Abstract
本发明公开了一种提高虾青素透皮吸收的组合物及其应用,该组合物含有水解透明质酸或其盐、乙酰化透明质酸或其盐和寡肽。这三者的组合能促进虾青素的透皮吸收,提高虾青素在活性皮肤层中的储留量,与化学类促渗剂相比,具有成分明确、天然来源、对皮肤刺激小等优势。

技术领域
本发明涉及一种提高虾青素透皮吸收的组合物,还涉及以该组合物和虾青素为有效成分的具有抗氧化功能的化妆品,属于化妆品技术领域。
背景技术
皮肤产生的分泌物跟空气中的氧原子接触后会发生氧化反应,使肌肤就犹如裸露在空气中的苹果一样变得暗沉,时间久了还会使肌肤变得松弛、出现表情纹等,而抗氧化剂能缓解皮肤的氧化,缓解皮肤过早产生皱纹、色斑等衰老现象,还具有提亮肤色的作用,受到了广大爱美人士的关注。为此,市场上出现了很多抗氧化产品。
虾青素亦称虾黄素,是一种酮或类胡萝卜素,色泽为橘红色,具脂溶性,不溶于水,可溶于有机溶剂。由于虾青素具有很强的还原性,且是一种天然的抗氧化剂,近年来作为新型化妆品原料广泛应用于膏霜、乳剂、唇用香脂、护肤品等各类化妆品中,特别是在高级化妆品领域有较多的应用。天然虾青素以其独特的分子结构,通过其抗氧作用,可以高效地猝灭紫外线引起的自由基,防止皮肤光老化、减少UVA和UVB对皮肤的伤害、防止皮肤癌的产生、延缓细胞衰老、减少皮肤皱纹、减少黑色素沉积、减少雀斑产生,还可保持水分,让皮肤更有弹性、张力和润泽感。虾青素通过进入到活性皮肤层中清除氧自由基、减少紫外线、雾霾、化学试剂对皮肤的伤害,对皮肤形成第一层保护。但要想达到理想的抗氧化作用,虾青素要在活性皮肤层中有足够的浓度才可以。目前多数含有虾青素的化妆品由于制剂问题,虾青素不易透过皮肤的角质层进入到活性皮肤层中,导致虾青素在活性皮肤层内的浓量不高,生物利用度低,达不到有效浓度,因此其抗氧化的效果并不是很理想。
现在的公知技术中,一般会选用丙二醇、氮酮等来作为化学促渗剂来促进虾青素的皮肤透过性,但是该类促渗剂对皮肤会有刺激,限制了其在护肤品领域的应用。随着科技的发展,在护肤品领域兴起了微针、纳晶、离子导入、高压水氧导入的方式,来促进营养成分的吸收,但是这种促进吸收的方式一是需要专业人员操作,二是以破坏皮肤屏障来实现,增加了感染风险。因此,在化妆品领域急需一种生物活性来源的、对皮肤无刺激的、使用方便的促渗技术,来帮助营养成分吸收,又不对皮肤造成不良影响。
发明内容
针对虾青素不易于透过皮肤角质层的不足,本发明提供了一种提高虾青素透皮吸收的组合物,该组合物能够很好的提高抗氧化剂虾青素的透皮吸收率,使其在活性皮肤层中储留量提高,从而使虾青素在化妆品中能够起到更为有效的抗氧化作用,此外,该组合物中的各成分也是对皮肤有利的成分,在提高虾青素透皮吸收的同时也能对皮肤起到很好的保湿、修复、美白、抗氧化、清除自由基等作用,从而使皮肤健康有光泽。
本发明还提供了一种具有抗氧化功能的化妆品,该化妆品有效成分包括虾青素和上述提高虾青素透皮吸收的组合物,这两者联用可以很好的提高虾青素的皮肤透过性,使抗氧化功能更为显著。
本发明具体技术方案如下:
一种提高虾青素透皮吸收的组合物,该组合物包括以下重量份的组分:水解透明质酸或其盐0.01-1份,乙酰化透明质酸或其盐0.01-1份,寡肽0.01-0.5份。
优选的,上述提高虾青素透皮吸收的组合物中,包括以下重量份的组分:水解透明质酸或其盐0.05-0.5份,乙酰化透明质酸或其盐0.05-0.5份,寡肽0.1-0.3份。
本发明经过试验,惊奇的发现水解透明质酸或其盐、乙酰化透明质酸或其盐和寡肽的组合能够促进抗氧化剂虾青素的透皮吸收率,提高其在活性皮肤层中的储留量,可以作为虾青素的促渗剂使用。该组合物中的各成分都是生物活性成分,也是常用于化妆品中的有效活性成分,对皮肤养护有利,不会对皮肤产生刺激,避免了化学促渗剂带来的不足。该组合物中,水解透明质酸或其盐由酶切法得到,分子量≤100kDa,可透皮吸收,并具有高抗氧化活性、高保湿性和晒后修复作用,能快速透皮吸收,直达表皮和真皮层,深层保湿,肌底修护。乙酰化透明质酸或其盐是由天然保湿因子透明质酸或其盐经乙酰化反应得到的,乙酰基的引入给透明质酸或其盐带来了亲脂性,增强了透明质酸或其盐对皮肤的亲和性和吸附性,提高了保湿性能。因此,乙酰化透明质酸或其盐可以牢牢吸附角质层,双倍保水,修复受损角质层,并充分水合皮肤。寡肽是由特定序列氨基酸组成的肽类物质,具有营养和修复的作用。上述两种透明质酸和寡肽具有水合作用,有助于打开皮肤通道,促进营养物质的渗透,同时细胞膜上有透明质酸和寡肽的受体,可以使水解透明质酸或其盐、乙酰化透明质酸或其盐以及寡肽和虾青素在氢键引力作用下,引导主动转运,进而使虾青素可以透过角质层进入到活性皮肤层中,提高虾青素在皮肤活性层的浓度,从而更好的发挥抗氧化和抗衰老的作用。
进一步的,本发明所用的水解透明质酸或其盐的分子量为2kDa-100kDa,该水解透明质酸或其盐可以直接采用酶切法得到,优选采用芽孢杆菌A50CGMCC NO.5744发酵培养得到的芽孢杆菌透明质酸酶对透明质酸或其盐进行降解,得到的相应分子量的酶切寡聚透明质酸或其盐即为本发明水解透明质酸或其盐。
进一步的,本发明所用的乙酰化透明质酸或其盐是将透明质酸或其盐进行乙酰化修饰得到的产物,其分子量为10kDa-200kDa,乙酰化程度或取代度为50-80%。
进一步的,上述水解透明质酸盐和乙酰化透明质酸盐指的是它们的可以用于化妆品中的金属离子盐,例如钠盐、钾盐、钙盐、锌盐等,常用的为钠盐。
进一步的,本发明所用的寡肽为由特定序列氨基酸组成的肽类物质,分子量段一般在1000道尔顿以下,也称作小肽,低聚肽或小分子活性肽,氨基酸个数为4—10个,常用的有寡肽-1、寡肽-2、寡肽-3、寡肽-4、寡肽-5、寡肽-6等,各寡肽在本发明中的作用类似。
进一步的,本发明还提供了一种具有抗氧化功能的化妆品,该化妆品包括虾青素和虾青素促渗剂,所述虾青素促渗剂为本发明上述提高虾青素透皮吸收的组合物。
进一步的,虾青素和虾青素促渗剂中的各成分在化妆品中的质量百分含量为:水解透明质酸或其盐0.01-1%、乙酰化透明质酸或其盐0.01-1%、寡肽0.01-0.5%、虾青素0.001-0.1%。
优选的,虾青素和虾青素促渗剂中的各成分在化妆品中的质量百分含量为:水解透明质酸或其盐0.05-0.5%、乙酰化透明质酸或其盐0.05-0.5%、寡肽0.1-0.3%、虾青素0.005-0.02%。
在上述化妆品中,水解透明质酸或其盐、乙酰化透明质酸或其盐和寡肽的存在大大提高了虾青素的透皮吸收,明显提高了产品的抗氧化作用。
进一步的,除了以上成分外,化妆品中还可以含有其他有效成分或者辅助成分,例如还包括下述成分A和成分B中的至少一种:
成分A:发酵产物和/或植物提取物;
成分B:调理剂、辅助抗氧化剂、保湿剂、乳化剂、增稠剂、防腐剂、稳定剂和pH值调节剂中的至少一种。
进一步的,上述成分A和成分B可以单独存在,也可以组合存在,成分A和成分B可以为单一的物质,也可以是混合的物质。
上述发酵产物、植物提取物、调理剂、辅助抗氧化剂、保湿剂、乳化剂、增稠剂、防腐剂、稳定剂、pH值调节剂以及它们的添加量均为本领域公知技术。本领域技术人员可以从现有技术中进行选择。
进一步的,所述发酵产物可以选自酵母提取物、二裂酵母发酵产物溶胞物、乳酸杆菌发酵产物中的至少一种。发酵产物在化妆品中所占质量百分比含量一般为0.1-10%,优选的,质量百分比含量为1-3%。
进一步的,所述植物提取物可以选自仙人掌花提取物、黄芩提取物、马齿苋提取物、越橘叶提取物、木棉花提取物、玫瑰花水、咖啡因、红没药醇、白花春黄菊提取物、紫松果菊提取物中的至少一种。植物提取物在化妆品中所占质量百分比含量一般为0.01-5%,优选的,质量百分比含量为0.1-1%。
进一步的,所述调理剂用于溶解虾青素。所述调理剂可以选自角鲨烷、橄榄油、蓖麻油、杏仁油、棉籽油、花生油、霍霍巴油、茶籽油、椰子油、棕榈油、鳄梨油、米糠油、杏仁油、小麦胚芽油、玉米胚芽油、芝麻油、澳洲坚果油、石栗子油、沙棘油、乳木果油、芥花油,硬脂酸丁酯、肉豆蔻酸肉豆蔻酯、肉豆蔻酸异丙酯、棕榈酸异丙酯、硬脂酸异辛酯、棕榈酸异辛酯、油酸甲酯、油酸癸酯、亚油酸异丙酯、异硬脂酸异丙酯、硬脂酸单甘油酯、异硬脂酸甘油酯、辛酸/癸酸甘油三酯、C12-15烷基苯甲酸酯和碳酸二辛酯中的至少一种。
进一步的,所述辅助抗氧化剂可以选自生育酚、生育酚乙酸酯、抗坏血酸棕榈酸酯、抗坏血酸乙基醚、生育酚棕榈酸酯、泛醌、半胱氨酸、组氨酸、阿魏酸、白藜芦醇、肌肽、麦角硫因中的至少一种。
进一步的,所述保湿剂可以选自透明质酸或其盐、银耳多糖、海藻糖、乙酰壳糖胺、聚谷氨酸、甜菜碱、肌酸、尿囊素中的至少一种。
进一步的,所述乳化剂可以选自卵磷脂、聚山梨醇酯-80、甘油硬脂酸酯和PEG-100硬脂酸酯、氢化卵磷脂、泊洛沙姆、聚甘油-3聚蓖麻醇酸酯、聚甘油-3蓖麻醇酸酯、菊粉月桂基氨基甲酸酯、丙烯酰基二甲基牛磺酸酯基聚合物、双-PEG/PEG-20/5PEG/PEG-20/5聚二甲基硅氧烷,甲氧基PEG/PEG-25/4聚二甲基硅氧中的至少一种。
进一步的,所述增稠剂可以选自黄原胶、小核菌胶、海藻酸钠、卡波姆、丙烯酸酯类、C10-30烷醇丙烯酸酯交联聚合物、羧甲基纤维素、羟乙基纤维素、羟丙基纤维素中的至少一种。
所述防腐剂可以选自丁二醇、戊二醇、丙二醇、乙二醇、辛甘醇、甘油、甘露醇、山梨醇、苯甲酸、苯甲酸钠、辛酰羟肟酸、对羟基苯乙酮、甘草酸二钾、水杨酸中的至少一种。
进一步的,本发明化妆品可以制成各种可行的类型,例如护肤水、肌底液、乳液、精华液、面霜、隔离霜、防晒霜、面膜、爽肤水、喷雾、唇膏、唇蜜等类型。
进一步的,上述化妆品中,按照各成分的性质以及化妆品的类型,根据现有技术中公开的方式将各成分进行混合,得到不同类型的化妆品。
本发明的有益效果:
1.本发明以水解透明质酸或其盐、乙酰化透明质酸或其盐和寡肽的组合来促进虾青素的透皮吸收性能,提高虾青素在活性皮肤层中的储留量,与化学类促渗剂相比,具有成分明确、天然来源、对皮肤刺激小等优势。
2.水解透明质酸或其盐、乙酰化透明质酸或其盐和寡肽的组合产品应用简便、安全,仅通过简单的涂抹方式就可以使各有效成分充分的吸收,无需借助专业设备及刺破皮肤的方法,充分发挥护肤美容效果。
3.水解透明质酸或其盐、乙酰化透明质酸或其盐和寡肽不仅能够作为虾青素的促渗剂,并且还具有很好的保湿、补水、修复等护肤作用,将水解透明质酸或其盐、乙酰化透明质酸或其盐、寡肽、虾青素混合用作化妆品的有效成分可以使其具有很好的抗氧化作用,起到美白、提亮的效果,此外,还具有提高虾青素的有效浓度、清除自由基、改善皮肤弹性、使皮肤健康有光泽的效果。
具体实施方式
下面结合具体实施例对本发明做进一步说明,下述说明仅是示例性的,并不对本发明保护范围进行限制。
下述实施例中,如无特别说明,所述浓度均为质量百分浓度。
下述实施例中,所用水解透明质酸或其盐来自于华熙生物科技股份有限公司,所述乙酰化透明质酸或其盐来自于华熙生物科技股份有限公司。其他材料、试剂如无特别说明,均为市购产品。
实施例1
一种抗氧化肌底液,包括虾青素、促渗剂、调理剂、辅助抗氧化剂、保湿剂、乳化剂、增稠剂、发酵产物、植物提取物、防腐剂、稳定剂、pH值调节剂和纯化水。其中,虾青素含量为0.02%,促渗剂成分及含量为:分子量2kDa-100kDa的水解透明质酸钠0.1%,分子量10kDa-200kDa、乙酰化程度50-80%的乙酰化透明质酸钠0.2%,寡肽-1 0.2%,其他成分含量如下表1所示。

制备方法如下:
(1)依次将调理剂、辅助抗氧化剂加入一容器中,加热至70-80℃备用;
(2)依次将纯化水、增稠剂、乳化剂、防腐剂、稳定剂、水解透明质酸钠、乙酰化透明质酸钠、保湿剂加入另一种容器中,加热至至70-80℃备用;
(3)依次将发酵产物、植物提取物、虾青素加入一容器中,混合,备用。
(4)在70-80℃温度下,将步骤(1)得到的混合物缓慢加入步骤(2)得到的溶液中,设定2000-3000rpm,均质8-10min,抽真空,排气泡,然后降温至50-60℃,加入步骤(3)得到的混合物,设定2000-3000rpm,均质8-10min,抽真空,排气泡,待温度降至40℃以下时,加入寡肽,搅拌均匀,用pH值调节剂调节pH值,pH值控制在5.0-7.0范围内,得到抗氧化肌底液。
实施例2-8
按照实施例1的配方和方法制备抗氧化肌底液,不同的是:虾青素和促渗剂的含量不同,具体如下表2所示。

1.2标准曲线的绘制
精密称取虾青素对照品5.0mg,加入1ml二氯甲烷,再用甲醇稀释至50ml定容,备用,避光-4℃保存。分别移取适量储备液适量,用甲醇稀释成浓度分别为0.5、1.0、2.0、5.0、10.0和20.0μg/ml的标准工作溶液,现配现用,取20μL经HPLC测定各组在475nm处的峰面积(A),以A值对浓度(ρ)进行线性回归,计算标准曲线。
1.3、样品
实验样品:实施例1-15的抗氧化肌底液;对照样品:对比例1的抗氧化肌底液。
2.实验方法:
取SD大鼠腹部皮肤,去除脂肪、肌肉、黏膜等组织后,夹在Franz扩散池中间,32℃水浴。对照组供给池给予对照样品溶液1ml,样品组供给池给予实验样品各1ml,接收池均装适量1%Tween 80溶液,搅拌进行透皮试验。对照组和样品组各设4个时间点,每个时间点3组平行样品,分别于试验开始后的第4.0h、8.0h、12.0h、24.0h取出各自三组组织,用生理盐水清洗表面残留物5次,用滤纸吸干,剪取有效渗透皮肤,加入4ml溶剂(甲醇:二氯甲烷3:1体积比),用组织粉碎机粉碎2min,超声提取30min,加入1ml 1%KOH甲醇溶液,摇匀,30℃反应30min,在4℃条件下5000r/min离心5min,0.22μm滤膜过滤,然后按照上述色谱条件进行测试。
3.结果计算方式
按1.1色谱条件检测,将各组峰面积代入回归方程求出虾青素质量浓度,计算滞留量。
虾青素滞留量Qn(单位μg·cm-2)=ρV/S
其中:ρ:由标准曲线计算得到的浓度(μg/ml)
V:提取液的体积(ml)
S:实验皮肤的面积(cm2)
4.试验结果
各样品进行上述透皮吸收实验后测得的虾青素滞留量结果如下表4所示:

验证例2不同的促渗剂效果对比
按照实施例1的配方和方法制备抗氧化肌底液,不同的是:促渗剂成分及含量如下表5所示。

将上述样品按照验证例1中的方法进行虾青素透皮吸收实验,结果如下表6所示。

根据已有报道,在虾青素添加量0.02%的浓度下,加入促渗剂的体系24h透皮吸收实验的虾青素滞留量Qn一般在0.5-1.6μg·cm-2,不加入促渗剂的体系24h透皮吸收实验的虾青素滞留量Qn一般在0.08-0.3μg·cm-2之间,促渗剂能够使虾青素的透皮渗透量提升5-6倍。样品2-9~2-12、2-15~2-18、2-21~2-30的促渗剂是本发明的促渗剂,其促渗作用在此范围之内,是一种合格的促渗剂。而参照其他样品的数据,只有这三者成分按照特定的含量搭配才能起到很好的促渗效果。
Claims (10)
1.一种提高虾青素透皮吸收的组合物,其特征是包括以下重量份的组分:水解透明质酸或其盐0.01-1份,乙酰化透明质酸或其盐0.01-1份,寡肽0.01-0.5份。
2.根据权利要求1所述的组合物,其特征是包括以下重量份的组分:水解透明质酸或其盐0.05-0.5份,乙酰化透明质酸或其盐0.05-0.5份,寡肽0.1-0.3份。
3.根据权利要求1或2所述的组合物,其特征是:水解透明质酸或其盐的分子量为2kDa-100kDa;乙酰化透明质酸或其盐的分子量为10kDa-200kDa,乙酰化程度为50-80%。
4.一种具有抗氧化功能的化妆品,其特征是:包括虾青素和虾青素促渗剂,所述虾青素促渗剂为权利要求1、2或3所述的提高虾青素透皮吸收的组合物。
5.根据权利要求4所述的化妆品,其特征是:虾青素和虾青素促渗剂中各成分的质量百分含量为:水解透明质酸或其盐0.01-1%、乙酰化透明质酸或其盐0.01-1%、寡肽 0.01-0.5%、虾青素0.001-0.1%;优选的,虾青素和虾青素促渗剂中各成分的质量百分含量为:水解透明质酸或其盐0.05-0.5%、乙酰化透明质酸或其盐0.05-0.5%、寡肽0.1-0.3%、虾青素0.005-0.02%。
6.根据权利要求4或5所述的化妆品,其特征是:还包括下述成分A和成分B中的至少一种:
成分A:发酵产物和/或植物提取物;
成分B:调理剂、辅助抗氧化剂、保湿剂、乳化剂、增稠剂、防腐剂、稳定剂和pH值调节剂中的至少一种。
7.根据权利要求6所述的化妆品,其特征是:所述调理剂选自角鲨烷、橄榄油、蓖麻油、杏仁油、棉籽油、花生油、霍霍巴油、茶籽油、椰子油、棕榈油、鳄梨油、米糠油、杏仁油、小麦胚芽油、玉米胚芽油、芝麻油、澳洲坚果油、石栗子油、沙棘油、乳木果油、芥花油,硬脂酸丁酯、肉豆蔻酸肉豆蔻酯、肉豆蔻酸异丙酯、棕榈酸异丙酯、硬脂酸异辛酯、棕榈酸异辛酯、油酸甲酯、油酸癸酯、亚油酸异丙酯、异硬脂酸异丙酯、硬脂酸单甘油酯、异硬脂酸甘油酯、辛酸/癸酸甘油三酯、C12-15烷基苯甲酸酯和碳酸二辛酯中的至少一种;所述辅助抗氧化剂选自生育酚、生育酚乙酸酯、抗坏血酸棕榈酸酯、抗坏血酸乙基醚、生育酚棕榈酸酯、泛醌、半胱氨酸、组氨酸、阿魏酸、白藜芦醇、肌肽和麦角硫因中的至少一种;所述保湿剂选自透明质酸或其盐、银耳多糖、海藻糖、乙酰壳糖胺、聚谷氨酸、甜菜碱、肌酸和尿囊素中的至少一种。
8.根据权利要求6所述的化妆品,其特征是:所述乳化剂选自卵磷脂、聚山梨醇酯-80、甘油硬脂酸酯和PEG-100硬脂酸酯、氢化卵磷脂、泊洛沙姆、聚甘油-3聚蓖麻醇酸酯、聚甘油-3蓖麻醇酸酯、菊粉月桂基氨基甲酸酯、丙烯酰基二甲基牛磺酸酯基聚合物、双-PEG/PEG-20/5PEG/PEG-20/5聚二甲基硅氧烷和甲氧基PEG/PEG-25/4聚二甲基硅氧中的至少一种;所述增稠剂选自黄原胶、小核菌胶、海藻酸钠、卡波姆、丙烯酸酯类、C10-30 烷醇丙烯酸酯交联聚合物、羧甲基纤维素、羟乙基纤维素和羟丙基纤维素中的至少一种;所述防腐剂选自丁二醇、戊二醇、丙二醇、乙二醇、辛甘醇、甘油、甘露醇、山梨醇、苯甲酸、苯甲酸钠、辛酰羟肟酸、对羟基苯乙酮、甘草酸二钾和水杨酸中的至少一种。
9.根据权利要求6所述的化妆品,其特征是:所述发酵产物选自酵母提取物、二裂酵母发酵产物溶胞物和乳酸杆菌发酵产物中的至少一种;优选的,所述植物提取物选自仙人掌花提取物、黄芩提取物、马齿苋提取物、越橘叶提取物、木棉花提取物、玫瑰花水、咖啡因、红没药醇、白花春黄菊提取物和紫松果菊提取物中的至少一种。
10.根据权利要求4或5所述的化妆品,其特征是:类型为护肤水、肌底液、乳液、精华液、面霜、隔离霜、防晒霜、面膜、爽肤水、喷雾、唇膏或唇蜜。
