Abstract
本发明公开了一种橄榄晶冲剂及其制备方法,属于药物组合物技术领域,所述橄榄晶冲剂以青果为主要原料,辅以含有枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香的中药浸膏和蔗糖制备得到。该制备方法制得的橄榄晶冲剂饮用方便,适于长期饮用,能够有效提高胃蛋白酶活性,增加胃排空率,明显减少胃液分泌量、总酸度和总酸排出量,避免胃酸分泌过多,造成对胃、肠。食道等器官的损伤;同时,该橄榄晶冲剂具有防醉酒作用,对急性酒精性肝损伤具有一定的保护作用,适于长期饮酒,胃功能紊乱,消化不良的人群饮用。

技术领域
本发明属于药物组合物技术领域,具体涉及一种橄榄晶冲剂及其制备方法。
背景技术
随着中国经济的发展,物质水平的逐步提高,现代人的饮食习惯和膳食结构也发生了巨大的变化。现代人饮食习惯和膳食结构普遍存在两个问题:一,摄入热量过多,且摄入食物中肉类过量,蔬菜、谷物和水果摄入不足;第二,饮酒过度。正常情况下,人体摄入食物后,食物刺激胃壁运动,同时引起胃窦粘膜释放胃泌素,胃泌素加速胃酸分泌,同时提高幽门泵活动,幽门括约肌开放,胃运动加强,当胃内压大于十二指肠压时,胃内容物即可进入十二指肠,完成胃内食物排空。但当摄入过多的食物后,会造成胃酸分泌过多,引起消化功能紊乱,造成消化不良的问题,胃酸过多还会伤及胃、十二指肠黏膜,造成胃溃疡或十二指肠溃疡等疾病。而过度的饮酒,酒中的乙醇具有亲脂性和溶脂性能,会导致胃黏膜糜烂和黏膜出血,轻则引起酒后呕吐,反酸,重者出现急性胃黏膜损伤,引起上消化道出血。
针对上述人群,西医治疗多以促胃动力药物为主,如胃复安、吗丁啉、西沙必利等,但其成分单一,具有一定的副作用,不适于较长疗程的治疗。而中药材药源丰富,多数为药食同源,较西药安全有效、毒副作用少、无耐药性。
发明内容
为了克服上述现有技术的缺陷,本发明所要解决的技术问题是:提供一种无便于饮用且可长期饮用的橄榄晶冲剂及其制备方法。
为了解决上述技术问题,本发明采用的技术方案为:一种橄榄晶冲剂,包括以下质量比的组分:青果肉、中药浸膏和蔗糖,所述青果肉、中药浸膏和蔗糖的质量比为90:4:135;
所述中药浸膏由枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香组成;
所述枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香的质量比为1.5:1:2:0.5:2:1:1:1:2:1.5:1:1.5:1:0.5:1:0.5:0.5。
本发明还提供了一种橄榄晶冲剂的制备方法,包括以下步骤:
步骤1、青果肉中加入纯化水,煎煮1h后过滤、浓缩得到青果浓缩液,备用;将蔗糖干燥、粉碎,然后过80目筛,称重备用;
步骤2、将枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香分别粉碎为粒径小于0.85mm的粗粉,加入乙醇,搅拌混匀后浸润,再加入乙醇静置24h,然后进行渗漉,收集渗漉液,对渗漉液进行减压浓缩,回收乙醇,制得中药浸膏;
步骤3、将青果浓缩液与中药浸膏混合均匀,加入蔗糖和95%的乙醇溶液,混合均匀后进行制粒、干燥和整粒,得到橄榄晶冲剂;
其中,所述青果肉、中药浸膏和蔗糖的质量比为90:4:135;所述枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香的质量比为1.5:1:2:0.5:2:1:1:1:2:1.5:1:1.5:1:0.5:1:0.5:0.5。

本发明最关键的构思在于:使用青果肉与不会产生抑制作用的中药浸膏复配,利用中药浸膏中的中药与青果肉协同增效,制备得到橄榄晶冲剂。
本发明提供一种橄榄晶冲剂,包括以下质量比的组分:
青果肉、中药浸膏和蔗糖,所述青果肉、中药浸膏和蔗糖的质量比为90:4:135;
所述中药浸膏由枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香组成;

本发明还提供了一种橄榄晶冲剂的制备方法,具体包括以下步骤:
步骤1、青果肉中加入纯化水,煎煮1h后过滤、浓缩得到青果浓缩液,备用;将蔗糖干燥、粉碎,然后过80目筛,称重备用;
步骤2、将枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香分别粉碎为粒径小于0.85mm的粗粉,加入乙醇,搅拌混匀后浸润,再加入乙醇静置24h,然后进行渗漉,收集渗漉液,对渗漉液进行减压浓缩,回收乙醇,制得中药浸膏;
步骤3、将青果浓缩液与中药浸膏混合均匀,加入蔗糖和95%的乙醇溶液,混合均匀后进行制粒、干燥和整粒,得到橄榄晶冲剂;

进一步的,所述步骤3中95%的乙醇溶液的重量和青果肉的质量比为9:1。
进一步的,所述步骤1为:青果肉中加入纯化水,使饮用水浸没青果肉,保持沸腾状态煎煮1h,得到第一次青果提取液,再向青果肉中加入纯化水使纯化水浸没青果肉,保持沸腾状态煎煮1h,得到第二次青果提取液,再重复一次得到第三次青果提取液;
将第一次青果提取液、第二次青果提取液和第三次青果提取液混合,然后过滤、浓缩得到青果浓缩液。
进一步的,所述步骤3为:将青果浓缩液、食用盐和中药浸膏混合均匀,加入蔗糖和95%的乙醇溶液,混合均匀后进行制粒、干燥和整粒,得到橄榄晶冲剂;
其中,所述食用盐的添加量为青果肉重量的1%。

所述中药浸膏由枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香组成;
所述枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香的质量比为1.5:1:2:0.5:2:1:1:1:2:1.5:1:1.5:1:0.5:1:0.5:0.5。
本发明的实施例二为:
一种橄榄晶冲剂的制备方法,具体包括以下步骤:
步骤1、90kg的青果肉置于提取罐中,加入纯化水,使饮用水浸没青果肉,并高出20cm,保持沸腾状态煎煮1h,将提取液经粗、精过滤器过滤后,利用减压真空系统,抽至储液罐中,得到第一次青果提取液,再向剩余青果肉中加入纯化水使纯化水浸没青果肉,保持沸腾状态煎煮1h,将提取液经粗、精过滤器过滤后,利用减压真空系统,抽至储液罐中,得到第二次青果提取液,再重复一次得到第三次青果提取液;
将第一次青果提取液、第二次青果提取液和第三次青果提取液混合,然后抽至浓缩罐中,控制温度80℃以下和真空度-0.09MPa,对提取液进行浓缩,浓缩后得到青果浓缩液;
将蔗糖干燥、粉碎,然后过80目筛,称重135kg备用;
步骤2、将11.25kg枇杷叶、7.5kg苍耳子、15kg甜杏仁、3.75kg干姜、15kg山楂、7.5kg羌活、7.5kg杜仲叶、7.5kg绿衣枳实、15kg肉桂、11.25kg陈皮、7.5kg麦冬、11.25kg厚朴花、7.5kg薄荷、3.75kg薏苡仁、7.5kg土党参、3.75kg鸡矢藤和3.75kg广藿香分别粉碎为粒径小于0.85mm的粗粉,加入50%的乙醇溶液,搅拌混匀后浸润,再加入50%的乙醇溶液,使液面高出药材面5cm,静置24h,然后装入渗漉罐中进行渗漉,收集渗漉液,对渗漉液进行减压浓缩,回收乙醇,制得中药浸膏,称取4kg中药浸膏;
步骤3、将青果浓缩液、中药浸膏和食用盐混合均匀,加入蔗糖和10kg的乙醇配置成的95%的乙醇溶液,混合均匀后进行制粒、干燥和整粒,得到橄榄晶冲剂,将上述降糖茶检测后分装为10000袋,得到成品橄榄晶冲剂;
其中,所述食用盐的添加量为青果肉重量的1%;
其中,所述步骤3中95%的乙醇溶液的重量和青果肉的质量比为9:1;
所述纯化水为将饮用水采用二级反渗透法制备得到,即,将饮用水依次通过石英砂过滤器、活性炭过滤器,保安过滤器和一级反渗透装置、二级反渗透装置,当制得的纯化水电导率、酸碱度、氨达到控制标准时经紫外线灭菌后进入纯化水贮罐贮存备用。
实验例1:
检测实施例2所制得的橄榄晶冲剂对胃肠动力障碍小鼠胃排空的影响:
将小鼠随机分为6组,分别为正常对照组、模型组、阳性对照组、橄榄晶高、中、低剂量组,每组10只小鼠。连续给药7d后,除正常对照组外,其余各组均腹腔注射硫酸阿托品(2mg/kg)复制胃肠动力障碍小鼠模型,20min后每组小鼠均灌胃葡聚糖蓝(BD)2000溶液0.4ml,再20min后收集胃内容物测其吸光度值,并计算胃内色素排空率。
检测实施例2所制得的橄榄晶冲剂对胃液分泌量的影响:
将大鼠随机分为5组,分别为正常对照组、阳性对照组、橄榄晶高、中、低剂量组,每组10只大鼠。连续给药7d后,麻醉大鼠,打开腹腔结扎大鼠幽门部,再由十二指肠给药一次,缝合腹壁,5h后摘取全胃,收集胃内容物,过滤胃液,记录胃液量,酸度计测其pH,计算总酸排出量;并采用ELISA法检测胃蛋白酶的活性。
结果显示:实施例2所制得的橄榄晶冲剂可明显降低胃肠动力障碍小鼠胃内容物的OD值和增加胃内色素排空率(P<0.05或0.01);尚可明显增加正常大鼠胃蛋白酶活性,但胃液分泌量、总酸度和总酸排出量均明显低于正常对照组(P<0.05或0.01)。
实验例2:
检测实施例2所制得的橄榄晶冲剂对醉酒小鼠酒精代谢的影响:
将雄性小鼠,随机分为6组,分别为正常对照组、模型组、阳性对照组、橄榄晶高、中、低剂量组,每组10只小鼠。给药30min后,除正常对照组外,其余各组小鼠按18ml/kg剂量灌胃56度红星二锅头白酒,给酒后1.5h,摘眼球取血,检测血液中乙醇浓度,并检测肝组织中乙醇脱氢酶(ADH)、乙醛脱氢酶(ALDH)活性。
检测实施例2所制得的橄榄晶冲剂对小鼠急性酒精性肝损伤的保护:
将雄性小鼠,随机分为6组,分别为正常对照组、模型组、阳性对照组、橄榄晶高、中、低剂量组,每组10只小鼠。从给药第1d起,每次给药1h后,除正常对照组外,其余各组动物均灌胃56度红星二锅头白酒15ml/kg。连续7d后,检测血清中门冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)活性。
结果显示:实施例2所制得的橄榄晶冲剂可明显延长小鼠的醉酒潜伏期,明显缩短醉酒小鼠的醉酒持续期、降低醉酒率和死亡率(P<0.05或0.01)。橄榄晶冲剂可明显降低醉酒小鼠血液中的乙醇浓度,增加醉酒小鼠肝组织中ADH、ALDH的活性,可明显降低急性酒精性肝损伤小鼠血清中AST、ALT的活性(P<0.05或0.01)。
在本试验条件下,可观察到本发明提供的橄榄晶冲剂可明显增加胃排空率和胃蛋白酶活性,但可明显减少胃液分泌量、总酸度和总酸排出量。橄榄晶冲剂具有一定防醉酒作用,并对小鼠急性酒精性肝损伤具有一定的保护作用。
综上所述,本发明提供的橄榄晶冲剂,以青果为主要原料,辅以含有枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香的中药浸膏,通过使用青果肉与中药浸膏配合,青果肉与上述成分制备得到的中药浸膏均无毒副作用,不会对中药浸膏产生抑制作用,其中青果肉与中药浸膏中的枇杷叶、麦冬、绿衣枳实、鸡矢藤、干姜、山楂、厚朴花和广藿香共用,能够有效起到生津助消化的作用,杜仲叶、肉桂、甜杏仁、陈皮、干姜、土党参与青果肉起到养肝护胃,促进酒精代谢的作用,羌活、土党参与山楂、杜仲叶和青果肉的共用,能够有效调节脾胃,上述成分配置所得的橄榄晶冲剂能够有效提高胃蛋白酶活性,增加胃排空率,明显减少胃液分泌量、总酸度和总酸排出量,避免胃酸分泌过多,造成对胃、肠、食道等器官的损伤;同时,该橄榄晶冲剂具有防醉酒作用,对急性酒精性肝损伤具有一定的保护作用,适于长期饮酒,胃功能紊乱,消化不良的人群饮用,且该橄榄晶冲剂使用开水即可冲服,饮用方便。

Claims (6)
1.一种橄榄晶冲剂,其特征在于,包括以下质量比的组分:青果肉、中药浸膏和蔗糖,所述青果肉、中药浸膏和蔗糖的质量比为90:4:135;
所述中药浸膏由枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香组成;
所述枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香的质量比为1.5:1:2:0.5:2:1:1:1:2:1.5:1:1.5:1:0.5:1:0.5:0.5。
2.根据权利要求1所述的橄榄晶冲剂,其特征在于,还包括食用盐,所述食用盐的添加量为青果肉重量的1%。
3.一种橄榄晶冲剂的制备方法,其特征在于,包括以下步骤:
步骤1、青果肉中加入纯化水,煎煮1h后过滤、浓缩得到青果浓缩液,备用;将蔗糖干燥、粉碎,然后过80目筛,称重备用;
步骤2、将枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香分别粉碎为粒径小于0.85mm的粗粉,加入50%的乙醇溶液,搅拌混匀后浸润,再加入50%的乙醇溶液静置24h,然后进行渗漉,收集渗漉液,对渗漉液进行减压浓缩,回收乙醇,制得中药浸膏;
步骤3、将青果浓缩液与中药浸膏混合均匀,加入蔗糖和95%的乙醇溶液,混合均匀后进行制粒、干燥和整粒,得到橄榄晶冲剂;
其中,所述青果肉、中药浸膏和蔗糖的质量比为90:4:135;所述枇杷叶、苍耳子、甜杏仁、干姜、山楂、羌活、杜仲叶、绿衣枳实、肉桂、陈皮、麦冬、厚朴花、薄荷、薏苡仁、土党参、鸡矢藤和广藿香的质量比为1.5:1:2:0.5:2:1:1:1:2:1.5:1:1.5:1:0.5:1:0.5:0.5。
4.根据权利要求3所述的橄榄晶冲剂的制备方法,其特征在于,所述步骤3中95%的乙醇溶液的重量和青果肉的质量比为9:1。
5.根据权利要求3所述的橄榄晶冲剂的制备方法,其特征在于,所述步骤1为:青果肉中加入纯化水,使饮用水浸没青果肉,保持沸腾状态煎煮1h,得到第一次青果提取液,再向青果肉中加入纯化水使纯化水浸没青果肉,保持沸腾状态煎煮1h,得到第二次青果提取液,再重复一次得到第三次青果提取液;
将第一次青果提取液、第二次青果提取液和第三次青果提取液混合,然后过滤、浓缩得到青果浓缩液。
6.根据权利要求3所述的橄榄晶冲剂的制备方法,其特征在于,所述步骤3为:将青果浓缩液、食用盐和中药浸膏混合均匀,加入蔗糖和95%的乙醇溶液,混合均匀后进行制粒、干燥和整粒,得到橄榄晶冲剂;
其中,所述食用盐的添加量为青果肉重量的1%。
