一种固体饮料及其制备方法,尤其涉及圆苞车前子壳粉、山楂、低聚木糖、食品添加剂组成的复配固体饮料。所述固体饮料由重量份数285~374.7份的圆苞车前子壳粉、重量份数427.5~749.5份的山楂、重量份数47.5~187.4份的低聚木糖、重量份数0.47~3.1份的食品添加剂组成。其中:圆苞车前子壳粉系采用元苞车前种子外壳为原料,经碾磨过60~80目筛制得,膳食纤维含量为82%~88%;山楂经抽提制成含有机酸13%~19%、总黄酮3%~8%的抽提物;低聚木糖采用含木二糖‑木七糖70%~95%、木二糖‑木四糖50%~65%的食品原料;食品添加剂为三氯蔗糖、微晶纤维素、木糖醇、羟丙基淀粉、α‑环状糊精中的一种或几种。实验表明:本发明所述固体饮料具有显著促进肠道益生菌生长、抑制肠道有害菌生长、改善肠道菌群结构的保健功能。

圆苞车前子壳是人工种植的车前科车前属圆苞车前种子的外壳,于2014年被国家卫计委(2014年第10号公告)批为新食品原料,因其富含可溶性膳食纤维被广泛用于药品、保健食品、食品、饮料以及饲料中。山楂是传统药食同源药材,含有黄酮、三萜、有机酸、氨基酸、维生素、多糖等多种营养及功能成分,具有降血脂、降血压、强心、抗心律不齐等作用,同时也是健脾开胃、消食化滞、活血化痰的良药,对胸膈脾满、疝气、血淤、闭经等症有很好的疗效。低聚木糖又称木寡糖,于2014年被国家卫计委(2014年第20号公告)批为新食品原料,是目前国内外竞相研究开发的功能性低聚糖之一。
随着人们生活质量的提高和保健意识的增强,开发纯天然、绿色功能性健康食品是科研人员、营养学家的追求和目标。《一种减肥酸奶及其制备方法》(专利申请公布号:CN201210361849.2)公开了一种减肥酸奶,系采用草决明粉、乌梅粉、车前子粉、荷叶粉、山楂粉以及脱脂生牛乳进行混合、发酵制得。本发明旨在采用新食品原料圆苞车前子壳粉、低聚木糖及传统药食同源山楂、GB2760允许使用的食品添加剂组方,通过本发明特定比例、特有工艺及质量控制技术方案,制备一种显著促进肠道益生菌生长、改善肠道菌群结构、双向调节胃肠道、减肥美容的固体饮料。
发明内容
本发明的目的是提供一种显著促进肠道益生菌生长的固体饮料及其制备方法。
本发明的目的是通过下述技术方案来实现的。
一种显著促进肠道益生菌生长的固体饮料,它包括下列重量份的原料:
圆苞车前子壳粉285~374.7份,山楂427.5~749.5份,低聚木糖47.5~187.4份,食品添加剂0.47~3.1份。
作为优选,它包括下列重量份的原料:
圆苞车前子壳粉300~350份,山楂500~650份,低聚木糖60~100份,食品添加剂1~3份。
进一步优选,它包括下列重量份的原料:
圆苞车前子壳粉337~339份,山楂580~582份,低聚木糖77~80份,食品添加剂1~2份。
其次,圆苞车前子壳粉系采用圆苞车前种子外壳为原料,经碾磨过60~80目筛制得。其中,膳食纤维含量为82%~88%,优选为83%~87%,更优选为84%~86%。
山楂粉粹成20目粗粉,采用50%乙醇渗漉法提取,提取液浓缩、干燥、粉碎过60~80目筛,得山楂提取物。其中,山楂提取物总黄酮的含量为3%~8%,优选为4%~7%,更优选为5%~6%;有机酸含量为13%~19%,优选为14%~18%,更优选为15%~17%。
低聚木糖直接使用新食品原料。其中,木二糖-木七糖含量为70%~95%,优选为71%~80%,更优选为72%~76%;木二糖-木四糖含量为50%~65%,优选为52%~63%,更优选为55%~60%。
再其次,食品添加剂包含三氯蔗糖、微晶纤维素、木糖醇、羟丙基淀粉、α-环状糊精中的一种或几种。
最后,按本发明重量份数及优选重量份数称取山楂,按本发明提取方法制备并检测山楂提取物,将其移至洁净混合容器中,按本发明重量份数及优选重量份数称取圆苞车前子壳粉、低聚木糖、食品添加剂添加至混合容器中,充分搅拌混合均匀,经干压、过60目筛、包装,制得显著促进肠道益生菌生长的固体饮料。
具体实施内容
下面结合实施例对本发明做进一步说明,但不以任何方式对本发明加以限制,基于本发明教导所作出的任何变换或替换,均属于本发明的保护范围。
实施例1:圆苞车前子壳粉的制备
取干燥圆苞车前种子外壳200g,经粉碎过60~80目筛,得圆苞车前子壳粉196g,经检测:膳食纤维含量为86.6%。
实施例2:山楂提取物的制备
取干燥山楂500g,粉碎成20目粗粉,按料液质量体积比1:8(g/ml)添加50%的乙醇将粗粉润湿装桶,浸泡60h,浸泡后缓慢添加余量50%乙醇并以3ml/min的速度收集渗漉液,减压浓缩、干燥、粉碎过60~80目筛,得山楂提取物123g。经检测有机酸含量为15.6%,总黄酮含量为5.7%。
实施例3:低聚木糖的准备
直接购置食品级或药品级低聚木糖100g,经检测低聚木糖中木二糖-木七糖含量为72.3%,木二糖-木四糖含量为56.4%
实施例4:固体饮料制备
取实施例1制备的圆苞车前子壳粉29g,实施例2制备的山楂提取物12g,实施例3准备的低聚木糖6g、三氯蔗糖0.05g置洁净容器中,充分搅拌使混合均匀,干压过60目筛,制得固体饮料46.1g。
实施例5:固体饮料制备
取实施例1制备的圆苞车前子壳粉35g,实施例2制备的山楂提取物16g,实施例3准备的低聚木糖10g置洁净容器中,添加微晶纤维素、三氯蔗糖、羟丙基淀粉混合物共计0.3g,充分搅拌使混合均匀,干压过60目筛,制得固体饮料60.1g。
实施例6:固体饮料制备
取实施例1制备的圆苞车前子壳粉33.8g,实施例2制备的山楂提取物14.5g,实施例3准备的低聚木糖7.8g置洁净容器中,添加三氯蔗糖、微晶纤维素混合物共计0.15g,充分搅拌使混合均匀,干压过60目筛,制得固体饮料55.1g。
实施例7:固体饮料制备(比例外)
取实施例1制备的圆苞车前子壳粉20g,实施例2制备的山楂提取物20g,实施例3购买的低聚木糖4g置洁净容器中,添加三氯蔗糖、微晶纤维素、α-环状糊精混合物共计0.4g,充分搅拌使混合均匀,干压过60目筛,制得固体饮料43.5g。
实施例8:半固体饮料制备
取实施例6制备的固体饮料10g,添加100℃开水150ml,充分搅拌冲调成糊状,灭菌包装制得半固体饮料。
实施例9:制剂制备
取实施例6制备的固体饮料,灭菌、定量包装即得粉剂;取实施例6制备的固体饮料,添加预胶化淀粉适量,经制粒、干燥、灭菌、定量包装即得颗粒剂;取实施例6制备的固体饮料,添加预胶化淀粉、聚维酮K30适量,经制粒、压片、定量包装即得片剂;取实施例6制备的固体饮料,添加硬脂酸镁适量,经充填、定量包装即得硬胶囊剂。
为了对本发明固体饮料技术效果进行说明,本发明通过动物实验,选取实施例6制备的固体饮料作为试验样品,编号为⑤。选取实施例1制备的圆苞车前子壳粉、实施例2制备的山楂提取物、实施例3购置的低聚木糖、实施例7非本发明特定比例制备的固体饮料作为对照样品,编号依次为①、②、③、④,同时设空白对照组,开展下述试验研究:
1.材料和方法
1.1试验动物与分组:体重为20±2g的SPF级昆明种雌性小鼠60只。每10只分为一组,共6组。
1.2剂量选择及样品处理:本发明固体饮料(试验样品⑤)根据60kg成年人推荐量为每日10g,折合小鼠给样剂量为1.3g/kg.bw(相当于人体推荐剂量的7.8倍),依据等剂量给样可比原则,确定对照样品④给样剂量为1.3g/kg.bw;依据等计量给样可比原则,结合国家规定的圆苞车前子壳粉、山楂、低聚木糖日最高推荐剂量,确定对照样品①给样剂量为1.3g/kg.bw;对照样品②给样剂量为1.3g/kg.bw;对照样品③为0.39g/kg.bw(均相当于人体推荐最高剂量的7.8倍)。以灌胃方式给予受试动物,每日灌胃1次,灌胃体积为0.2ml/10g.bw,空白对照组灌给予等量蒸馏水,连续灌胃14d。
1.3试验方法:肠道菌群的计数:给予受试物前及最后一次给受试药物后24h,无菌采取小鼠粪便,放入已灭菌的装有3ml稀释液的试管中,称重后震荡摇匀,使粪便完全均质于稀释液中。采取10倍系列稀释至10-8。选择合适的稀释度分别接种在各培养基上培养双歧杆菌(BS培养基)、乳杆菌(LC培养基)、肠杆菌(EMB培养基)及肠球菌(EC培养基)。培养48~72h后,以菌落形态,革兰氏染色镜检计数菌落,计算每克湿便中的双歧杆菌、肠球菌、肠杆菌及乳杆菌菌落数,去对数后进行统计处理。
2结果与分析
2.1对小鼠肠道双歧杆菌的影响

图1可看出:在连续给小鼠灌胃14d后,小鼠肠道内的双歧杆菌数量发生了变化。灌胃前后空白对照组、样品对照组①、②小鼠肠道内的双歧杆菌数量无差异显著性(P>0.05)。与空白对照组相比,③组双歧杆菌数量增长13.5%(P<0.01);④组双歧杆菌数量增长12.7%(P<0.01);⑤组双歧杆菌数量增长26.7%(P<0.01)。⑤组相较于③、④组具有显著性差异(P<0.01)。
2.2对小鼠肠道乳杆菌的影响

图2可看出:在连续给小鼠灌胃14d后,小鼠肠道内的乳杆菌数量发生了变化。灌胃前后空白对照组、样品对照组①、②小鼠肠道内的肠道乳杆菌数量无差异显著性(P>0.05)。与空白对照组相比,③组肠道乳杆菌数量增长8.5%(P<0.05);④组肠道乳杆菌数量增长6.7%(P<0.05);⑤组肠道乳杆菌数量增长19.7%(P<0.01)。⑤组相较于③、④组肠道乳杆菌数量增长具有显著性差异(P<0.01)。
2.3对小鼠肠道肠杆菌的影响

图3看出:在连续给小鼠灌胃14d后,小鼠肠道内的肠杆菌数量发生了变化。灌胃前后空白对照组、样品对照组①、②小鼠肠道内的肠道肠杆菌数量无显著性差异(P>0.05)。与空白对照组相比,③组肠道肠杆菌数量减少5%(P<0.05);④组肠道肠杆菌数量减少5%(P<0.05);⑤组肠道肠杆菌数量减少10%(P<0.01)。⑤组相较于③、④组肠道肠杆菌减少具有显著性差异(P<0.01)。
2.4对小鼠肠道肠球菌的影响

图4看出:在连续给小鼠灌胃14d后,小鼠肠道内的肠球菌数量发生了变化。灌胃前后空白对照组、样品对照组①、②小鼠肠道内的肠球菌数量无差异显著性(P>0.05)。与空白对照组相比,③组肠道肠球菌数量减少4.6%(P<0.01);④组肠道肠球菌数量减少5.6%(P<0.01);⑤组肠道肠球菌数量减少9.6%(P<0.01)。⑤组相较于③、④组肠道肠球菌减少具有显著性差异(P<0.01)。
3结论
本发明所述固体饮料能显著促进肠道双歧杆菌、乳杆菌等有益菌生长,显著抑制肠道肠杆菌、肠球菌等有害菌生长,从而显著改善肠道菌群结构。
Claims (9)
1.一种显著促进肠道益生菌生长、改善肠道菌群结构的固体饮料。其是由重量份数为285~374.7份的圆苞车前子壳粉、重量份数为427.5~749.5份的山楂、重量份数为47.5~187.4份的低聚木糖、重量份数为0.47~3.1份的食品添加剂组成。
2.根据权利要求1所述固体饮料,其特征在于:其是由重量份数为300~350份的圆苞车前子壳粉、重量份数为500~650份的山楂、重量份数为60~100份的低聚木糖、重量份数为1~3份的食品添加剂组成。
3.根据权利要求1所述的固体饮料,其特征在于:其是由重量份数为337~339份的圆苞车前子壳粉、重量份数为580~582份的山楂、重量份数为77~80份的低聚木糖、重量份数为1~2份的食品添加剂组成。
4.根据权利要求1~3所述的固体饮料,其特征在于:所述的圆苞车前子壳粉系采用元苞车前种子外壳为原料,经碾磨过60~80目筛制得。其中,膳食纤维含量为82%~88%,优选为83%~87%,更优选为84%~86%。
5.根据权利要求1~4所述固体饮料,其特征在于:所述山楂经粉碎成20目粗粉,采用50%乙醇渗漉提取、浓缩、干燥、粉碎过60~80目筛得山楂提取物。其中,有机酸含量为13%~19%,优选为14%~18%,更优选为15%~17%;总黄酮含量为3%~8%,优选为4%~7%,更优选为5%~6%。
6.根据权利1~5所述固体饮料,其特征在于:所述低聚木糖为食品原料。其中,木二糖-木七糖含量为70%~95%,优选为71%~80%,更优选为72%~76%;木二糖-木四糖含量为50%~65%,优选为52%~63%,更优选为55%~60%。
7.根据权利要求1~6所述固体饮料,其特征在于:所述食品添加剂包含三氯蔗糖、微晶纤维素、木糖醇、羟丙基淀粉、α-环状糊精中的一种或几种。
8.根据权利要求1~7所述固体饮料,其特征在于:所述固体饮料具有促进肠道益生菌生长、抑制有害菌生长、改善肠道菌群结构、双向调节胃肠道、减肥美容等功能。
9.根据权利要求1~8所述固体饮料,其特征在于:所述固体饮料可用开水冲调后直接食用,也可加水冲调进一步制成粘稠状、凝胶状半固体口服制剂,还可以添加或不添加药品、保健食品、功能性普通食品、特殊医学食品、特殊膳食食品可接受的辅料制成粉剂、颗粒剂、片剂、胶囊剂等固体口服制剂。
