Abstract
本发明公开了一种具有补肺健脾益气功效的红粬缓释片,其是以黄酒酿造后剩余的红粬醪糟与经典中药药对——黄芪、党参为主要原料,经红粬匀浆、脱水、药对处理、填料入罐、封罐发酵、酶解、喷雾干燥、低温研磨、辅料处理、压片成型而制得。本发明所得产品能够为人体提供充足的蛋白质、短肽和人体必须的氨基酸,并具有醒脾开胃、滋阴养血、养气定神之功效,可协同发挥补肺健脾益气的功效。

技术领域
本发明属于保健品技术领域,具体涉及一种具有补肺健脾益气功效的红粬缓释片。
背景技术
红粬是我国黄酒酿造的一种特殊粬种,其以大米为原料,经接种粬母培养而成。近年来,我国黄酒行业发展迅速,产量历年上升,这也使酿造后剩余的副产物红粬醪糟产量大量攀升。据统计报道,我国年产酒糟量约在3000万t以上,并且仍以逐年递增的趋势发展,然而,大部分的醪糟未被再利用,而是直接排放到环境中,仅有少部分作为动物饲料,这样不仅浪费了大量的资源,而且对环境造成了巨大的压力。因此,如何实现废弃红粬醪糟的有效利用是一个亟待解决的问题。
红粬醪糟中含丰富营养物质,以干物质计算,醪糟中含有的粗蛋白质含量高达30-40.5%,且氨基酸种类齐全,其中人体必需氨基酸的含量占氨基酸总量高达36.60%,其氨基酸组成接近FAO/WHO及美国科学院提出的“2~5岁儿童的氨基酸模式”,因此,可将醪糟开发为良好的氨基酸及蛋白质补充剂。
黄芪和党参作为中药经典药对,常常一起使用以增强补气效果。党参是常用的补气中药,其性平、味甘,入脾、肺经,具补中益气、和胃养血等功效,最宜用于平素倦怠乏力、精神不振、自觉气短,稍一活动就喘促的气虚患者。而黄芪味甘性温,入脾、肺经,具补气固表利水、养血生肌托毒之功。党参偏于阴而补中,黄芪偏于阳而实表。二药相合,一里一表,一阴一阳,相互为用,其功益彰,共奏补肺健脾补气之功。
银耳性中平,味甘,还富含人体所需的蛋白质、脂肪、多糖氨基酸和多种维生素,有很高的营养价值,并具有滋阴养胃、补脑强心、益气和血、生津止咳的功效。救心菜是中国农科院利用传统稀有野菜费菜与景天三七杂交而成。每100克食用部分含蛋白质2.1克、脂肪0.7克、碳水化合物8克、粗纤维1.5克、胡萝卜素2.8毫克、维生素B2 0.31毫克、维生素B10.05毫克、烟酸0.9毫克、维生素C 95毫克、钙500毫克、磷29毫克、铁3.2毫克,属于典型的高钙低脂食物,其主要功效成分为景天庚糖、齐墩果酸、谷甾醇、蛋白质和有机酸等,具有宁心平肝、安神补血、清热解毒、止血化瘀的功效。
发明内容
本发明的目的在于提供一种具有补肺健脾益气功效的红粬缓释片。
为实现上述目的,本发明采用如下技术方案:
一种具有补肺健脾益气功效的红粬缓释片,其制备方法包括以下步骤:
1)红粬匀浆:将酿造好的黄酒过滤,取剩下的红粬醪糟均匀打碎,滤去谷壳;
2)脱水:将步骤1)处理后的醪糟放入卧式螺旋卸料沉降离心机,脱水至形成含水量70%的泥状物;
3)药对处理:按经典中药药对黄芪与党参的重量比为5:2,分别称取黄芪和党参,并将黄芪、党参洗净,切薄片,干燥至含水量10-15%,贮存备用;
4)填料入罐:在陶罐中按一层醪糟、一层黄芪、一层党参的方式间隔铺好,每层铺设的高度为5cm,再在最上面加铺一层5-7cm厚的醪糟;
5)封罐发酵:将陶罐密封,隔绝空气,于10-15℃下低温发酵10d,然后开罐,弃除最上端的一层醪糟,再将罐内药对和醪糟分离备用;
6)酶解:在步骤5)所得醪糟内加入其重量0.3%的胃蛋白酶,充分搅拌30min后静止1h,使醪糟内的蛋白质分解为易于人体吸收的短肽;再在搅拌条件下在酶解产物中加入醪糟重量0.5%的葡萄糖,90-100℃搅拌反应30min;
7)喷雾干燥:将步骤6)所得酶解液进行喷雾干燥,得醪糟粉;
8)低温研磨:将步骤5)所得药对真空干燥至含水量5%,然后低温研磨成200目的药对微粉;
9)辅料处理:将银耳洗净,干燥,低温研磨成200目的银耳微粉;将救心菜洗净,干燥,低温研磨成200目的救心菜微粉;
10)压片成型:将步骤7)-9)中所得醪糟粉、药对微粉、银耳微粉、救心菜微粉混合,并加入海藻酸钠、壳聚糖及赋形剂葛根淀粉,混合均匀后,经压片制得所述红粬缓释片;各物质的用量按重量份计为:醪糟粉3份、药对微粉2份、银耳微粉3份、救心菜微粉2份、海藻酸钠0.2-0.5份、壳聚糖0.1-0.3份、葛根淀粉0.2-0.5份。
黄芪、党参为中医经典药对,为益气、补肺健脾益气要药。本发明将黄芪、党参与红粬醪糟共同进行发酵,可使黄芪中的总黄酮含量和党参中的党参炔苷含量明显升高,药效增强。经发明人检测,发酵后黄芪中毛蕊异黄酮的含量为140±15μg/g,毛蕊异黄酮-7-O-β-D-葡萄糖苷的含量为430±30μg/g,较生黄芪明显提高(生黄芪中毛蕊异黄酮的含量为104±10μg/g,毛蕊异黄酮-7-O-β-D-葡萄糖苷的含量为401±30μg/g);同时,发酵后党参中的党参炔苷含量为0.13m/g,也高于未发酵的党参(未发酵党参中的党参炔苷含量为0.09mg/g)。
本发明经发酵后所得醪糟中含有30%左右的蛋白质,但蛋白质为大分子物质,无法被人体直接吸收。本发明在所得醪糟中加入胃蛋白酶进行酶解,可以将醪糟中所含的蛋白质定向分解成易于人体吸收的短肽,以使产品适宜脾胃虚弱的人群;而加入的少量葡萄糖可与醪糟发生美拉德反应,使所得产品具红粬风味,醇香浓郁,并可促进醪糟中氨基酸类物质发生转化,使产品风味独特甘甜美味。同时,在处理过程中加热到90℃以上,不仅可使醪糟内残留的醇、酸类等具刺激性气味的易挥发物质挥发,保留具有芳香风味的酯类,同时还可使醪糟内原有的根霉、曲霉以及加入的胃蛋白酶等酶菌失活,并可以有效降解醪糟内可能含有的橘青霉素。
本发明针对脾肺气虚症的亚健康患者,以红粬醪糟结合经典中药药对——黄芪、党参,以使所得产品能够为人体提供充足的蛋白质、短肽和人体必须的氨基酸,并具有醒脾开胃消食之功效,且配以银耳、救心菜,利用银耳中含有的银耳多糖具有滋阴润肺的功效,及救心菜中含有的景天苷和谷甾酸齐墩果酸具有滋阴养血、养气定神、提高免疫力之功效,可协同发挥补肺健脾益气的功效。
具体实施方式
一种具有补肺健脾益气功效的红粬缓释片,其制备方法包括以下步骤:

7)喷雾干燥:将步骤6)所得酶解液进行喷雾干燥,得醪糟粉;
8)低温研磨:将步骤5)所得药对真空干燥至含水量5%,然后低温研磨成200目的药对微粉;
9)辅料处理:将银耳洗净,干燥,低温研磨成200目的银耳微粉;将救心菜洗净,干燥,低温研磨成200目的救心菜微粉;
10)压片成型:将步骤7)-9)中所得醪糟粉、药对微粉、银耳微粉、救心菜微粉混合,并加入海藻酸钠、壳聚糖及赋形剂葛根淀粉,混合均匀后,经压片制得所述红粬缓释片;各物质的用量按重量份计为:醪糟粉3份、药对微粉2份、银耳微粉3份、救心菜微粉2份、海藻酸钠0.2-0.5份、壳聚糖0.1-0.3份、葛根淀粉0.2-0.5份。

实施例1
1)红粬匀浆:将酿造好的黄酒过滤,取剩下的红粬醪糟均匀打碎,滤去谷壳;

4)填料入罐:在陶罐中按一层醪糟、一层黄芪、一层党参的方式间隔铺好,每层铺设的高度为5cm,连铺4层,再在最上面加铺一层5cm厚的醪糟;
5)封罐发酵:将陶罐密封,隔绝空气,于15℃下低温发酵10d,然后开罐,弃除最上端的一层醪糟,再将罐内药对和醪糟分离备用;
6)酶解:将胃蛋白酶预先用pH=2的食品级乙酸溶解,然后在步骤5)所得醪糟内加入其重量0.3%的胃蛋白酶,充分搅拌30min后静止1h;再在搅拌条件下在酶解产物中加入醪糟重量0.5%的葡萄糖,90℃搅拌反应30min;
7)喷雾干燥:将步骤6)所得酶解液进行喷雾干燥,得醪糟粉;
8)低温研磨:将步骤5)所得药对真空干燥至含水量5%,然后低温研磨成200目的药对微粉;
9)辅料处理:将银耳洗净,干燥,低温研磨成200目的银耳微粉;将救心菜洗净,干燥,低温研磨成200目的救心菜微粉;
10)压片成型:取3重量份醪糟粉、2重量份药对微粉、3重量份银耳微粉、2重量份救心菜微粉混合,并加入0.3重量份海藻酸钠、0.1重量份壳聚糖及0.2重量份赋形剂葛根淀粉,混合均匀后,经压片制得红粬缓释片。
实施例2

4)填料入罐:在陶罐中按一层醪糟、一层黄芪、一层党参的方式间隔铺好,每层铺设的高度为5cm,连铺4层,再在最上面加铺一层6cm厚的醪糟;
5)封罐发酵:将陶罐密封,隔绝空气,于13℃下低温发酵10d,然后开罐,弃除最上端的一层醪糟,再将罐内药对和醪糟分离备用;
6)酶解:将胃蛋白酶预先用pH=2的食品级乙酸溶解,然后在步骤5)所得醪糟内加入其重量0.3%的胃蛋白酶,充分搅拌30min后静止1h;再在搅拌条件下在酶解产物中加入醪糟重量0.5%的葡萄糖,100℃搅拌反应30min;
7)喷雾干燥:将步骤6)所得酶解液进行喷雾干燥,得醪糟粉;
8)低温研磨:将步骤5)所得药对真空干燥至含水量5%,然后低温研磨成200目的药对微粉;
9)辅料处理:将银耳洗净,干燥,低温研磨成200目的银耳微粉;将救心菜洗净,干燥,低温研磨成200目的救心菜微粉;
10)压片成型:取3重量份醪糟粉、2重量份药对微粉、3重量份银耳微粉、2重量份救心菜微粉混合,并加入0.2重量份海藻酸钠、0.3重量份壳聚糖及0.5重量份赋形剂葛根淀粉,混合均匀后,经压片制得红粬缓释片。

5)封罐发酵:将陶罐密封,隔绝空气,于10℃下低温发酵10d,然后开罐,弃除最上端的一层醪糟,再将罐内药对和醪糟分离备用;
6)酶解:将胃蛋白酶预先用pH=2的食品级乙酸溶解,然后在步骤5)所得醪糟内加入其重量0.3%的胃蛋白酶,充分搅拌30min后静止1h;再在搅拌条件下在酶解产物中加入醪糟重量0.5%的葡萄糖,95℃搅拌反应30min;
7)喷雾干燥:将步骤6)所得酶解液进行喷雾干燥,得醪糟粉;
8)低温研磨:将步骤5)所得药对真空干燥至含水量5%,然后低温研磨成200目的药对微粉;
9)辅料处理:将银耳洗净,干燥,低温研磨成200目的银耳微粉;将救心菜洗净,干燥,低温研磨成200目的救心菜微粉;
10)压片成型:取3重量份醪糟粉、2重量份药对微粉、3重量份银耳微粉、2重量份救心菜微粉混合,并加入0.5重量份海藻酸钠、0.2重量份壳聚糖及0.3重量份赋形剂葛根淀粉,混合均匀后,经压片制得红粬缓释片。
取健康SD大鼠60只,从中随机选取10只作为正常组,其余作为造模组,采用烟熏、气管滴入LPS双因素复合制作慢性柱塞性肺疾病(COPD)肺气虚证大鼠模型,然后将造模成功的大鼠随机分为模型组、药对组、醪糟组、微粉组、试验组(其中,药对组喂食材料为实施例1所制备药对微粉,醪糟组喂食材料为实施例1所制备醪糟粉,微粉组喂食材料为实施例1中银耳微粉与救心菜微粉按3:2配制的混合物,试验组喂食材料为实施例1用于压片制备缓释的混合粉料),每组10只。药对组、醪糟组、微粉组、试验组按照15g/(kg·d)的剂量加入2mL水中进行灌胃,正常组和模型组灌胃给予等量清水,每日1次,连续10d。灌胃期间,除正常组外,其余各组每日继续给予烟熏处理。最后一次给药2h后,将各组大鼠称重后麻醉,剥离胸腺和脾脏进行称重,计算胸腺指数和脾脏指数,计算公式如下:
胸腺(脾脏)指数(g/100g)=胸腺(脾脏)质量/体重×100,
结果见表1。

由表1可见,与单独采用药对微粉、醪糟粉或银耳微粉与救心菜微粉的混合物相比,采用本发明缓释片配方进行饲喂的大鼠胸腺指数和脾脏指数显著增加,证明其可明显改善肺气虚证。
以上所述仅为本发明的较佳实施例,凡依本发明申请专利范围所做的均等变化与修饰,皆应属本发明的涵盖范围。
Claims (5)
1.一种具有补肺健脾益气功效的红粬缓释片,其特征在于:其制备方法包括以下步骤:
1)红粬匀浆:将酿造好的黄酒过滤,取剩下的红粬醪糟均匀打碎,滤去谷壳;
2)脱水:将步骤1)处理后的醪糟脱水至含水量70%;
3)药对处理:按经典中药药对比例,分别称取黄芪和党参,并将黄芪、党参洗净,切薄片,干燥至含水量10-15%,贮存备用;
4)填料入罐:在陶罐中按一层醪糟、一层黄芪、一层党参的方式间隔铺好,再在最上面加铺一层5-7cm厚的醪糟;
5)封罐发酵:将陶罐密封,隔绝空气,于10-15℃下低温发酵10d,然后开罐,弃除最上端的一层醪糟,再将罐内药对和醪糟分离备用;
6)酶解:在步骤5)所得醪糟内加入胃蛋白酶,充分搅拌30min后静止1h,再在搅拌条件下在酶解产物中加入葡萄糖,90-100℃搅拌反应30min;
7)喷雾干燥:将步骤6)所得酶解液进行喷雾干燥,得醪糟粉;
8)低温研磨:将步骤5)所得药对真空干燥至含水量5%,然后低温研磨成200目的药对微粉;
9)辅料处理:将银耳洗净,干燥,低温研磨成200目的银耳微粉;将救心菜洗净,干燥,低温研磨成200目的救心菜微粉;
10)压片成型:将步骤7)-9)中所得醪糟粉、药对微粉、银耳微粉、救心菜微粉混合,并加入海藻酸钠、壳聚糖及赋形剂葛根淀粉,混合均匀后,经压片制得所述红粬缓释片。
2.根据权利要求1所述的红粬缓释片,其特征在于:步骤3)中所述经典中药药对比例为黄芪和党参的重量比为5:2。
3.根据权利要求1所述的红粬缓释片,其特征在于:步骤4)中每层的铺设高度为5cm。
4.根据权利要求1所述的红粬缓释片,其特征在于:步骤6)中胃蛋白酶的加入量为醪糟重量的0.3%,葡萄糖的加入量为醪糟重量的0.5%。
5.根据权利要求1所述的红粬缓释片,其特征在于:步骤10)中各物质的用量按重量份计为:醪糟粉3份、药对微粉2份、银耳微粉3份、救心菜微粉2份、海藻酸钠0.2-0.5份、壳聚糖0.1-0.3份、葛根淀粉0.2-0.5份。
