芝的桑椹培养基;将乳酸菌菌种活化培养后转接于接种灵芝的桑椹培养基中,放入摇床中,于培养温度25℃,摇床转速150rpm,发酵周期7天,得乳酸菌发酵灵芝桑椹产物;将上述乳酸菌发酵灵芝桑椹产物在冻结温度为-30℃,冷阱温度为-28℃,真空度为16Pa,冻干最终温度为30℃条件下冷冻干燥,粉碎过60目筛,即得桑椹灵芝乳酸菌共发酵产物。
实施方式2:
称取葡萄糖1.2g、玉米粉0.3g、磷酸氢二钾0.13g、酵母膏0.25g,装入250ml摇瓶中,加入50ml蒸馏水摇匀,纱布及牛皮纸封口,于蒸汽灭菌器中126℃灭菌25min,即得灵芝种子液培养基。在制作好的灵芝种子液培养基中接入灵芝菌丝,纱布及牛皮纸封口,放入摇床中,23℃,150rpm摇瓶振荡培养,4天后即得灵芝种子液。取桑椹,清洗榨汁,量取桑椹汁加入摇瓶中,加入蒸馏水,混匀;桑椹汁与蒸馏水质量比为1.5:5,混匀,调整Ph值为7。瓶口用纱布及报纸封口,在126℃条件下灭菌25min后,即得桑椹培养基。将灵芝种子液按照8%接种量转接于桑椹培养基中,得到接种灵芝的桑椹培养基;将乳酸菌菌种活化培养后转接于接种灵芝的桑椹培养基中,放入摇床中,于培养温度27℃,摇床转速150rpm,发酵周期7天,得乳酸菌发酵灵芝桑椹产物;将上述乳酸菌发酵灵芝桑椹产物在冻结温度为-30℃,冷阱温度为-28℃,真空度为16Pa,冻干最终温度为30℃条件下冷冻干燥,粉碎过60目筛,即得桑椹灵芝乳酸菌共发酵产物。
实施方式3:
称取葡萄糖0.9g、玉米粉0.7g、磷酸氢二钾0.5g、酵母膏0.35g,装入250ml摇瓶中,加入50ml蒸馏水摇匀,纱布及牛皮纸封口,于蒸汽灭菌器中126℃灭菌25min,即得灵芝种子液培养基。在制作好的灵芝种子液培养基中接入灵芝菌丝,纱布及牛皮纸封口,放入摇床中,25℃,150rpm摇瓶振荡培养,4天后即得灵芝种子液。取桑椹,清洗榨汁,量取桑椹汁加入摇瓶中,加入蒸馏水,混匀;桑椹汁与蒸馏水质量比为1:3,混匀,调整Ph值为5。瓶口用纱布及报纸封口,在126℃条件下灭菌25min后,即得桑椹培养基。将灵芝种子液按照10%的接种量转接于桑椹培养基中,得到接种灵芝的桑椹培养基;将乳酸菌菌种活化培养后转接于接种灵芝的桑椹培养基中,放入摇床中,于培养温度29℃,摇床转速150rpm,发酵周期7天,得乳酸菌发酵桑椹灵芝产物;将上述乳酸菌发酵桑椹灵芝产物在冻结温度为-34℃,冷阱温度为-30℃,真空度为18Pa,冻干最终温度为30℃条件下冷冻干燥,粉碎过60目筛,即得桑椹灵芝乳酸菌共发酵产物。
实施方式4:
称取葡萄糖1.5g、玉米粉0.5g、磷酸氢二钾0.5g、酵母膏0.35g,装入250ml摇瓶中,加入50ml蒸馏水摇匀,纱布及牛皮纸封口,于蒸汽灭菌器中126℃灭菌25min,即得灵芝种子液培养基。在制作好的灵芝种子液培养基中接入灵芝菌丝,纱布及牛皮纸封口,放入摇床中,2,5℃,150 rpm摇瓶振荡培养,4天后即得灵芝种子液。取桑椹,清洗榨汁,量取桑椹汁加入摇瓶中,加入蒸馏水,混匀;桑椹汁与蒸馏水质量比为1:7,混匀,调整Ph值为6。瓶口用纱布封口,在126℃条件下灭菌25min后,即得桑椹培养基。将灵芝种子液按照6%的接种量转接于桑椹培养基中,得到接种灵芝的桑椹培养基;将乳酸菌菌种活化培养后转接于接种灵芝的桑椹培养基中,放入摇床中,于培养温度25℃,摇床转速150rpm,发酵周期7天,得乳酸菌发酵桑椹灵芝产物;将上述乳酸菌发酵桑椹灵芝产物在冻结温度为-30℃,冷阱温度为-28℃,真空度为18Pa,冻干最终温度为30℃条件下冷冻干燥,粉碎过60目筛,即得桑椹灵芝乳酸菌共发酵产物。

称取实施方式2制备的桑椹灵芝乳酸菌共发酵产物,置于40~60℃烘箱内干燥,干燥产物过50~90目筛。取桑椹灵芝乳酸菌共发酵产物粉末,置于圆底烧瓶中,加7倍量蒸馏水,浸泡25min,加热回流1h,用8层纱布过滤,滤渣再加7倍量蒸馏水回流1h,滤过,合并滤液。浓缩至1.135 g/ml生药浓度,即得桑椹灵芝乳酸菌共发酵产物提取液;提取液浓缩至干,即得桑椹灵芝乳酸菌共发酵产物提取物,可用于以下实验。
实施方式6
桑椹灵芝乳酸菌共发酵饼干的制备方法
取实施方式3制备的桑椹灵芝乳酸菌共发酵产物20g,加入20g绵白糖、5g全脂奶粉、13g黄油(加热融化)、1g食盐混合揽拌,揽拌后混合物温度降至25℃,再加入2.5g小苏打混合。将称量好的80g低筋面粉(过筛)、8g玉米淀粉与预调的配料混合后,放入和面机进行调制面团,面团成型后静置5min。辊印面团,使面胚成型.最后送入烘箱设以面火175℃、底火145℃,烘烤10~15min。烘烤完成后,冷却饼干,即得桑椹灵芝乳酸菌共发酵饼干成品。

桑椹灵芝乳酸菌共发酵产物保健饮品的制备方法
(1)按实施方式5中制备得到桑椹灵芝乳酸菌共发酵产物提取液;
(2)调配:为了使口感更为甘甜可口,进行调配,配方为:取桑椹灵芝乳酸菌共发酵产物提取液(浓度为0.3g/mL)为原料1000mL,再以其总体积计,加入白砂糖:4%;柠檬酸:0.05%;木糖醇:2.5%;β~环状糊精:2.0%;麦芽糊精:1.5%;蜂蜜:2%;混合均匀后经过灌装,灭菌,即得桑椹灵芝乳酸菌共发酵保健饮品。

取实施方式5制备的桑椹灵芝乳酸菌共发酵产物提取物100g,糊精200g,混合均匀,制粒,过筛,干燥,即得桑椹灵芝乳酸菌发酵提取物的颗粒剂;或将制得的颗粒经进一步压片,干燥,即得桑椹灵芝乳酸菌共发酵产物片剂。

桑椹灵芝乳酸菌共发酵产物胶囊制备方法
取实施方式5制备的桑椹灵芝乳酸菌共发酵产物提取物100g,微粉硅胶适量,混合均匀,采用胶囊制备工艺制备,即得桑椹灵芝乳酸菌共发酵产物胶囊。
上述实施方式只为说明本发明的技术构思及特点,其目的在于让熟悉此项技术的人能够了解本发明的内容并据以实施,并不能以此限制本发明的保护范围。凡根据本发明精神实质所做的等效变换或修饰,都应涵盖在本发明的保护范围之内。
Claims (8)
1.一种桑椹灵芝乳酸菌共发酵产物的制备方法,其特征在于,包括如下步骤:
1)称取葡萄糖、玉米粉、磷酸氢二钾、酵母膏装入摇瓶中,加入蒸馏水摇匀,用纱布及牛皮纸封口,于蒸汽灭菌器中126℃灭菌25min,即得灵芝种子液培养基;
2)在所述灵芝种子液培养基中接入灵芝菌丝,用纱布及牛皮纸封口,放入摇床中,于23~31℃,150rpm条件下,摇瓶避光振荡培养2~6天,即得灵芝种子液;
3)取桑椹,清洗榨汁,量取桑椹汁加入摇瓶中,加入蒸馏水,混匀;用纱布及牛皮纸封口,在126℃条件下灭菌25min后,即得桑椹培养基;所述桑椹培养基中桑椹汁与蒸馏水的质量比为(0.6~1.6)︰(2.5~7.5);所述桑椹培养基的PH值为5~7;
4)将步骤2)制得的所述灵芝种子液按照4~12%的接种量转接于所述桑椹培养基中,得接种灵芝的桑椹培养基;
5)进行乳酸菌菌种活化培养,转接于步骤4)所得的接种灵芝的桑椹培养基中,将所得培养基放入摇床中,于培养温度23~31℃,摇床转速150rpm,避光发酵2~7天,得乳酸菌发酵桑椹灵芝产物;
6)将步骤5)所得乳酸菌发酵桑椹灵芝产物,冷冻干燥,粉碎过筛,即得桑椹灵芝乳酸菌共发酵产物。
2.根据权利要求1所述的桑椹灵芝乳酸菌共发酵产物的制备方法,其特征在于:步骤1)中所述灵芝种子液培养中,葡萄糖、玉米粉、磷酸氢二钾、酵母膏、蒸馏水的质量比为(0.2~1.5)︰(0.3~0.8):(0.13~0.38)︰(0.15~0.35)︰(24~76)。
3.根据权利要求1所述的桑椹灵芝乳酸菌共发酵产物的制备方法,其特征在于:步骤6)中所述冷冻干燥条件为:冻结温度为-10~-35℃,冷阱温度为-28~-32℃,真空度为8~18Pa,冻干最终温度为30~75℃。
4.根据权利要求1所述的桑椹灵芝乳酸菌共发酵产物的制备方法,其特征在于:在所述步骤6)之后,还将所述桑椹灵芝乳酸菌共发酵产物粉碎后过30目~100目筛。
5.一种如权利要求1至4所述任一项所述的桑椹灵芝乳酸菌共发酵产物的制备方法制备的桑椹灵芝乳酸菌共发酵产物的提取物的制备方法,其特征在于,包括以下步骤:
将桑椹灵芝乳酸菌共发酵产物置于40~60℃烘箱内干燥,干燥产物过50~90目筛,得桑椹灵芝乳酸菌共发酵产物粉末;
取桑椹灵芝乳酸菌共发酵产物粉末,置于圆底烧瓶中,加5~9倍量蒸馏水,浸泡20~30min,加热回流0.5~1.5h,用5~10层纱布过滤,滤渣再加5~9倍量蒸馏水回流0.5~1.5h,滤过,合并滤液;
将滤液浓缩至1.135 g/ml生药浓度,得桑椹灵芝乳酸菌共发酵产物提取液;
将所述桑椹灵芝乳酸菌共发酵产物提取液浓缩至干,即得桑椹灵芝乳酸菌共发酵产物提取物。
6.一种如权利要求5所述的桑椹灵芝乳酸菌共发酵产物提取物在制备提高免疫力保健食品中的应用。
7.一种如权利要求5所述的桑椹灵芝乳酸菌共发酵产物提取物在制备提高免疫力功能型食品中的应用。
8.一种如权利要求5所述的桑椹灵芝乳酸菌共发酵产物提取物在制备提高免疫力药物中的应用。
