背景技术
中草药饲料添加剂应用我国传统中兽医理论、中草药的物性(阴阳、寒凉、温热)、物味(酸、苦、甘、辛、咸)及物间关系,对动物起到保健、增加采食量、促进营养物质吸收、提高生产性能及降低饲料消耗的作用。目前,动物性食品遭受药物污染及对人类健康的影响越来越受重视。随着人们对抗菌药和激素等常用药物毒副作用的认识不断加深,以及人们对食品安全的广泛关注,中草药饲料添加剂因具有普通饲料添加剂无可比拟的优势而受到国内外饲料行业的普遍关注。而针对泌乳牛的中草药饲料添加剂产品在市场上很少,其在泌乳牛生产实际中产业化应用也鲜有报道。
孙凤俊等试验表明,用中草药添加剂饲喂奶牛,试验组每头平均日增重比对照组提高20.93%,增加产奶量4.5kg/(头·d);樊振华等在泌乳牛日粮中添加中草药添加剂,经过60d的饲喂试验使奶牛日产奶量平均提高了4.3kg,乳脂率提高了0.171个百分点;麻廷峰等在奶牛的日粮中添加一定量的中草药添加剂,可以明显提高奶牛产奶量,并且有效减少奶牛乳房炎的发病率。但这些研究多数处于试验阶段,作用机制不太明确;而目前市售制剂往往存在制作工艺落后、添加量偏大、适口性差、使用效果不稳定等问题,成为制约其产业化的瓶颈。

(5)提高奶产量5%-15%,提高乳脂率和乳蛋白率各0.1-0.2个百分点,实现奶牛健康高产;
(6)改善血液pH和肝功能相关指标,提高奶牛健康水平。
本发明的中药制剂,通过对泌乳牛的饲喂试验,使得健康添加组及病牛添加组日均单产分别提高0.80kg和2.08kg;隐性乳房炎牛恢复健康,且乳蛋白率平均提高了0.4个百分点;健康添加组的乳脂率和乳蛋白率分别提高了0.42个百分点和0.19个百分点。在显著改善乳指标、增强夏季对热应激的耐受力情况下每头牛日增加经济效益3.2-11.2元。

本发明中,各原料的作用机理如下:
黄芪:其具有增强机体免疫功能、保肝、利尿、抗衰老、抗应激、降压等临床功效;其性甘,微温,入、肺、脾、肝、肾经,具有补气固表,且大补脾阳功能,可治诸虚;本方用其固表补脾、提高免疫力以及保肝抗应激作用以为主药、故用量大而为方中之君药。
党参:党参有增强免疫力、扩张血管、降压、改善微循环、增强造血功能等作用;党参味甘性平。有补中益气,和胃生津,祛痰止咳等功效,用于脾胃虚弱,气血两亏,体倦无力,食少,口渴,久泻,脱肛等症。本发明中辅助黄芪,益气和胃,提高机体免疫力,是为辅药。
当归:具有补血和血,调经止痛,润燥滑肠、抗癌、抗老防老、提高免疫之功效。当归味甘而重,故专能补血,其气轻而辛,故又能行血,补中有动,行中有补,为血中之要药。因而,它既能补血,又能活血,既可通经,又能活络;而归尾的通经活络功能更强。方中使用归尾,以其补血活血功能协助主药补益气血、提高免疫力,又以其通经活络功能,改善微循环,增强乳腺细胞对损伤的修复能力。
熟地:具有补血滋阴功效,可用于血虚萎黄,眩晕,心悸失眠,月经不调,崩漏等症,亦可用于肾阴不足的潮热骨蒸、盗汗、遗精、消渴等症,是虚证类非处方药药品六味地黄丸主要成分之一。本发明用其补血养阴功效,辅助主药气血双补,提高免疫力,增强肝肾功能。
白术:具有健脾益气,燥湿利水,止汗,安胎的功效,用于脾虚食少,腹胀泄泻,痰饮眩悸,水肿,自汗,胎动不安。除湿益燥,和中益气,温中,去脾胃中湿,除胃热,强脾胃,进饮食,安胎。性味归经味苦,甘,性温。归脾、胃经。健脾益气,燥湿利水,止汗,安胎。用于脾虚食少,腹胀泄泻,痰饮眩悸,水肿,自汗,胎动不安。土白术健脾,和胃,安胎。用于脾虚食少,泄泻便溏,胎动不安。本方用其健脾益气燥湿强胃、增进饮食之功效,辅助黄芪益气固表,提高机体免疫力,是为臣药。
陈皮:味苦、辛,性温,归肺、脾经,理气健脾,燥湿化痰,理气健脾,燥湿化痰,用于脘腹胀满,食少吐泻,咳嗽痰多。在本发明,主要用其健脾理气功效,一方面调理脾胃功能,开胃增食,另一方面,解除补气补血诸药的滋腻,辅助其发挥正常功效。
甘草:味甘、性平,其功效益气健脾、润肺止咳、缓急止痛、清热解毒、调和诸药等;本发明主要用其益气健脾、甘缓和中、解疮疡肿毒以及调和诸药的特性,是为使药(奶牛作为高产动物,其脾胃负担重,常有挛痛不适,乳腺问题属于疮疡之疾)。
川芎:用于活血行气,祛风止痛。川芎辛温香燥,走而不守,既能行散,上行可达巅顶;又入血分,下行可达血海。活血祛瘀作用广泛,适宜瘀血阻滞各种病症;祛风止痛,效用甚佳,可治头风头痛、风湿痹痛等症。昔人谓川芎为血中之气药,殆言其寓辛散、解郁、通达、止痛等功能。本发明用其引领诸药,共达乳腺,是为药中使者。
肉苁蓉:味甘;咸;性温、归肾;大肠经,补肾阳;益精血;润肠道。主肾阳虚衰;精血不足之阳痿;遗精;白浊;尿频余沥;腰痛脚弱;耳鸣目花;月经衍期;宫寒不孕;肠燥便秘;肉苁蓉,滋肾补精血之要药。甘能除热补中,酸能入肝,咸能滋肾,肾肝为阴,阴气滋长,则五脏之劳热自退。肾肝足,则精血日盛,精血盛则多子。妇人症瘕,病在血分,血盛则行,行则症瘕自消矣。膀胱虚,则邪客之,得补则邪气自散,腰痛自止。久服则肥健而轻身,益肾肝补精血之效也,若曰治痢,岂滑以导滞之意乎,此亦必不能之说也。主五劳七伤,补中,除茎中寒热痛,养五脏,强阴,益精气,妇人症瘕。本发明,用其强阴壮阳,制五劳七伤、润肠道、治腰痛脚弱功效,益气补血活血、提高奶牛非特异免疫力、调和黄芪等补益气血药物的滋腻,改善胃肠功能,是重要辅助药物。
麦冬:甘、微苦,微寒。归肺、心、胃经。养阴生津,润肺止咳:用于肺胃阴虚之津少口渴、干咳咯血;心阴不足之心悸易惊及热病后期热伤津液等证。配沙参、川贝可治肺阴虚干咳。与乌梅、五味子、等配伍治疗胃液损耗,口干胃热症。清热养阴:用于热伤胃阴,口渴咽干、大便燥结等。清心除烦:用于温病热邪扰及心营,心阴不足,心烦不眠、心悸、心慌等,润肺利咽:用于肺热阴伤,咽喉干痛,声哑失音、舌燥口渴等。本发明麦冬主要用其养阴生津、清热功效,调理脾胃功能,增强奶牛瘤胃动力,清胃肠虚热,同时缓解热应激对奶牛的伤害;提高免疫力,增强抗病能力。
白芍:味甘、酸,性微寒,有养血的作用;平肝止痛,养血调经,敛阴止汗。用于头痛眩晕,胁痛,腹痛,四肢挛痛,血虚萎黄,月经不调,自汗,盗汗。用于肝炎:白芍具有保肝、降酶的作用,常与柴胡、当归、生地黄等配伍,有人认为白芍能促进白蛋白的合成。可以疏肝理气、柔肝养血、缓中止痛、平肝敛阴,常用于肝胃不和所致的胸胁胀痛、脘腹疼痛,月经不调、经行腹痛、崩漏,以及自汗、盗汗,头痛、眩晕等病症。
白芍还有调节免疫、抗炎等作用,临床上治疗慢性胃炎、消化性溃疡、慢性肠炎、结肠易激综合征、急性黄疸型肝炎、慢性乙型肝炎、肝纤维化和肝硬化、治疗坐骨神经痛、头痛、癫痫、冠心病、类风湿关节炎有一定作用。白芍的功效与作用主要是抗菌止痛。本发明白芍主要用其两方面功能:1.与当归、熟地黄、甘草等药一起生血活血,辅助黄芪,气血双补,提高奶牛的免疫功能,扶正祛邪;2.用其柔肝敛阴,疏肝理气与缓中止痛作用,与茵陈、栀子、大黄等共解肝胆之毒并排出代谢毒素以及外源性有毒有害物质,同时,白芍可缓解腹部各种不适,提高奶牛的舒适感受,有利于泌乳和修复组织损伤。
山楂:酸甘,微温。入脾、胃、肝经。消食健胃,行气散瘀。是健脾开胃、消食化滞、活血化痰的良药,对胸膈脾满、疝气、血淤、闭经等症有很好的疗效,同时具有降血脂、血压、强心、抗心律不齐等作用。现代医学研究:山楂对非特异性和特异性免疫功能均有促进作用。山楂可显著增加小鼠胸腺及脾脏重量、血清溶菌酶含量,提高血清血凝抗体滴度、T淋巴细胞转化率及外周血T淋巴细胞百分率。山楂对小鼠红细胞免疫黏附功能也有促进作用。本发明主要用其消食健胃、行气散淤以及提高免疫力功效,辅助诸药起到预防保健作用。加工上炒至焦黑,减少了对消化道刺激,增加了预防消化道出血、修复损伤的功效,对奶牛的瘤胃积食以及拌发的泻痢有很好的治疗作用,对保护瘤胃技能和和消化到粘膜有很好的作用。
莱菔子:味辛、甘,性平。归肺、脾、胃经。功能消食除胀,降气化痰。用于饮食停滞,脘腹胀痛,大便秘结,积滞泻痢,痰壅喘咳。莱菔子之功,长于利气。生能升,熟能降,升则吐风痰,散风寒,发疮疹;降则定痰喘咳嗽,调下痢后重,止内痛,皆是利气之效。
方中用炒莱菔子,用其消食除胀,降气化痰,舒缓胃肠不适,通调胃肠道。与山楂、陈皮等药,共同辅佐主药健脾理中(胃肠以通为用),扶正祛邪。
侧柏叶:味苦、涩,性寒。归肺、肝、脾经。功能凉血止血,化痰止咳,生发乌发。用于吐血,衄血,咯血,便血,崩漏下血,肺热咳嗽,血热脱发,须发早白。本发明用以凉血止血修复瘤胃和消化道损伤,维护消化机能正常。
大黄:苦,寒。归脾、胃、大肠、肝、心包经。泻下攻积,清热泻火,凉血解毒,逐瘀通经,利湿退黄。用于实热积滞便秘,血热吐衄,目赤咽肿,痈肿疔疮,肠痈腹痛,瘀血经闭,产后瘀阻,跌打损伤,湿热痢疾,黄疸尿赤,淋证,水肿;本发明用其凉血解毒,逐瘀通经,利湿退黄等功效用于预防乳腺疾病,并配合茵陈、栀子清肝胆湿热,解毒排毒,提高机体免疫力。
栀子:清热,泻火,凉血。治热病虚烦不眠,黄疸,淋病,消渴,目赤,咽痛,吐血,衄血,血痢,尿血,热毒疮疡,扭伤肿痛。方中栀子与大黄、茵陈共同清肝胆湿热,凉血解毒,辅助主药在扶正固本基础上,祛热邪,提高机体免疫力。
茵陈:性味归经:苦、辛,微寒。归脾、胃、肝、胆经。功能清热利湿;退黄。主治:黄疸、小便不利、湿疮瘙痒、传染性黄疸型肝炎等。药理学研究有利胆,保护肝功能,解热,抗炎,降血脂,降压,扩冠等作用。在本发明,用其清热利湿、保肝利胆功效,与栀子、大黄共同保肝护肝,排毒解毒,解除代谢毒素和饲料中有毒物质对肝脏的损害,和诸药共同完成扶正固本,排毒解毒,提高机体免疫力,预防疾病,提高生产性能的作用。
猪蹄甲:味咸,性微寒。归胃、大肠经。化痰定喘,解毒生肌。用于咳嗽喘息,肠痈,痔漏,疝气偏坠,白秃疮,冻疮。其味咸,性平,无毒,入手足阳明经,能清热凉血,行气逐淤,滋阴补肾。猪蹄甲有较好的促进乳汁分泌功能,与穿山甲对比差异不显著。穿山甲和猪蹄甲高剂量均能不同程度地使脓肿平均直径缩小、湿重减轻,提示高剂量猪蹄甲与穿山甲均可局限、缩小阑尾肿块,即消痈排脓作用类似;穿山甲和猪蹄甲均能使大鼠阑尾脓肿病理过程向痊愈方向发展,猪蹄甲高剂量组与穿山甲效果相类似,中、低剂量组作用较弱;各用药组均能不同程度地提高阑尾脓肿大鼠的白细胞吞噬功能,各用药组白细胞吞噬百分率均高于模型对照组。这一结果说明在排脓消痈作用上,猪蹄甲似可替代穿山甲,其抗炎作用与增强机体的免疫功能有关。更多的研究都证明,猪蹄甲在对通乳和乳痈治疗方面的治疗和预防作用与穿山甲类似,本发明用其通乳功能,作为全方的引经之药,使药性走向泌乳系统,起到对乳腺损伤的修复和促进作用;同时,用酒浸后炙炒,强化了其通乳活血化瘀排脓消痈功能(亦提高其生物利用度),用醋浸泡,强化其敛肝养阴,增强肌体免疫力效果。
本发明中,黄芪以补气固表,带诸药扶正祛邪,通过提高机体免疫力,达到预防奶牛乳房炎发病作用,故用量大而为君药;白术健脾利湿、党参益气健脾、当归补血和血、熟地补血养阴,麦冬养阴清热,肉苁蓉强阴壮阳,益气补血活血、提高奶牛非特异免疫力,共同辅助主药气血双补、健脾和胃、提高机体健康水平,共为臣药;莱菔子、陈皮、茵陈、山楂、侧柏叶、大黄、栀子、白芍等,分别理气健脾、清热泻火、改善消化系统功能、清肝胆湿热、排毒解毒,共同辅佐主药完成提高机体免疫力、防病等功能,共为佐药;甘草益气健脾、甘缓和中、解疮疡肿毒以及调和诸药、川芎活血行气,祛风止痛,并可引领诸药共达病所;猪蹄甲在对通乳和乳痈治疗方面的治疗和预防作用与穿山甲类似,可引导药性走向泌乳系统,起到对乳腺损伤的修复和促进作用,是方中引经之药,三药共同完成调和诸药药性并引导其走向泌乳系统之功能,共为方中使药。诸药合用,补气养血、活血、理气、疏肝健脾、和中清热。通过固本培元,调和营卫,提高机体免疫力,达到改善奶牛健康、预防乳房炎发病、降低体细胞数的、抗应激、提高生产性能的目的。

本发明临床试验及效果
1.1试验动物及饲养管理
试验于2014年8月1日至2014年10月1日在河北省宁晋县集斌奶牛养殖场进行。在250头泌乳牛中选取年龄、胎次、泌乳天数相近的泌乳奶牛28头,其中隐性乳房炎阳性牛和健康牛各14头。按照试验目的分为4组,每组7头:阳性对照组和健康对照组饲喂常规日粮,阳性添加组和健康添加组饲喂含有100g本发明中药制剂的日粮。试验牛按照分组情况饲喂于不同牛舍,每天饲喂2次,自由采食和饮水,由同一饲养员饲喂。在试验开始后一个月(第31天)、两个月(第61天)的时间,分别于早饲前空腹采血于肝素钠真空管标记待用;采集中心检测奶样送河北省DHI中心检测。
1.2试验设计
试验预试期为1个月;自2014年7月15日正式试验,正式期61天。预试期间对泌乳牛分组编号、分圈舍;使奶牛适应环境和日粮。不同试验组日粮除是否含有中药添加剂外,使用同一日粮配方。
1.3测定项目及方法
1.3.1采血及样品测定方法
在规定的时间点,空腹颈静脉采血两份,每份4ml。一份用于在离心机上2000r/min离心12min分离血清,全自动生化分析仪检测谷丙转氨酶、谷草转氨酶、乳酸脱氢酶、白蛋白等含量;一份离心处理后用于血液pH的检测。
1.3.2奶样采集方法及检测:
在预定的时间点,每头试验牛采集的奶样混匀后取40ml置于采样瓶中,送往河北DHI中心检测。根据检测结果统计试验牛的产奶量、体细胞、乳脂率、乳蛋白率。
1.4数据统计分析
试验数据采用SPSS19.0进行统计分析。P<0.05表示差异显著。
2结果与分析
2.1本发明中药制剂对奶牛血液指标的影响
通过检测试牛血清中与乳腺炎有关的酶的含量,间接反应本发明中药制加剂的效果。不同处理组试牛血清中酶、白蛋白及pH变化如表1所示。

由表1可以看出,病牛添加组饲用本发明中药制剂饲料后第1月后,SA、GTP的含量开始下降,至第2个月其SA、GPT的含量与阳性对照组有显著性差异(p<0.05)。与健康对照组、健康添加组对比,基本恢复正常。而健康对照组的SA、GPT含量在试验期内逐步上升,可能与夏季炎热所引起的乳腺炎发病或热应激有关。健康添加组的SA、GPT含量较为稳定,表明本发明中药制剂可以提高机体对热应激的耐受性。在整个试验期间,GOT、LDH的变化量不大,不同组别之间没有显著性差异。
表1显示,健康对照组、阳性对照的血液pH在试验期间基本没有变化,分别在6.75、6.5左右;病牛添加组和健康添加组的血液pH均呈上升趋势,试验两个月后病牛添加组的血液pH接近健康对照组。说明本发明中药制剂可以有效改善机体内环境,改善体液酸碱度,有效增加机体对热应激及疾病的抵抗能力。
2.2本发明中药制剂对奶牛生产性能的影响
由表2可以看出,健康添加组及病牛添加组在食用本发明中药制剂后产奶牛呈现逐步上升的趋势,饲喂两个月后日均单产分别提高0.80、2.08公斤。另外,病牛添加组的乳脂率变化不大,乳蛋白率由平均2.9%提高到3.3%,平均提高了0.4个百分点。健康添加组的乳脂率由3.3%提高到3.72%,平均提高了0.42个百分点;乳蛋白率由2.9提高到3.19,平均提高了0.29个百分点。阳性牛添加组的体细胞数呈明显下降趋势(下降36.5万),而对照组变化不明显(下降4.3),健康添加组和对照组体细胞变化均不明显;阳性对照组、健康对照组的产奶量均呈现下降的趋势;各组之间乳脂率没有明显差异,而阳性添加组和健康添加组在饲喂中药制剂后乳蛋白率均显著提高。

3、结论
试验结果表明,本发明中药制剂可以提高泌乳牛日均单产,显著改善乳指标,控制乳房炎的发生,改善机体内环境,减少酸中毒的发生;提高奶牛机体免疫力,降低体细胞数,减少夏季热应激,提高奶牛生产性能。为开辟新的高效饲养技术提供理论和实践基础。
三、本发明中药制剂使用效果
1:2010年11月9日呼和浩特市某牧场,54头牛,体细胞加权平均数54万/ml,隐性乳房炎牛67%,临床乳房炎牛14.8%。用药后,12月6日监测结果:体细胞数加权值33万/ml,隐性乳房炎40%,临床乳房炎0;乳脂率用药前3.35%,用药后3.54%;乳蛋白率用药前3.01%,用药后3.17%,产奶量比对照组增加0.8kg。
2:北京某牧场:2012年3月110头牛在做DHI检测时,发现牛群平均体细胞数高达88万/ml。排除其中11头亚临床乳房炎牛外,其余99头牛的体细胞均在72万/ml以上。然后应用中药治疗方案:⑴在全泌乳牛群中每天每头投放本发明中药制剂30克,一次性投入在TMR饲料中,作为保健预防。⑵体细胞在60-200万/ml以上的牛,每天每头投药量50克,作为预防治疗。投药20天后,牛场113头牛参加了4月初DHI的检测,检测结果:牛群平均体细胞数为26万/ml。排除上月遗留和新生的5头体细胞在100-200/ml万/ml的牛外,其余108头牛平均体细胞为17.5万。6月份检测结果:60-80万/ml的仅3例,牛群中体细胞10万左右的占50%,20万左右的占30%,平均体细胞数16万,牛群体况良好,乳房炎发病率为0。到12月底,牛群单产平均29kg/d,比3月份提高3kg。

1.提高奶牛免疫力、预防奶牛乳房炎的中药制剂,其特征在于,所述中药制剂由以下重量份数的原料制成:黄芪150-200份,党参40-80份,当归40-80份,熟地40-80份,白术30-60份,陈皮40-80份,甘草15-45份,川芎15-45份,肉苁蓉30-50份,麦冬30-50份,白芍30-50份,山楂60-150份,莱菔子40-80份,侧柏叶60-100份,大黄5-15份,栀子10-30份,茵陈40-80份,猪蹄甲40-80份。
2.根据权利要求1所述的提高奶牛免疫力、预防奶牛乳房炎的中药制剂,其特征在于,所述中药制剂由以下重量份数的原料制成:黄芪160-190份,党参50-70份,当归50-70份,熟地50-70份,白术40-50份,陈皮50-70份,甘草20-40份,川芎20-40份,肉苁蓉35-45份,麦冬35-45份,白芍35-45份,山楂70-140份,莱菔子50-70份,侧柏叶70-90份,大黄8-12份,栀子15-25份,茵陈50-70份,猪蹄甲50-70份。
3.根据权利要求1所述的提高奶牛免疫力、预防奶牛乳房炎的中药制剂,其特征在于,所述中药制剂由以下重量份数的原料制成:黄芪170-180份,党参55-65份,当归55-65份,熟地55-65份,白术42-47份,陈皮55-65份,甘草25-35份,川芎25-35份,肉苁蓉38-42份,麦冬38-42份,白芍38-42份,山楂80-130份,莱菔子55-65份,侧柏叶75-85份,大黄9-11份,栀子18-23份,茵陈55-65份,猪蹄甲55-65份。
