Abstract
本发明公开了一种酒制黄精多糖及其在脾虚免疫功能调节方面的用途。该酒制黄精多糖的平均分子量为2.78×103Da~1.65×104Da;其单糖组成及比例为:鼠李糖(1.82~2.91%)、甘露糖(5.29~6.16%)、半乳糖(76.51~79.44%)以及半乳糖醛酸(13.43~14.66%)。通过动物实验,明确了该酒制黄精多糖能够调节脾虚免疫功能,且较黄精生品多糖功效更强。这一特点可使其应用于增强调节脾虚免疫功能的药品或保健品,且可供临床实际需要进行选择该酒制黄精多糖。
Description
技术领域
本发明涉及一种酒制黄精多糖及其医药方面的用途。
背景技术
中医理论认为“脾主运化而归属于土,为后天之本,气血生化之源”。李杲《脾胃论》“百病皆由脾胃衰而生”,可见脾胃虚弱是疾病发生的基础。脾为后天之本、脾为之卫等有关脾胃中医理论反映了脾与机体防御机能的关系。近年来基础免疫学和临床免疫学研究证明脾功能状况与人体免疫密切相关,免疫系统和免疫功能的改变是脾虚证本质研究的重要内容之一。脾虚证发生涉及免疫器官、免疫细胞及免疫分子的改变,对特异性免疫及非特异性免疫应答均有一定的影响,用于治疗脾虚证的健脾方药在改善脾虚症状的同时伴随着机体免疫功能的改善或恢复。
黄精为百合科黄精属植物黄精Polygonatum sibiritum Red.的干燥根茎。黄精始载于《名医别录》,其性平、味甘,入脾、肺、肾经,具有补脾益气、养阴润肺、滋阴填髓的功效。明代缪希雍《神农本草经疏》中记载“黄精君,纯得土之冲气,其色正黄,味厚气薄,土位乎中,脾治中焦,故补中,脾土为后天生气之源,故益气”。黄精药用历史悠久,疗效确切,是一味重要的药食两用中药。
中药炮制为中国特有的一门制药技术,中药在用于临床前必须要经过炮制加工,以保证用药的安全性和有效性。由于生黄精具有麻舌、刺激咽喉副作用,所以临床上必须使用炮制品。黄精炮制最早见于南北朝雷敩的《雷公炮炙论》,经过历代的衍变,黄精炮制方法主要有水煮、单蒸、重蒸、加辅料蒸制等,在蒸制程度上要求“九蒸九晒”,黄精炮制方法经过历代的衍变,目前2015版《中国药典》沿用了“酒蒸”或“酒炖”法。汪绂《医林纂要探源》曰:“生黄精实有辛莶之味,戟人喉吻,惟蒸晒久,庶几补养滋肾耳”。可见,黄精炮制后不仅去除了麻舌刺激副作用,更重要的是增强了其临床补益作用。
黄精主要含有多糖、皂苷、黄酮、蒽醌及氨基酸类等成分,其中多糖为黄精中最主要成分。据报道黄精多糖有一系列生物活性,如免疫调节、降血糖、降血脂、抗骨质疏松、抗氧化、抗病毒及保肝等活性,其中免疫调节为黄精最主要的生物活性。
近年来国内外学者对黄精生药材中多糖类成分的提取、分离纯化及活性等方面展开一系列研究。Liufen等从中药黄精中分离得到一种中性糖PD,其活性保留片段B3结构主要为高度分支的果聚糖。刘柳等从黄精中提取分离得到含以半乳糖为主的中性糖、酸性糖和糖蛋白,所分离组分具有促进小鼠脾细胞增殖作用;丁侃等从黄精中分离得到结构以高度分支的半乳甘露聚糖和同型半乳聚糖为主的中性糖组分。刘娜等研究发现黄精粗多糖具抗炎活性,可促进环磷酰胺致免疫抑制小鼠T、B淋巴细胞增殖和巨噬细胞吞噬作用并恢复小鼠血清中IL-2、IL-8等因子水平。Yelithao K等从黄精中分离得到以高度分支的葡甘露聚糖为主的多糖组分,该组分能显著促进巨噬细胞分泌NO能力,对其结构进行衍生化处理后免疫调节活性显著增强。专利文献CN101255199A公开了一种黄精多糖的提取方法,该专利中采用沸水提取乙醇沉淀的方法提取得到黄精多糖提取物,黄精多糖对小鼠细胞免疫、体液免疫和小鼠单核-巨噬细胞免疫功能调节方面具有促进作用。专利文献CN1796418A公开了一种黄精药材中多糖提取方法、不同组分多糖复合物配比及促进小鼠脾细胞增殖,该专利在肿瘤免疫功能低下疾病的辅助治疗药物或作为抗衰老作用的保健品具有重要意义。以上均对黄精生药材中所得黄精多糖进行研究。
从90年代开始,国内学者从黄精酒制工艺、酒制前后小分子类物质成分变化及药理活性等方面对黄精酒制进行了研究。有研究发现黄精酒制后粗多糖收率降低5.7%,总糖含量降低25.75%,还原糖含量增加82%,游离氨基酸增加6个。常亮等研究发现黄精生品含大量黏液质,酒制过程中黏液质被水解为果糖、葡萄糖等单糖。钟凌云、杨云等研究发现黄精酒制后其小分子糖类、5-羟甲基糠醛含量相应增加,该类小分子对健康小鼠和环磷酰胺致免疫抑制小鼠的非特异性免疫及特异性免疫功能均有促进作用;由于具体成分的不同,虽然同属黄精制品,但目前难以确定酒制黄精多糖仍然具有同样的功效。例如,半夏常见炮制品有生半夏、清半夏、姜半夏及法半夏,临床实践已证实不同方法炮制的半夏作用和功效也不相同。专利文献CN108164615A公开了一种黄精多糖的制备提取方法和对环磷酰胺诱导的小鼠免疫功能评价作用,该专利中对黄精进行清蒸加工得到黄精清蒸饮片,采用水提醇沉法得到黄精多糖,该多糖对正常小鼠脾细胞增殖有促进作用,另对环磷酰胺诱导小鼠脾脏、胸腺指数,胸腺细胞体外增殖能力,小鼠巨噬细胞的吞噬功能和IL-6和TNF-α分泌的能力有促进作用,从而在免疫缺陷病和肿瘤等疾病的治疗方面具有实际应用价值。以上仅对黄精酒制后小分子类物及清蒸后黄精多糖免疫调节作用进行研究,但并不涉及酒制黄精多糖的提取分离、理化性质及相关功效研究。
综上,尽管国内外有黄精多糖成分的提取、分离纯化、结构解析及活性等方面的研究,但是这些研究主要集中于对黄精生药材中多糖提取分离和免疫活性评价。黄精临床应用形式以酒制品为主,历代医家认为黄精具有重要的补脾益气作用,但黄精酒制前后多糖类成分提取、理化性质及是否仍对脾虚免疫调节作用未见有深入研究。
发明内容
本发明首次深入研究分析了黄精酒制前后多糖类成分提取、理化性质及其药用功效,明确了酒制黄精多糖的成分及其在增强调节脾虚免疫功能方面的效用。具体来说:
一种酒制黄精多糖的制备方法,包括以下步骤:
1)将黄精酒制品饮片粉碎,过100目筛,按照体积比加入10~15倍量水,70~90℃热水搅拌提取2~3次,每次1~2小时,过滤弃药渣,滤液回收溶剂;
2)将回收的溶剂按照体积比加入4倍量体积无水乙醇进行醇沉,静置24h,过滤弃上清;
3)将过滤所得沉淀加水复溶,用Sevage法脱蛋白,沉淀用3×103Da透析袋透析,透析液冷冻干燥得酒制黄精多糖。
该酒制黄精多糖,其平均分子量为2.78×103Da~1.65×104Da;其单糖组成及比例如下:
通过动物实验,明确了该酒制黄精多糖能够调节脾虚免疫功能,且较黄精生品多糖功效更强。这一特点可使其应用于增强调节脾虚免疫功能的药品或保健品,且可供临床实际需要选择该酒制黄精多糖。

具体实施方式
下面结合附图和实施例对本发明做详细描述。
(一)、黄精生品多糖和酒制品多糖提取
1、黄精酒制品饮片制备:取黄精饮片生品适量,置适宜容器中,按照体积比5:1加入黄酒拌匀闷润10~15h,置高压蒸锅中蒸4~8h,蒸至内外滋润黑色,取出,晾干,得黄精酒制饮片。
2、分别将黄精生品及酒制品饮片粉碎,过100目筛,得黄精生品及酒制品饮片粉末,备用。
3、分别称取黄精生品及酒制品饮片粉末适量,按照体积比加入12倍量水,80℃热水搅拌提取2~3次,每次1~2小时,过滤弃药渣,滤液回收溶剂,按照体积比加入4倍量无水乙醇进行醇沉,静置24h,过滤弃上清,沉淀加水复溶,用Sevage法脱蛋白(正丁醇:二氯甲烷=1:4),沉淀用3×103Da透析袋透析,透析液冷冻干燥得黄精生品粗多糖PSPC和酒制品粗多糖PSPW。
(二)、PSPC和PSPW理化性质比较
1、采用蒽酮-浓硫酸法、考马斯亮蓝法、间羟联苯法等对PSPC和PSPW糖含量、蛋白含量、糖醛酸含量等进行测定,结果如下表1。
注:与PSPC进行比较,aP<0.05表示差异显著,bP<0.01表示差异极显著。
2、采用傅里叶变换红外光谱(FT-IR)对PSPC和PSPW红外光谱特征进行分析,结果如下图1所示。
如图1所示,PSPC和PSPW有类似的红外光谱,均含有多糖的典型吸收峰。两者在3400cm-1附近均有宽的强的吸收峰,3418.02cm-1处为多糖O-H伸缩振动吸收峰。2926.37cm-1处为甲基或次甲基的C-H伸缩振动吸收峰。1645.22cm-1和1738.76cm-1处为C=O键的伸缩振动吸收峰,推断样品中含有糖醛酸。PSPW红外图谱中出现1433.94cm-1吸收峰,推测为N-H弯曲伸缩振动吸收峰,表明黄精多糖为糖蛋白复合物,PSPC中该峰不显著。1030.92cm-1和1254.90cm-1处为吡喃糖特征吸收峰。
3、采用气相色谱法对PSPC和PSPW单糖组成进行测定,如下表2、图2所示。
由结果可知PSPC和PSPW中均检测到糖醛酸,表明两者均为酸性糖。如图2、表2所示,PSPC和PSPW单糖组成和比例发生显著变化,其中PSPC主要为半乳糖(29.63%)、甘露糖(36.10%)、葡萄糖(15.09%)和半乳糖醛酸(10.20%)。PSPW中半乳糖比例最高,达到76.83%,另外还含有甘露糖(5.37%)和半乳糖醛酸(13.5%)。

表2中,“-”表示未检测到。
4、采用HPGPC法对PSPC和PSPW分子量进行测定,结果如下图3所示。
根据图3所示结果,PSPC的平均分子量为4.01×103Da,PSPW的平均分子量为1.42×104Da。
另外,在酒制品多糖提取环节,通过调整工艺参数,分别考察70℃、80℃及90℃热水提取多糖,发现最终得到的PSPW的组分平均分子量范围为2.78×103~1.65×104Da,其单糖组成及比例为:鼠李糖(1.82~2.91%)、甘露糖(5.29~6.16%)、半乳糖(76.51~79.44%)以及半乳糖醛酸(13.43~14.66%)。
(三)、PSPC和PSPW脾虚免疫功能评价
1、脾虚模型建立及给药
造模所用番泻叶水煎液制备方法:称取一定量番泻叶药材,加入12倍量水,煎煮15分钟,煎煮1次,过滤,滤液回收溶剂(45~55℃条件)至浓度为1g/mL。
SPF级雄性昆明小鼠,分为9组。除空白组灌胃生理盐水外,其余各组小鼠均采用灌胃番泻叶水煎液(10g/kg)加饥饱失常法建立脾气虚模型,连续造模15天。从第8天开始,除空白组和模型组灌服生理盐水外,其余各给药组分别灌胃PSPC/PSPW低、中、高剂量水溶液(200mg/mL、400mg/mL和800mg/mL),阳性组灌服香菇多糖片水溶液(100mg/mL),连续给药7天。
2、小鼠体重、胸腺指数及脾脏指数测定,如表3所示。
如表3所示,与空白组相比,模型组小鼠体重及免疫器官指数显著降低,说明脾虚显著影响小鼠体重和免疫器官指数,该模型建立成功。与模型组相比,PSPC、PSPW及LNT组小鼠体重、胸腺指数和脾脏指数均有不同程度的增加,并呈一定的浓度依赖性。其中LPS组、PSPW高中低剂量组和PSPC高中剂量组小鼠体重较模型组显著增加,PSPC低剂量组与模型组相比无显著性差异。PSPW高剂量组胸腺指数和脾脏指数与空白组相比无显著性差异,说明PSPW高剂量可恢复小鼠免疫器官指数至正常水平。两者对比发现,PSPW对脾虚小鼠体重和免疫器官恢复作用显著强于PSPC(其中,体重直接反映中医意义上的“脾虚”,可通过胸腺指数和脾脏指数来表征免疫器官的情况)。
注:与正常组相比,aP<0.05表示差异有显著性,bP<0.01表示差异有极显著性;组内进行比较,cP<0.05表示差异有显著性,dP<0.01表示差异有极显著性;与PSPC进行比较,eP<0.05表示差异有显著性,fP<0.05表示差异有极显著性。
3、对小鼠胸腺、脾脏组织病理切片影响,如图4、5所示。
如图4所示,与正常组相比,模型组小鼠胸腺组织的皮质髓质分界不清,皮质变薄,胸腺内淋巴细胞分布松散且数量减少。与模型组小鼠相比,PSPC、PSPW各剂量组和LNT组小鼠胸腺皮质较模型组小鼠变厚,淋巴细胞增多。说明PSPC和PSPW在一定程度上缓解脾虚引起的小鼠免疫器官损伤。如图5所示,与正常组相比,模型组小鼠脾脏组织红髓和白髓分界不清,白髓结构被破坏,淋巴细胞减少。与模型组小鼠相比,PSPC和PSPW各剂量组和LNT组小鼠脾脏红髓和白髓分界相对清晰,淋巴细胞有增多的趋势,其中PSPC和PSPW组高、中剂量组相比于低剂量组均有明显的恢复趋势,且PSPW恢复作用强于PSPC。
4、对外周血中白细胞、淋巴细胞及红细胞影响,如表4所示。
如表4所示,与正常组比较,模型组白细胞、淋巴细胞及红细胞数量显著减少。与模型组比较,PSPC、PSPW及LNT各组小鼠白细胞、淋巴细胞和红细胞数量显著增多,PSPC各剂量组以量效关系分别恢复至正常小鼠白细胞、淋巴细胞和红细胞数量的67.22%-74.43%,66.67-79.32%及85.30%-90.25%,PSPW各剂量组以量效关系恢复至正常组小鼠的73.81-92.90%,75.74-93.46%及75.74-93.46%。可见,PSPC和PSPW对脾虚致白细胞、淋巴细胞和红细胞数量减少有恢复作用,且PSPW组恢复作用显著优于PSPC组。

5、对血清中IL-2、IL-6、TNF-α及IFN-γ分泌量的影响,如图6所示。
由图6A-6D可以看出,与正常组相比,模型组小鼠血清中IL-2、IL-6、TNF-α及IFN-γ因子从92.83±9.69ng/L,112.34±13.11ng/L,132.73±8.75ng/L,150.15±9.43ng/L降低为40.85±4.63ng/L,52.33±4.18ng/L,49.21±8.48ng/L,79.02±5.59ng/L。与模型组相比,LNT、PSPW及PSPC组对IL-2、IL-6、TNF-α及IFN-γ细胞因子水平降低有一定的恢复作用,PSPW和PSPC调节作用具一定的剂量依赖性,PSPC低剂量组对IL-2水平影响无显著性,PSPC及PSPW组间进行比较发现PSPW调节作用优于PSPC。

2.根据权利要求1所述的用途,其特征在于,所述酒制黄精多糖的制备方法包括以下步骤:
1)将黄精酒制品饮片粉碎,过100目筛,按照体积比加入10~15倍量水,70~90℃热水搅拌提取2~3次,每次1~2小时,过滤弃药渣,滤液回收溶剂;
2)将回收的溶剂按照体积比加入4倍量体积无水乙醇进行醇沉,静置24h,过滤弃上清;
3)将过滤所得沉淀加水复溶,用Sevage法脱蛋白,沉淀用3×103Da透析袋透析,透析液冷冻干燥得酒制黄精多糖。
3.一种用于调节脾虚免疫功能的药品或保健品,其特征在于:其有效成分为权利要求1中所述酒制黄精多糖,其指明具有较黄精生品多糖更强的药效。
