Technical Field
The invention relates to the technical field of cosmetics, in particular to a liposome structure, components and a preparation method for realizing skin cleaning and anti-aging.
Background
In recent years, the cosmetic market has developed very rapidly. The cosmetics are products which are applied, sprayed or the like on the surface of a human body such as skin and the like to achieve the purposes of cleaning, maintaining, beautifying and resisting aging, and comprise facial cleanser, essence, facial mask and the like. With the improvement of living standard of people, the requirement of consumers on the efficacy of cosmetics is higher and higher. This has led to active ingredients with high potency being continually explored and used in the development of cosmetic formulations. The existence of two problems in this process limits the effective use of many active ingredients with specific efficacy. Firstly, a plurality of active components, such as vitamin A, vitamin E, coenzyme Q10 and other components with antioxidant and whitening effects are easy to be inactivated under external environment due to self instability or poor compatibility of formula raw materials; secondly, many active substances can not effectively permeate into the stratum corneum of the skin to play the actual effect in the using process of consumers. Under the condition, the liposome technology in the coating system represents important application value due to the unique penetration promoting effect.
The microcapsule is a nano-scale phospholipid bead with a special structure, and the outside of the bead is respectively provided with a hydrophilic end capable of being dissolved in water and a hydrophobic end capable of being dissolved in oil. Therefore, the microcapsules have good permeability and solubility in both aqueous and oily substances. The phospholipid component in the liposome is the same as the cell membrane molecules of our human. The phospholipid membrane is composed of lipophilic and hydrophilic molecules which are arranged oppositely and form a structure which is coiled like an onion. When it reaches the deep part of horny layer, it is gradually damaged like onion skin peeling, so it can quickly penetrate into skin to make skin keep moist for a long time, and the key of its effect lies in the structure of liposome.
With the continuous development and innovation of liposome technology, the research of liposome in dermatology and cosmetology is developing vigorously, and the cosmetics containing liposome have the advantages over the traditional cosmetics in the aspects of penetration aiding, slow release, moisture retention, safety and the like. The microcapsule is applied to cosmetics, and the advantages are mainly shown as follows: the stability of the effective components in the product can be improved, and the effective components are prevented from being oxidized (changed and deteriorated) and volatilized, such as vitamin C microencapsulation and enzyme preparation microencapsulation; solidifying the liquid component microcapsule to facilitate the preparation of the product, or solidifying the liquid product to facilitate the use, storage and transportation; thirdly, the water-insoluble materials can be uniformly dispersed in the water medium; fourthly, various components which are forbidden to be compatible can be isolated in the same product; fifthly, the release speed of the effective components can be controlled, so that the effective components of the product have long effect. At present, cosmetics containing various kinds of microcapsules are common in developed countries, but the research on the aspect of the aspects in China is still in the initial stage, and a perfect market is not formed.
Disclosure of Invention
The invention aims to provide a liposome structure, a liposome component and a preparation method for realizing skin cleaning and anti-aging. The invention adopts the liposome A and the liposome B with two different particle size structures to respectively wrap the skin cleaning composition and the skin anti-aging composition, which respectively act on the surface and the deep layer of the skin, and mutually promote through the mutual synergistic action of various active substances, thereby obtaining remarkable skin cleaning and anti-aging effects.
In order to realize the purpose of the invention, the invention adopts the following technical scheme:
a mixture of liposome for cleaning skin and resisting aging comprises two kinds of liposome A and B with different structures at a weight ratio of 1: 1; the skin cleaning composition comprises a liposome A, a liposome B, a liposome C, a liposome B, a liposome C, a liposome B, a liposome C, a vitamin E, a superoxide dismutase; the microcapsule B is of a single-layer microcapsule structure and is formed by coating a skin anti-aging composition with a wall material, the wall material of the microcapsule B comprises sphingomyelin, cholesterol, fatty acid, phosphatidylcholine and cholesterol sulfate, and the skin anti-aging composition contains glutathione, tartaric acid, vitamin C, retinol and coenzyme Q10.
Preferably, the average particle size of the liposome A is 500 to 2000nm, and the polydispersity is 0.30 to 0.40.
Preferably, the average particle size of the liposome B is 20 to 50nm, and the polydispersity is 0.10 to 0.15.
Preferably, the microcapsule A wall material comprises the following components in parts by weight: 15 parts of lecithin and 85 parts of soybean lecithin; the skin cleansing composition comprises the following components in parts by weight: 30-40 parts of jojoba oil, 25-30 parts of vitamin E, 20-30 parts of superoxide dismutase and 10-20 parts of dipalmitoyl hydroxyproline.
Preferably, the microcapsule B wall material comprises the following components in parts by weight: 45 parts of sphingomyelin, 15 parts of cholesterol, 15 parts of fatty acid, 15 parts of phosphatidylcholine and 10 parts of cholesterol sulfate; the skin anti-aging composition comprises the following components in parts by weight: 10-15 parts of glutathione, 25-30 parts of fruit acid, 35-45 parts of vitamin C, 10-15 parts of retinol and 5-10 parts of coenzyme Q10.
Further, the invention also provides a preparation method of the microcapsule mixture for realizing skin cleaning and anti-aging, which comprises the following steps:
(1) weighing lecithin and soybean lecithin, dissolving in organic solvent, adding jojoba oil, vitamin E, superoxide dismutase and dipalmitoyl hydroxyproline, and evaporating under reduced pressure to remove organic solvent to obtain film. Adding a buffer solution with the pH value of 6.5, mechanically stirring and hydrating for 3h at the temperature of 45-50 ℃ to obtain a suspension, standing for 24h, and performing centrifugal separation to obtain the liposome A.
(2) Dissolving sphingomyelin, cholesterol, fatty acid, phosphatidylcholine and cholesterol sulfate in organic solvent, adding glutathione, fruit acid, vitamin C, retinol and coenzyme Q10, and evaporating under reduced pressure to remove organic solvent to obtain film. Adding a buffer solution with the pH value of 6.5, mechanically stirring and hydrating for 5 hours at the temperature of 40-45 ℃, homogenizing for 1 hour at the temperature of 30-40 ℃, carrying out ultrasonic treatment for 0.5-1 hour, and filtering through a microporous filter membrane to obtain the liposome B.
(3) Weighing the liposome A and the liposome B according to a certain proportion, adding the liposome A and the liposome B into glycerol, mixing and stirring uniformly, filling nitrogen gas, sealing, and refrigerating for storage.
Preferably, the organic solvent is a mixture of chloroform and methanol, wherein the volume ratio of chloroform to methanol is 1: 1.
Preferably, the buffer solution is a phosphate buffer solution with a concentration of 8 mmol/L.
Furthermore, the invention also provides an application of the microcapsule mixture for realizing skin cleaning and anti-aging, in particular an application of the microcapsule mixture in skin cleaning and anti-aging cosmetics.
After the technical scheme is adopted, the invention has the following beneficial effects:
1. the invention provides a liposome mixture for realizing skin cleaning and anti-aging, wherein the active ingredients of a skin cleaning composition comprise jojoba oil, vitamin E, superoxide dismutase and dipalmitoyl hydroxyproline, and the liposome mixture can play roles in resisting allergy and purifying the skin surface; the active ingredients of the skin anti-aging composition comprise glutathione, tartaric acid, vitamin C, retinol and coenzyme Q10, and the skin anti-aging composition has the effects of eliminating wrinkles, improving skin elasticity and resisting aging, and particularly, the vitamin C is used as an in-vivo antioxidant, can oxidize the glutathione and reduce hydrogen peroxide generated by organism metabolism, so when the glutathione is used together with the vitamin C, the effect of removing free radicals can be improved.
2. The microcapsule structure can protect the wrapped nutrient components and provide a sustained-release system, wherein the invention adopts two kinds of microcapsule A and microcapsule B with different particle size structures to respectively wrap the skin cleaning composition and the skin anti-aging composition, the microcapsule A is a multilayer microcapsule structure and can isolate the active components to prevent the active components from being oxidized and volatilized or irritating the skin and increase the stability of the active components, and because of the large particle size structure of the microcapsule A, the microcapsule A can be remained on the surface of the skin, and the larger specific surface is beneficial to the release of the components in the skin cleaning composition and directly acts on the surface of the skin, so that the effective components of the product are highly efficient. Due to the nano-scale particle size structure of the liposome B, the liposome B can smoothly pass through the stratum corneum of the skin, so that the anti-aging composition for the skin enters the deep part of the dermis to release the functional components in the stratum, and the skin is deeply moistened and repaired.
3. The components of the microcapsule B wall material comprise 45 parts of sphingomyelin, 15 parts of cholesterol, 15 parts of fatty acid, 15 parts of phosphatidylcholine and 10 parts of cholesterol sulfate, and the components of the microcapsule wall film formed by the microcapsule B wall material are close to skin lipid components, so that the microcapsule B wall material has good compatibility with skin, has good skin permeation effect and is suitable for delivery through the skin.
4. Due to the small volume of the liposome B, molecules of the outer barrier of the liposome B can be exchanged with molecules of cell membranes, and phosphatidylcholine in the outer wall of the liposome B can be exchanged and substituted, so that the aged cell membranes have a structure rich in phosphatidylcholine, the cells can recover the young activity, and the aging is delayed.
5. The microcapsules a encapsulating the skin cleansing composition act as an abrasive due to their larger microcapsule structure, increasing the cleaning effect on the skin surface.
6. Compared with the prior art, the prepared liposome mixture for realizing skin cleaning and anti-aging can achieve remarkable skin cleaning and anti-aging effects through mutual promotion and mutual synergistic action among various active substances.
Detailed Description
The present invention will be described in further detail with reference to examples, but the embodiments of the present invention are not limited thereto.
Example 1
Weighing 15 parts of lecithin and 85 parts of soybean phospholipid by weight, dissolving the lecithin and the soybean phospholipid in a mixed solution of chloroform and methanol (v: v ═ 1:1), adding 30 parts of jojoba oil, 25 parts of vitamin E, 20 parts of superoxide dismutase and 10 parts of dipalmitoyl hydroxyproline, and removing the organic solvent by reduced pressure evaporation to form a film. Adding phosphate buffer solution with pH of 6.5 and concentration of 8mmol/L, mechanically stirring and hydrating at 50 deg.C for 3 hr to obtain suspension, standing for 24 hr, and centrifuging to obtain microcapsule A. 45 parts of sphingomyelin, 15 parts of cholesterol, 15 parts of fatty acid, 15 parts of phosphatidylcholine and 10 parts of cholesterol sulfate are dissolved in a mixture of chloroform and methanol (v: v ═ 1:1), 10 parts of glutathione, 25 parts of fruit acid, 35 parts of vitamin C, 10 parts of retinol and 5 parts of coenzyme Q10 are added, and the organic solvent is removed by evaporation under reduced pressure to form a film. Adding phosphate buffer solution with pH of 6.5 and concentration of 8mmol/L, mechanically stirring and hydrating at 50 deg.C for 5 hr, homogenizing at 40 deg.C for 1 hr, ultrasonic treating for 1 hr, and filtering with microporous membrane to obtain microcapsule B. Weighing the liposome A and the liposome B according to the weight ratio of 1:1, adding the liposome A and the liposome B into glycerol, mixing and stirring uniformly, filling nitrogen gas, sealing, and refrigerating for storage.
Example 2
Weighing 15 parts of lecithin and 85 parts of soybean phospholipid by weight, dissolving the lecithin and the soybean phospholipid in a mixed solution of chloroform and methanol (v: v ═ 1:1), adding 40 parts of jojoba oil, 30 parts of vitamin E, 30 parts of superoxide dismutase and 20 parts of dipalmitoyl hydroxyproline, and removing the organic solvent by reduced pressure evaporation to form a film. Adding phosphate buffer solution with pH of 6.5 and concentration of 8mmol/L, mechanically stirring and hydrating at 45 deg.C for 3 hr to obtain suspension, standing for 24 hr, and centrifuging to obtain microcapsule A. 45 parts of sphingomyelin, 15 parts of cholesterol, 15 parts of fatty acid, 15 parts of phosphatidylcholine and 10 parts of cholesterol sulfate are weighed and dissolved in a mixed solution of chloroform and methanol (v: v ═ 1:1), 15 parts of glutathione, 30 parts of fruit acid, 45 parts of vitamin C, 10 parts of retinol and 10 parts of coenzyme Q10 are added, and the organic solvent is removed by evaporation under reduced pressure to form a film. Adding phosphate buffer solution with pH of 6.5 and concentration of 8mmol/L, mechanically stirring and hydrating at 45 deg.C for 5 hr, homogenizing at 30 deg.C for 1 hr, ultrasonic treating for 1 hr, and filtering with microporous membrane to obtain microcapsule B. Weighing the liposome A and the liposome B according to the weight ratio of 1:1, adding the liposome A and the liposome B into glycerol, mixing and stirring uniformly, filling nitrogen gas, sealing, and refrigerating for storage.
Structural testing
The particle sizes of the microcapsules A and B were measured by laser light scattering. The scattering angle was 90 ° using a Zeta PALS particle size analyzer. The sample to be tested is put into a transparent cuvette (refractive index 1.33), the temperature is kept for 3min at the temperature of 25.0 +/-0.1 ℃, the sample injection measurement is carried out, and the average particle size (Dz) and the particle size distribution condition (polydispersity PDI) are recorded. The test time for each sample was 180s, and 3 replicates were tested, with the results shown in Table 1.

Effect testing
The mixtures of microcapsules of examples 1 and 2 were used as samples for testing.
1. Skin cleansing test
The subject is a 30-year-old female, has healthy skin and no history of skin sensitivity, and is coated with 3cm × 1cm oily lipstick and eyebrow pencil in a range of 4cm × 5cm inside the left arm of the subject. Samples were applied to the spots and gently rubbed for 1min, and washed with water. The cleaning effect was observed and recorded and analyzed by a digital camera. The significant effect is that the skin oil color is obviously removed, generally the skin oil color is removed by more than 1/2, the ineffective effect is that the skin oil color is not removed, and the result is shown in table 2.

The experimental results show that the microcapsule mixture of the present invention has a remarkable ability to achieve skin cleansing.
2. Skin anti-aging test
(1) Test object
And (3) human body experiments, namely selecting 40 female subjects with the ages of 40-45 years, and testing the change of skin textures of the female subjects before and after the female subjects use the sample, so as to determine the anti-aging effect of the sample.
(2) Laboratory apparatus
A MicroSkin II multifunctional skin mirror image analysis system; skin elasticity tester MPA 580.
(3) Experimental methods
Blank group: the area tested did not use any cosmetic.
Applying a sample on a test area (the same area) cleaned by clean water, cleaning the face after 15 minutes, and testing, wherein the sample is applied 3 times per week, and no sample is used in a blank area; during the experiment, the subject was unable to smear or remove any cosmetic other than the sample being tested at the experimental site. After the subjects continuously used the samples for two weeks and four weeks, the subjects cleaned the experimental sites at the same time each time, and the skin texture before the samples were applied to the experimental sites and in the blank group was measured by the technician using the MicroSkinII multifunctional skin mirror image analysis system. The results are given in Table 3 below.

The skin elasticity of the test sites was measured using the skin elasticity tester MPA580, and the results are shown in table 4.

As can be seen from the test results, the anti-aging effects of the samples of examples 1 and 2 were significant, the average surface wrinkles of the skin were significantly reduced, and the elasticity of the skin was significantly increased, and it was found that the microcapsule mixture of the present invention has significant anti-aging ability of the skin.
It should be noted that those skilled in the art should realize that the addition and the replacement of technical features of the present invention without departing from the technical features and scope of the present invention, and that the technical features and the replacement of technical features of the present invention belong to the protection scope of the present invention.
