Abstract
本发明涉及一种治疗痛风和高尿酸血症的发酵中药组合物,属于中医药领域。所述治疗痛风和高尿酸血症的发酵中药组合物包括以下组分:蛹虫草,葛根,茯苓、淡竹叶、薏米、骨碎补、黑老虎与川芎。本发明以传统中医方剂为基础,结合现代微生物学技术,使用乳酸菌对中药进行发酵处理,得到发酵后的药剂;所述药剂对高尿酸血症和痛风具有很好的治疗效果,具有见效快,无毒副作用的特点;本发明的药剂,采用微生物发酵技术,克服了传统药剂药效慢、药效利用率低等缺陷,疗效比传统中药配方优越。

技术领域
本发明涉及药剂,尤其涉及一种治疗痛风和高尿酸血症的发酵中药组合物。
背景技术
痛风是高尿酸血症的一种并发症,痛风是以血尿酸升高为主要生化指标的病症,与饮食结构、生活方式关系密切。随着国内生活水平提高,饮食结构的变化,糖、脂肪、蛋白质的摄入明显增加,痛风发病日益增多,已经成为一种常见的疾病。在全球范围内,痛风每年400万人的速度递增,在我国尤其严重,痛风的发病率为6.89%,东南沿海甚至高达15.09%。痛风病的发生常伴有四肢及身体不适等症状,并可诱发肾功能不全、高血压、糖尿病、冠心病等综合病情、重症者可引起关节炎症反复发作,疼痛难忍,并形成痛风石造成关节畸形,给患者带来巨大的痛苦,直接影响生活质量,严重者可危及生命。
目前,临床上一般采用秋水仙碱非甾体类抗炎药、糖皮质激素、促肾上腺皮质激素和抑制剂等药物居多,但此类药物对肝肾有较大副作用,不能多用,也无法根除,严重限制了该类药物的广泛使用和长期应用。
中医治疗痛风有着悠久的历史,在长期的临床实践过程中积累了极为丰富的治疗经验。中药大多数是属于多成分的天然药物,一般认为中药药性平和、具有毒副作用小的特点。
现代微生物学研究表明,乳酸菌具有参与细胞免疫应答,减少痛风尤其在急性痛风发作期的炎性反应,减少由于炎症反应造成的局部红肿热痛不良症状的作用。导致痛风疾病的高浓度尿酸主要来自于嘌呤的代谢。乳酸菌可以分解食物中摄取的嘌呤,降低嘌呤的吸收和血清尿酸水平。同时还可以干预嘌呤的代谢通路,减少尿酸的形成。
2016年9月21日公开的中国专利申请公开号为CN105943958A的专利申请中公开了一种治疗痛风的桦褐孔菌复合固体颗粒及其制备方法。该发明由以下原料组成:麦绿素10-30份、桦褐孔菌5-25份、黑木耳4-8份、葛根0.1-0.5份、茯苓0.1-0.5份、淡竹叶0.1-0.3份、薏苡仁0.1-0.3份,猫须草5-15份、白芷1-10份、牛初乳0.2-2份、黄精0.1-0.3份、蛹虫草1-3份、茶树花0.1-0.3份、甜菊糖0.2-1份、山梨糖醇30-50份。
2017年6月30日公开的中国专利申请公开号为CN106902187A的专利申请中公开了一种预防和治疗痛风的袋泡茶,该袋泡茶由以下重量份的原料制成:黄芪6-12份、葛根6-12份、绞股蓝6-12份、茯苓4-10份、三七1-5份、蛹虫草0.5-2份、绿茶2-8份。上述公开的专利有预防和治疗痛风的疗效,但由于传统中药存在药效慢、药效利用率低等缺陷。
发明内容
本发明的目的是提供一种治疗痛风和高尿酸血症的发酵中药组合物,以解决传统中药存在药效慢、药效利用率低的问题。
根据本发明的一个方面,本发明提供的一种治疗痛风和高尿酸血症的发酵中药组合物,包括以下组分:蛹虫草,葛根,茯苓、淡竹叶、薏米、骨碎补、黑老虎虎与川芎。
进一步地,所述药剂包括以下重量份的组分:蛹虫草0.5-3份、葛根4-30份、茯苓5-40份、淡竹叶5-20份、薏米5-30份、骨碎补2-20份、黑老虎2-20份、川芎2-20份。
进一步地,所述药剂包括以下重量份的组分:蛹虫草0.5-2份、葛根6-25份、茯苓8-30份、淡竹叶8-15份、薏米10-25份、骨碎补3.5-15份、黑老虎3.5-15份、川芎3.5-15份。
进一步地,所述药剂包括以下重量份的组分:蛹虫草1份、葛根10份、茯苓12份、淡竹叶10份、薏米15份、骨碎补4份、黑老虎4份、川芎4份。
根据本发明的一个方面,本发明提供的一种治疗痛风和高尿酸血症的发酵中药组合物的制备方法,包括以下步骤:称取蛹虫草,葛根,茯苓、淡竹叶、薏米、骨碎补、黑老虎与川芎,将其混匀并碾磨成粉;将纯净水加入碾磨后的粉中,灭菌,之后接入乳酸菌,在35-40℃下静置培养20-40h;最后将发酵后的产物干燥。
进一步地,经碾磨成粉的粒度为150-200目。
进一步地,灭菌的温度和时间分别为110-130℃、20-40min。
将所述药剂制成片剂、丸剂、颗粒剂、胶囊、口服液中任一。
本发明与现有技术相比,具有以下优点:
1.本发明以传统中医方剂为基础,结合现代微生物学技术,使用乳酸菌对中药进行发酵处理,得到发酵后的药剂;
2.所述药剂对高尿酸血症和痛风具有很好的治疗效果,具有见效快,无毒副作用的特点;
3.本发明的药剂,采用微生物发酵技术,克服了传统药剂药效慢、药效利用率低等缺陷,疗效比传统中药配方优越。

具体实施方式
下面将结合实施例对本发明的技术方案进行清楚、完整地描述,显然,所描述的实施例是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
本发明的治疗痛风和高尿酸血症的发酵中药组合物,包括以下组分:蛹虫草,葛根,茯苓、淡竹叶、薏米、骨碎补、黑老虎与川芎。
下面详细说明一下本发明所用各种原料。
蛹虫草:子囊菌亚门,又名北冬虫夏草、北虫草等。中医认为,虫草入肺肾二经,既能补肺阴,又能补肾阳,主治肾虚,阳痿遗精,腰膝酸痛,病后虚弱,久咳虚弱,劳咳痰血,自汗盗汗等,是唯一的一种能同时平衡、调节阴阳的中药。
葛根:性凉,味辛甘,归脾胃经。主解肌退热,生津,透疹,升阳止泻。用于外感发热头痛、高血压颈项强痛、口渴、消渴、麻疹不透、热痢、泄泻。研究表明葛根对学习记忆障碍有明显的治疗作用,可用于治疗老年性痴呆,智力障碍,记忆力差等病症。
茯苓:性甘、淡,平。归心、肺脾、肾经。利水渗湿,健脾,宁心。用于水肿尿少,痰饮眩悸,脾虚食少,便溏泄泻,心神不安,惊悸失眠。所含茯苓酸具有增强免疫力、抗肿瘤以及镇静、降血糖等的作用。
淡竹叶:性甘、淡,寒。归心、胃、小肠经。清凉,解热,利尿及催产之效。主治:胸中疾热、咳逆上气、吐血、热毒风、止消渴、压丹石毒、消痰、治热狂烦闷、中风失音不语、痛头风、止惊悸、瘟疫迷闷、杀小虫、除热缓脾。
薏米:性凉,味甘、淡。微寒、无毒。归脾、胃、肺、大肠经。有健脾、利尿、清热、镇咳之效。主治风湿身痛,湿热脚气,湿热筋急拘挛,湿痹,水肿,肺萎肺痈,咳吐脓血,喉痹痈肿,肠痈热淋。
骨碎补:性苦、温。归肾、肝经。具有补肾强骨,续伤止痛之功效。主治肾虚腰痛,耳鸣耳聋,牙齿松动,跌扑闪挫,筋骨折伤;外治斑秃,白癜风。
黑老虎:辛;微苦;性温。能行气活血,消肿止痛,治胃病,风湿骨痛,跌打瘀痛。
川芎:辛,温。归肝、胆、心包经。具有活血行气,祛风止痛之功效。用于安抚神经,正头风头痛,症瘕腹痛,胸胁刺痛,跌扑肿痛,头痛,风湿痹痛。
下面通过具体的实施例来阐述本发明,本领域技术人员应当理解的是,这不应被理解为对本发明权利要求范围的限制。以下各实施例的份数为重量份。
实施例1
蛹虫草0.5份、葛根5份、茯苓10份、淡竹叶5份、薏米10份、骨碎补1份、黑老虎1份、川芎1份;
将上述中草药混合均匀后研磨成粒度为150目的粉末,取32g粉末置于1000ml三角瓶中,加入320ml纯净水,121℃灭菌30min,接入乳酸杆菌,其浓度1%,37℃静置培养30h;将发酵后的产物进行喷雾干燥,得到本发明发酵后的颗粒药剂。
实施例2
蛹虫草0.5份、葛根30份、茯苓5份、淡竹叶20份、薏米5份、骨碎补20份、黑老虎2份、川芎20份;
将上述中草药混合均匀后研磨成粒度为160目的粉末,取39g粉末置于1000ml三角瓶中,加入390ml纯净水,128℃灭菌36min,接入乳酸杆菌,其浓度2%,40℃静置培养20h;将发酵后的产物进行喷雾干燥,得到本发明发酵后的颗粒药剂。
实施例3
蛹虫草3份、葛根4份、茯苓40份、淡竹叶5份、薏米30份、骨碎补2份、黑老虎20份、川芎2份;
将上述中草药混合均匀后研磨成粒度为170目的粉末,取42g粉末置于1000ml三角瓶中,加入420ml纯净水,125℃灭菌26min,接入乳酸杆菌,其浓度10%,40℃静置培养35h;将发酵后的产物进行喷雾干燥,得到本发明发酵后的颗粒药剂。
实施例4
蛹虫草0.5份、葛根25份、茯苓8份、淡竹叶15份、薏米10份、骨碎补15份、黑老虎3.5份、川芎15份;
将上述中草药混合均匀后研磨成粒度为180目的粉末,取40g粉末置于1000ml三角瓶中,加入400ml纯净水,122℃灭菌30min,接入乳酸杆菌,其浓度9%,39℃静置培养28h;将发酵后的产物进行喷雾干燥,得到本发明发酵后的颗粒药剂。
实施例5
蛹虫草2份、葛根6份、茯苓30份、淡竹叶8份、薏米25份、骨碎补3.5份、黑老虎15份、川芎3.5份;
将上述中草药混合均匀后研磨成粒度为190目的粉末,取39g粉末置于1000ml三角瓶中,加入390ml纯净水,121℃灭菌28min,接入乳酸杆菌,其浓度6%,38℃静置培养27h;将发酵后的产物进行喷雾干燥,得到本发明发酵后的颗粒药剂。
实施例6
蛹虫草1份、葛根10份、茯苓12份、淡竹叶10份、薏米15份、骨碎补4份、黑老虎4份、川芎4份;
将上述中草药混合均匀后研磨成粒度为200目的粉末,取38g粉末置于1000ml三角瓶中,加入380ml纯净水,120℃灭菌25min,接入乳酸杆菌,其浓度4%,37℃静置培养28h;将发酵后的产物进行喷雾干燥,得到本发明发酵后的颗粒药剂。
实施例7
蛹虫草0.5份、葛根28份、茯苓7份、淡竹叶17份、薏米7份、骨碎补16份、黑老虎3份、川芎18份;
将上述中草药混合均匀后研磨成粒度为185目的粉末,取35g粉末置于1000ml三角瓶中,加入350ml纯净水,115℃灭菌33min,接入乳酸杆菌,其浓度8%,36℃静置培养35h;将发酵后的产物进行喷雾干燥,得到本发明发酵后的颗粒药剂。
实施例8
蛹虫草2.5份、葛根5份、茯苓35份、淡竹叶6份、薏米27份、骨碎补3份、黑老虎18份、川芎3份;
将上述中草药混合均匀后研磨成粒度为175目的粉末,取40g粉末置于1000ml三角瓶中,加入400ml纯净水,110℃灭菌30min,接入乳酸杆菌,其浓度3%,35℃静置培养33h;将发酵后的产物进行喷雾干燥,得到本发明发酵后的颗粒药剂。
实施例9(水提法)
蛹虫草0.5份、葛根28份、茯苓7份、淡竹叶17份、薏米7份、骨碎补16份、黑老虎3份、川芎18份;
取上述中草药粉碎成粗粉,置于容器中,加水浸没药材,浸泡1h后,加热至煮沸,保持微沸0.5h,分离煎出液;药渣煎出2次,至煎液味淡为止,合并各次煎出液,浓缩至相对密度为1.00浸膏。
为阐明本发明的发酵后的药剂治疗高尿酸血症及其并发症,用上述实施例制得的药物进行了下列试验:
1动物实验
1.1试验材料
(1)药物及试剂:
本发明药物为根据实施例3所述原料药重量配比及方法制成的提取物。
秋水仙碱,为云南植物药业有限公司产品。
(2)动物:SD大鼠。
1.2试验方法与结果
对高尿酸血症小鼠血尿酸水平的影响
取昆明种小鼠60只,雌雄皆用,体重18~22g,随机分为生理盐水组、本发明发酵药剂和药剂(水提)低剂量组(4g/kg)、本发明药物中剂量组(8g/kg)、本发明药物高剂量组(16g/kg)、阳性对照组,分别灌服生理盐水、本发明药物和秋水仙碱,每日1次,连续4d。末次给药1h后,除生理盐水组外,其余各组动物腹腔注射次黄嘌呤1mg/kg造成小鼠高血尿酸,生理盐水组注射等容量空白对照液。注射后0.5h,各组动物摘眼球取血,3000r/min离心15min,取血清测血尿酸值。

结论:本发明发酵药剂可以显著降低高尿酸血症小鼠血尿酸水平,且效果明显优于未发酵药剂(水提法)。

Claims (8)
1.一种治疗痛风和高尿酸血症的发酵中药组合物,其特征在于,包括以下组分:蛹虫草,葛根,茯苓、淡竹叶、薏米、骨碎补、黑老虎与川芎。
2.根据权利要求1所述的治疗痛风和高尿酸血症的发酵中药组合物,其特征在于,包括以下重量份的组分:蛹虫草0.5-3份、葛根4-30份、茯苓5-40份、淡竹叶5-20份、薏米5-30份、骨碎补2-20份、黑老虎2-20份、川芎2-20份。
3.根据权利要求2所述的治疗痛风和高尿酸血症的发酵中药组合物,其特征在于,包括以下重量份的组分:蛹虫草0.5-2份、葛根6-25份、茯苓8-30份、淡竹叶8-15份、薏米10-25份、骨碎补3.5-15份、黑老虎3.5-15份、川芎3.5-15份。
4.根据权利要求3所述的治疗痛风和高尿酸血症的发酵中药组合物,其特征在于,包括以下重量份的组分:蛹虫草1份、葛根10份、茯苓12份、淡竹叶10份、薏米15份、骨碎补4份、黑老虎4份、川芎4份。
5.一种根据权利要求1-4中任一项所述治疗痛风和高尿酸血症的发酵中药组合物的制备方法,其特征在于,包括步骤:
称取蛹虫草,葛根,茯苓、淡竹叶、薏米、骨碎补、黑老虎与川芎,将其混匀并碾磨成粉;
将纯净水加入碾磨后的粉中,灭菌,之后接入乳酸菌,在35-40℃下静置培养20-40h;最后将发酵后的产物干燥得到药剂。
6.根据权利要求5所述的治疗痛风和高尿酸血症的发酵中药组合物的制备方法,其特征在于,经碾磨成粉的粒度为150-200目。
7.根据权利要求5所述的治疗痛风和高尿酸血症的发酵中药组合物的制备方法,其特征在于,灭菌的温度和时间分别为110-130℃、20-40min。
8.根据权利要求5所述的治疗痛风和高尿酸血症的发酵中药组合物的制备方法,其特征在于,将所述药剂制成片剂、丸剂、颗粒剂、胶囊、口服液中任一。
