一种以乙醇提取南瓜果胶的方法,农业加工技术领域,在于解决果胶的提取问题,其特征是按下面的步骤进行:一、将南瓜100℃以上高温加热熟化;二、将熟化南瓜置于布袋中缓慢压榨,除去水分得到干原料;三、淀粉水解:将干原料放进塑料容器中,加水浸泡浸润,逐渐加入盐酸加热水解1~2分钟,再将稀糊装入布袋压榨,得到二次榨渣和水解液;四、再将二次榨渣加水和盐酸重复水解压榨过滤1次,合并两次的滤液;五、在合并滤液用活性炭脱色后过滤出活性碳,将滤液浓缩;六、在浓缩液中加入95%的乙醇,搅拌均匀后果胶絮凝出,过滤出果胶;七、将过滤的果胶用95%的乙醇溶液洗涤,再过滤、压榨得较为纯净的南瓜果胶。

本发明涉及提取南瓜果胶的方法,特别涉及以乙醇提取南瓜果胶的方法,属于农业加工技术领域。
背景技术
南瓜(拉丁学名:Cucurbita moschata (Duch. ex Lam.) Duch. ex Poiret),饭瓜,番南瓜,山东地区称作番瓜,东北地区称作倭(wō)瓜。河北地区有称作北瓜。可食用,有橘黄色和青色两种,外形呈扁圆或不规则葫芦形状,未成熟果实皮脆肉质致密,可配菜、做馅,成熟果实甜面,可熬粥。葫芦科、南瓜属的植物。一年生蔓生草本,茎常节部生根,伸长达2-5米,密被白色短刚毛。叶柄粗壮,原产墨西哥的中美洲一带,世界各地普遍栽培。明代传入我国,现南北各地广泛种植。可以做成南瓜泥有助于宝宝消化,对大人也有治疗便秘的效果。
南瓜是 葫芦科 南瓜属的植物。“南瓜”一词可以特指南瓜属 中的 ,也可以泛指包括笋瓜(又称 印度南瓜)、北方又称倭瓜,西葫芦(又称美洲南瓜)等在内的其他南瓜属栽培种。其中前者因产地叫法各异,台湾话称为 金瓜,原产于 北美洲。按颜色分可以分为 橙色、橙红色、 黄色、 白色、双色混合等。
南瓜果实有圆、 扁圆、 长圆、 纺锤形或 葫芦形,先端多凹陷,表面光滑或有瘤状突起和纵沟,成熟后有白霜。种皮灰白色或茶褐色,边缘明显粗糙。肉厚,黄白色,老熟后有特殊香气,味甜而面。种子扁,嫩瓜可作蔬菜,是夏秋季的瓜菜之一,味甘适口。 老瓜作 饲料或代糖,不少地方称之为“饭瓜”。种子可炒食或入药。
南瓜为一年生双子叶 草本植物,能爬蔓,茎的横断面呈五角形。叶子呈心脏形,花黄色,果实一般扁圆形或梨形,嫩时绿色,成熟时赤褐色果实可做蔬菜,种子可以吃。
葫芦科(Cucurbitaceae) 西葫芦种(Cucurbita pep)或南瓜种(C. moschata)的果实。该两个种的一些品种均有称为pumpkin与squash者,称法并不一致,尤其是在美国,在美洲则为西葫芦的俗称, 灌木或非蔓生小果品种称为小果南瓜(squash);生长季节长,且具长蔓的大果品种称为大果南瓜(pumpkin)。
果大型,一般重4~8公斤或更多,淡黄至橘黄色,扁球形球形至长圆形。果皮光滑,通常具浅沟或肋。果柄硬而木质化,具棱角。西葫芦的果柄与果实连接处不膨大。称作冬南瓜(butternut squash)的极大品种,果实可重34公斤或更多。植株极长蔓,通常在相隔约2.5-3米的小土堆上一次植1-3株。果实于早秋成熟,冬南瓜置于干燥,可在冰点以上妥存数月。
南瓜内所含的果胶有很好的吸附性,既能保护胃肠道粘膜,又能消除体内细菌毒素和重金属中的铅、汞和放射性元素毒害,起到解毒作用;果胶对胃肠道粘膜免受粗糙食品刺激,促进溃疡面愈合,特别适宜于胃病患者。果胶还能和体内多余的胆固醇结合在一起,使胆固醉吸收减少,血液胆固醇浓度下降。
南瓜果胶在南瓜中只占较少量,从南瓜中提取果胶会浪费大量的南瓜原料,获得的经济效益不大,所以提取南瓜果胶必须与南瓜作为原料的食品加工一体化进行。
发明内容
为了克服一船果胶提取技术繁杂,提取成本高的缺点,本人发明了一种从南瓜中提取南瓜果胶的方法,这种方法以酒精参与提取,提取出的果胶纯度高,品质好,能满足人们食品和各领域的需求。
一种以乙醇提取南瓜果胶的方法解决问题所采取的技术方案是按下面的步骤进行:一、熟化:将南瓜切碎,100℃以上高温加热熟化,以破坏组织细胞壁结构和杀灭果胶酶,利于压榨出细胞中的水分;二、将熟化南瓜置于布袋中缓慢压榨,除去水分得到干原料;三、淀粉水解:将干原料放进塑料容器中,加水浸泡浸润,逐渐加入盐酸搅拌均匀成稀糊,测得2≤pH值≤3,加热至90℃~100℃约1~2分钟后降温至常温,再将稀糊装入布袋压榨,得到二次榨渣和水解液;四、再将二次榨渣加水2倍,加入盐酸重复水解压榨过滤1次,合并两次的滤液;五、在在合并滤液中加入0.3%-0.5%的活性炭,保持55-60℃,脱色30分钟后过滤出活性碳,将滤液用外置干燥剂浓缩至原体积的3%左右;六、在浓缩液中加入95%的乙醇,加入量为浓缩体积的2倍,搅拌均匀后有果胶絮凝出,过滤出果胶,回收乙醇;七、将过滤的果胶用95%的乙醇溶液洗涤,再过滤、压榨得较为纯净的南瓜果胶。
一种以乙醇提取南瓜果胶的方法的有益效果是:一种以乙醇提取南瓜果胶的方法通过将南瓜高温破坏组织细胞,压榨除水,酸化水解,活性碳脱色净化,浓缩干燥,再用乙醇稀释提取,再次纯化提取等流程,提取出一流纯度的南瓜果胶,提取过程中产生的南瓜辅料还可以用于其它食品生产,相比于其它提取方法,本方法提取的南瓜果胶纯度较高、无色清纯。
具体实施方式
实施例:一种以乙醇提取南瓜果胶的方法主要是按下面的步骤实施进行:一、熟化:将南瓜切碎,100℃以上高温加热熟化,以破坏组织细胞壁结构和杀灭果胶酶,利于压榨出细胞中的水分;二、将熟化南瓜置于布袋中缓慢压榨,除去水分得到干原料;三、淀粉水解:将干原料放进塑料容器中,加水浸泡浸润,逐渐加入盐酸搅拌均匀成稀糊,测得2≤pH值≤3,加热至90℃~100℃约1~2分钟后降温至常温,再将稀糊装入布袋压榨,得到二次榨渣和水解液;四、。再将二次榨渣加水2倍,加入盐酸重复水解压榨过滤1次,合并两次的滤液;五、在在合并滤液中加入0.3%-0.5%的活性炭,保持55-60℃,脱色30分钟后过滤出活性碳,将滤液用外置干燥剂浓缩至原体积的3%左右;六、在浓缩液中加入95%的乙醇,加入量为浓缩体积的2倍,搅拌均匀后有果胶絮凝出,过滤出果胶,回收乙醇;七、将过滤的果胶用95%的乙醇溶液洗涤,再过滤、压榨得较为纯净的南瓜果胶。
