无名葵,锦葵科,草本一年生,在200多个秋菊植物中最具食用,药用,保健功效,有极高的利用价值。除观赏绿化环境外,珍惜之处在于全株无一废弃物,芽、苗、嫩尖、花蕾、嫩果富含蛋白质、高聚合原胶、Ve、Vc及矿物质,可作为新品种供应市场,家庭及饭店可炒菜,过水凉拌,煲汤等,荤素皆宜。口感风味独特,粘甜滑溜,无任何蔬菜与其相雷同,食之助消化,润肠理气使胃肠中废祛物通畅排出,有效的抑制毒素被吸收,控制低密度蛋白质提高,或脂肪堆积对人体的危害;尤其治疗老年人便秘有立竿见影的效果,解脱排便困难之苦,强力清洗肠胃过盛的脂肪,利于老年人的身心健康。
无名葵初放的花朵和籽仁提取生物总黄酮是一项重要新发现,无名葵黄酮含量居所有现在已知含黄酮植物之首,在此之前世界公认银杏和大豆黄酮含量最高,而新发现的无名葵却高出银杏和大豆500倍。生物黄酮对人体的重要性,早已被提高到非常重要的地位,有效抑制杀灭癌细胞和止血消炎作用,可全面预防和除治各种“混血”疾病,对人体产生的强大深远影响,且具有显著的补肾壮阳功效,因为无名葵被誉为天然“伟哥”;同时,VE的含量也比其它物种胜出,高出牛奶2000倍、鲜橙汁3000倍,独占鳌头;此外,Vc、蛋白、聚糖胶、植物纤维、矿物硒、铁、锌等人体必须的微量元素也极其丰富。
无名葵具有多种功能活性,其中黄酮、多糖等的提取是无名葵资源化利用的关键所在。回流法是多糖传统提取方法,其具有提取时间较长、能耗高、溶剂消耗量大、得率低和多糖活性相对较低等缺点。近年来,闪式提取(HE)、超声提取(UE)、超声波提取(ME)等方法广泛应用于植物多糖的提取,这些方法具有时间短、溶剂用量较小、提取得率高等特点,但不同的多糖提取方法不仅影响多糖的提取效率,而且影响多糖的结构和活性,从而影响无名葵提取液的营养特性。建立无名葵高抗氧化性和营养性的提取方法是本领域的最主要的技术弊端。
发明内容
为了克服上述现有技术的不足,实现无名葵的高值化利用,本发明提供一种高值化利用无名葵的工艺,集成现代生物分离技术、生物酶解技术、混菌微生物发酵技术、中草药配伍应用技术,研制开发出黄酮、多糖含量高和抗氧化能力强的无名葵提取液,实现无名葵提取液在保健、医疗和食品等领域的综合利用。该工艺能够有效提高无名葵黄酮、多糖提取率和抗氧化能力,生产工艺简单高效,生产成本低,易于工业化生产。
本发明通过下述技术方案实现上述技术效果:
一种高值化利用无名葵的工艺,其特征在于所述无名葵通过超声波辅助酶解和复合微生物发酵工艺制备,其中所述超声波辅助酶解采用纤维素酶和蛋白酶,复合微生物发酵包括好氧发酵和厌氧发酵二步发酵。
上述超声波辅助酶解工艺具体包括如下步骤:将无名葵原料清除杂质后粉碎,过80-150目筛,然后加入无名葵原料质量比10倍的纯净水,在40~60℃条件下浸泡40~60min;加入中草药原料质量比2~3%的纤维素酶、1~2%的非淀粉多糖酶,在超声波辅助提取10min,超声波功率为250~400W,然后静置30min以上。
上述好氧发酵工艺具体包括如下步骤:向酶解后的无名葵提取液中继续加入10倍纯净水,然后加入发酵液质量比3~5%葡萄糖、0.3~0.5%硫酸铵、0.1~0.2%磷酸二氢钾、0.01%硫酸镁,搅拌均匀;向发酵液中接种米曲霉和枯草芽孢杆菌,接种量分别为发酵液体积比的2~3%、3~5%;控温28~35℃进行好氧发酵,摇床转速100~150r/min,发酵时间12~24h。
优选的,好氧发酵过程在发酵罐中进行时通风量为0.1~0.2m3/m3.min。
上述米曲霉的种子液的制备方法为:在无菌条件下,采用土豆培养基在35~40℃条件下培养24h,制得米曲霉种子液,要求种子液的有效活菌数≥109cfu/mL。
上述枯草芽孢杆菌种子液制备方法为:在无菌条件下,采用LB培养基在28~32℃条件下培养24h,制得液体芽孢杆菌种子液,要求种子液的有效活菌数≥109cfu/mL。
上述厌氧发酵工艺包括如下步骤:好氧发酵结束后接种发酵液质量比2%的乳酸杆菌,控温28~35℃进行厌氧发酵,摇床转速100~150r/min,发酵时间12~24h,发酵结束后固液分离得无名葵提取液。
优选的,超声波辅助酶解时加入无名葵质量比3~5%的玛卡。
本发明提供一种高值化利用无名葵的工艺,所得无名葵提取液中无名葵多糖、黄酮含量显著提升,具有较强的抗氧化能力;提取液可根据应用途径做浓缩、吸附干燥、醇析或树脂纯化等处理,满足在保健、食品、医药、保鲜等领域的高效利用。
本发明提供一种高值化利用无名葵的工艺,与现有技术相比,具有以下显著优势:
(1)本发明集成现代生物分离技术、生物酶解技术、混菌微生物发酵技术、中草药配伍应用技术,开发出黄酮、黄酮、多糖含量高和抗氧化能力强的无名葵提取液,实现无名葵提取液在保健、医疗和食品等领域的综合利用;
(2)本发明利用好氧菌(米曲霉、枯草芽孢杆菌)和厌氧菌(乳酸杆菌)共同发酵中草药原料提取无名葵多糖,充分利用微生物间的协同作用,提升无名葵黄酮、多糖的提取率和体外抗氧化能力,其中:米曲霉能分泌纤维素酶等分解纤维素,打破纤维素对无名葵多糖逸出的束缚;枯草芽孢杆菌在高温发酵过程中能够杀灭发酵液中的致病菌,并具有较强的产淀粉酶、蛋白酶等能力,可以将大分子淀粉、蛋白物质降解为微生物所利用;试验结果表明本发明中好氧发酵和厌氧发酵取得了显著的协同增效作用;
(3)采本发明在无名葵提取过程中选择性的添加玛卡,可以显著无名葵提取液的抗氧化能力,并取得了预料不到的技术效果。
具体实施方式
以下实施例中好氧菌种的活化均采用以下方法:
(1)米曲霉的种子液的制备方法为:在无菌条件下,采用土豆培养基在35~40℃条件下培养24h,制得米曲霉种子液,要求种子液的有效活菌数≥109cfu/mL。
(2)枯草芽孢杆菌种子液制备方法为:在无菌条件下,采用LB培养基在28~32℃条件下培养24h,制得液体芽孢杆菌种子液,要求种子液的有效活菌数≥109cfu/mL。
实施例1
一种高值化利用无名葵的工艺,具体工艺步骤包括:
(1)超声波辅助酶解:将无名葵原料清除杂质后粉碎,过80-150目筛,然后加入无名葵原料质量比10倍的纯净水,在40~60℃条件下浸泡50min;加入中草药原料质量比2.5%的纤维素酶、1.5%的非淀粉多糖酶,在超声波辅助提取10min,超声波功率为250~400W,然后静置30min以上;
(2)好氧发酵:向酶解后的无名葵提取液中继续加入10倍纯净水,然后加入发酵液质量比4%葡萄糖、0.4%硫酸铵、0.15%磷酸二氢钾、0.01%硫酸镁,搅拌均匀;向发酵液中接种米曲霉和枯草芽孢杆菌,接种量分别为发酵液体积比的2.5%、4%;控温28~35℃进行好氧发酵,摇床转速100~150r/min,发酵时间18h;
(3)厌氧发酵:好氧发酵结束后接种发酵液质量比2%的乳酸杆菌,控温28~35℃进行厌氧发酵,摇床转速100~150r/min,发酵时间12h,发酵结束后固液分离得无名葵提取液。
实施例2
一种高值化利用无名葵的工艺,具体工艺步骤包括:
(1)超声波辅助酶解:将无名葵原料清除杂质后粉碎,过80-150目筛,然后加入无名葵原料质量比10倍的纯净水,并加入无名葵质量比3%的玛卡,在40~60℃条件下浸泡50min;加入中草药原料质量比2.5%的纤维素酶、1.5%的非淀粉多糖酶,在超声波辅助提取10min,超声波功率为250~400W,然后静置30min以上;
(2)好氧发酵:向酶解后的无名葵提取液中继续加入10倍纯净水,然后加入发酵液质量比4%葡萄糖、0.4%硫酸铵、0.15%磷酸二氢钾、0.01%硫酸镁,搅拌均匀;向发酵液中接种米曲霉和枯草芽孢杆菌,接种量分别为发酵液体积比的2.5%、4%;控温28~35℃进行好氧发酵,摇床转速100~150r/min,发酵时间18h;
(3)厌氧发酵:好氧发酵结束后接种发酵液质量比2%的乳酸杆菌,控温28~35℃进行厌氧发酵,摇床转速100~150r/min,发酵时间12h,发酵结束后固液分离得无名葵提取液。
实施例3
一种高值化利用无名葵的工艺,具体工艺步骤包括:
(1)超声波辅助酶解:将无名葵原料清除杂质后粉碎,过80-150目筛,然后加入无名葵原料质量比10倍的纯净水,并加入无名葵质量比5%的玛卡,在40~60℃条件下浸泡50min;加入中草药原料质量比2.5%的纤维素酶、1.5%的非淀粉多糖酶,在超声波辅助提取10min,超声波功率为250~400W,然后静置30min以上;
(2)好氧发酵:向酶解后的无名葵提取液中继续加入10倍纯净水,然后加入发酵液质量比4%葡萄糖、0.4%硫酸铵、0.15%磷酸二氢钾、0.01%硫酸镁,搅拌均匀;向发酵液中接种米曲霉和枯草芽孢杆菌,接种量分别为发酵液体积比的2.5%、4%;控温28~35℃进行好氧发酵,摇床转速100~150r/min,发酵时间18h;
(3)厌氧发酵:好氧发酵结束后接种发酵液质量比2%的乳酸杆菌,控温28~35℃进行厌氧发酵,摇床转速100~150r/min,发酵时间12h,发酵结束后固液分离得无名葵提取液。
实施例4 不同工艺条件对无名葵提取液营养特性的影响
试验分组:试验分为实施例1-3试验组、对照1组(实施例1好氧发酵前发酵液)、对照2组(实施例1工艺中不含厌氧发酵,其余制备工艺同实施例1)、对照3组(实施例1工艺中不含好氧发酵,其余制备工艺同实施例1)、对照4组(实施例2好氧发酵前发酵液)、对照5组(实施例2工艺中不含厌氧发酵,其余制备工艺同实施例1)、对照6组(实施例2工艺中不含好氧发酵,其余制备工艺同实施例1)、对照7组(实施例3好氧发酵前发酵液)、对照8组(实施例3工艺中不含厌氧发酵,其余制备工艺同实施例1)、对照9组(实施例3工艺中不含好氧发酵,其余制备工艺同实施例1);
测定指标:
(1)黄酮含量的测定采用硝酸铝法;
(2)DPPH清除能力测定:取不同的样品溶液于96微孔板中,分别加入0.4mmol/L DPPH溶液15µL,再加入100µL水,混匀后避光保存30min,置于酶标仪517 nm波长处测定吸光度值。清除率(%)=(1-(A1-A2)/A0)×100。式中:A0为只加DPPH溶液的吸光度;A1为加样品和DPPH溶液的吸光度;A2为无水乙醇代替DPPH溶液吸光度;
(3)·O2 -清除能力测定:取不同浓度的样品溶液于96微孔板中,分别加入0.15 mmol/LNBT溶液50µL、0.46 mmol/L还原型辅酶Ι溶液50µL和0.06 mmol/L PMS溶液50 µL。混匀后于25 ℃水浴5min,置于酶标仪560nm波长处测定吸光度。清除率(%)=(1-(A1-A2)/A0)×100。式中:A0为空白对照实验(水代替多糖溶液)的吸光度;A1为样品实验的吸光度;A2为0.2 mol/L磷酸盐缓冲溶液(pH7.0)代替NBT溶液的吸光度;
(4)·OH清除能力的测定:取不同的样品溶液于96微孔板中,分别加入0.75 mmol/L邻二氮菲50µL、pH7.4的0.2mol/L PBS缓冲液100µL、0.75 mmol/L FeSO4溶液50µL以及体积分数0.01% H2O2溶液50µL。混匀后于37 ℃水浴下放置30 min,在536 nm波长处检测吸光度。清除率(%)=(1-(A1-A2)/A0)×100。式中:A0为蒸馏水代替样品的吸光度;A1为样品反应后的吸光度;A2为样品和蒸馏水替代H2O2的吸光度;

上述试验结果表明,微生物好氧发酵和厌氧发酵的二步发酵工艺显著提升了无名葵提取液中的黄酮含量和抗氧化能力,尤其是无名葵提取过程中玛卡的加入显著提升了产品的抗氧化能力。综合比较,以实施例2为本发明最佳实施例。
以上实施例仅用于说明本发明的技术方案,而非对其进行限制;尽管参照前述实施例对被发明进行了详细的说明,但对于本领域的普通技术人员来说,依然可以对前述实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换;而对这些修改或者替换,并不使相应技术方案的本质脱离本发明所要求保护的技术方案的精神和范围。
Claims (10)
1.一种高值化利用无名葵的工艺,其特征在于所述无名葵通过超声波辅助酶解和复合微生物发酵工艺制备,其中所述超声波辅助酶解采用纤维素酶和蛋白酶,复合微生物发酵包括好氧发酵和厌氧发酵二步发酵。
2.根据权利要求1所述的一种高值化利用无名葵的工艺,其特征在于所述超声波辅助酶解工艺具体包括如下步骤:将无名葵原料清除杂质后粉碎,过80-150目筛,然后加入无名葵原料质量比10倍的纯净水,在40~60℃条件下浸泡40~60min;加入中草药原料质量比2~3%的纤维素酶、1~2%的非淀粉多糖酶,在超声波辅助提取10min,超声波功率为250~400W,然后静置30min以上。
3.根据权利要求1所述的一种高值化利用无名葵的工艺,其特征在于所述好氧发酵工艺具体包括如下步骤:向酶解后的无名葵提取液中继续加入10倍纯净水,然后加入发酵液质量比3~5%葡萄糖、0.3~0.5%硫酸铵、0.1~0.2%磷酸二氢钾、0.01%硫酸镁,搅拌均匀;向发酵液中接种米曲霉和枯草芽孢杆菌,接种量分别为发酵液体积比的2~3%、3~5%;控温28~35℃进行好氧发酵,摇床转速100~150r/min,发酵时间12~24h。
4.根据权利要求3所述的一种高值化利用无名葵的工艺,其特征在于所述好氧发酵过程中发酵罐通风量为0.1~0.2m3/m3.min。
5.根据权利要求3所述的一种高值化利用无名葵的工艺,其特征在于所述米曲霉的种子液的制备方法为:在无菌条件下,采用土豆培养基在35~40℃条件下培养24h,制得米曲霉种子液,要求种子液的有效活菌数≥109cfu/mL。
6.根据权利要求1所述的一种高值化利用无名葵的工艺,其特征在于所述枯草芽孢杆菌种子液制备方法为:在无菌条件下,采用LB培养基在28~32℃条件下培养24h,制得液体芽孢杆菌种子液,要求种子液的有效活菌数≥109cfu/mL。
7.根据权利要求1所述的一种高值化利用无名葵的工艺,其特征在于所述厌氧发酵工艺包括如下步骤:好氧发酵结束后接种发酵液质量比2%的乳酸杆菌,控温28~35℃进行厌氧发酵,摇床转速100~150r/min,发酵时间12~24h,发酵结束后固液分离得无名葵提取液。
8.根据权利要求1所述的一种高值化利用无名葵的工艺,其特征在于所述超声波辅助酶解时加入无名葵质量比3~5%的玛卡。
9.按权利要求1-8所述工艺制备的无名葵提取液。
10.权利要求9所述无名葵提取液在保健、食品、医药、保鲜等领域中的应用。
