Abstract
本发明涉及蜂产品加工技术领域,公开了一种蜂王浆组合物,按重量份数计,包括以下组分,蜂王浆85‑95份;罗汉果糖2‑5份;水溶性壳聚糖1‑1.5份;甘油2‑4份;铵磷脂1‑3份;乙酰化二淀粉磷酸酯0.5‑2份;蜂花粉1‑10份。本发明具有以下优点和效果:通过在蜂王浆组合物内加入罗汉果糖、水溶性壳聚糖、甘油、铵磷脂、乙酰化二淀粉磷酸酯,能有效提高蜂王浆组合物内有效成分如王浆酸、水溶性蛋白、氨基酸、酶类等的稳定性,延长蜂王浆组合物的贮藏时间,便于在蜂王浆的采收、运输、贮藏、加工及消费者保存和食用过程中保证蜂王浆的质量,发挥其应有的营养保健和医疗作用。

技术领域
本发明属于蜂产品加工领域,特别涉及一种蜂王浆组合物。
技术背景
蜂王浆是5日龄-15日龄工蜂头部营养腺分泌出来的,用于饲喂蜂王及幼虫的一种特殊乳状物质,又称蜂乳,其珍稀名贵,出产奇特,成分复杂,有极强的保健功能和医疗效用,是介于食品与药品之间的纯天然保健食品。
蜂王浆研究结果表明,鲜蜂王浆功效的稳定性与其营养价值、保健功效是正相关,蜂王浆功效成分的稳定性是蜂王浆质量和营养保健作用的外在表现,是衡量鲜王浆好坏的一种有效的评定方法。但蜂王浆是极不稳定的天然产物,如空气对蜂王浆起到氧化作用;水蒸气对蜂王浆有水解作用;光对蜂王浆犹如催化剂,催化还原其醛、酮基团等等。
现有技术中,蜂王浆的低温贮藏方式大体分为两种,一种是冷藏,温度高于冰点温度,冷藏贮藏时,蜂王浆的有效成分仍会随着时间的延长而逐渐减少,冷藏时蜂王浆保存时间较短。一种是冷冻,温度低于冰点温度。冷冻贮藏时,温度较低,蜂王浆的有效成分变化较慢,贮藏时间长,但在解冻过程中体系温度升高,冰晶体消解,产生溶质浓缩和脱水损伤,对贮藏品都会产生品质和鲜活度上的影响,影响蜂王浆内有效成分的稳定和蜂王浆的品质。
发明内容
本发明的目的是提供一种蜂王浆组合物,具有便于低温贮藏、稳定性好的效果。
本发明的上述技术目的是通过以下技术方案得以实现的:一种蜂王浆组合物,按重量份数计,包括以下组分,蜂王浆85-95份;罗汉果糖2-5份;水溶性壳聚糖1-1.5份。
通过采用上述技术方案,蜂王浆组合物包括蜂王浆、罗汉果糖、水溶性壳聚糖,蜂王浆成分复杂,含有水溶性蛋白、氨基酸类等活性成分,这些活性成分具有不少生理功能,如增强免疫力、抗癌、增强消化系统功能等。蜂王浆内的酶类如胆碱酯酶、葡萄糖氧化酶、淀粉酶、脂肪酶、转氨酶和超氧化物歧化酶(SOD)等,这些酶都对人类身体有极其重要的生理功能。
罗汉果糖是一种天然健康性甜味品,易溶于水。水溶性壳聚糖是能溶于水的壳聚糖,罗汉果糖和水溶性壳聚糖分散于蜂王浆组合物内,均能溶于蜂王浆含有的水分中,当蜂王浆组合物冷藏时,罗汉果糖和水溶性壳聚糖两者共同作用,能有效提高蜂王浆中水溶性蛋白、氨基酸和酶类等活性成分的稳定性,延长蜂王浆组合物在冷藏时的贮藏时间。当蜂王浆组合物冷冻时,罗汉果糖和水溶性壳聚糖溶于蜂王浆中的水分内,解冻过程中能明显改善蜂王浆的品质,减小解冻过程对蜂王浆组合物内有效成分的影响,提高蜂王浆组合物解冻后的鲜活度和品质,且解冻后蜂王浆内的有效成分高,达到了便于低温贮藏、稳定性好的效果。
本发明的进一步设置为:按重量份数计,包括罗汉果糖3-5份。
本发明的进一步设置为:按重量份数计,包括水溶性壳聚糖1-1.3份。
本发明的进一步设置为:按重量份数计,还包括乙酰化二淀粉磷酸酯0.5-2份。
通过采用上述技术方案,乙酰化二淀粉磷酸酯用作食品添加剂中的增稠剂或稳定剂,易溶于水,分散于蜂王浆组合物中。当蜂王浆组合物冷冻贮藏时,解冻过程中能提高蜂王浆组合物中糖、水溶性蛋白的稳定性。
本发明的进一步设置为:按重量份数计,还包括甘油2-4份。
通过采用上述技术方案,甘油作为防冻剂,当蜂王浆组合物冷冻贮藏时,解冻过程中甘油能对蜂王浆组合物内的糖、蛋白、功能性脂肪酸的生理活性有良好的保护作用。
本发明的进一步设置为:按重量份数计,还包括铵磷脂1-3份。
通过采用上述技术方案,铵磷脂是一种食品添加剂,能与蜂王浆组合物内的水溶性壳聚糖共同作用,提高蜂王浆组合物内功能性脂肪酸和水溶性蛋白的稳定性。
本发明的进一步设置为:按重量份数计,还包括竹叶抗氧化物0.5-1份。
通过采用上述技术方案,竹叶抗氧化物是一种有独特天然竹香的天然抗氧化剂,有效成分包括黄酮类、内脂类和酚酸类。竹叶抗氧化物分散于蜂王浆组合物内,起抗氧化作用,有利于提高蜂王浆组合物内水溶性蛋白、氨基酸、功能性脂肪酸的稳定性。
本发明的进一步设置为:按重量份数计,还包括糠酸甲酯1-1.5份。
通过采用上述技术方案,糠酸甲酯是一种天然物质,糠酸甲酯分散于蜂王浆组合物内,在蜂王浆冷冻贮藏时,糠酸甲酯能与竹叶抗氧化物起到协同增效的作用解冻过程中对蜂王浆组合物内的功能性脂肪酸和水溶性蛋白起到保护作用。
本发明的进一步设置为:按重量份数计,还包括蜂花粉1-10份。
本发明的进一步设置为:按重量份数计,还包括亚油酸0.1-0.5份。
通过采用上述技术方案,王浆酸又称10-羟基-2-葵烯酸,是从蜂王浆中分离出的一种有机酸,且在自然界中只存在于蜂王浆中,是蜂王浆质量的重要指标之一。蜂王浆本身组分内含有亚油酸,亚油酸是人体必需脂肪酸,能降低血液胆固醇,预防动脉粥样硬化。蜂王浆组合物内还包括0.1-0.5份亚油酸,在蜂王浆组合物低温贮藏时,有利于提高王浆酸的稳定性。
综上所述,本发明具有以下有益效果:
1、蜂王浆组合物包括罗汉果糖和水溶性壳聚糖,不仅能在冷藏时有效提高蜂王浆内水溶性蛋白、氨基酸和酶的稳定性,而且在冷冻贮藏时,解冻过程中有效减少蜂王浆组合物内有效成分的损失,提高冷冻后蜂王浆组合物的品质。
2、甘油作为防冻剂,在蜂王浆组合物冷冻贮藏时,解冻过程中对蜂王浆组合物内的功能性脂肪酸、水溶性蛋白起到良好的保护作用。
3、竹叶抗氧化物起抗氧化的作用,且能与糠酸甲酯起到协同增效的作用,在蜂王浆组合物冷冻贮藏后,解冻过程中对功能性脂肪酸起到保护作用
4、蜂花粉分散于蜂王浆组合物内,增强蜂王浆组合物的营养;
5、亚油酸作为人体必需氨基酸,降低血液胆固醇,预防动脉粥样硬化,既能进一步增强蜂王浆组合物的功能性和营养性,且在蜂王浆组合物冷藏时,亚油酸还能起到提高王浆酸稳定性的作用。

实验部分
主要试剂:Folin酚试剂、10-羟基-2-葵烯酸标准品、盐酸(优级纯)、浓硫酸(分析纯)、无水乙醇、亚铁氰化钾(分析纯)、氢氧化钠(分析纯)、氢氧化钠(分析纯)、2,6-二氯靛酚(分析纯)、硫酸铜(分析纯)、抗坏血酸、邻苯三酚溶液(分析纯)、中性甲醛(分析纯)、甲醇(色谱纯)、牛血清蛋白标准品。
主要仪器:高效液相色谱仪、恒温烘箱、离心机、高速冷冻离心机、酸度测定仪、紫外分光光度计、电子天平、隔膜真空泵、超纯水设备、电冰箱。
蜂王浆氨基酸总量的测定方法:采用双指示甲醛滴定法,蛋白质经水解或酶解可由大分子变成小的蛋白质分子,如水解后的中间产物肽、胨等,最后彻底水解成为氨基酸,氨基酸是构成蛋白质最基本的物质。水解后的蛋白质和水解前的蛋白质在物理特性、化学结构以及被吸收消化的程度上是很不同的,其差别与水解的程度有密切的关系,分析氨基酸的含量就可以知道水解程度,进一步评价食品的营养价值。具体方法是,称取蜂王浆组合物约3.00g两份,分别置于250mL三角瓶中,加水50mL。一份加入中性红指示剂2-3滴,用0.100N的氢氧化钠滴定至终点(由红色变琥珀色),记录用量,另一份加入麝香草酚酞3滴和中性甲醛20mL,以0.100N氢氧化钠标准溶液滴定至淡蓝色,记录用量。按照以下公式计算,氨基酸%=N(V2-V1)×0.014/W,其中V1是用中性红作指示剂时碱液的消耗量,V2是用麝香酚酞作指示剂时碱液的消耗量,N是标准碱液当量浓度,W是样品重量。
蜂王浆中王浆酸含量的测定方法:样品处理,10-羟基-2-葵烯酸标准溶液:每毫升含量1mg。取10-羟基-2-葵烯酸0.0250g置于抽真空硫酸干燥器内放置24h的10-羟基-2-葵烯酸,称准至0.0001g,溶于无水乙醇,移入25mL容量瓶中,用无水乙醇定容,过0.45μm滤膜待测。蜂王浆组合物:试样解冻至室温,用玻璃棒充分搅匀,称取0.5g样品,准确至0.0002g,加2mL0.01mol/L盐酸溶液,搅匀加7mL乙醇搅匀,移入50mL容量瓶中,加25mL乙醇,用乙醇定容,立即在超声波浴上混合15min,取出,在3000r/min离心10min,过0.45μm滤膜等测定。
液相色谱条件:色谱柱Kromosil C18(250×4.6mm,5μm),瑞典EKA CHENICALS公司;检测波长:210nm;流动相:甲醇-0.01mol/L盐酸溶液(55+45),过0.45μm滤膜;流速:1mL/min;柱箱温度:40℃。
10-羟基-2-葵烯酸标准曲线绘制方法,准确移取0.5,1.0,2.0,3.0,4.0,5.0mL10-羟基-2-葵烯酸标准溶液至10mL容量瓶中,用无水乙醇定容,分别进样5μL,用蜂面积比值计算,应呈线性。
试样测定方法,吸取试样5μL,注入色谱柱,按外标法定量。
蜂王浆中水溶性蛋白的测定方法:标准曲线的绘制,250μg/ml的牛血清白蛋白,取6支普通试管,配成试管1(蒸馏水1ml)、试管2(0.2mL牛血清蛋白、0.8ml蒸馏水)、试管3(0.4ml牛血清蛋白、0.6ml蒸馏水)、试管4(0.6ml牛血清蛋白、0.4ml蒸馏水)、试管5(0.8ml牛血清蛋白、0.2ml蒸馏水)、试管6(1.0ml牛血清蛋白)。然后各加入试剂甲5ml,混合后再室温下放置10min,再各加0.5ml试剂乙,立即混合均匀。30min后,以不含蛋白质的试管1位对照,于650nm波长下测定各试管中溶液的吸光度并记录数据。以蛋白质浓度为横坐标,以吸光度为纵坐标绘制标准曲线。
准确提取蜂王浆组合物样品0.2g,准确到0.0001g,于离心管,加蒸馏水1mL,在4℃,12000r/min离心20min。上清液即为蜂王浆的水溶性蛋白,取普通试管2支,各加入待测溶液1mL,分别加入试剂甲5mL,混匀后放置10min,再各加入试剂乙0.5mL,迅速混匀,室温放置30min,于650nm波长下测定吸光度,并记录结果,根据标准曲线计算出水溶性蛋白的含量。
蜂王浆中超氧化物歧化酶(SOD)的测定方法:用改进的邻苯三酚自氧化法测定蜂王浆中SOD酶的活性,终止剂选择5%的抗坏血酸。其测定原理是:邻苯三酚在碱性条件下,能迅速自氧化,生成带色的中间产物。在自氧化过程的初始阶段,黄色中间物的积累在滞后30S-40S后就与时间成线性关系。中间产物在420nm处有强烈的光吸收,在有SOD存在时由于它能催化生成O2和H2O2,从而阻止了中间物的积累,通过计算就可以求出SOD的活力。
-18℃条件下贮藏:取各实施例及对比例中的蜂王浆组合物,置于-18℃条件下贮藏,每隔8个月取各实施例及对比例中的蜂王浆组合物,解冻后测定氨基酸总量、水溶性蛋白、王浆酸、超氧化物歧化酶,将实验结果在表2-1、表2-2中列出。
4℃条件下贮藏:取各实施例及对比例中的蜂王浆组合物,置于4℃条件下贮藏,45天为贮藏时间,每隔15天测定一次蜂王浆组合物中的总糖、王浆酸、氨基酸总量、超氧化物歧化酶,将实验结果在表3-1、表3-2中列出。

通过表2-1、表2-2、表3-1、表3-2的实验结果可知,蜂王浆组合物在-18℃条件下冷冻贮藏或者在4℃条件下冷藏贮藏时,罗汉果糖、水溶性壳聚糖有利于提高蜂王浆组合物内的氨基酸、水溶性蛋白、王浆酸的稳定性。铵磷脂、竹叶抗氧化物、糠酸甲酯均能提高蜂王浆组合物中氨基酸、水溶性蛋白、王浆酸的稳定性,延长蜂王浆组合物的贮藏时间。

蜂王浆 85-95份;
罗汉果糖 2-5份;
水溶性壳聚糖 1-1.5份。
