本发明公开了一种中药型医用耦合剂及其制备方法,其中所述中药型医用耦合剂由如下原料制备而成:瓜尔胶、卡波姆、薄荷脑、赖氨酸、三乙酯二胺四乙酸钾、大茴香醛、茶螺烷、黄芩苷、复合中药提取物、三异丙醇胺、磷酸、甘油、蒸馏水。本发明所提供的医用耦合剂对细菌和致病性真菌均具有良好的灭菌效果,抗菌广,灭菌快。此外医用耦合剂的超声衰减值小,并且无任何受刺激现象,不腐蚀和损伤探头,室温存放一年,各项性能指标几乎无变化,稳定性佳,同时迟发性超敏试验物致敏性,各项性能指标良好。

技术领域
本发明属于临床医学检测技术领域,具体涉及一种中药型医用耦合剂及其制备方法。
背景技术
超声波入射到两种不同媒质的分界面上时,二者拮抗越大,穿过界面进入另一媒质的声能越少。如果让超声探头与皮肤直接接触,有空气存在,所发超声波根本无法到达并进入人体,故而通常会使用医用超声耦合剂,将其涂抹于探头和皮肤表面之间,可以起到隔绝空气的作用,使超声波能够顺畅的传播。对于医用超声耦合剂有几项基本要求:1)良好的生物相容性,符合医疗用品的相关规定,即在24h以内接触无细胞毒性,同时对皮肤无刺激、无致敏;2)使用安全无毒,对于成分的限定:必须排除羧甲基纤维素和石蜡油制品;以及含醇类仅限于丙二醇等无毒化合物。
在超声检测过程中,由于超声探头与患者皮肤或或者黏膜进行直接接触,带菌量相对较多,且种类也比较繁杂,很可能带来各种潜在的感染,因此相对于非抗菌型医用超声耦合剂,抗菌型医用超声耦合剂发展前景更好。目前,基于抗菌的活性成分的不同,可将医用超声耦合剂分为三类,即中药型超声耦合剂、有机小分子型超声耦合剂和高分子型超声耦合剂。其中中药型超声耦合剂由于其使用更安全、抗菌广和抗过敏性佳而备受关注。然而,大部分的中药提取物不溶于水,因此往往需要添加非水性溶剂和乳化剂,如此在一定程度上会腐蚀超声探头和影响超声诊断的准确度。
发明内容
本发明的目的是避免上述现有技术中的至少一种不足之处,提供一种中药型医用耦合剂及其制备方法。
本发明的目的通过以下技术方案来实现:
一种中药型医用耦合剂,由如下重量份的原料制备而成:瓜尔胶4-9份、卡波姆2.2-7.5份、薄荷脑1.4-5份、赖氨酸0.6-2.5份、三乙酯二胺四乙酸钾1.5-3.2份、大茴香醛1.2-2.4份、茶螺烷0.8-2.1份、黄芩苷1.3-2.7份、复合中药提取物1.8-2.9份、三异丙醇胺0.6-2份、磷酸0.3-1.2份、甘油16-22份、蒸馏水38-50份。

上述中药型医用耦合剂的制备方法,包括如下步骤:
S1. 将复合中药提取物加入1-4倍重量份的60wt.%乙醇中先浸泡24-48小时,再转移至3-6倍重量份的40wt.%乙醇中再浸泡24-72小时,将两次提取的上清液混合备用;
S2. 将三乙酯二胺四乙酸钾、薄荷脑、赖氨酸和磷酸加入蒸馏水中搅拌均匀;
S3. 将瓜尔胶和卡波姆加入甘油和大茴香醛中充分研磨均匀,然后加入茶螺烷和黄芩苷,研磨均匀;
S4. 将复合中药提取物和步骤S2中的溶液依次缓慢加入步骤S3中,研磨均匀,然后加入三异丙醇胺,研磨均匀,即制得中药型医用耦合剂。
进一步的,步骤S1中先浸泡时间为30小时,再浸泡时间为48小时。
进一步的,步骤S2中搅拌速度为100-300转/分钟,搅拌时间为30-60分钟。
进一步的,步骤S4中所述缓慢加入时间控制在20-40分钟。
由于采用了以上技术方案,本发明的有益效果:
本发明所提供的医用耦合剂对不同的菌种作用1min后,对细菌灭菌率高达97.4及以上,且对致病性真菌灭菌率高达95.8及以上,显示对细菌和致病性真菌均具有良好的灭菌效果,抗菌广,灭菌快。此外医用耦合剂的超声衰减值小,仅介于0.018-0.027dB/(cm·MHz)的范围内,并且无任何受刺激现象,不腐蚀和损伤探头,室温存放一年,各项性能指标几乎无变化,稳定性佳,同时迟发性超敏试验物致敏性,各项性能指标良好。

下面结合具体实施例,对本发明作进一步详细描述。以下实施例用于说明本发明,但不用来限制本发明的范围。
实施例1
一种中药型医用耦合剂,由如下重量份的原料制备而成:瓜尔胶4份、卡波姆2.2份、薄荷脑1.4份、赖氨酸0.6份、三乙酯二胺四乙酸钾1.5份、大茴香醛1.2份、茶螺烷0.8份、黄芩苷1.3份、复合中药提取物1.8份、三异丙醇胺0.6份、磷酸0.3份、甘油16份、蒸馏水38份。
其中,所述复合中药提取物的原料为质量比为1:1.3:1.8:0.4:1.2:2.7:1.3:0.9:1.6的川滇香薷、芸香叶、荞麦花、珊瑚姜、莳萝、罗勒、蛤蟆衣、狗尾巴草和半枝莲。
其中,所述复合中药提取物的提取步骤为:按重量份称重各中药原料,碎成粗粉颗粒,过20目筛,然后用15倍重量的水浸泡煎煮10小时以上,药液过滤浓缩成浸膏,将浸膏脱水干燥制成植物提取物干粉。
所述中药型医用耦合剂的制备方法,包括如下步骤:
S1. 将复合中药提取物加入1倍重量份的60wt.%乙醇中先浸泡24小时,再转移至3-6倍重量份的40wt.%乙醇中再浸泡24小时,将两次提取的上清液混合备用;
S2. 将三乙酯二胺四乙酸钾、薄荷脑、赖氨酸和磷酸加入蒸馏水中搅拌均匀,其搅拌速度为100转/分钟,搅拌时间为30分钟;
S3. 将瓜尔胶和卡波姆加入甘油和大茴香醛中充分研磨均匀,然后加入茶螺烷和黄芩苷,研磨均匀;
S4. 将复合中药提取物和步骤S2中的溶液依次缓慢加入(控制在20分钟)步骤S3中,研磨均匀,然后加入三异丙醇胺,研磨均匀,即制得中药型医用耦合剂。
实施例2
一种中药型医用耦合剂,由如下重量份的原料制备而成:瓜尔胶9份、卡波姆7.5份、薄荷脑5份、赖氨酸2.5份、三乙酯二胺四乙酸钾3.2份、大茴香醛2.4份、茶螺烷2.1份、黄芩苷2.7份、复合中药提取物2.9份、三异丙醇胺2份、磷酸1.2份、甘油22份、蒸馏水50份。
其中,所述复合中药提取物的原料为质量比为1:1.3:1.8:0.4:1.2:2.7:1.3:0.9:1.6的川滇香薷、芸香叶、荞麦花、珊瑚姜、莳萝、罗勒、蛤蟆衣、狗尾巴草和半枝莲。
其中,所述复合中药提取物的提取步骤为:按重量份称重各中药原料,碎成粗粉颗粒,过20目筛,然后用15倍重量的水浸泡煎煮10小时以上,药液过滤浓缩成浸膏,将浸膏脱水干燥制成植物提取物干粉。

S4. 将复合中药提取物和步骤S2中的溶液依次缓慢加入(控制在40分钟)步骤S3中,研磨均匀,然后加入三异丙醇胺,研磨均匀,即制得中药型医用耦合剂。
实施例3
一种中药型医用耦合剂,由如下重量份的原料制备而成:瓜尔胶4.5份、卡波姆2.4份、薄荷脑1.8份、赖氨酸0.8份、三乙酯二胺四乙酸钾1.7份、大茴香醛1.5份、茶螺烷1.1份、黄芩苷1.5份、复合中药提取物2.1份、三异丙醇胺0.8份、磷酸0.5份、甘油18份、蒸馏水40份。
其中,所述复合中药提取物的原料为质量比为1:1.3:1.8:0.4:1.2:2.7:1.3:0.9:1.6的川滇香薷、芸香叶、荞麦花、珊瑚姜、莳萝、罗勒、蛤蟆衣、狗尾巴草和半枝莲。
其中,所述复合中药提取物的提取步骤为:按重量份称重各中药原料,碎成粗粉颗粒,过20目筛,然后用15倍重量的水浸泡煎煮10小时以上,药液过滤浓缩成浸膏,将浸膏脱水干燥制成植物提取物干粉。

S3. 将瓜尔胶和卡波姆加入甘油和大茴香醛中充分研磨均匀,然后加入茶螺烷和黄芩苷,研磨均匀;
S4. 将复合中药提取物和步骤S2中的溶液依次缓慢加入(控制在30分钟)步骤S3中,研磨均匀,然后加入三异丙醇胺,研磨均匀,即制得中药型医用耦合剂。
实施例4
一种中药型医用耦合剂,由如下重量份的原料制备而成:瓜尔胶8.6份、卡波姆7.6份、薄荷脑4.2份、赖氨酸2.3份、三乙酯二胺四乙酸钾3份、大茴香醛2.2份、茶螺烷2份、黄芩苷2.4份、复合中药提取物2.7份、三异丙醇胺1.7份、磷酸0.9份、甘油21份、蒸馏水47份。
其中,所述复合中药提取物的原料为质量比为1:1.3:1.8:0.4:1.2:2.7:1.3:0.9:1.6的川滇香薷、芸香叶、荞麦花、珊瑚姜、莳萝、罗勒、蛤蟆衣、狗尾巴草和半枝莲。
其中,所述复合中药提取物的提取步骤为:按重量份称重各中药原料,碎成粗粉颗粒,过20目筛,然后用15倍重量的水浸泡煎煮10小时以上,药液过滤浓缩成浸膏,将浸膏脱水干燥制成植物提取物干粉。
所述中药型医用耦合剂的制备方法,包括如下步骤:

S3. 将瓜尔胶和卡波姆加入甘油和大茴香醛中充分研磨均匀,然后加入茶螺烷和黄芩苷,研磨均匀;
S4. 将复合中药提取物和步骤S2中的溶液依次缓慢加入(控制在25分钟)步骤S3中,研磨均匀,然后加入三异丙醇胺,研磨均匀,即制得中药型医用耦合剂。
实施例5
一种中药型医用耦合剂,由如下重量份的原料制备而成:瓜尔胶5.3份、卡波姆4.6份、薄荷脑2.8份、赖氨酸1.6份、三乙酯二胺四乙酸钾2.4份、大茴香醛1.9份、茶螺烷1.8份、黄芩苷1.6份、复合中药提取物2.5份、三异丙醇胺1.3份、磷酸0.6份、甘油20份、蒸馏水42份。
其中,所述复合中药提取物的原料为质量比为1:1.3:1.8:0.4:1.2:2.7:1.3:0.9:1.6的川滇香薷、芸香叶、荞麦花、珊瑚姜、莳萝、罗勒、蛤蟆衣、狗尾巴草和半枝莲。
其中,所述复合中药提取物的提取步骤为:按重量份称重各中药原料,碎成粗粉颗粒,过20目筛,然后用15倍重量的水浸泡煎煮10小时以上,药液过滤浓缩成浸膏,将浸膏脱水干燥制成植物提取物干粉。

本对比例与实施例1的区别之处为:不包含赖氨酸和大茴香醛,且所述复合中药提取物的原料中不添加川滇香薷和莳萝。
对比例2
本对比例与实施例1的区别之处为:不包含黄芩苷和薄荷脑,且所述复合中药提取物的原料中不添加芸香叶、蛤蟆衣和罗勒。
性能测试
下面为实施例1-5和对比例1、2所制备的产品的理化性能和灭菌性能(1mL待测耦合剂)进行测试,其测试结果详见下表:

Claims (9)
1.一种中药型医用耦合剂,其特征在于,由如下重量份的原料制备而成:瓜尔胶4-9份、卡波姆2.2-7.5份、薄荷脑1.4-5份、赖氨酸0.6-2.5份、三乙酯二胺四乙酸钾1.5-3.2份、大茴香醛1.2-2.4份、茶螺烷0.8-2.1份、黄芩苷1.3-2.7份、复合中药提取物1.8-2.9份、三异丙醇胺0.6-2份、磷酸0.3-1.2份、甘油16-22份、蒸馏水38-50份。
2.根据权利要求1所述的中药型医用耦合剂,其特征在于,所述复合中药提取物的原料为川滇香薷、芸香叶、荞麦花、珊瑚姜、莳萝、罗勒、蛤蟆衣、狗尾巴草和半枝莲。
3.根据权利要求1所述的中药型医用耦合剂,其特征在于,所述复合中药提取物的原料为质量比为1:1.3:1.8:0.4:1.2:2.7:1.3:0.9:1.6的川滇香薷、芸香叶、荞麦花、珊瑚姜、莳萝、罗勒、蛤蟆衣、狗尾巴草和半枝莲。
4.根据权利要求2或3所述的中药型医用耦合剂,其特征在于,所述复合中药提取物的提取步骤为:按重量份称重各中药原料,碎成粗粉颗粒,过20目筛,然后用15倍重量的水浸泡煎煮10小时以上,药液过滤浓缩成浸膏,将浸膏脱水干燥制成植物提取物干粉。

6.一种中药型医用耦合剂的制备方法,其特征在于,包括如下步骤:
S1. 将复合中药提取物加入1-4倍重量份的60wt.%乙醇中先浸泡24-48小时,再转移至3-6倍重量份的40wt.%乙醇中再浸泡24-72小时,将两次提取的上清液混合备用;
S2. 将三乙酯二胺四乙酸钾、薄荷脑、赖氨酸和磷酸加入蒸馏水中搅拌均匀;
S3. 将瓜尔胶和卡波姆加入甘油和大茴香醛中充分研磨均匀,然后加入茶螺烷和黄芩苷,研磨均匀;
S4. 将复合中药提取物和步骤S2中的溶液依次缓慢加入步骤S3中,研磨均匀,然后加入三异丙醇胺,研磨均匀,即制得中药型医用耦合剂。
7.根据权利要求6所述的中药型医用耦合剂的制备方法,其特征在于,步骤S1中先浸泡时间为30小时,再浸泡时间为48小时。
8.根据权利要求6所述的中药型医用耦合剂的制备方法,其特征在于,步骤S2中搅拌速度为100-300转/分钟,搅拌时间为30-60分钟。
9.根据权利要求6所述的中药型医用耦合剂的制备方法,其特征在于,步骤S4中所述缓慢加入时间控制在20-40分钟。
