Abstract
本发明公开了一种临床微生物检验用中药消毒剂,其制备原料由以下重量份数的组分组成:黄连15‑25份、白千层叶10‑20份、龙葵10‑20份、粉萆薢10‑20份、荨麻子10‑20份、醡浆草5‑10份、肉桂5‑10份、繁缕5‑10份、楮叶3‑5份、大枫子3‑5份。此外,本发明还提供了一种临床微生物检验用中药消毒剂的制备方法。本发明药物为纯中药制剂,所选药材配伍相宜,并采用现代先进的医药制药技术制成,具有杀菌能力强,作用速度快,稳定性好的显著优点,适应于检验科以及医院其他科室的杀菌消毒,不污染环境,对人和动物无刺激,无毒副作用,是一种值得推广的理想的杀菌消毒药物。

技术领域
本发明属于中药及检验技术领域,具体涉及一种临床微生物检验用中药消毒剂及其制备方法。
背景技术
近年来,随着医学生物学和现代信息技术,特别是医学分子生物学的不断进步,医学微生物学检验技术也取得了显著进步,为指导临床感染的诊断和治疗提供了重要依据。检验科作为医院进行医学检验的科室,检验工作需要在无菌条件下进行,以防止细菌、真菌以及病毒等混入检验标本,影响检验效果,使检验结果不精确,为此,检验科的杀菌消毒工作十分重要。
目前用于临床微生物检验用的消毒剂主要包括含氯消毒剂、过氧化物类消毒剂、醛类消毒剂、季铵盐类消毒剂、含碘消毒剂、乙醇等,但是这些消毒剂均存在弊端,如化学类消毒剂消毒效果好,但是毒性、腐蚀性较大;含碘消毒剂杀菌效果好,但是成本较高;乙醇对细菌芽孢无效。这些弊端制约了上述消毒剂的广泛应用,因此,急需开发出一种消毒效果好又绿色环保的消毒剂。
发明内容
本发明提供了一种临床微生物检验用中药消毒剂,解决了现有技术中化学类消毒剂消毒效果好,但是毒性、腐蚀性较大;含碘消毒剂杀菌效果好,但是成本较高;乙醇对细菌芽孢无效的问题。
本发明的第一个目的是提供一种临床微生物检验用中药消毒剂,其制备原料由以下重量份数的组分组成:黄连15-25份、白千层叶10-20份、龙葵10-20份、粉萆薢10-20份、荨麻子10-20份、醡浆草5-10份、肉桂5-10份、繁缕5-10份、楮叶3-5份、大枫子3-5份。
优选的,所述临床微生物检验用中药消毒剂的制备原料由以下重量份数的组分组成:黄连20份、白千层叶15份、龙葵15份、粉萆薢15份、荨麻子15份、醡浆草5份、肉桂10份、繁缕5份、楮叶4份、大枫子4份。
本发明的第二个目的是提供一种临床微生物检验用中药消毒剂的制备方法,具体按照以下步骤实施:
步骤1,按照重量份数称取黄连15-25份、白千层叶10-20份、龙葵10-20份、粉萆薢10-20份、荨麻子10-20份、醡浆草5-10份、肉桂5-10份、繁缕5-10份、楮叶3-5份、大枫子3-5份;
步骤2,将步骤1中称取的黄连、粉萆薢、荨麻子、繁缕、大枫子混合后粉碎,过60目筛,以超临界二氧化碳循环逆流萃取3h,萃取压力为35Mpa,萃取温度为50℃,得到萃取液;
步骤3,将步骤1中称取的白千层叶、醡浆草混合在一起,往其中加入各组分重量和10倍的水,浸泡60min,然后武火煎开,再文火煎煮30min,过滤,得滤渣和第一次滤液;往滤渣中加入滤渣重量10倍的水,先武火煎开,再文火煎煮30min,过滤,得第二次滤液,合并第一次滤液和第二次滤液,混匀,减压浓缩至相对密度为1.2,得混合浓缩液;
步骤4,将步骤1中称取的龙葵粉碎后往其中加入龙葵重量3倍的石油醚以及与龙葵等重量的乙醇,浸泡3h,浸泡完毕后再往其中加入龙葵重量8倍的水,然后在100℃下循环提取6h,提取完毕后过滤,得第一次龙葵滤液和龙葵滤渣;
往龙葵滤渣中加入龙葵滤渣重量2倍的石油醚、龙葵滤渣重量0.5倍的乙醇以及龙葵滤渣重量5倍的水,然后在100℃下循环提取1.5h,提取完毕后过滤,得第二次龙葵滤液,合并第一次龙葵滤液和第二次龙葵滤液,混匀,减压浓缩除去石油醚、乙醇和水,得到龙葵提取液;
步骤5,将步骤1中称取的肉桂粉碎后过60目筛,得到肉桂粉,将肉桂粉在-20℃下冷冻干燥12h,得冷冻肉桂粉;将冷冻肉桂粉放入盐水中浸泡6h后过滤,得到滤渣,将滤渣蒸馏3h,收集混合蒸汽,将混合蒸汽冷凝后油水分离,得到肉桂油;
步骤6,将步骤1中称取的楮叶粉碎后往其中加入楮叶重量6倍的丙酮溶液浸泡2h,再加热回流提取3次,每次提取3h,合并所有的提取液,过滤,滤液浓缩除去乙醇,得到楮叶提取液;
步骤7,将步骤2中的萃取液、步骤3中的混合浓缩液、步骤4中的龙葵提取液、步骤5中的肉桂油、步骤6中的楮叶提取液混合后搅拌均匀,得到混合物料,然后将混合物料冷冻干燥,得到冻干粉,所述冻干粉即为所述临床微生物检验用中药消毒剂。
优选的,所述步骤4中所用石油醚的沸程规格为60-90℃。
优选的,所述步骤5中所用盐水的质量浓度为2%。
优选的,所述冻干粉能够按照常规方法制成药学上可接受的中药制剂。
优选的,所述步骤7中冷冻干燥的具体操作步骤如下:
将混合物料于-30℃下预冷3h,然后放入冻干机中,在2h内迅速降温至-30℃,待混合物料变为块状结构后,在3h内升温至0℃,然后在0.5h内迅速降温至-30℃,待混合物料变为松散冰状结构后,开启真空系统,30min内使真空度缓慢升至30Pa,然后开启加热系统,使混合物料在4h内缓慢升温至40℃,在5h内维持真空度和温度不变,然后关闭真空系统,使压力缓慢恢复到常压,即得到混合物料的冻干粉。
本发明所用中药药性如下:
黄连:苦,寒,无毒。具有清热燥湿、泻火解毒的功效。用于湿热痞满、呕吐吞酸、泻痢、黄疸、高热神昏、心火亢盛、心烦不寐、血热吐衄、目赤、牙痛、消渴、痈肿疔疮;外治湿疹、湿疮、耳道流脓。黄连对金黄色葡萄球菌、溶血性链球菌、肺炎球菌、脑膜炎双球菌、痢疾杆菌、炭疽杆菌等有抗菌作用,还能抑制流感病毒、乙肝病毒等。体外还能抑制阿米巴原虫、阴道滴虫、锥虫。
白千层叶:味苦,性微寒。具有祛风止痛的功效。用于风湿骨痛、神经痛、肠炎、腹泻、过敏性皮炎、湿疹等症。白千层叶对金黄色葡萄球菌和大肠杆菌有抑制作用。
龙葵:甘、酸,凉。具有清热解毒、化瘀止痛、催乳的功效。用于肠炎、痢疾、肝炎、阑尾炎、产后瘀血腹痛、子宫收缩痛、牙痛、头发早白、乳汁不下、乳腺炎、跌打损伤、疮疡肿毒。龙葵对金黄色葡萄球菌、痢疾杆菌、伤寒杆菌、变形杆菌、大肠杆菌、绿脓杆菌、猪霍乱杆菌均有一定的抑菌作用。
粉萆薢:苦,平。具有利水、祛风、利湿的功效。主治治风湿顽痹、腰膝疼痛、小便不利、淋浊、遗精、湿热疮毒。粉萆薢对支原体具有很好的抑菌作用。
荨麻子:苦、辛,温,有小毒。具有祛风湿、凉血、定痉的功效。用于高血压病。外用治荨麻疹初起,风湿关节炎,毒蛇咬伤,小儿惊风。
醡浆草:酸,寒。具有清热利湿、凉血散瘀、消肿解毒的功效。治泄泻,痢疾,黄疸,淋病,赤白带下,麻疹,吐血,衄血,咽喉肿痛,疔疮,痈肿,疥癣,痔疾,脱肛,跌打损伤,汤火伤。醡浆草能有效抑制黄曲霉毒素。
肉桂:性大热,味辛、甘。治命门火衰、肢冷脉微、亡阳虚脱、腹痛泄泻、寒疝奔豚、腰膝冷痛、经闭症瘕、阴疽、流注、及虚阳浮越、上热下寒。桂皮油有强大杀菌作用,对革兰氏染色刚性菌的效果比阴性者好。
繁缕:辛,热。具有散寒燥湿、解酒毒、化湿消痞、行气温中、开胃消食的功效。用于湿浊中阻、不思饮食、湿温初起、胸闷不饥、寒湿呕逆、胸腹胀痛、食积不消。繁缕对金葡球菌、白葡球菌、大肠杆菌、痢疾杆菌和绿脓杆菌表现出抑菌现象。
楮叶:味甘,性凉。具有凉血止血、利尿解毒的功效。主治吐血、衄血、崩血、金疮出血、水肿、疝气、痢疾、毒疮。现代医学研究证实,楮叶的丙酮提取物对葡萄球菌有抑制作用。
大枫子:辛,热,有毒。具有祛风燥湿、攻毒杀虫的功效。主治麻风。外用治梅疮。疥癣。手背皲裂等症。大枫子对金黄色葡萄球菌和其他球菌有抑制作用。
与现有技术相比,本发明的有益效果在于:
1)本发明中各原料根据其具体性质,采用了不同的方法提取其中有效成分,避免了传统的药物提取多采用高温煎煮的方法,从而导致有效成分被破坏,致使其抑菌活性降低,MIC值升高的问题。
2)本发明药物为纯中药制剂,所选药材配伍相宜,并采用现代先进的医药制药技术制成,具有杀菌能力强,作用速度快,稳定性好的显著优点,适应于检验科以及医院其他科室的杀菌消毒,不污染环境,对人和动物无刺激,无毒副作用,是一种值得推广的理想杀菌药物。
具体实施方式
为了使本领域技术人员更好地理解本发明的技术方案能予以实施,下面结合具体实施例对本发明作进一步说明,但所举实施例不作为对本发明的限定。
下面各实施例中未注明具体条件的试验方法,均按照本领域的常规方法和条件进行,所用的材料若无特殊说明均为市售。
实施例1
一种临床微生物检验用中药消毒剂,其制备原料由以下重量份数的组分组成:黄连15份、白千层叶20份、龙葵20份、粉萆薢10份、荨麻子10份、醡浆草10份、肉桂5份、繁缕10份、楮叶3份、大枫子5份。
具体按照以下步骤实施:
步骤1,按照重量份数称取黄连15份、白千层叶20份、龙葵20份、粉萆薢10份、荨麻子10份、醡浆草10份、肉桂5份、繁缕10份、楮叶3份、大枫子5份;
步骤2,将步骤1中称取的黄连、粉萆薢、荨麻子、繁缕、大枫子混合后粉碎,过60目筛,以超临界二氧化碳循环逆流萃取3h,萃取压力为35Mpa,萃取温度为50℃,得到萃取液;
步骤3,将步骤1中称取的白千层叶、醡浆草混合在一起,往其中加入各组分重量和10倍的水,浸泡60min,然后武火煎开,再文火煎煮30min,过滤,得滤渣和第一次滤液;往滤渣中加入滤渣重量10倍的水,先武火煎开,再文火煎煮30min,过滤,得第二次滤液,合并第一次滤液和第二次滤液,混匀,减压浓缩至相对密度为1.2,得混合浓缩液;
步骤4,将步骤1中称取的龙葵粉碎后往其中加入龙葵重量3倍的石油醚以及与龙葵等重量的乙醇,浸泡3h,浸泡完毕后再往其中加入龙葵重量8倍的水,然后在100℃下循环提取6h,提取完毕后过滤,得第一次龙葵滤液和第一次龙葵滤渣;
往第一次龙葵滤渣中加入龙葵滤渣重量2倍的石油醚、龙葵滤渣重量0.5倍的乙醇以及龙葵滤渣重量5倍的水,然后在100℃下循环提取1.5h,提取完毕后过滤,得第二次龙葵滤液,合并第一次龙葵滤液和第二次龙葵滤液,混匀,减压浓缩除去石油醚、乙醇和水,得到龙葵提取液;
步骤5,将步骤1中称取的肉桂粉碎后过60目筛,得到肉桂粉,将肉桂粉在-20℃下冷冻干燥12h,得冷冻肉桂粉;将冷冻肉桂粉放入质量浓度为2%的盐水中浸泡6h后过滤,得到滤渣,将滤渣置于蒸馏罐中通入水蒸汽蒸馏3h,得到混合蒸汽,将混合蒸汽冷凝后油水分离,得到肉桂油;
步骤6,将步骤1中称取的楮叶粉碎后往其中加入楮叶重量6倍的丙酮溶液浸泡2h,再加热回流提取3次,每次提取3h,合并所有的提取液,过滤,滤液浓缩除去乙醇,得到楮叶提取液;
步骤7,将步骤2中的萃取液、步骤3中的混合浓缩液、步骤4中的龙葵提取液、步骤5中的肉桂油、步骤6中的楮叶提取液混合后搅拌均匀,得到混合物料,然后将混合物料冷冻干燥,得到冻干粉,该冻干粉即为所述临床微生物检验用中药消毒剂。
需要说明的是,步骤4中所用石油醚的沸程规格为60-90℃。
进一步需要说明的是,冻干粉能够按照常规方法制成药学上可接受的中药制剂。
进一步需需要说明的是,步骤7中冷冻干燥的具体操作步骤如下:

实施例2
一种临床微生物检验用中药消毒剂,其制备原料由以下重量份数的组分组成:黄连20份、白千层叶15份、龙葵15份、粉萆薢15份、荨麻子15份、醡浆草5份、肉桂10份、繁缕5份、楮叶4份、大枫子4份。
实施例2的制备方法同实施例1,区别在于将配方改为实施例2的配方。
实施例3
一种临床微生物检验用中药消毒剂,其制备原料由以下重量份数的组分组成:黄连25份、白千层叶10份、龙葵10份、粉萆薢20份、荨麻子20份、醡浆草8份、肉桂8份、繁缕5份、楮叶5份、大枫子3份。

实施例4

实施例5
一种临床微生物检验用中药消毒剂,其制备原料由以下重量份数的组分组成:黄连15份、白千层叶10份、龙葵20份、粉萆薢20份、荨麻子15份、醡浆草10份、肉桂5份、繁缕8份、楮叶5份、大枫子4份。
实施例5的制备方法同实施例1,区别在于将配方改为实施例5的配方。
实施例1-5均制备出了临床微生物检验用中药消毒剂,下面对本发明制备出的中药消毒液进行抑菌实验,以说明实施例1-5的效果。
一、抑菌效果测定
利用实施例1-5制得的中药消毒剂进行抑菌实验,实验菌种为表皮葡萄球菌、白色念珠菌、铜绿假单胞菌、大肠杆菌、肺炎克雷伯菌,抑菌实验的对照组1采用质量浓度为0.5%的碘酊,对照组2采用84消毒液,菌种培养基为牛肉膏蛋白胨培养基。具体实验步骤如下:
(1)制备菌液:将上述五组菌种分别接种于牛肉膏蛋白胨培养基上,在37℃下培养24h,用接种环挑取少许菌体于装有无菌生理盐水的试管中,震荡均匀,制成菌悬液;
(2)分别取实施例1-5制得的中药消毒剂、碘酊以及84消毒液各20mL,然后分别置于无菌培养皿中,得到测试培养皿,备用;
(3)取制成的菌悬液0.1mL分别置于各测试培养皿中,混合均匀,分别标记为实验组和对照组,具体实验结果见表1。

从表1中可以看出,实施例1-5制得的中药消毒剂相比于一般的碘酊和84消毒液来说具有更好的杀菌效果。
二、空气杀菌效果测定
分别在面积约10m2大小的房间,实验组采用实施例1制得的中药消毒剂于50-80℃熏蒸消毒2h;对照组采用紫外消毒等消毒(灯管功率>0.7W/m2,照射2h)。各组在消毒前后分别进行沉降菌落数的检测,根据消毒前后沉降菌落数的差异来评价空气杀菌效果。熏蒸或照射8h、24h后再次分别测定沉降菌落数,评价不同方法空气消毒的延续性效果。具体结果见表2。

从表2可以看出,实施例1制备出的中药消毒剂对空气的净化杀菌作用与紫外线照射杀菌的效果基本一致。
三、皮肤刺激性试验
多次完整皮肤刺激试验:选择体重20-25kg健康白色家兔3只,试验前24h将白色家兔背部脊柱两侧的毛剪掉,去毛范围左、右各约3cm*3cm。次日,将实施例1的中药消毒剂原液0.5mL均匀涂抹于一侧去毛皮肤表面,另一侧去毛皮肤作为空白对照。涂抹4h,用蒸馏水清除残留受试物,每天涂抹一次,连续涂抹14d,必要时剪毛,不损伤皮肤。在每次涂抹后24h观察结果。
一次性破损皮肤刺激试验:选择体重20-25kg健康白色家兔3只,试验前24h将家兔背部脊柱两侧的毛剪掉,去毛范围左、右各约3cm*3cm。次日,先用浓度为75%酒精擦拭消毒去毛部位皮肤,用注射针头在皮区内划一个“井”形的破损伤口,将实施例1的中药消毒剂原液滴于破损部位皮肤上覆盖的4层纱布上,然后用一层油纸覆盖,再用无刺激性胶布固定。另一侧去毛破损皮肤用等量灭菌蒸馏水作为对照,与前作同样处理。敷用4h后,用蒸馏水除去残留实施例1的中药消毒剂原液,分别于去除受试物后1、24、48h,观察皮肤局部反应。
两组皮肤刺激性试验的结论是:皮肤刺激性试验结果表明,实施例1的中药消毒剂对家兔完整皮肤多次刺激试验后,家兔表皮无明显变化,判定为无刺激性;对家兔破损皮肤一次性刺激试验后,家兔表皮无明显变化判定为无刺激性。
需要说明的是,本发明权利要求书中涉及数值范围时,应理解为每个数值范围的两个端点以及两个端点之间任何一个数值均可选用,由于采用的步骤方法与实施例1-5相同,为了防止赘述,本发明的描述了优选的实施例,但本领域内的技术人员一旦得知了基本创造性概念,则可对这些实施例作出另外的变更和修改。所以,所附权利要求意欲解释为包括优选实施例以及落入本发明范围的所有变更和修改。
显然,本领域的技术人员可以对本发明进行各种改动和变型而不脱离本发明的精神和范围。这样,倘若本发明的这些修改和变型属于本发明权利要求及其等同技术的范围之内,则本发明也意图包含这些改动和变型在内。
Claims (7)
1.一种临床微生物检验用中药消毒剂,其特征在于,其制备原料由以下重量份数的组分组成:黄连15-25份、白千层叶10-20份、龙葵10-20份、粉萆薢10-20份、荨麻子10-20份、醡浆草5-10份、肉桂5-10份、繁缕5-10份、楮叶3-5份、大枫子3-5份。
2.根据权利要求1所述的临床微生物检验用中药消毒剂,其特征在于,其制备原料由以下重量份数的组分组成:黄连20份、白千层叶15份、龙葵15份、粉萆薢15份、荨麻子15份、醡浆草5份、肉桂10份、繁缕5份、楮叶4份、大枫子4份。
3.根据权利要求1所述的临床微生物检验用中药消毒剂的制备方法,其特征在于,具体按照以下步骤实施:
步骤1,按照重量份数称取黄连15-25份、白千层叶10-20份、龙葵10-20份、粉萆薢10-20份、荨麻子10-20份、醡浆草5-10份、肉桂5-10份、繁缕5-10份、楮叶3-5份、大枫子3-5份;
步骤2,将步骤1中称取的黄连、粉萆薢、荨麻子、大枫子混合后粉碎,过60目筛,以超临界二氧化碳循环逆流萃取3h,萃取压力为35Mpa,萃取温度为50℃,得到萃取液;
步骤3,将步骤1中称取的白千层叶、醡浆草,繁缕混合在一起,往其中加入各组分重量和10倍的水,浸泡60min,然后武火煎开,再文火煎煮30min,过滤,得滤渣和第一次滤液;往滤渣中加入滤渣重量10倍的水,先武火煎开,再文火煎煮30min,过滤,得第二次滤液,合并第一次滤液和第二次滤液,混匀,减压浓缩至相对密度为1.2,得混合浓缩液;
步骤4,将步骤1中称取的龙葵粉碎后往其中加入龙葵重量3倍的石油醚以及与龙葵等重量的乙醇,浸泡3h,浸泡完毕后再往其中加入龙葵重量8倍的水,然后在100℃下循环提取6h,提取完毕后过滤,得第一次龙葵滤液和龙葵滤渣;
往龙葵滤渣中同时加入龙葵滤渣重量2倍的石油醚、龙葵滤渣重量0.5倍的乙醇以及龙葵滤渣重量5倍的水,然后在100℃下循环提取1.5h,提取完毕后过滤,得第二次龙葵滤液,合并第一次龙葵滤液和第二次龙葵滤液,混匀,减压浓缩除去石油醚、乙醇和水,得到龙葵提取液;
步骤5,将步骤1中称取的肉桂粉碎后过60目筛,得到肉桂粉,将肉桂粉在-20℃下冷冻干燥12h,得冷冻肉桂粉;将冷冻肉桂粉放入盐水中浸泡6h后过滤,得到滤渣,将滤渣蒸馏3h,并收集混合蒸汽,将混合蒸汽冷凝后油水分离,得到肉桂油;
步骤6,将步骤1中称取的楮叶粉碎后往其中加入楮叶重量6倍的丙酮溶液浸泡2h,再加热回流提取3次,每次提取3h,合并所有的提取液,过滤、浓缩,得到楮叶提取液;
步骤7,将步骤2中的萃取液、步骤3中的混合浓缩液、步骤4中的龙葵提取液、步骤5中的肉桂油、步骤6中的楮叶提取液混合后搅拌均匀,得到混合物料,然后将混合物料冷冻干燥,得到冻干粉,所述冻干粉即为所述临床微生物检验用中药消毒剂。
4.根据权利要求3所述的临床微生物检验用中药消毒剂的制备方法,其特征在于,所述步骤4中所用石油醚的沸程规格为60-90℃。
5.根据权利要求3所述的临床微生物检验用中药消毒剂的制备方法,其特征在于,所述步骤5中所用盐水的质量浓度为2%。
6.根据权利要求3所述的临床微生物检验用中药消毒剂的制备方法,其特征在于,将所述冻干粉按照常规方法制成药学上可接受的中药制剂。
7.根据权利要求3所述的临床微生物检验用中药消毒剂的制备方法,其特征在于,所述步骤7中冷冻干燥的具体操作步骤如下:
将混合物料于-30℃下预冷3h,然后放入冻干机中,在2h内迅速降温至-30℃,待混合物料变为块状结构后,在3h内升温至0℃,然后在0.5h内迅速降温至-30℃,待混合物料变为松散冰状结构后,开启真空系统,30min内使真空度缓慢升至30Pa,然后开启加热系统,使混合物料在4h内缓慢升温至40℃,在5h内维持真空度和温度不变,然后关闭真空系统,使压力缓慢恢复到常压,即得到混合物料的冻干粉。
