Abstract
本发明公开了一种抗菌食品保鲜袋方法及其制备,该抗菌食品保鲜袋包括抗菌层;所述抗菌层包括纳米银、纳米二氧化钛、分散剂和抗菌植物精油纳米脂质体。纳米银、纳米二氧化钛和抗菌植物精油纳米脂质体均具有抗菌的功效,加入分散剂混合后,可以大大降低纳米银颗粒团聚程度,使纳米粒子尽可能的分散均匀,提高抗菌食品保鲜袋的抗菌效果。

技术领域
本发明公开了一种抗菌食品保鲜袋,涉及纳米结构材料类技术领域。
背景技术
目前市场上流通的绝大多数食品保鲜袋,并不具备抗菌能力,它在给生活带来便利的同时。也成了一个潜在的污染源,只要有部分食品存有脏污,就会滋生大量细菌,影响人体健康。加上近些年人们生活质量的提高,健康意识的普遍增强,开发一款具备抗菌能力的食品保鲜袋迫在眉睫。
食品保鲜袋抗菌,关键在于抗菌材料。根据化学组成,抗菌材料可以分为天然、无机和有机三大类。天然抗菌材料主要是沸石类矿物,组成为架状结构的水铝硅酸盐,具有纳米孔隙和孔道结构,对于分子尺寸的粒子具有较强的吸附能力和吸附选择,缺点是抗菌能力一般,耐热性差、寿命短,生产条件繁琐,不能批量化生产。无机抗菌性主要有两种,一种是离子型抗菌剂,主要有Ag+、Zn2+等金属离子;另一种是光催化型抗菌剂。有机抗菌剂以有机酸、酚、醇为主要成分,以破坏细胞膜,使蛋白质变性、代谢受阻等为抗菌机理,其优点是杀菌力强、即效好、来源广泛,缺点是毒性大、会产生微生物耐药性、耐热性较差、易迁移等。
纳米银,是利用前沿纳米技术将银纳米化,纳米技术出现,使银在纳米状态下的杀菌能力产生了质的飞跃,极少的纳米银可产生强大的杀菌作用,可在数分钟内杀死650多种细菌,广谱杀菌且无任何的耐药性,能够促进伤口的愈合、细胞的生长及受损细胞的修复,无任何毒性反应,对皮肤也未发现任何刺激反应,这给广泛应用纳米银来抗菌开辟了广阔的前景,是最新一代的天然抗菌剂。纳米银颗粒直接进入菌体与氧代谢酶(-SH)结合,使菌体窒息而死的独特作用机制,可杀死与其接触的大多数细菌、真菌、霉菌、孢子等微生物。经国内八大权威机构研究发现:其对耐药病原菌如耐药大肠杆菌、耐药金葡萄球菌、耐药绿脓杆菌、化脓链球菌、耐药肠球菌,厌氧菌等有全面的抗菌活性;对烧烫伤及创伤表面常见的细菌如金黄色葡萄球菌、大肠杆菌、绿脓杆菌、白色念珠菌及其它G+、G-性致病菌都有杀菌作用;对沙眼衣原体、引起性传播性疾病的淋球菌也有强大的杀菌作用。纳米银颗粒具有超强的渗透性,可迅速渗入皮下2mm杀菌,对普通细菌、顽固细菌、耐药细菌以及真菌引起的较深处的组织感染均有良好的杀菌作用。
纳米二氧化钛是白色疏松粉末,屏蔽紫外线作用强,具有良好的分散性、很高的化学稳定性、热稳定性、无毒性、超亲水性、非迁移性等特点,且完全可以与食品接触,所以被广泛应用于抗紫外材料、纺织、光催化触媒、自洁玻璃、防晒霜、涂料、油墨、食品包装材料、造纸工业、航天工业中、锂电池中。纳米二氧化钛对枯草杆菌黑色变种芽孢、绿脓杆菌、大肠杆菌、金色葡萄球菌、沙门氏菌、牙枝菌和曲霉的杀灭率均达到98%以上;用纳米二氧化钛光催化氧化深度处理自来水,可大大减少水中的细菌数,饮用后无致突变作用,达到安全饮用水的标准;在涂料中添加纳米二氧化钛可以制造出杀菌、防污、除臭、自洁的抗菌防污涂料,应用于医院病房、手术室及家庭卫生间等细菌密集、易繁殖的场所,可净化空气、防止感染、除臭除味。
将纳米银和纳米二氧化钛结合起来制备抗菌材料,大大提高了整体的抗菌效果,但是高比表面积带来的高表面活性易造成纳米抗菌材料间的团聚,影响抗菌效果的均匀性。
发明内容
本发明目的在于提高纳米抗菌材料在食品保鲜袋本体上的分散性。
本方案中的基础方案:抗菌食品保鲜袋,其特征在于,包括抗菌层;所述抗菌层包括纳米银、纳米二氧化钛、分散剂和抗菌植物精油纳米脂质体。
纳米银、纳米二氧化钛均具有显著的抗菌效果,但是高比表面积带来的高表面活性易造成纳米抗菌材料间的团聚,影响抗菌效果的均匀性。
分散剂是能提高和改善固体或液体物料分散性能的助剂,加入分散剂可以在一定程度上降低纳米银和纳米二氧化钛的团聚程度。
抗菌植物精油具有体积小,抗菌效果好的特点。纳米脂质体是由磷脂双分子层构成的双分子层囊泡,具有类生物膜结构。纳米脂质体属于一种缓释制剂,可以使被包裹的物质缓慢释放,并且保护所包埋的物质,避免了抗菌植物精油的挥发,另外,抗菌植物精油纳米脂质体由于其纳米尺寸还具有高附着力的特点,极大地增加了抗菌植物精油与细菌的接触,从而提高了抗菌植物精油的杀菌活性。
在纳米银和纳米二氧化钛中加入抗菌植物精油纳米脂质体,不仅使抗菌效果叠加,而且抗菌植物精油纳米脂质体充斥在纳米银纳米二氧化钛颗粒之间,在物理空间上拉开了纳米颗粒之间的距离,能够减少纳米颗粒的团聚;而纳米颗粒同样充斥在纳米脂质体之间,拉大了纳米脂质体之间的距离,也使得纳米脂质体分散的较为均匀。
优选方案一:作为对基础方案的优化,所述分散剂为聚乙烯基吡咯烷酮、聚乙二醇、聚马来酸中的一种或几种。
聚乙烯基吡咯烷酮简称PVP,是一种非离子型高分子化合物,有工业级、医药级、食品级3种规格,本发明中使用食品级的聚乙烯基吡咯烷酮。PVP本身不会致癌,有良好的食物安全性,能与特定多酚化合物(如单宁)形成络合物,在食品加工方面主要作为啤酒、果汁、葡萄酒等食品澄清剂和稳定剂。PVP有优良的生理惰性,不参与人体新陈代谢,又具有优良的生物相容性,对皮肤、粘膜、眼等不形成任何刺激。PVP能与特定多酚化合物(如单宁)形成络合物,使其在果汁饮料中起到澄清作用和防凝作用。PVP作为高分子表面活性剂,在不同的分散体系中,可作为分散剂、乳化剂、增稠剂、流平剂、粒度调节剂、抗再沉淀剂、凝聚剂、助溶剂和洗涤剂。
聚乙二醇无毒、无刺激性,具有良好的水溶性,并与许多有机物组份有良好的相溶性。它们具有优良的润滑性、保湿性、分散性、粘接剂、抗静电剂及柔软剂等,在化妆品、制药、化纤、橡胶、塑料、造纸、油漆、电镀、农药、金属加工及食品加工等行业中均有着极为广泛的应用。比如用作分散剂,改进树脂的水分散性、柔韧性等。
聚马来酸又叫HPMA,是一种低分子量聚电解质,无毒。化学稳定性及热稳定性高,分解温度在330℃以上,可作为分散剂使用。
优选方案二:作为对基础方案的进一步优化,所述抗菌植物精油纳米脂质体为胎菊精油纳米脂质体、大蒜精油纳米脂质体或鱼腥草精油纳米脂质体中的一种。
胎菊对体外的大肠杆菌、痢疾杆菌、伤寒杆菌、副伤寒杆菌、变形菌以及绿脓杆菌和霍乱弧菌等细菌具有完全的抑制作用,对人体的结核杆菌有某些抑制效果。
大蒜是天然的植物广谱抗菌素,大蒜约含2%的大蒜素,并且含硫化合物具有奇强的抗菌消炎作用,对多种球菌、杆菌、真菌和病毒等均有抑制和杀灭作用。
鱼腥草对酵母菌、霉菌、溶血性的链球菌、金黄色葡萄球菌、流感杆菌、卡他球菌、肺炎球菌有明显的抑制作用,对大肠杆菌、痢疾杆菌、伤寒杆菌也有抑制作用。
抗菌食品保鲜袋的制备方法,其特征在于,包括以下步骤:
(1)制备二氧化钛溶胶:取无水乙醇30份,依次滴加钛酸丁酯10份,冰醋酸2份,边滴边搅拌,滴毕后继续搅拌25min,得均匀透明的淡黄色溶液;取硝酸0.5份,无水乙醇10份,去离子水1份,混合成硝酸乙醇溶液;然后将硝酸乙醇溶液滴加到所述淡黄色溶液中,边滴加边搅拌;滴毕,继续搅拌1.5h,得均匀透明的淡黄色二氧化钛溶胶;
(2)制备纳米银粒子:配置硝酸银的无水乙醇溶液,溶解,得溶液A;配置分散剂的无水乙醇溶液,即溶液B;然后将溶液A逐滴滴入到溶液B中得混合液,溶液A与溶液B的体积比为的1:2;再将混合液转入聚四氟乙烯内衬高压釜中,置于180℃烘箱中反应24h;冷却至室温,离心分离;所得产物经去离子水和无水乙醇洗涤后,真空干燥,得到纳米银粒子;
(3)制备含银离子的二氧化钛溶胶:将纳米银粒子加入到无水乙醇中,室温下超声20min,得纳米银溶液;然后将纳米银溶液滴入纳米二氧化钛溶胶中,纳米银溶液与纳米二氧化钛溶胶的体积比为1:5,超声震荡,制得含银离子的二氧化钛溶胶;
(4)制备抗菌植物精油纳米脂质体:取1份大豆卵磷脂,0.2份胆固醇和0.2份的植物精油,加氯仿使其溶解,大豆卵磷脂与氯仿的质量体积比为1g/40mL;将溶剂蒸干,得光滑的薄膜;然后将薄膜放入真空干燥箱中干燥;取适量聚乙烯吡咯烷酮加入醋酸盐缓冲液中,聚乙烯吡咯烷酮与醋酸盐缓冲液的质量浓度为0.05g/50mL;然后将聚乙烯吡咯烷酮的醋酸盐缓冲液在超声波条件下水化,将水化后的混合液于细胞超微粉碎仪中以工作10s,间隙5s的频率粉碎30min;将所得产品进行离心,4000rpm,15min,取上层液体;将所得液体用0.2μm滤膜进行过滤,滤液为抗菌植物精油纳米脂质体;
(5)将1份抗菌植物精油纳米脂质体与2份含银离子的二氧化钛溶胶混合,超声波混合均匀,得混合纳米抗菌剂;然后将所述混合纳米抗菌剂1份与塑料粒子7份混合,用来制造食品保鲜袋。
塑料粒子是指用于生产食品保鲜袋的常用塑料粒子,属于公知常识,在此不再一一列举。
具体实施方式
下面通过具体实施方式对本发明作进一步详细的说明:
实施例一:
(1)准备原料:无水乙醇,冰醋酸,硝酸,钛酸丁酯,聚乙烯基吡咯烷酮,硝酸银,去离子水,大豆卵磷脂,胆固醇,胎菊精油,氯仿。
(2)制备二氧化钛溶胶:室温下(25-30摄氏度)取无水乙醇30份,依次滴加钛酸丁酯10份(29mmol),冰醋酸2份,边滴边搅拌,滴毕后继续搅拌25min,得均匀透明的淡黄色溶液。取硝酸0.5份,无水乙醇10份,去离子水1份,混合成硝酸乙醇溶液。然后将硝酸乙醇溶液缓慢滴加到上述淡黄色溶液中,边滴加边搅拌。滴毕,继续搅拌1.5h,得均匀透明的淡黄色二氧化钛溶胶。
(3)制备纳米银粒子:首先配置硝酸银的无水乙醇溶液(0.017g/10ml),搅拌至完全溶解,得溶液A;配置聚乙烯基吡咯烷酮的无水乙醇溶液(0.05g/10ml),即溶液B。然后将溶液A逐滴滴入到溶液B中得混合液,溶液A与溶液B的体积比为的1:2。再将混合液转入聚四氟乙烯内衬高压釜中,置于180℃烘箱中反应24h。自然冷却至室温,离心分离。所得产物用去离子水洗涤3次,然后用无水乙醇洗涤3次,50℃真空干燥,得到银灰色的粉末,即纳米银粒子。
(4)制备含银离子的二氧化钛溶胶:将纳米银粒子加入到无水乙醇中(0.05g/10ml),室温下超声20min,使其分散均匀,得纳米银溶液。然后将纳米银溶液缓慢滴入纳米二氧化钛溶胶中,纳米银溶液与纳米二氧化钛溶胶的体积比为1:5,超声震荡30min,制得含银离子的二氧化钛溶胶。
(5)制备胎菊精油纳米脂质体:取1份大豆卵磷脂,0.2份胆固醇和0.2份的胎菊精油,加氯仿使其溶解,大豆卵磷脂与氯仿的质量体积比为1g/40mL。然后在旋转蒸发仪中(蒸发温度为20-30℃)蒸发至溶剂蒸干,得光滑的薄膜;然后将薄膜放入真空干燥箱中,30℃,真空状态下干燥24h。取适量PVP加入醋酸盐缓冲液中(0.05g/50mL)超声波条件下扩散,然后将聚乙烯吡咯烷酮的醋酸盐缓冲液在超声波条件下水化,将水化后的混合液于细胞超微粉碎仪中以工作10s,间隙5s的频率粉碎30min。将所得产品进行离心,4000rpm,15min,取上层液体。将所得液体用0.2μm滤膜进行过滤,滤液为胎菊精油纳米脂质体。
(6)将1份胎菊精油纳米脂质体与2份含银离子的二氧化钛溶胶混合,超声波混合均匀,得混合纳米抗菌剂。然后将得到的混合纳米抗菌剂1份与塑料粒子7份混合,用来制造食品保鲜袋I。
另外利用上述步骤(1)至(4),得到含银离子的二氧化钛溶胶,用该溶胶与1份与上述步骤(6)中的塑料粒子7份混合,用来制造食品保鲜袋II。
用上述步骤(5)制备的胎菊精油纳米脂质体1份与上述步骤(6)中的塑料粒子7份混合,用来制造食品保鲜袋III。
用上述步骤(6)中的塑料粒子直接制造食品保鲜袋IV。
以食品保鲜袋I、食品保鲜袋II、食品保鲜袋III、食品保鲜袋IV为材料,测试抗菌性能。将食品保鲜袋I、食品保鲜袋II、食品保鲜袋III、食品保鲜袋IV分别裁成2cm×2cm大小的方片,将200μl浓度为105-106个/ml的大肠杆菌溶液、黄金色葡萄球菌溶液滴在方片上,涂抹均匀,然后平铺在培养皿中,放入37℃的湿度适宜的培养箱中培养24h。然后取出,首先置于显微镜下观察菌落分布,然后用3mlPBS缓冲液冲洗方片,得悬浊液,将悬浊液混匀后,用血球计数板计数。
经显微镜观察,食品保鲜袋I方片上的菌落不明显,菌落小且数量少,分布较均匀;食品保鲜袋II方片上的菌落较为明显,菌落大小不一且数量稍多,分布较不均匀;食品保鲜袋III方片上的菌落较为明显,菌落大小不一且数量稍多,分布较不均匀;食品保鲜袋IV方片上的菌落明显,菌落大且数量多,分布较均匀。

由此可以看出,食品保鲜袋I的抗菌效果明显好于另外三种材质的食品保鲜袋。
实施例二:本实施例与实施例一的不同之处在于用聚乙二醇替代聚乙烯基吡咯烷酮,用大蒜精油替代胎菊精油。实验结果与实施例一一致。
实施例三:本实施例与实施例一的不同之处在于用聚马来酸代聚乙烯基吡咯烷酮,用鱼腥草精油替代胎菊精油。实验结果与实施例一一致。
以上所述的仅是本发明的优选实施方式,应当指出,对于本领域的技术人员来说,在不脱离本发明结构的前提下,还可以作出若干变形和改进,这些也应该视为本发明的保护范围,这些都不会影响本发明实施的效果和专利的实用性。
Claims (4)
1.抗菌食品保鲜袋,其特征在于,包括抗菌层;所述抗菌层包括纳米银、纳米二氧化钛、分散剂和抗菌植物精油纳米脂质体。
2.根据权利要求1所述的抗菌食品保鲜袋,其特征在于,所述分散剂为聚乙烯基吡咯烷酮、聚乙二醇、聚马来酸中的一种或几种。
3.根据权利要求2所述的抗菌食品保鲜袋,其特征在于,所述抗菌植物精油纳米脂质体为胎菊精油纳米脂质体、大蒜精油纳米脂质体或鱼腥草精油纳米脂质体中的一种。
4.根据权利要求1至3中任一项所述的抗菌食品保鲜袋的制备方法,其特征在于,包括以下步骤:
(1)制备二氧化钛溶胶:取无水乙醇30份,依次滴加钛酸丁酯10份,冰醋酸2份,边滴边搅拌,滴毕后继续搅拌25min,得均匀透明的淡黄色溶液;取硝酸0.5份,无水乙醇10份,去离子水1份,混合成硝酸乙醇溶液;然后将硝酸乙醇溶液滴加到所述淡黄色溶液中,边滴加边搅拌;滴毕,继续搅拌1.5h,得均匀透明的淡黄色二氧化钛溶胶;
(2)制备纳米银粒子:配置硝酸银的无水乙醇溶液,溶解,得溶液A;配置分散剂的无水乙醇溶液,即溶液B;然后将溶液A逐滴滴入到溶液B中得混合液,溶液A与溶液B的体积比为的1:2;再将混合液转入聚四氟乙烯内衬高压釜中,置于180℃烘箱中反应24h;冷却至室温,离心分离;所得产物经去离子水和无水乙醇洗涤后,真空干燥,得到纳米银粒子;
(3)制备含银离子的二氧化钛溶胶:将纳米银粒子加入到无水乙醇中,室温下超声20min,得纳米银溶液;然后将纳米银溶液滴入纳米二氧化钛溶胶中,纳米银溶液与纳米二氧化钛溶胶的体积比为1:5,超声震荡,制得含银离子的二氧化钛溶胶;
(4)制备抗菌植物精油纳米脂质体:取1份大豆卵磷脂,0.2份胆固醇和0.2份的植物精油,加氯仿使其溶解,大豆卵磷脂与氯仿的质量体积比为1g/40mL;将溶剂蒸干,得光滑的薄膜;然后将薄膜放入真空干燥箱中干燥;取适量聚乙烯吡咯烷酮加入醋酸盐缓冲液中,聚乙烯吡咯烷酮与醋酸盐缓冲液的质量浓度为0.05g/50mL;然后将聚乙烯吡咯烷酮的醋酸盐缓冲液在超声波条件下水化,将水化后的混合液于细胞超微粉碎仪中以工作10s,间隙5s的频率粉碎30min;将所得产品进行离心,4000rpm,15min,取上层液体;将所得液体用0.2μm滤膜进行过滤,滤液为抗菌植物精油纳米脂质体;
(5)将1份抗菌植物精油纳米脂质体与2份含银离子的二氧化钛溶胶混合,超声波混合均匀,得混合纳米抗菌剂;然后将所述混合纳米抗菌剂1份与塑料粒子7份混合,用来制造食品保鲜袋。
