Abstract
本发明公开了一种消炎镇痛中药组合物及其制备方法和应用，其消炎镇痛中药组合物是由活性组分单独制成或由活性组分与药物制剂常用辅料混合制成外用剂型。其中所述的活性成分是由艾叶、凤仙花、花椒和黑蚂蚁按一定的重量百分数配比制备而成。本发明的消炎镇痛中药组合物采用纯天然的中药为原料，无任何副作用，安全可靠，药材分布广，制备工艺简单、提取溶剂价格低廉，质量稳定可控，易于规模化。加之本发明提供的中药组合物，将艾叶、凤仙花、花椒和黑蚂蚁四味中药组合使用后，发挥了协同增效作用，对骨关节炎症具有明显的镇痛消炎效果，可显著改善骨关节肿痛、屈伸不利等病变，为临床用药提供了新的选择。

技术领域
本发明涉及中药领域，具体涉及一种消炎镇痛中药组合物及其制备方法和应用。
背景技术
艾叶、凤仙花、花椒和黑蚂蚁均具有祛风寒除湿、通经透骨、抗炎镇痛、活血止痛之功效，在我国民间被用于治疗风湿、类风湿性关节炎引起的关节肿痛、屈伸不利等虚损性疑难杂症。另外，艾叶、凤仙花、花椒和黑蚂蚁四味药材还具有抗病毒、平喘、镇咳、祛痰、抗过敏、增强免疫、活血化瘀、利尿解毒、杀虫、防治乙肝、预防糖尿病、延缓衰老等医疗保健功效。
骨关节炎已成为严重危害人类健康及其生活质量的广发疾病，临床表现为关节疼痛、肿胀、变形及功能障碍，严重者导致关节残疾及功能丧失。我国的关节炎患者达1亿以上，且人数在不断增加。目前对骨关节炎没有很好的根治方法，主要采用消炎镇痛药物加以治疗。但抗炎镇痛的药物大多具有副作用，如阿片类药物虽然具有镇痛效果好，但长期服用易成瘾。其他激素类药物易造成人体代谢机能紊乱、并发症等，加之严重的副作用和昂贵的费用，不易被人接收。传统的中药作用慢，镇痛时间短，远远不能满足人们消炎镇痛的需求。外用药物作为一种经皮肤吸收的制剂，不仅能减轻药物的副作用，还集速效、舒适、给药方法简单于一体。这就迫切需要研制一种由纯天然的中药为原料制成的消炎镇痛中药组合物作为治疗骨关节炎的药物。
发明内容
为了解决现有技术中存在的问题，本发明提供一种消炎镇痛中药组合物及其制备方法和应用。
为了实现本发明的目的，本发明采用以下技术方案：
一种消炎镇痛中药组合物，它的活性组分是由下列重量百分数的原料药制成：艾叶10~60%，凤仙花15~60%，花椒10~40%，黑蚂蚁10~40%。
所述的消炎镇痛中药组合物的制备方法：该方法将艾叶、凤仙花、花椒和黑蚂蚁四味药材按重量百分数混合并切碎，并将所述的原料与乙醇按料液体积比1:1~1:10在浓度为40~95%的医用乙醇条件下浸提10~90天，过滤后得乙醇浸提液，即该消炎镇痛中药组合物的活性成分。
进一步，所述的消炎镇痛中药组合物的活性组分单独制成或由消炎镇痛中药组合物的活性组分与药物制剂常用辅料混合制成外用剂型。
进一步，所述的外用剂型为搽剂、洗剂、气雾剂、喷雾剂或酊剂等。
相应的，本发明还提供了消炎镇痛中药组合物在制备治疗骨关节炎的药物中的应用。
与现有技术相比，本发明的有益效果在于：（1）本发明所用药材分布广，制备工艺简单、提取溶剂价格低廉，质量稳定可控，易于规模化。（2）本发明提供的中药组合物，将艾叶、凤仙花、花椒和黑蚂蚁四味中药组合使用后，发挥了协同增效作用，对骨关节炎症具有明显的镇痛消炎效果，可显著改善骨关节肿痛、屈伸不利等病变，为临床用药提供了新的选择。（3）本发明所获得的中药组合物给药方便，经小鼠试验，消炎镇痛疗效快作用显著，无任何副作用，安全可靠。
具体实施方式
实施例1
搽剂的制备：取艾叶10%、凤仙花25%、花椒25%、黑蚂蚁40%四种中药原料混合并切碎，按1:1的料液比例加入浓度为95%的医用乙醇浸泡10天，过滤后即得中药组合物乙醇浸提液，装瓶，即得搽剂。
实施例2
搽剂的制备：取艾叶60%、凤仙花20%、花椒10%、黑蚂蚁10%四种中药原料混合并切碎，按1:10的料液比例加入浓度为40%的医用乙醇浸泡90天，过滤后得乙醇浸提液，装瓶，即得搽剂。
实施例3
搽剂的制备：取艾叶10%、凤仙花60%、花椒10%、黑蚂蚁20%四种中药原料混合并切碎，按1:5的料液比例加入浓度为70%的医用乙醇浸泡60天，过滤后得中药组合物乙醇浸提液，装瓶，即得搽剂。

搽剂的制备：取艾叶30%、凤仙花15%、花椒40%、黑蚂蚁15%，将四种中药原料混合并切碎，按1:7的料液比例加入浓度为50%的医用乙醇浸泡40天，过滤后即得中药组合物乙醇浸提液，装瓶，即得搽剂。
实施例5
洗剂的制备：取艾叶40%、凤仙花35%、花椒15%、黑蚂蚁10%四种中药原料混合并切碎，按1:6的料液比例加入浓度为55%的医用乙醇浸泡80天，过滤后得乙醇浸提液，即该中药组合物的活性成分，在活性成分中加入干油，制成本发明消炎镇痛中药组合物的洗剂。
实施例6
气雾剂的制备：取艾叶50%、凤仙花30%、花椒10%、黑蚂蚁10%四种中药原料混合并切碎，按1:4的料液比例加入浓度为60%的医用乙醇浸泡50天，过滤后得中药组合物乙醇浸提液，即该中药组合物的活性成分，将所得活性成分与适量抗氧剂、抛射剂混合均匀，即得本发明消炎镇痛中药组合物的气雾剂。
实施例7
喷雾剂的制备：取艾叶20%、凤仙花30%、花椒20%、黑蚂蚁30%四种中药原料混合并切碎，按1:9的料液比例加入浓度为80%的医用乙醇浸泡30天，过滤后即得中药组合物乙醇浸提液，即该中药组合物的活性成分，向所得的活性成分中加入常规辅料，按照常规工艺制成本发明消炎镇痛中药组合物的喷雾剂。
实施例8
酊剂的制备：取艾叶20%、凤仙花45%、花椒15%、黑蚂蚁20%，将四种中药原料混合并切碎，按1:3的料液比例加入浓度为65%的医用乙醇浸泡70天，过滤后即得中药组合物乙醇浸提液，分装，即得酊剂。
实施例9
本发明消炎镇痛中药组合物对小鼠巴豆油性耳肿的抑制实验：
取体重20±2克的昆明雄性小鼠56只，分组及给药按表1所述处理，各组动物每日按剂量右耳涂药，连续3 d，与末次给药后30 min每只小鼠右耳2面均匀涂抹0.05 mL巴豆油性致炎剂致炎，致炎2h后将小鼠脱臼致死，剪下两耳，用直径6 mm的打孔器打下同一部位耳片，立即称重。以左右两耳片的重量之差作为肿胀度，取平均数与对照组比较，实验结果见表1。

上述实验结果表明：本发明消炎镇痛中药组合物可抑制巴豆油引起的小鼠耳肿，实施例1组、实施例2组、实施例3组、实施例4组与对照组的效果相比，具有显著差异（P＜0.05）；实施例1组、实施例2组、实施例3组、实施例4组与溶媒组的效果相比，具有显著差异（P＜0.01）。
实施例10
本发明消炎镇痛中药组合物对小鼠醋酸扭体反应的影响实验：
取体重20±2克的昆明小鼠56只，雌雄各半，分组及给药按表2所述处理。各组小鼠腹部去毛每天按剂量涂药，连续给药3 d，于末次给药后30 min 腹腔注射0.6%的冰醋酸0.2mL/只，观察记录15 min 内小鼠扭体反应的次数，取平均数与空白对照组比较，实验结果见表2。

上述实验结果表明：本发明消炎镇痛中药组合物对腹腔注射冰醋酸所致小鼠出现扭体反应有明显的抑制作用。
实施例11
本发明消炎镇痛中药组合物治疗的典型案例：
1. 患者，男，48岁，患关节炎10余年，遇气候变化，关节肿胀，疼痛加剧。用本发明实施例3的药物一日三次，一周后肿胀消失，疼痛感明显减轻。
2.患者，女，63岁，患风湿性关节炎20余年，关节变形，生活不能自理，先后服用过多种药物，时好时坏，用本发明实施例7的药物一日三次，半月后消炎止痛，用药二个月后，能够下地行动，生活能够自理。
3.患者，女，38岁，右腕关节拉伤，红肿疼痛，用本发明实施例8的药物一日三次，当日红肿疼痛症状减轻，三日康复。
4.患者，男，56岁，患风湿性关节炎20余年，行动困难，用本发明实施例4的药物一日三次，半月后消炎止痛，用药一个月后，能自主活动。
5.患者，男，42岁，踝关节频繁扭伤，红肿充血，疼痛难忍，用本发明实施例6的药物一日三次，五日肿胀全部消失，至今一年未见复发。

1.一种消炎镇痛外用中药组合物，其特征在于，它的活性组分是由下列重量百分数的原料药制成：艾叶10～60％，凤仙花15～60％，花椒10～40％，黑蚂蚁10～40％，所述活性组分的制备方法为：将艾叶、凤仙花、花椒和黑蚂蚁按重量百分数混合并切碎，并将所述的原料与乙醇按料液体积比1:1～1:10在浓度为40～95％的医用乙醇条件下浸提10～90天，过滤后，得到中药组合物乙醇浸提液，即该消炎镇痛外用中药组合物的活性组分。
2.根据权利要求1所述的消炎镇痛外用中药组合物的制备方法，其特征在于，活性组分的制备方法是将艾叶、凤仙花、花椒和黑蚂蚁按重量百分数混合并切碎，并将所得的原料与乙醇按料液体积比1:1～1:10在浓度为40～95％的医用乙醇条件下浸提10～90天，过滤后，得到中药组合物乙醇浸提液，即该消炎镇痛外用中药组合物的活性组分。
3.根据权利要求2所述的消炎镇痛外用中药组合物的制备方法，其特征在于将得到的消炎镇痛外用中药组合物的活性组分单独制成或由消炎镇痛外用中药组合物的活性组分与药物制剂常用辅料混合制成外用剂型。
4.根据权要求3所述的消炎镇痛外用中药组合物的制备方法，其特征在于外用剂型为搽剂、洗剂、气雾剂、喷雾剂或酊剂。
5.根据权利要求1所述的消炎镇痛外用中药组合物在制备治疗骨关节炎的药物中的应用。
