本发明涉及一种食药两用药材组合,具体涉及一种在消食养胃方面具有良好活性的复方鸡内金超微粉药物组合及其制备方法。
技术背景
中药超微粉碎技术在20世纪90年代中后期兴起,它在遵循中医药理论的前提下,结合中药物料的特点,采用现代粉体技术,将中药材、中药提取物及中药制剂微粉化。
超微粉碎技术用于医药、食品、化妆及陶瓷等行业,国内应用起步较晚,但随着此技术在中药行业的应用优势逐渐显现,人们对它的关注度越来越高,该技术是近年来国内外发展非常迅速的一项新技术,是中药现代化发展的重要方向之一。此技术现已越来越广泛地用于其他相关行业,如中药保健品行业、食品深加工行业等,并且取得了较好的成果,成为开发新产品的巨大动力。
中药材中80%是以植物药为主的,植物类药材的有效成分多贮存在细胞内,超微粉碎技术(中药超微粉碎技术是指利用机械或流体动力将3mm以上物料颗粒粉碎至粒径为30um以下的过程)可打破植物药材细胞壁,使细胞内的有效成分充分释放出来,从而提高药材的生物利用度,有利于畜禽机体对药物的吸收利用,使中草药临床治疗效果产生质的飞跃。超微粉碎技术打破了沿用千百年的“药罐子”,被誉为中药饮片史上的一次具有里程碑意义的革命。中药材(植物)的细胞直径通常在10-200μm左右,中草药超微粉的粒径一般可粉碎到细胞粒径为48μm(300目)~25μm(500目)的粉末,甚至可溶于水中饮用。超微中药材颗粒的粒径范围为1-75um,在此范围内颗粒中药所含药效物质基础与传统中药饮片、中药制剂相比较,将不会发生明显的分子结构上的变化,不会导致中药属性、药效特征和功能主治的改变,为中药药效和稳定提供了基本保证。
超微粉碎的药材,具有质量可控、节省药材、增强药效、促进吸收及方便调配、便于服用的特点,且中药超微粉天然物无毒副作用或毒性较小可控。
目前超微粉技术的最新特点:
(1)超微粉碎
根据被粉碎物的不同特性,一次将被粉碎物粉碎至中心度为10微米左右的超微粉。量产细度最高可达1-2微米约5500目。根据被粉碎物特性的某种粉碎细度,可直接制成高纯度的片剂,若用于提取可大幅提高提取率及效率。以玛咔为例,粉碎后检测到的有效生物碱含量是普通粉碎的20倍(中科院过程工程研究所检测)。
(2)低温冷却
粉碎过程必然产生高热,过高温度会使被粉碎物变色,变味,品质降低甚至完全变质。可以根据被粉碎物的特性,设定粉碎限制温度,通过整体式冷却系统控制粉碎温度,并通过温度传感器实时监控粉碎温度,在无人看管状态下,如果超温,则会自动报警并停机。通常被粉碎物可以保持低于38度下粉碎。对于有特殊要求的被粉碎物,最低粉碎温度可以在20度以下。
(3)气流粉碎
超级气流粉碎机其基本原理是通过机器内部产生高速回旋气流并由此长生振动波及冲击波将被粉碎物粉碎至微米级粉末。由于没有采用普通刀片式或锤击式因此不会产生刀破片,或重金属污染。符合出口到国际市场的技术要求。另外高速气流来自机器内部因此不会被污染,能耗比也低于其它压缩式气流粉碎机。
(4)可彻底清洗
所有与被粉碎物有接触的机械零件均可拆卸分解并清洗。机器是可以简单移动的,整台机器可以推入洗净室高压冲洗。
(5)对高含油含糖及黏性物体粉碎能力强
可对大量高含油含糖及黏性物体进行超微粉碎。
本组方的药材组合均为药食两用中药,安全性高,功效明确,主要介绍如下:
鸡内金为雉科动物家鸡Gallus gallus domesticus Brisson的干燥沙囊内壁。杀鸡后,取出鸡肫,立即囊下内壁,洗净,干燥,即得。炒鸡内金表面暗黄褐色或焦黄色,用放大镜观察,显颗粒状或微细泡状。轻折即断,断面有光泽。具有徤胃消食,淫精止遗,通淋化石的功效。用于食积不消,呕吐泻痢,小儿疳积,遗尿,遗精,石淋涩痛,胆胀胁痛等。
薏苡仁(麸炒)为禾本科植物薏苡Coix lacryma-jobi L.var.mayuen(Roman.)Stapf的干燥成熟种仁。秋季果实成熟时采割植株,晒干,打下果实,再晒干,除去外壳、黄褐色种皮和杂质,收集种仁。麸炒薏苡仁微鼓起,表面徽黄色。本品具有利水渗湿,健脾止泻,除痹,排脓,解毒散结的功效。用于水肿,脚气,小便不利,脾虚泄泻,湿痹拘挛,肺痈,肠痈,赘疣,癌肿。
赤小豆为豆科植物赤小豆Vigna umbellata Ohwi et Ohashi或赤豆Vignaangularis Ohwi et Ohashi的干燥成熟种子。秋季果实成熟而未开製时拔取全株,晒干,打下种子,除去杂质,再晒干,即得。具有利水消肿,解毒排脓的功效。用于水肿胀满,脚气浮肿,黄疸尿赤,风湿热痹,痈肿疮毒,肠痈腹痛等。
芡实(麸炒)为睡莲科植物芡Euryaleferox Salisb.的干燥成熟种仁。秋末冬初采收成熟果实,除去果皮,取出种子,洗净,再除去硬壳(外种皮),晒干,即得。麸炒芡实表面黄色或微黄色。具有益肾固精,补脾止泻,除湿止带的功效。用于遗精滑精,遗尿尿频,脾虛久泻,白浊,带下等。
莱菔子为十字花科植物萝卜Raphanus sativus L.的干燥成熟种子。夏季果实成熟时采割植株,晒干,搓出种子,除去杂质,再晒干,即得。炒莱菔子表面微鼓起,色泽加深,质酥脆,气微香。本品具有消食除胀,降气化痰的功效。用于饮食停滞,脘腹胀痛,大便秘结,积滞泻痢,痰壅喘咳。
发明内容
一种复方鸡内金消食养胃超微粉药材组合,所述复方的组成由鸡内金、薏苡仁、赤小豆、芡实和莱菔子的原药材或提取物组成。
2.根据权利要求1所述的一种复方鸡内金消食养胃超微粉药材组合,所述复方鸡内金消食养胃超微粉药材组合由鸡内金、薏苡仁、赤小豆、芡实和莱菔子的超微粉组成。
所述鸡内金、薏苡仁、赤小豆、芡实和莱菔子的超微粉组成的重量比1-10:3-30:3-15:3-15:2-12。
所述鸡内金、薏苡仁、赤小豆、芡实和莱菔子的超微粉组成的重量比3:9:9:9:5。
所述鸡内金、薏苡仁、赤小豆、芡实和莱菔子的超微粉为300-2000目。
复方鸡内金消食养胃超微粉药材组的制备方法,包括以下步骤:
1)根据配方称取药材,将按照《中国药典》规定的炒作方法炮制方法进行炒或麸炒,其中鸡内金、莱菔子为炒制,薏苡仁、芡实为麸炒,赤小豆按照《中国药典》附录清炒法进行炒制;
2)将上述炒制过的药材,按照如下两种方式进行粉碎及混合,得到复方鸡内金消食养胃超微粉药物组。
方式一:将鸡内金(炒)、薏苡仁(麸炒)、赤小豆(炒)、芡实(麸炒)和莱菔子(炒)按照比例混合后进行超微粉碎,过300目筛;
方式二:将鸡内金(炒)、薏苡仁(麸炒)、赤小豆(炒)、芡实(麸炒)和莱菔子(炒)分别进行超微粉碎,过300目筛,再按照比例混合。
将复方鸡内金消食养胃超微粉药材组添加必要的辅料可以制备成颗粒剂、片剂、胶囊、糖果、冲调食品。
上述所述超微粉碎所用机器为气流涡旋超微粉碎机。
所述的复方鸡内金药材组在制备消食养胃食品或保健食品中应用。
本发明的有益技术效果是:本发明提供了一种鸡内金消食养胃药材组,其超微粉的评价优于普通粉,并且该复方在消食养胃方面具有明显的活性,结果与健胃消食片相当。
具体实施例方式
实施例1复方鸡内金微粉(粉末)感官评价试验
试验样品:取鸡内金(炒)90g、薏苡仁(麸炒)270g、赤小豆(炒)270g、芡实(麸炒)270g和莱菔子(炒)150g各5份,混合进行粉碎或超微粉碎成100目、200目、300目、500目、1000目的5个混合粉末。
试验方法:取上述100目、200目、300目、500目、1000目的混合粉末,由100名志愿者分别观察、品尝,对粒度、颜色、口感进行评价。
试验结果:按照上述方法进行评价,评价结果见表1。

通过上述研究结果表明,当颗粒粒径为48μm(300目)以下时,样品感官效果评价比较好。
实施例2复方鸡内金微粉制备工艺考察试验
工艺1试验样品(先混合再打粉):取鸡内金(炒)90g、薏苡仁(麸炒)270g、赤小豆(炒)270g、芡实(麸炒)270g和莱菔子(炒)150g各3份,混合进行超微粉碎成300目、500目、1000目的3个混合粉末。
工艺2试验样品(先打粉再混合):取鸡内金(炒)1000g、薏苡仁(麸炒)1000g、赤小豆(炒)1000g、芡实(麸炒)1000g和莱菔子(炒)1000g g,分别进行超微粉碎成300目、500目、1000目的三种粉末,再分别取鸡内金30g、薏苡仁90g、赤小豆90g、芡实90g和莱菔子50g微粉混合成3个混合粉末。
试验方法:取上述六份300目、500目、1000目的混合粉末,由100名志愿者分别观察、品尝,对粒度、颜色、口感进行评价。
试验结果:按照上述方法进行评价,评价结果见表2。

通过上述研究结果表明,两种工艺都得到了比较满意的评价,两种工艺没有差异。
实施例3复方鸡内金微粉的消食养胃活性考察
1、试验样品:
取鸡内金(炒)90g、薏苡仁(麸炒)270g、赤小豆(炒)270g、芡实(麸炒)270g和莱菔子(炒)150g,混合进行超微粉碎成微粉,过300目筛。得到复方鸡内金微粉实验样品。
2、复方鸡内金微粉对大鼠胃酸、胃蛋白酶分泌的影响评价试验:
取SD大鼠一批,适应环境5天,选择体重在200~240g大鼠50只,雌雄各半,随机分组。给复方鸡内金微粉样品、健胃消食片或水,连续3天,于第3天给药后禁食不禁水24h,乙醚麻醉,碘酊消毒,切开腹腔,暴露胃,结扎幽门,于十二指肠给药1次,缝合腹腔切口,消毒,覆盖伤口,5h后处死,开腹,结扎胃贲门,取胃,收集胃内容物,3000rpm离心15min,记录上清液量,吸取2ml,进行胃液总酸度、游离酸、胃蛋白酶活性的测定,实验结果见表3。

注:与溶媒对照组比较,*为P<0.05,**为P<0.01。
结果显示复方鸡内金微粉能明显促进大鼠胃液分泌、增加胃酸排出量,与健胃消食片比较无显著差异,与健胃消食片基本一致。
3、复方鸡内金微粉对小鼠胃排空的评价试验
取昆明小鼠50只,体重20g~22g,雌雄各半,随机分为五组,每组9~10只,即生理盐水组(给与复方鸡内金微粉中剂量组相当量的生理盐水),复方鸡内金微粉大剂量组(2.0g生药/Kg)、中剂量组(1.0g生药/Kg)和小剂量组(0.5g生药/Kg)及阳性对照组(健胃消食片1.0g/Kg)。均为灌胃给药,每天1次,连续7d。于末次给药后即将小鼠禁食不禁水15h,随即各组小鼠再给一次药,1h后按0.3ml/10g体重灌服酚红糊,20min后处死动物,开腹,取胃,置于小烧杯内,剪碎,用蒸馏水冲洗,然后各管加0.3N Ba(OH)21ml,混匀后3min~5min再加1ml 5%ZnSO4摇匀,以3000r/min,离心10min,取上清液3ml,加10%NaOH溶液0.5ml,在560nm处测光密度值,计算酚红残留量,各组进行比较,结果见表4。
实验结果:

结果显示复方鸡内金微粉能促进小鼠胃排空的速度,无明显剂量关系,与健胃消食片比较无显著差异,与健胃消食片基本一致。

1.一种复方鸡内金消食养胃超微粉药材组合,其特征在于:所述复方的组成由鸡内金、薏苡仁、赤小豆、芡实和莱菔子的原药材或提取物组成。
2.根据权利要求1所述的一种复方鸡内金消食养胃超微粉药材组合,其特征在于:所述复方鸡内金消食养胃超微粉药材组合由鸡内金、薏苡仁、赤小豆、芡实和莱菔子的超微粉组成。
3.根据权利要求2所述的一种复方鸡内金消食养胃超微粉药材组合,其特征在于:所述鸡内金、薏苡仁、赤小豆、芡实和莱菔子的超微粉组成的重量比1-10:3-30:3-15:3-15:2-12。
4.根据权利要求3所述的一种复方鸡内金消食养胃超微粉药材组合,其特征在于:所述鸡内金、薏苡仁、赤小豆、芡实和莱菔子的超微粉组成的重量比3:9:9:9:5。
5.根据权利要求2所述的一种复方鸡内金消食养胃超微粉药食两用药材超微粉组合,其特征在于:所述鸡内金、薏苡仁、赤小豆、芡实和莱菔子的超微粉为300-2000目。
6.一种复方鸡内金消食养胃超微粉药材组的制备方法,其特征在于包括以下步骤:
1)根据配方称取药材,将按照中国药典规定的炒作方法炮制方法进行炒或麸炒,其中鸡内金、莱菔子为炒制,薏苡仁、芡实为麸炒,赤小豆按照中国药典附录清炒法进行炒制;
2)将上述炒制过的药材,按照如下两种方式进行粉碎及混合,得到复方鸡内金消食养胃超微粉药物组;
方式一:将炒制鸡内金、麸炒薏苡仁、炒制赤小豆、麸炒芡实和炒制莱菔子按照比例混合进行超微粉碎,过300目筛;
方式二:将炒制鸡内金、麸炒薏苡仁、炒制赤小豆、麸炒芡实和炒制莱菔子分别进行超微粉碎,过300目筛,再按照比例混合。
7.一种复方鸡内金消食养胃超微粉药材组的制备方法,其特征在于:将复方鸡内金消食养胃超微粉药材组添加必要的辅料可以制备成颗粒剂、片剂、胶囊、糖果、冲调食品。
8.根据权利要求6所述的一种复方鸡内金消食养胃超微粉药材组的制备方法,其特征在于:根据权利要求6所述超微粉碎所用机器为气流涡旋超微粉碎机。
9.权利要求1-5任一项所述的一种复方鸡内金药材组在制备消食养胃食品或保健食品中应用。
