Abstract
本发明涉及保健药品技术领域,具体涉及一种治疗风湿疼痛的糖果及其制备方法。该糖果主要由以下重量份数的原料制备而成:乌药叶20~30份、木瓜20~30份、雪莲培养物5~10份、蝮蛇提取物5~10份、柳叶蜡梅20~30份、甘草15~25份、食用葡萄糖100~120份。该糖果中的中药成分协同配伍使制备的产品对风湿疼痛具有一定的治疗和缓解作用,且药食同源,无毒副作用,安全高效。本发明制备方法,通过限定各中药原料有效成分的提取方式,最大限度的提取和保留中药中的有效成分,提高原料的利用率,进而提高产品对风湿疼痛的治疗和缓解作用。

技术领域
本发明涉及保健药品技术领域,具体涉及一种治疗风湿疼痛的糖果及其制备方法。
背景技术
风湿痛指主要侵犯关节、肌肉、骨骼及关节周围的软组织(肌腱、韧带、滑囊、筋膜等部位)的疾病所引起的局部性或全身性疼痛,其病症属麻痹症一类,中医认为是风和湿两种病邪结合所致的病症。常见症状有头痛、发热、微汗、恶风、身重、小便不利、骨节酸痛、不能屈伸等。关节病变除有疼痛外尚伴有肿胀和活动障碍,呈发作与缓解交替的慢性病程。由于患者的血液循环不通常,导致肌肉或者组织所需要的营养无法通过血液循环来输送,致使患者肌肉缺少营养而加速老化变得僵硬,严重的会导致患者肌肉和血管萎缩,部分患者且可出现关节致残和内脏功能衰竭。
中医认为风湿病的发生,主要与正虚、邪侵及痰浊瘀血有关。在人体正气不足时,风、湿、热外邪侵袭,痹阻肌肉、关节、经络之间,致使气血运行不畅,则出现肌肉筋骨关节疼痛、麻木、展伸不利,甚至关节肿大、灼热、畸形。病久不愈,可致关节肿大、变形,疼痛如剧,皮下结节,肢体僵硬,麻木不仁,且诸症顽固难愈。中医治疗风湿病遵循辨证治疗原则,首先辨明虚实寒热。病属实者,以肢体关节肿胀、疼痛、麻木为主症,无正气虚弱表现;病属虚者,伴气血损伤、脏腑亏虚证候。临床治疗风湿病通常按病证寒热性质主要分为风寒湿痹与风湿热痹两型,风寒湿痹在治疗过程中需要实现散寒、祛风、除湿、通经活络的功效,风湿热痹在治疗过程中需要实现清热、祛风、除湿、通经活络的功效。
基于以上认识,人们以中药成分为原料药,研究开发出多种对风湿疼痛具有治疗和缓解作用的中成药,如疏风定痛丸、风湿骨痛丸、麝香风湿片、护骨酒、湿热痹冲剂等。上述现有的中成药虽然对风湿疼痛均具有一定的治疗和缓解功效,但是这些中成药大多都苦味和药味很重,适口性差,患者在服用过程中通常还需要承受喝药的痛苦,服用不便。那么如何将对风湿痛具有治疗作用的药物与食品结合起来,制备药食同源的食物,减轻患者服用药物痛苦感的同时,实现对风湿疼痛的治疗和缓解作用,成为了目前亟待解决的问题。
发明内容
为了克服现有技术的缺陷,本发明的目的是提供一种治疗风湿疼痛的糖果,中药成分与葡萄糖结合制成休闲糖果食品,减轻患者服用药物的痛苦感的同时,达到治疗和缓解风湿疼痛的目的。
同时,本发明还在于提供一种治疗风湿疼痛的糖果的制备方法。
为了实现以上目的,本发明所采用的技术方案是:
一种治疗风湿疼痛的糖果,主要由以下重量份数的原料制备而成:乌药叶20~30份、木瓜20~30份、雪莲培养物5~10份、蝮蛇提取物5~10份、柳叶蜡梅20~30份、甘草15~25份、食用葡萄糖100~120份。
优选的,上述治疗风湿疼痛的糖果,主要由以下重量份数的原料制备而成:乌药叶25份、木瓜25份、雪莲培养物8份、蝮蛇提取物8份、柳叶蜡梅25份、甘草20份、食用葡糖糖110份。
上述治疗风湿疼痛的糖果的制备方法,包括以下操作步骤:
1)中药成分预处理:
A:制备乌药叶、木瓜、甘草提取物:取乌药叶、木瓜、甘草混合粉碎后,加入固体物料2~3倍重量的水,依次经过微波加压浸提和双向动态逆流提取,固液分离得提取液2,将提取液2依次经过过滤、浓缩得浸膏1,将浸膏1冻干、粉碎,得粉状提取物1,备用;
其中微波加压浸提的具体方法为:温度为55~65℃,压力为0.1~0.3MPa,微波浸提30~40min;之后加入乳酸10~20份,调整温度为40~50℃,压力为0.1~0.3MPa,微波浸提45~60min;之后再调整温度为35~40℃,压力为0.1~0.3MPa,微波浸提40~50min;微波浸提完成后进行固液分离得药渣1和浸提液1;
双向动态逆流提取的具体方法为:在浸提液1中加入药渣1 2~3倍重量的乙醇和丙醇混合溶剂,得混合物1,以混合物1作为双向动态逆流提取的浸提剂对药渣1进行浸提;
浸膏1冻干的具体方法为:在浸膏1中加入重量份0.2~0.3份牛血清白蛋白、0.3~0.4份甘露醇、0.2~0.4份海藻糖、0.1~0.3份甲基纤维素、0.1~0.3份谷氨酸、0.2~0.4份组氨酸、0.2~0.3份卵磷脂,混合均匀后进行冻干;
B:制备柳叶蜡梅提取物:取柳叶蜡梅粉碎后,加入固体物料1~2倍重量的水,依次经过微波加压浸提和双向动态逆流提取,固液分离得提取液4,将提取液4依次经过过滤、浓缩得浸膏2,将浸膏2冻干、粉碎,得粉状提取物2,备用;
其中微波加压浸提的具体方法为: 温度为50~60℃,压力为0.5~0.6MPa,微波浸提60~70min;加入乳酸5~8份,调整温度为40~50℃,压力为0.3~0.4MPa,微波浸提50~60min;之后再调整温度为35~40℃,压力为0.2~0.3MPa,微波浸提40~50min;微波浸提完成后进行固液分离得药渣2和提取液3;
双向动态逆流提取的具体方法为:在提取液3中加入药渣2 1~2倍重量的乙醇和丙醇混合溶剂,得混合物2,以混合物2作为双向动态逆流提取的浸提剂对药渣2进行浸提;
浸膏2冻干的具体方法为:在浸膏2中加入重量份0.2~0.4份棉子糖、0.2~0.5份麦芽糖、0.3~0.5份β-环式糊精、0.1~0.3份吐温80、02~0.4份精氨酸、0.2~0.3份卵磷脂、0.1~0.3份组氨酸,混合均匀后进行冻干;
2)取葡萄糖加热熔融后,加入步骤1)制备的粉状提取物、粉状提取物2、雪莲培养物和蝮蛇提取物,混合均匀后,冷却,切块,即得所述的治疗风湿疼痛的糖果。
可选的,步骤A中双向动态逆流提取过程中加入的乙醇和丙醇的用量比例为3~4:3。
可选的,步骤B中双向动态逆流提取过程中加入的乙醇和丙醇的用量比例为3~4:2。
可选的,步骤A和步骤B中双向动态逆流提取过程中左侧逆向进浸提剂的速度为500~600L/h,右侧逆向进浸提剂的速度为150~180L/h,药渣的加料速度为80~90kg/h。
可选的,步骤A中双向动态逆流提取的温度为35~45℃,提取时间为1~2小时。
可选的,步骤B中双向动态逆流提取的温度为50~60℃,提取时间为1~2小时。
可选的,浸膏1和浸膏2的相对密度为1.1~1.2。
本发明治疗风湿疼痛的糖果,以乌药叶、木瓜、雪莲培养物、蝮蛇提取物、柳叶蜡梅和甘草为中药成分,结合葡萄糖制备而成;其中乌药叶温中理气、消肿止痛;木瓜增加机体免疫力,帮助机体修复组织;雪莲培养物温肾助阳、祛风胜湿、通经活血;蝮蛇提取物祛风、通络、止痛、解毒;柳叶蜡梅解表祛风、理气化痰、镇痛、抗炎、抗风湿,具有调节免疫系统,治疗血瘀阻络之风寒湿痹痛;甘草调和诸药。各种中药成分协同配伍,以柳叶蜡梅为君药,以雪莲提取物、乌药叶为臣药,以蝮蛇提取物、木瓜和甘草为佐使药,诸药共奏抗风胜湿,活血通络止痛的功效。同时,本发明将中药成分与葡萄糖结合制备成糖果,减轻服用药物的痛苦感,同时实现对风湿疼痛的治疗和缓解作用。
本发明治疗风湿疼痛的制备方法,通过限定各中药原料有效成分的提取方式,例如通过分三个温度阶段进行微波浸提,微波浸提过程中加压,微波浸提过程中加入醋酸或乳酸等成分,双向逆流提取过程中加入乙醇和丙醇混合溶剂等方式,促进原料中有效成分的溶出,同时在冻干过程中加入复配冻干保护剂,最大限度的提取和保留中药中的有效成分,提高原料的利用率,进而提高产品对风湿疼痛的治疗和缓解作用。
具体实施方式
下面通过具体实施例对本发明的技术方案进行详细说明。
实施例1
一种治疗风湿疼痛的糖果,主要由以下重量份数的原料制备而成:乌药叶25份、木瓜25份、雪莲培养物8份、蝮蛇提取物8份、柳叶蜡梅25份、甘草20份、食用葡糖糖110份。
上述治疗风湿疼痛的糖果的制备方法,包括以下操作步骤:
1)中药成分预处理:
A:制备乌药叶、木瓜、甘草提取物:取乌药叶、木瓜、甘草混合粉碎后,加入固体物料2.5倍重量的水,在温度为60℃,压力为0.2MPa的条件下,微波加压浸提35min后,加入乳酸15份,调整温度为45℃,压力为0.2MPa,持续微波加热浸提50min后,调整温度为38℃,压力为0.2MPa,微波浸提45min,微波浸提完成后进行固液分离得药渣1和浸提液1;在浸提液1中加入药渣1 2.5倍重量的乙醇和丙醇按照3.5:3的混合溶剂,得混合物1,以混合物1为浸提剂对药渣1进行双向动态逆流提取,调整左侧逆流进浸提剂的速度为550L/h,右侧逆向进浸提剂的速度为160L/h,药渣的加料速度为85kg/h,浸提温度为40℃,逆流提取1.5小时;动态逆流提取完成后进行固液分离,得提取液2,将提取液2依次经过过滤、浓缩得相对密度为1.1的浸膏1,在浸膏1中加入重量份0.25份牛血清白蛋白、0.35份甘氨酸、0.3份海藻糖、0.2份甲基纤维素、0.2份谷氨酸、0.3份组氨酸、0.25份卵磷脂,混合均匀后进行冻干,粉碎,得粉状提取物1,备用;
B:制备柳叶蜡梅提取物:取柳叶蜡梅粉碎后,加入固体物料1.5倍重量的水,在温度为55℃,压力为0.5MPa的条件下,微波加压浸提65min后,加入7份乳酸后,调整温度为45℃,压力为0.3MPa,持续微波加热浸提55min后,调整温度为38℃,压力为0.2MPa,微波浸提45min,微波浸提完成后进行固液分离得药渣2和浸提液3;在浸提液3中加入药渣2 1.5倍重量的乙醇和丙醇按照3.5:2的混合溶剂,得混合物2,以混合物2为浸提剂对药渣2进行双向动态逆流提取,调整左侧逆流进浸提剂的速度为550L/h,右侧逆向进浸提剂的速度为160L/h,药渣的加料速度为85kg/h,浸提温度为55℃,逆流提取1.5小时;动态逆流提取完成后进行固液分离,得提取液4,将提取液4依次经过过滤、浓缩得相对密度为1.1的浸膏2,在浸膏2中加入重量份0.3份棉子糖、0.3份麦芽糖、0.4份β-环式糊精、0.2份吐温80、0.3份精氨酸、0.25份卵磷脂、0.2份组氨酸,混合均匀后进行冻干,粉碎,得粉状提取物2,备用;
2)取食用葡萄糖加热熔融后,加入步骤1)制备的粉状提取物1、粉状提取物2、雪莲提取物和蝮蛇提取物,混合均匀后,冷却,切块,即得所述的治疗风湿病的糖果。
实施例2
一种治疗风湿疼痛的糖果,主要由以下重量份数的原料制备而成:乌药叶20份、木瓜20份、雪莲培养物5份、蝮蛇提取物5份、柳叶蜡梅20份、甘草15份、食用葡糖糖100份。

2)取食用葡萄糖加热熔融后,加入步骤1)制备的粉状提取物1、粉状提取物2、雪莲提取物和蝮蛇提取物,混合均匀后,冷却,切块,即得所述的治疗风湿病的糖果。
对比例1
本对比例治疗风湿疼痛的糖果,其配方同实施例1,其制备方法与实施例1不同的是,采用实施例1中制备粉状提取1的方法同时提取乌药叶、木瓜、柳叶蜡梅和甘草,制备获得粉状提取物,其他同实施例1。
对比例2
本对比例治疗风湿疼痛的糖果,其配方同实施例1,其制备方法与实施例1不同的是在中药成分预处理过程中省去添加使用乳酸;在制备粉状提取物1的过程中,在温度为50℃,压力为0.2MPa,微波浸提130min,得浸提液1后,在浸提液1中加入药渣1 2.5倍重量的乙醇,得混合物1;在制备粉状提取物2的过程中,调整温度为50℃,压力为0.5MPa,微波浸提165min,得浸提液3后,在浸提液3中加入药渣2 1.5倍重量的乙醇,得混合物2;其他同实施例1。
对比例3
本对比例治疗风湿疼痛的糖果,其配方同实施例1,其制备方法与实施例1不同的是在制备粉状提取物1的过程中,在调整微波浸提温度为38℃的过程中,加入乳酸;在制备粉状提取物2的过程中,在调整微波浸提温度为38℃的过程中,加入乳酸;其他同实施例1。
对比例4
本对比例治疗风湿痛的糖果,其配方同实施例1,其制备方法与实施例1不同的是在制备粉状提取物1的过程中,在浸膏1中加入 重量份0.25份牛血清白蛋白、0.85份甘氨酸、0.5份海藻糖、0.25份卵磷脂;在制备粉状提取物2的过程中,在浸膏2中加入重量份0.3份棉子糖、0.7份麦芽糖、0.45份吐温80。
试验例1
1、试验动物:构建类风湿性关节炎小鼠模型,具体方法可参见黄乃艳等在激光生物学报中发表的名称为《类风湿性关节炎的动物模型》中关于佐剂性关节炎模型的构建方法,构建类风湿因子IgM值在90~100的类风湿性关节炎小鼠模型。
2、试验方法:取合格的类风湿性关节炎小鼠模型100只,随机分为5组,每组20只,分别对每组给药,连续给药14天后,测定各组的类风湿因子IgM值,其中第一组给药为实施例1制备的粉状提取物1、粉状提取物2、雪莲营养物和蝮蛇提取物的混合物,加入注射用水制成的注射剂;第二组给药为对比例1制备的粉状提取物、雪莲营养物和蝮蛇提取物的混合物,加入注射用水制成的注射剂;第三组给药为对比例2制备的粉状提取物1、粉状提取物2、雪莲营养物和蝮蛇提取物的混合物,加入注射用水制成的注射剂;第四组给药为对比例3制备的粉状提取物1、粉状提取物2、雪莲营养物和蝮蛇提取物的混合物,加入注射用水制成的注射剂;第五组给药为对比例4制备的粉状提取物1、粉状提取物2、雪莲营养物和蝮蛇提取物的混合物,加入注射用水制成的注射剂;其中五组给药量相同;给药前后类风湿因子IgM值变化结果如下表1所示:

由上述表1所示的数据可知,对于类风湿性关节炎的治疗效果为第一组优于第三组优于第四组优于第五组优于第二组,由此可见,实施例1制备的粉状提取物中的中药有效成分疗效最显著,其余的顺序依次为对比例2、对比例3、对比例4和对比例1,因此本发明制备治疗风湿疼痛的糖果限定的制备方法,能够将中药中的有效成分充分提取,并最大限度的保留有效成分的活性,提高制备的产品对风湿疼痛的治疗和缓解效果。
试验例2 临床病例效果统计
1、一般资料
对200例患有不同程度的类风湿性关节炎,伴随风湿疼痛的患者,进行追踪观察治疗效果。
2、治疗方法:
每日服用实施例1制备的治疗风湿疼痛的糖果,每日3次,每次两粒,每粒单重15±0.05g;持续服用60天后,统计患者风湿疼痛的改善状况。
3、治疗标准:
显效:风湿疼痛明显缓解,无明显的疼痛感;
有效:风湿疼痛缓解,偶尔伴随有间歇性的疼痛;
无效:风湿疼痛无缓解。
4、统计结果显示,经过60天的治疗后,200例患者中,显效140例,有效50例,无效10例,总有效率达到95%。并且在整个治疗过程中,所有患者中未见不良反应。由此可见,本发明制备的治疗风湿疼痛的糖果对风湿疼痛的治疗和缓解效果显著,且所用的原料药食同源,无毒副作用,安全高效。
Claims (9)
1.一种治疗风湿疼痛的糖果,其特征在于,主要由以下重量份数的原料制备而成:乌药叶20~30份、木瓜20~30份、雪莲培养物5~10份、蝮蛇提取物5~10份、柳叶蜡梅20~30份、甘草15~25份、食用葡萄糖100~120份。
2.如权利要求1所述的治疗风湿疼痛的糖果,其特征在于,主要由以下重量份数的原料制备而成:乌药叶25份、木瓜25份、雪莲培养物8份、蝮蛇提取物8份、柳叶蜡梅25份、甘草20份、食用葡糖糖110份。
3.一种如权利要求1或2所述的治疗风湿疼痛的糖果的制备方法,其特征在于,包括以下操作步骤:
1)中药成分预处理:
A:制备乌药叶、木瓜、甘草提取物:取乌药叶、木瓜、甘草混合粉碎后,加入固体物料2~3倍重量的水,依次经过微波加压浸提和双向动态逆流提取,固液分离得提取液2,将提取液2依次经过过滤、浓缩得浸膏1,将浸膏1冻干、粉碎,得粉状提取物1,备用;
其中微波加压浸提的具体方法为:温度为55~65℃,压力为0.1~0.3MPa,微波浸提30~40min;之后加入乳酸10~20份,调整温度为40~50℃,压力为0.1~0.3MPa,微波浸提45~60min;之后再调整温度为35~40℃,压力为0.1~0.3MPa,微波浸提40~50min;微波浸提完成后进行固液分离得药渣1和浸提液1;
双向动态逆流提取的具体方法为:在浸提液1中加入药渣1 2~3倍重量的乙醇和丙醇混合溶剂,得混合物1,以混合物1作为双向动态逆流提取的浸提剂对药渣1进行浸提;
浸膏1冻干的具体方法为:在浸膏1中加入重量份0.2~0.3份牛血清白蛋白、0.3~0.4份甘露醇、0.2~0.4份海藻糖、0.1~0.3份甲基纤维素、0.1~0.3份谷氨酸、0.2~0.4份组氨酸、0.2~0.3份卵磷脂,混合均匀后进行冻干;
B:制备柳叶蜡梅提取物:取柳叶蜡梅粉碎后,加入固体物料1~2倍重量的水,依次经过微波加压浸提和双向动态逆流提取,固液分离得提取液4,将提取液4依次经过过滤、浓缩得浸膏2,将浸膏2冻干、粉碎,得粉状提取物2,备用;
其中微波加压浸提的具体方法为: 温度为50~60℃,压力为0.5~0.6MPa,微波浸提60~70min;加入乳酸5~8份,调整温度为40~50℃,压力为0.3~0.4MPa,微波浸提50~60min;之后再调整温度为35~40℃,压力为0.2~0.3MPa,微波浸提40~50min;微波浸提完成后进行固液分离得药渣2和提取液3;
双向动态逆流提取的具体方法为:在提取液3中加入药渣2 1~2倍重量的乙醇和丙醇混合溶剂,得混合物2,以混合物2作为双向动态逆流提取的浸提剂对药渣2进行浸提;
浸膏2冻干的具体方法为:在浸膏2中加入重量份0.2~0.4份棉子糖、0.2~0.5份麦芽糖、0.3~0.5份β-环式糊精、0.1~0.3份吐温80、02~0.4份精氨酸、0.2~0.3份卵磷脂、0.1~0.3份组氨酸,混合均匀后进行冻干;
2)取葡萄糖加热熔融后,加入步骤1)制备的粉状提取物、粉状提取物2、雪莲培养物和蝮蛇提取物,混合均匀后,冷却,切块,即得所述的治疗风湿疼痛的糖果。
4.如权利要求3所述的治疗风湿疼痛的糖果的制备方法,其特征在于,步骤A中双向动态逆流提取过程中加入的乙醇和丙醇的用量比例为3~4:3。
5.如权利要求3所述的治疗风湿疼痛的糖果的制备方法,其特征在于,步骤B中双向动态逆流提取过程中加入的乙醇和丙醇的用量比例为3~4:2。
6.如权利要求3所述的治疗风湿疼痛的糖果的制备方法,其特征在于,步骤A和步骤B中双向动态逆流提取过程中左侧逆向进浸提剂的速度为500~600L/h,右侧逆向进浸提剂的速度为150~180L/h,药渣的加料速度为80~90kg/h。
7.如权利要求3所述的治疗风湿疼痛的糖果的制备方法,其特征在于,步骤A中双向动态逆流提取的温度为35~45℃,提取时间为1~2小时。
8.如权利要求3所述的治疗风湿疼痛的糖果的制备方法,其特征在于,步骤B中双向动态逆流提取的温度为50~60℃,提取时间为1~2小时。
9.如权利要求3所述的治疗风湿疼痛的糖果的制备方法,其特征在于,浸膏1和浸膏2的相对密度为1.1~1.2。
