The screening medium used here is a beef extract peptone medium, which comprises the following components: 10g of peptone, 3g of beef extract and 5.0g of sodium chloride, adding 1000ml of water to obtain a mixture, and carrying out high-temperature sterilization treatment at 121 ℃ for 30min at a pH of 7.0-7.4.
(2) Selecting 5 bacterial colonies with different forms, vigorous growth, smooth and viscous surface, repeatedly performing streak culture on the 5 bacterial colonies for more than three times under the same culture conditions to obtain pure bacterial strains, respectively numbering as No.1, No. 2, No. 3, No. 4 and No. 5 bacterial strains, and preserving at 4 ℃ after slant culture. The five strains are respectively cultured by using an activation culture medium, 1ml of a bacterial liquid obtained by culture is inoculated into a 1L beaker filled with 100g of human excrement, and after the bacterial liquid acts for 3d at the temperature of 30 ℃, the odor intensity, the ammonia concentration and the hydrogen sulfide concentration in the five beakers are respectively measured. Wherein, the strength of odor in the beaker inoculated with the No. 3 strain is reduced by 26.8 percent, the concentration of ammonia gas is reduced by 39.5 percent, the concentration of hydrogen sulfide is reduced by 35.2 percent, and the odor is far higher than that of other four strains, and the primary judgment shows that the odor removal capability is strongest, which is named as FJN.
(3) Strain identification
1) And morphological characteristics: the cells are rod-shaped, gram-positive, 0.7-1 μm wide and 2-3 μm long; the capsule is not contained, and the flagella is generated in the week and can move; culturing at 37 deg.C for 2-3 days to form spore with size of 0.6-0.9 × 1-1.5 μm, which is elliptical or cylindrical and is located at the center or slightly deviated from the thallus.
2) And culture properties: the strain cells are in a long rod shape, and spores are in an oval shape; the colony surface is rough and opaque, is dirty white, and can form wrinkles during liquid culture.
3) And molecular biological identification: extracting the whole genome DNA of the strain, carrying out PCR amplification on a 16S rDNA fragment, sequencing, comparing the sequencing result in NCBI (national center for Biotechnology information) and displaying that the strain is Bacillus subtilis.
Example 2: application of bacillus subtilis CGMCC13714 in feces deodorization
1. Preparation of culture medium and inoculated bacterial liquid
Solid activation medium: 10g of peptone, 10g of beef extract, 5g of yeast extract, 5.0g of sodium chloride and 15g of agar, and adding water to 1000ml, wherein the pH value is 7.0-7.4. Liquid expanded fermentation medium: 5g of peptone, 5g of beef extract, 10g of glucose and 5.0g of sodium chloride, and adding water to 1000ml, wherein the pH value is 7.0-7.4.
Inoculating bacillus subtilis CGMCC13714 into an activation culture medium for activation culture for 24h to obtain an activated strain for later use; inoculating the activated strain to an expanded fermentation medium, and performing shake culture for 24h at 35 ℃ under the condition of 120rmp to obtain an inoculated bacterial liquid for later use.
2. Deodorizing excrement
Inoculating 5ml of the inoculated bacterial liquid into a 1L beaker filled with 500g of human excrement (the inoculation amount is 1.0 percent, V/W), and sealing the beaker by using a preservative film; meanwhile, a control without adding bacteria liquid is carried out, three times of experiments are set, and the temperature is kept at 30 ℃. After 5d, the odor intensity, ammonia concentration and hydrogen sulfide concentration were measured, and the measurement results are shown in table 2 below.

As can be seen from Table 2, when the temperature is 30 ℃, after Bacillus subtilis CGMCC13714 is added into feces to act for 5 days, the odor intensity, the ammonia gas concentration and the hydrogen sulfide concentration are all reduced compared with the illumination, wherein the odor intensity is reduced by 29.3%, the ammonia gas concentration is reduced by 41.0% and the hydrogen sulfide concentration is reduced by 37.0%.
Example 3: application of bacillus subtilis CGMCC13714 in feces deodorization
1. Preparation of culture medium and inoculated bacterial liquid
Activating a culture medium: 10g of peptone, 10g of beef extract, 5g of yeast extract, 5.0g of sodium chloride, and water until the volume is 1000ml, wherein the pH value is 7.0-7.4. Liquid expanded fermentation medium: 5g of peptone, 5g of beef extract, 10g of glucose and 5.0g of sodium chloride, and adding water to 1000ml, wherein the pH value is 7.0-7.4.
Inoculating bacillus subtilis CGMCC13714 into an activation culture medium for activation culture for 24h to obtain an activated strain for later use; inoculating the activated strain to an expanded fermentation medium, and performing shake culture at 30 ℃ and 120rmp for 24h to obtain an inoculated bacterial liquid for later use.
2. Deodorizing excrement
Inoculating 5ml of the inoculated bacterial liquid into a 1L beaker filled with 500g of human excrement (the inoculation amount is 1.0 percent, V/W), and sealing the beaker by using a preservative film; meanwhile, a control without adding a bacterial solution is carried out, three times of experiments are set for each group, and the temperature is kept at 20 ℃. After 5d, the odor intensity, ammonia concentration and hydrogen sulfide concentration were measured, and the measurement results are shown in table 3 below.

As can be seen from Table 3, when the temperature is 20 ℃, the feces are added with the Bacillus subtilis CGMCC13714 bacterial liquid to act for 5 days, the odor intensity is reduced by 11.8%, the ammonia concentration is reduced by 32.5%, and the hydrogen sulfide concentration is reduced by 17.8%. The deodorizing effect is reduced as compared with that at a temperature of 30 c, but a certain deodorizing effect is still exhibited.
Example 4: application of bacillus subtilis CGMCC13714 in feces deodorization
The preparation methods of the culture medium and the inoculated bacterial liquid are the same as that in the example 2, three groups of parallel experiments are set for fecal deodorization treatment, the inoculum sizes (V/W) of the bacterial liquids in the three groups are respectively 2%, 5% and 10%, the temperature is kept within the range of 25-35 ℃, the odor intensity, the ammonia concentration and the hydrogen sulfide concentration are measured after 5 days, and the measurement results are shown in the following table 4.

As can be seen from Table 4, after the feces are added with the bacillus subtilis CGMCC13714 bacterial liquid with different inoculum sizes and acted for 5 days, the deodorization effect shows a trend of increasing firstly and then decreasing along with the increase of the inoculum size, wherein when the inoculum size is 5%, the deodorization effect is best, the odor intensity is decreased by 35.4%, the ammonia gas concentration is decreased by 50.0%, and the hydrogen sulfide concentration is decreased by 47.5%.
Example 5: bacillus subtilis CGMCC13714 applied to deodorization in public toilets
The preparation method of the culture medium and the inoculated bacterial liquid is the same as that of the example 2, the environmental temperature is 30-35 ℃ in a certain public toilet in Wu-Hou district of metropolis, and the squatting pan is sprayed with the bacterial liquid of bacillus subtilis CGMCC13714 (the spraying amount can be adjusted according to the flow rate and the odor degree of the public toilet, for example, the spraying amount is 12 mL/squatting pan). The odor intensity, ammonia concentration and hydrogen sulfide concentration in the toilet before and after spraying at 5d were measured, respectively, and the measurement results are shown in Table 5 below.

As can be seen from Table 5, after the Bacillus subtilis CGMCC13714 bacterial liquid 5d is sprayed in the public lavatory, the odor intensity, the ammonia concentration and the hydrogen sulfide concentration are all obviously reduced compared with those before the spraying, wherein the odor intensity is reduced by 52.7%, the ammonia concentration is reduced by 40.0%, the hydrogen sulfide concentration is reduced by 61.1%, and the deodorization effect is equivalent to that of the small-scale deodorization effect of the first embodiment.
Example 6: preparation of deodorizing bacterial agent
1. Liquid deodorant bacterial agent: preparation and activation culture of the culture medium the same as in example 3, the activated strain was inoculated into an expanded fermentation medium and cultured under shaking at 30 ℃ under 120rmp for 48 hours to obtain a liquid deodorant microbial inoculum. Expanding a fermentation medium: 5g of peptone, 5g of beef extract, 10g of glucose and 5.0g of sodium chloride, and adding water to 1000ml, wherein the pH value is 7.0-7.4.
2. Solid deodorant bacterial agent: mixing the liquid deodorant microbial inoculum and sterilized bran according to the mass ratio of 1:1, and placing the mixture in a constant-temperature incubator at 40 ℃ for drying and culturing for 21h to reduce the water content to 15-20% to obtain the solid deodorant microbial inoculum.

Claims (7)
1. An application of bacillus subtilis in preparing a fecal deodorizing microbial inoculum is characterized in that: the bacillus subtilis (Bacillus subtilis) has a preservation number in the common microorganism center of China Committee for culture Collection of microorganisms as follows: CGMCC No. 13714; the preservation date is as follows: 3 months and 1 day in 2017;
the method specifically comprises the following steps:
1) preparing bacillus subtilis CGMCC No.13714 bacterial liquid: inoculating bacillus subtilis CGMCC No.13714 in an activation culture medium to be cultured to obtain an activation strain, and inoculating the activation strain in an amplification fermentation culture medium to be subjected to amplification culture;
2) and inoculation deodorization: inoculating the bacillus subtilis CGMCC No.13714 bacterial liquid obtained by the enlarged culture in human excrement, and keeping the temperature at 20-35 ℃.
2. Use according to claim 1, characterized in that: the expanding culture in the step 1) is to inoculate the bacillus subtilis CGMCC No.13714 in an expanding fermentation medium and shake culture for 24-48 h under the conditions of 30-35 ℃ and 120 rpm.
3. Use according to claim 1, characterized in that: the inoculation amount of the bacillus subtilis CGMCC No.13714 bacterial liquid in the step 2) is 1-10% V/W.
4. Use according to claim 3, characterized in that: the inoculation amount of the bacillus subtilis CGMCC No.13714 bacterial liquid in the step 2) is 5 percent V/W.

Use according to claim 1, characterized in that: the components of the activation medium comprise: 10g of peptone, 10g of beef extract, 5g of yeast extract and 5g of sodium chloride, and adding water to 1000ml, wherein the pH value is 7.0-7.4; the components of the expanded fermentation medium comprise: 5g of peptone, 5g of beef extract, 10g of glucose and 5g of sodium chloride, and adding water to 1000ml, wherein the pH value is 7.0-7.4.
6. Use according to claim 5, characterized in that: the activating culture medium and the expanding fermentation culture medium also comprise 1.0 to 2.0 percent of agar W/V.
7. Use according to claim 1, characterized in that: preparing a solid deodorant microbial inoculum by using bacillus subtilis CGMCC No. 13714: mixing the bacillus subtilis CGMCC No.13714 bacterial liquid and the filler according to the mass ratio of 1:1, and placing the mixture in a constant-temperature incubator at 40 ℃ for drying and culturing for 20-24 h until the water content is 15-20%; the filler is sterilized bran.
