Description
技术领域
本发明涉及一种清热利湿治疗泌尿结石的药茶配方,更具体的说,是涉及一种清热利湿治疗泌尿结石的药茶配方及其制备方法技术领域。
背景技术
泌尿结石在肾和膀胱内形成。上尿路结石与下尿路结石的形成机制、病因、结石成份和流行病学有显著差异。上尿路结石大多数为草酸钙结石。膀胱结石中磷酸镁铵结石较上尿路多见。虽然部分肾结石有明确的原因,如甲状旁腺机能亢进、肾小管酸中毒、海绵肾、痛风、异物、长期卧床、梗阻和感染等,但大多数钙结石的形成原因目前仍不能完满解释。成核作用、结石基质和晶体抑制物质学说是结石形成的三种最基本学说。根据上尿路结石形成机制的不同,有人将其分为与代谢因素有关的结石和感染性结石。代谢性结石是由于代谢紊乱所致,如甲状旁腺机能亢进,各种原因引起的高尿钙症、高尿酸尿症和高草酸尿症等。高浓度化学成份损害肾小管,使尿中基质物质增多,盐类析出,形成结石。感染性结石是由于产生尿素酶的细菌分解尿液中的尿素而产生氨,使尿液碱化,尿中磷酸盐及尿酸铵等处于相对过饱的状态,发生沉积所致。细菌、感染产物及坏死组织亦为形成结石之核心。

A、将石斛鲜条放入清洗池内,去除表面泥土污渍,筛选优质石斛鲜条,晾干备用;
B、将清洗干净的石斛鲜条放入微波加热炉,保持温度50-60℃,进行加热10分钟,再取出放入旋转离心榨汁机内,进行三次榨汁,第一次加入石斛鲜条重量0.1-0.5倍的酒精含量52-60℃的白酒,时间10-15min,榨汁机的转速为20~30r/s;静置10分钟后,加入石斛鲜条重量2-3倍的蒸馏水,第二次榨汁机时间10-15min,榨汁机转速为80~100r/s,静置10min;后进行第三次榨汁,加入石斛鲜条重量3-5倍的蒸馏水,时间20-30min,榨汁机转速为100~150r/s,将石斛鲜条汁液与纤维过滤分离,使石斛鲜条的纤维分离率达95%以上,细胞破损率达80%以上,取得新鲜石斛汁和石斛鲜条纤维;再将新鲜葡萄放入50-60℃的温水清洗干净后,将葡萄肉、葡萄皮、葡萄籽分离待用;
C、石斛鲜条汁液放入与葡萄肉放入高压罐中,罐中保持压力0.5MP,温度保持100-150度,进行熬煮10分钟,得基体溶液I备用;
D、将分离的石斛鲜条纤维和葡萄皮放入无光密封罐内,在密封罐内加入相当于石斛鲜条纤维和葡萄皮总重量20-30%的蜂蜜及5-10倍重量的蒸馏水,搅拌打碎至100目,在30-35℃温度下密封保存24h,灭菌,过滤,放入无光密封罐,得基体溶液备用;提取基体溶液后的葡萄皮和石斛鲜条纤维加入质量体积比为0.5-0.7%的纤维素酶,加入1:10的质量比的水,保持35℃,经过24h酶解后可进一步提取水溶性膳食纤维,将水溶性膳食纤维加入基体溶液得到基体溶液II;
E、取路路通、木通、大黄清洗消毒后混合均匀,分别以6倍重量85%摩尔比乙醇提取2次,每次2小时,合并2次路路通、木通、大黄乙醇提取液,静置醇沉,过滤,滤液浓缩至无醇味,并减压浓缩至40℃下相对密度为1.15-1.35,制得路路通、木通、大黄的提取物备用;
将珍珠母、鸡内金、小茴香、车前子、木通混合搅拌放入超微粉碎机,超微粉碎机加热保持温度80度5-6分钟,进行超微粉碎得到300-400目的超微混合细粉;入相对于超微混合细粉质量6~8倍的蒸馏水,先浸泡10min~15min,倒入挥发油提取器中,蒸馏法提取挥发油2~4h,获得挥发油和超微混合细粉药渣;
取丝瓜络、王不留行、车前子、金钱草、滑石、海金沙、猪苓、泽泻、麦冬、山药、鱼鳔,混合均匀加30-60℃温水浸泡10分钟后,放入冷冻柜内,在-10~-20℃冷冻10-20分钟,形成冷冻药块,将冷冻药块和超微混合细粉药渣加水煎煮提取3次,第一次加1-2倍量水,温度保持100-120℃,进行熬煮10-20分钟;第二次加4-6倍量水,温度保持80-100℃,进行熬煮10分钟;第三次加6-10倍量水,温度保持120-150℃,进行熬煮30分钟,过滤,浓缩至原来体积的1/2,加入路路通、木通、大黄的提取物和挥发油,得基体溶液III备用;
F、取基体溶液I经过瞬间高温110-130℃,1-2分钟,放入配料罐内;取基体溶液II经目数为100目的精滤器滤后放入配料罐内;再取基体溶液III经目数为60目的精滤器滤后放入配料罐内;
G、将葡萄籽经真空脱气,然后采用CO2超临界流体萃取,CO2超临界流体萃取的条件为:萃取压力45MPa,萃取温度35-45℃,萃取时间30min,分离得到葡萄籽油;
H、三种基体溶液在配料罐搅拌均匀后加入葡萄籽油,经过封瓶机密封装入玻璃瓶中,封装好的玻璃瓶放入温度100~120℃,1.5MP,15~21分钟蒸汽炉内消毒灭菌,包装,装箱。
进一步:步骤B)中,新鲜葡萄清洗干净后,放入冷冻柜内,在-10~-20℃冷冻5-10分钟,使葡萄结冰,取出自然解冻后,再进行葡萄肉和葡萄皮分离作业。
进一步:步骤H)中所述配料罐内基体溶液I、基体溶液II、基体溶液III、葡萄籽油的重量配比是10:0.2~0.8:5~10:0.5。
珍珠母,味甘、咸,性寒;归肝、心经;质重镇降;具有针心安神,平肝潜阳,清肝明目。用于心悸失眠,惊痫癫狂,眩晕头痛,肝热目赤,翳膜遮睛,吐衄,崩漏。
鸡内金,鸡的砂囊内壁,具有消食健胃,涩精止遗的功效。
路路通,祛风通络,利水除湿。
丝瓜络,味甘,性凉。归肺、肝、胃经。通经活络,清热解毒,利尿消肿,止血。
王不留行,具有活血通经,下乳消肿,利尿通淋的功效。
车前子,清热利尿;渗湿止泻;明目;祛痰。
木通,泻火行水,通利血脉,治小便赤涩,淋浊,水肿,胸中烦热,喉痹咽痛。

下面结合具体实施例对本发明作更进一步的说明,以便本领域的技术人员更了解本发明,但并不因此限制本发明。
实施例 1
一种清热利湿治疗泌尿结石的药茶,由以下重量份的原料制成:珍珠母10份、鸡内金10份、路路通8份、丝瓜络12份、王不留行12份、车前子12份、木通8份、金钱草8份、滑石8份、海金沙8份、大黄8份、猪苓8份、泽泻8份、麦冬8份、小茴香8份、山药10份、鱼鳔8份、新鲜石斛15份、葡萄15份、蜂蜜、水适量。
一种清热利湿治疗泌尿结石的药茶的制备加工方法,所述制备方法包括以下步骤;
A、将石斛鲜条放入清洗池内,去除表面泥土污渍,筛选优质石斛鲜条,晾干备用;
B将清洗干净的石斛鲜条放入微波加热炉,保持温度50-60℃,进行加热10分钟,再取出放入旋转离心榨汁机内,进行三次榨汁,第一次加入石斛鲜条重量0.1-0.5倍的酒精含量52℃的白酒,时间15min,榨汁机的转速为30r/s;静置10分钟后,加入石斛鲜条重量2.5倍的蒸馏水,第二次榨汁机时间15min,榨汁机转速为80r/s,静置10min;后进行第三次榨汁,加入石斛鲜条重量3-5倍的蒸馏水,时间30min,榨汁机转速为150r/s,将石斛鲜条汁液与纤维过滤分离,使石斛鲜条的纤维分离率达95%以上,细胞破损率达80%以上,取得新鲜石斛汁和石斛鲜条纤维;再将新鲜葡萄放入55℃的温水清洗干净后,将葡萄肉、葡萄皮、葡萄籽分离待用;新鲜葡萄清洗干净后,放入冷冻柜内,在-10~-20℃冷冻10分钟,使葡萄结冰,取出自然解冻后,再进行葡萄肉、葡萄皮、葡萄籽分离作业。
C、石斛鲜条汁液放入与葡萄肉放入高压罐中,罐中保持压力0.5MP,温度保持120度,进行熬煮10分钟,得基体溶液I备用;
D、将分离的石斛鲜条纤维和葡萄皮放入无光密封罐内,在密封罐内加入相当于石斛鲜条纤维和葡萄皮总重量20-30%的蜂蜜及6倍重量的蒸馏水,搅拌打碎至100目,在30-35℃温度下密封保存24h,灭菌,过滤,放入无光密封罐,得基体溶液备用;提取基体溶液后的葡萄皮和石斛鲜条纤维加入质量体积比为0.5-0.7%的纤维素酶,加入1:10的质量比的水,保持35℃,经过24h酶解后可进一步提取水溶性膳食纤维,将水溶性膳食纤维加入基体溶液得到基体溶液II;葡萄皮中富含花青素,能够保护人体免受自由基的损伤,能够增强血管弹性、松弛血管,增加全身血液循环,具有一定的降血压功效;能增强免疫系统能力,具有强抗氧化、抗突变、减轻肝机能障碍、保护心血管等功能,葡萄皮和石斛鲜条纤维一起搅拌打碎经蜂蜜水提起花青素,可最大程度的提取出花青素的同时可保护花青素的品质。将水溶性膳食纤维加入基体溶液得到基体溶液II,可增加肠胃的蠕动的功效,保证代谢废物正常排出体外;吸附各种代谢废物,使之正常排出体外,提高石斛的综合利用价值同时提高人体对中药成分的吸收。
E、取路路通、木通、大黄清洗消毒后混合均匀,分别以6倍重量85%摩尔比乙醇提取2次,每次2小时,合并2次路路通、木通、大黄乙醇提取液,静置醇沉,过滤,滤液浓缩至无醇味,并减压浓缩至40℃下相对密度为1.15-1.35,制得路路通、木通、大黄的提取物备用;
将珍珠母、鸡内金、小茴香、车前子、木通混合搅拌放入超微粉碎机,超微粉碎机加热保持温度80度5-6分钟,进行超微粉碎得到300-400目的超微混合细粉;入相对于超微混合细粉质量6~8倍的蒸馏水,先浸泡10min~15min,倒入挥发油提取器中,蒸馏法提取挥发油2~4h,获得挥发油和超微混合细粉药渣;
取丝瓜络、王不留行、车前子、金钱草、滑石、海金沙、猪苓、泽泻、麦冬、山药、鱼鳔,混合均匀加30-60℃温水浸泡10分钟后,放入冷冻柜内,在-10~-20℃冷冻10-20分钟,形成冷冻药块,将冷冻药块和超微混合细粉药渣加水煎煮提取3次,第一次加1-2倍量水,温度保持100-120℃,进行熬煮10-20分钟;第二次加4-6倍量水,温度保持80-100℃,进行熬煮10分钟;第三次加6-10倍量水,温度保持120-150℃,进行熬煮30分钟,过滤,浓缩至原来体积的1/2,加入路路通、木通、大黄的提取物和挥发油,得基体溶液III备用;
F、取基体溶液I经过瞬间高温120℃,1-2分钟,放入配料罐内;取基体溶液II经目数为100目的精滤器滤后放入配料罐内;再取基体溶液III经目数为60目的精滤器滤后放入配料罐内;
G、将葡萄籽经真空脱气,然后采用CO2超临界流体萃取,CO2超临界流体萃取的条件为:萃取压力45MPa,萃取温度35-45℃,萃取时间30min,分离得到葡萄籽油;超临界萃取可以在接近室温37℃及CO2气体笼罩下进行提取,有效地防止了热敏性物质的氧化和逸散。因此,在萃取物中保持着药用植物的有效成分,而且能把高沸点、低挥发性、易热解的物质在远低于其沸点温度下萃取出来;
H、三种基体溶液在配料罐搅拌均匀后加入葡萄籽油,所述配料罐内基体溶液I、基体溶液II、基体溶液III、葡萄籽油的重量配比是10:0.5:6:0.5。经过封瓶机密封装入玻璃瓶中,封装好的玻璃瓶放入温度100~120℃,1.5MP,15~21分钟蒸汽炉内消毒灭菌,包装,装箱,携带饮用方便,保质期长达1-2年。
实施例2
一种清热利湿治疗泌尿结石的药茶,由以下重量份的原料制成:珍珠母5份、鸡内金5份、路路通5份、丝瓜络10份、王不留行5份、车前子6份、木通5份、金钱草5份、滑石5份、海金沙5份、大黄5份、猪苓5份、泽泻5份、麦冬5份、小茴香5份、山药5份、鱼鳔5份、新鲜石斛10份、葡萄10份、蜂蜜、水适量。加工方法同实施例1。
实施例3
一种清热利湿治疗泌尿结石的药茶,由以下重量份的原料制成:珍珠母15份、鸡内金12份、路路通10份、丝瓜络16份、王不留行15份、车前子15份、木通10份、金钱草10份、滑石10份、海金沙10份、大黄10份、猪苓10份、泽泻10份、麦冬10份、小茴香10份、山药10份、鱼鳔10份、新鲜石斛20份、葡萄20份、蜂蜜、水适量。加工方法同实施例1。
临床实验分三部分:
(一)动物急慢性毒性实验:
1、动物急性毒性试验
药物:本发明实施例1、实施例2及实施例3药茶加入米粉制成的药茶粉糊。
试验动物 :普通级 NIH 小鼠,体重 18g±2g,雌雄各半。
方法与结果:
将三种药茶各50g,加入米粉。将实验小鼠随机分成三组雌雄各半,分别喂食三种药茶粉糊,容积1ml/10g 体重,密切观察8小时并作记录,继续观察7天;7 天中,小鼠无死亡,行为活泼,摄食量如常,毛色光亮无疏松污,无惊厥嗜唾,眼、口、鼻、耳及肛周无异常分泌物,无断尾烂趾。分别从三组中选择5只处死,肉眼所见:心、肝、肺、胃内及胸腺等重要脏器,未见异常改变,表明两种该营养粉均无急性毒性。
2、动物长期毒性试验
药物:本发明实施例1、实施例2及实施例3所得药茶加入米粉制成的药茶粉糊。
动物:普通级 SD 大鼠,体重 200g±10g,雌雄各半。
方法与结果:
三种药茶粉糊的配置:均分为高、中、低三个剂量组,分别为患者食用量的200、100、50g;将实验鼠随机分成三组,分别连续喂食调制好的药茶粉糊,连续喂食40 天,观察动物全身毒性反应和严重程度,处死后按操作规程检查各部位,并进行血液学、ALT、BUN 及心、肝、脾、肺、肾、胃等主要脏器的病理学检查;
经过长期喂食,两组大鼠均未出现毒性反应。生长发育良好。肉眼外观及主要脏器未见异常。外周血象及血清ALT、BUN与对照组比较无病理性改变。病理报告心、肝、脾、肺、肾、胃等均未有意义的改变,因此可以认为,经病理证实,三种药茶粉糊对动物无慢性毒性表现。
通过以上两个毒性实验,证明本发明按实施例1、实施例2及实施例3制作的药茶是安全的,无毒副作用的,可以用来对临床患者进行临床试验。
3、本发明实施例饮用效果调查:
(1)调查方式 :
① 饮用人群:普通人群
② 饮用时间:60天
③ 饮用人数:50 人
④ 饮用方法:每人每天600mL
⑤ 指标:饮用后明显尿血、尿疼、排尿中断、口渴不饮、少腹两侧针疼、尿急、精神倦怠、食欲不振等症状消失,且口感清酸甜为显效;饮用后尿血、尿疼、排尿中断、口渴不饮、少腹两侧针疼、尿急、精神倦怠、食欲不振等症状缓解,且口感清酸甜则记为有效。饮用后无明显舒适感则记为无效。
(2)调查结果
调查结果见表1。由表1实验结果表明,实施例1、2、3有98%的受调查者感觉本发明所提供的饮料口感清酸甜,且上述临床症状消失或得到缓解,只有2% 的受调查者饮用后无明显增加舒适感。

结论:
由表2和表1对比可看出:
实施例对比组A的无效率的人数为实施例1、2、3的10倍左右;这表明本发明中药材配方和水溶性膳食纤维及葡萄籽油等营养成分之间产生了相互协同的药理作用,水溶性膳食纤维增加肠胃的蠕动的功效促进人体对药物成分的吸收,保证代谢废物正常排出体外,另外葡萄皮中含有的花青素、白藜芦醇,具有强抗氧化、抗突变、减轻肝机能障碍、保护心血管等功能,增强免疫系统能力;与中药为伍提高吸收效果同时改善中药的口感,经过本发明的精心配置和最大程度药物成分的提取,本发明中药材配方和制备方法出现了出人意料的技术效果。
表2和表3对比可看出:
实施例对比组A的无效率的人数为实施例对比组B的2倍左右,实施例对比组B和传统的熬制方法的实施例对比组A在治疗效果相比有了显著的提高,这表明本发明中药材经过步骤E的制备加工方法比传统的熬制方法,药效成分的提取程度有了显著的提高。
表1和表4相比:
实施例对比组C的配方不含鱼鳔,实施例对比组C的无效人数为实施例1、2、3的5倍左右;且有效率人数有均有所下降;表明鱼鳔在配方中起到了协同和引药归经的作用,鱼鳔应用效果取得了预想不到的技术效果。
综上所述,本发明提供的一种清热利湿治疗泌尿结石的药茶,清热利湿、通淋逐石、补肾益气、利湿化瘀、散结通阻治疗泌尿结石,具有酸甜的口感,提高人体免疫力、抗衰老、排毒养颜、滋养脾胃、降三高、防治糖尿病和防癌、抗癌等方面都有功效。且所有原料均来自药食两用天然植物,无毒副作用。

5.一种清热利湿治疗泌尿结石的药茶的制备加工方法,其特征在于,包括以下步骤;
A、将石斛鲜条放入清洗池内,去除表面泥土污渍,筛选优质石斛鲜条,晾干备用;
B、将清洗干净的石斛鲜条放入微波加热炉,保持温度50-60℃,进行加热10分钟,再取出放入旋转离心榨汁机内,进行三次榨汁,第一次加入石斛鲜条重量0.1-0.5倍的酒精含量52-60℃的白酒,时间10-15min,榨汁机的转速为20~30r/s;静置10分钟后,加入石斛鲜条重量2-3倍的蒸馏水,第二次榨汁机时间10-15min,榨汁机转速为80~100r/s,静置10min;后进行第三次榨汁,加入石斛鲜条重量3-5倍的蒸馏水,时间20-30min,榨汁机转速为100~150r/s,将石斛鲜条汁液与纤维过滤分离,使石斛鲜条的纤维分离率达95%以上,细胞破损率达80%以上,取得新鲜石斛汁和石斛鲜条纤维;再将新鲜葡萄放入50-60℃的温水清洗干净后,将葡萄肉、葡萄皮、葡萄籽分离待用;
C、石斛鲜条汁液放入与葡萄肉放入高压罐中,罐中保持压力0.5MP,温度保持100-150度,进行熬煮10分钟,得基体溶液I备用;
D、将分离的石斛鲜条纤维和葡萄皮放入无光密封罐内,在密封罐内加入相当于石斛鲜条纤维和葡萄皮总重量20-30%的蜂蜜及5-10倍重量的蒸馏水,搅拌打碎至100目,在30-35℃温度下密封保存24h,灭菌,过滤,放入无光密封罐,得基体溶液备用;提取基体溶液后的葡萄皮和石斛鲜条纤维加入质量体积比为0.5-0.7%的纤维素酶,加入1:10的质量比的水,保持35℃,经过24h酶解后可进一步提取水溶性膳食纤维,将水溶性膳食纤维加入基体溶液得到基体溶液II;
E、取路路通、木通、大黄清洗消毒后混合均匀,分别以6倍重量85%摩尔比乙醇提取2次,每次2小时,合并2次路路通、木通、大黄乙醇提取液,静置醇沉,过滤,滤液浓缩至无醇味,并减压浓缩至40℃下相对密度为1.15-1.35,制得路路通、木通、大黄的提取物备用;
将珍珠母、鸡内金、小茴香、车前子、木通混合搅拌放入超微粉碎机,超微粉碎机加热保持温度80度5-6分钟,进行超微粉碎得到300-400目的超微混合细粉;入相对于超微混合细粉质量6~8倍的蒸馏水,先浸泡10min~15min,倒入挥发油提取器中,蒸馏法提取挥发油2~4h,获得挥发油和超微混合细粉药渣;
取丝瓜络、王不留行、车前子、金钱草、滑石、海金沙、猪苓、泽泻、麦冬、山药、鱼鳔,混合均匀加30-60℃温水浸泡10分钟后,放入冷冻柜内,在-10~-20℃冷冻10-20分钟,形成冷冻药块,将冷冻药块和超微混合细粉药渣加水煎煮提取3次,第一次加1-2倍量水,温度保持100-120℃,进行熬煮10-20分钟;第二次加4-6倍量水,温度保持80-100℃,进行熬煮10分钟;第三次加6-10倍量水,温度保持120-150℃,进行熬煮30分钟,过滤,浓缩至原来体积的1/2,加入路路通、木通、大黄的提取物和挥发油,得基体溶液III备用;
F、取基体溶液I经过瞬间高温110-130℃,1-2分钟,放入配料罐内;取基体溶液II经目数为100目的精滤器滤后放入配料罐内;再取基体溶液III经目数为60目的精滤器滤后放入配料罐内;
G、将葡萄籽经真空脱气,然后采用CO2超临界流体萃取,CO2超临界流体萃取的条件为:萃取压力45MPa,萃取温度35-45℃,萃取时间30min,分离得到葡萄籽油;
H、三种基体溶液在配料罐搅拌均匀后加入葡萄籽油,经过封瓶机密封装入玻璃瓶中,封装好的玻璃瓶放入温度100~120℃,1.5MP,15~21分钟蒸汽炉内消毒灭菌,包装,装箱。
6.根据权利要求5所述的一种清热利湿治疗泌尿结石的药茶的制备加工方法,其特征在于,步骤B)中,新鲜葡萄清洗干净后,放入冷冻柜内,在-10~-20℃冷冻5-10分钟,使葡萄结冰,取出自然解冻后,再进行葡萄肉、葡萄皮、葡萄籽分离作业。
7.根据权利要求5所述的一种清热利湿治疗泌尿结石的药茶的制备加工方法,其特征在于,步骤H)中所述配料罐内基体溶液I、基体溶液II、基体溶液III、葡萄籽油的重量配比是10:0.2~0.8:5~10:0.5。
