Abstract
本发明属于中药领域,具体涉及一种含艾叶的治疗习惯性流产的药物制剂及其制备方法,所述药物制剂由以下重量份的原料制备而成:艾叶10-22份、玉米须10-0份、苎麻根8-20份、竹茹8-16份、黄芩8-14份、山药6-18份、枸杞6-14份、当归8-14份、地骨皮6-14份、砂仁4-14份、枳壳6-12份和甘草4-10份。本发明药物制剂具有的疗效,用于治疗习惯性流产见效快,疗效好,无毒副作用,生产工艺简单,易于推广应用。
Description
技术领域
本发明属于中药领域,具体涉及一种含艾叶的治疗习惯性流产的药物制剂及其制备方法。
背景技术
堕胎或小产连续发生三次以上者,称为“滑胎”,现代医学称“习惯性流产”。其临床特点是屡孕屡堕,每次流产往往发生于同一妊娠月“应期而堕”。其流产过程与一般流产相同。习惯性流产的病因病机主要是胎元不固,导致胎元不固的原因有虚有实。虚者多因肾虚、气血虚弱,导致系胎、固胎、养胎之精、气、血亏虚,使胎元不固发为流产;实者常因血热、血瘀,而致养胎、载胎之气血失调,使胎元不固发为流产。临床上以虚证或虚中夹实者多见。
目前,由于西药治疗习惯性流产效果不佳,并容易复发,且长期服用西药有较多副作用,因此,人们对有效治疗习惯性流产的中药存在很大的需求。中医认为复发性流产是“血气不足,故不能养胎所以致胎数堕”。肾气不足包括两个方面,一是父母的精气,若不足则阴阳虽能结合成形,但是胎元不固,或有畸形,终致流产,甚或频坠;二是母体虚弱,肾气不足。脾胃虚弱主要导致气血的不足,气虚不能载胎,血虚不能养胎,故而流产。脾肾双亏,血气不足,是习惯性流产的主要病因。因此,中医治疗习惯性流产重在辨证。由于习惯性流产多因气血虚弱、肾虚、血热及外伤,导致内分泌不足、气血循环不佳流产。所以在治疗上必须以补气、固肾、养血、清热、活血等疗法处方用药。在中医来看,本症有虚有实。虚证是肾虚、气血虚弱,无法充养于胚胎;实证为血热、瘀血等病因损伤胎儿所致。依中医理论,虚证宜补肾、益气养血;实证宜滋阴降火、化瘀。
中国申请专利201310662360.3公开了一种治疗习惯性流产的药物,该药物包括如下制备原料:山楂、白芍药、杜仲、狗脊、川芎、鹿角霜、香附、仙鹤草、木耳、茜草、胡桃仁、益母草、、肉苁蓉、砂仁、续断、巴戟天、黄芩、茯神、大枣、淫羊藿、覆盆子、升麻、吴茱萸、茯苓、伏龙肝、白术、椿皮、麦门冬、柴胡、仙茅、海螵蛸、益智仁、龙眼肉、板蓝根、和紫苏梗。该发明药物具有补虚助阳,行气调中,补肝安胎的功能,但其制备方法复杂,而且在温肾健脾和疏调气血方面针对性不强,故仍需进一步发明更为有效的治疗药物。
发明内容
本发明的目的在于克服现有技术对治疗习惯性流产的不足,提供一种治疗习惯性流产的药物制剂。
本发明含艾叶的治疗习惯性流产的药物制剂由以下重量份的原料制备而成:艾叶10-22份、玉米须10-0份、苎麻根8-20份、竹茹8-16份、黄芩8-14份、山药6-18份、枸杞6-14份、当归8-14份、地骨皮6-14份、砂仁4-14份、枳壳6-12份和甘草4-10份。
在该重量份的范围内,优选了3个疗效较为显著的配方:
配方1:艾叶10份、玉米须10份、苎麻根8份、竹茹8份、黄芩8份、山药6份、枸杞64份、当归8份、地骨皮6份、砂仁4份、枳壳6份和甘草4份。
配方2:艾叶22份、玉米须20份、苎麻根20份、竹茹16份、黄芩14份、山药18份、枸杞14份、当归14份、地骨皮14份、砂仁14份、枳壳12份和甘草10份。
配方3:艾叶16份、玉米须14份、苎麻根14份、竹茹12份、黄芩10份、山药12份、枸杞10份、当归10份、地骨皮10份、砂仁9份、枳壳9份和甘草7份。
相应地,本发明还提供该药物制剂的制备方法,包括以下步骤:
S1:分别按比例称取各味药材,去杂质,洗净,烘干,切片后粉碎成粗粉,备用;
S2:取艾叶粗粉,置于超临界二氧化碳萃取装置中,加入粗粉总量30-45%体积分数为60-85%的乙醇,调控二氧化碳流量为15~20L/h,萃取压力为15~20MPa,萃取温度为40~60℃,萃取时间为1.5~2h,减压分离得到艾叶提取物,保留艾叶残渣;
S3:取玉米须、苎麻根、竹茹、黄芩、山药、枸杞、当归、地骨皮、砂仁、枳壳和甘草粗粉,合并艾叶残渣,加入药材总量10-12倍量的水和粗粉总量1-2.5%的纤维素酶,调节PH为4.50-5.0,在50-65℃下浸提3-4小时,然后在80-95℃回流提取3-5小时,过滤,滤液与银杏叶提取物合并,混合均匀后减压浓缩至60℃下相对密度为1.10-1.25的浸膏;
S4:将艾叶提取物与S3所得的浸膏混合,减压干燥,粉碎,过80-100目筛,即得。
进一步地,在中药原料药细粉中添加适当的辅料,以现代药物制剂常规工艺制成临床所需剂型,如胶囊剂、片剂、散剂、颗粒剂、口服液或丸剂。
本发明药物制剂所用组分的来源、性味、归经及功效:
艾叶:本品为菊科植物艾的干燥叶;味辛、苦,性温;归肝、脾、肾经;散寒止痛,温经止血。
安息香:本品为安息香科植物白花树的干燥树脂;味辛、苦,性平;归心、脾经;开窃清神,行气活血,止痛。
竹茹:本品为禾本科植物青秆竹、大头典竹或淡竹的茎秆的干燥中间层;味甘,性微寒;归肺、胃经;清热化痰,除烦止呕。
苎麻根:本品为荨麻科植物苎麻的根;味甘,性寒;清热利尿,安胎止血,解毒。
杜仲:本品为杜仲科植物杜仲的干燥树皮;味甘,性温;归肝、肾经;补肝肾,强筋骨,安胎。
地骨皮:本品为茄科植物枸杞或宁夏枸杞的干燥根皮;味甘,性寒;归肺、肝、肾经;凉血除蒸,清肺降火。
黄芩:本品为唇形科植物黄芩的干燥根;味苦,性寒;归肺、胆、脾、大肠、小肠经;清热燥湿,泻火解毒,止血,安胎。
云支花:本品为蓼科植物圆叶蓼的全草;味酸、涩,性平;归肝、肾经;活血调经。
当归:本品为伞形科植物当归的干燥根;味甘、辛,性温;归肝、心、脾经;补血活血,调经止痛,润肠通便。
陈皮:本品为芸香科植物橘及其栽培变种的干燥成熟果皮;味苦、辛,性温;归肺、脾经;理气健脾,燥湿化痰。
扁豆:本品为豆科植物扁豆的白色种子;味甘,性平;归脾、胃经;健脾和中,消暑化湿。
甘草:本品为豆科植物甘草、胀果甘草或光果甘草的干燥根;味甘,性平;归心、肺、脾、胃经;补脾益气,清热解毒,祛痰止咳,缓急止痛,调和诸药。
本发明药物制剂组方的配伍分析:
本发明药物制剂组方依据中医对治疗习惯性流产的辨证认知,遵循中药的配伍理论,为了制成方便易用且疗效稳定的制剂,强化了药材的使用量。该组方是以艾叶、安息香和苎麻根为君药,温经止血,开窃清神,安胎止血;以竹茹、杜仲、地骨皮和黄芩为臣药,除烦止呕,清热燥湿,安胎;以云支花、当归、陈皮和扁豆为佐药,活血调经,理气健脾,调经止痛,消暑化湿;以甘草为使药,调和诸药药性,诸药配合,相辅相成,协同作用,在治疗习惯性流产方面有十分显著的疗效。
与现有技术相比,本发明具有如下技术优势:
(1)与当前治疗习惯性流产的化学治疗药物相比,本发明药物制剂为天然纯药物制剂,不良反应和副作用显著降低,且本发明药物制剂作用全面,药物治疗效果更佳。
(2)药效学试验表明,本发明药物制剂能降低大鼠的流产率,效果比市售的阳性药保胎丸效果更优,临床试验结果表明,本发明药物制剂治疗习惯性流产效果显著,可作为临床治疗习惯性流产的药物使用,临床推广价值高。
具体实施方式
以下通过具体实施例进一步描述本发明,本发明不仅仅限于以下实施例,在本发明的范围内或者在不脱离本发明的内容、精神和范围内,对本发明进行的变更、组合或替换,对于本领域的技术人员来说是显而易见的,且包含在本发明的范围之内。
实施例1
本发明实施例1含艾叶的治疗习惯性流产的药物制剂由以下重量份的原料制备而成:
艾叶10份、玉米须10份、苎麻根8份、竹茹8份、黄芩8份、山药6份、枸杞64份、当归8份、地骨皮6份、砂仁4份、枳壳6份和甘草4份。
制备方法:
S1:分别按比例称取各味药材,去杂质,洗净,烘干,切片后粉碎成粗粉,备用;
S2:取艾叶粗粉,置于超临界二氧化碳萃取装置中,加入粗粉总量45%体积分数为75%的乙醇,调控二氧化碳流量为20L/h,萃取压力为20MPa,萃取温度为50℃,萃取时间为2h,减压分离得到艾叶提取物,保留艾叶残渣;
S3:取玉米须、苎麻根、竹茹、黄芩、山药、枸杞、当归、地骨皮、砂仁、枳壳和甘草粗粉,合并艾叶残渣,加入药材总量10倍量的水和粗粉总量1.5%的纤维素酶,调节PH为5.0,在65℃下浸提3小时,然后在85℃回流提取4小时,过滤,滤液与银杏叶提取物合并,混合均匀后减压浓缩至60℃下相对密度为1.10的浸膏;
S4:将艾叶提取物与S3所得的浸膏混合,减压干燥,粉碎,过100目筛,即得药物细粉;
S5:往所述药物细粉中加入适当辅料,利用中药制剂技术制成胶囊剂。
实施例2
本发明实施例2含艾叶的治疗习惯性流产的药物制剂由以下重量份的原料制备而成:
艾叶22份、玉米须20份、苎麻根20份、竹茹16份、黄芩14份、山药18份、枸杞14份、当归14份、地骨皮14份、砂仁14份、枳壳12份和甘草10份。
制备方法同实施例1。
实施例3
本发明实施例3含艾叶的治疗习惯性流产的药物制剂由以下重量份的原料制备而成:
艾叶16份、玉米须14份、苎麻根14份、竹茹12份、黄芩10份、山药12份、枸杞10份、当归10份、地骨皮10份、砂仁9份、枳壳9份和甘草7份。
制备方法同实施例1。
试验例一、本发明药物制剂对大鼠流产试验的观察
1、试验方法
取SPF级雌性Wistar大鼠50只,雄性30只,体重180-220g,随机分成5组,分别为空白对照组、阳性对照组、本发明实施例1药物胶囊高、中、低剂量组,每组雌性10只,雄性6只,各组分笼饲养、观察。从雌雄鼠同居开始,实施例1高、中、低剂量组给药剂量分别给予4.8g/kg、2.4g/kg和1.2g/kg。阳性对照组给予保胎丸,给药剂量为4.8g/kg,空白组给同体积蔗糖水。妊娠后期,雌鼠肌注催产素(2.5U/100g体重),注射2min后,见空白对照组的大鼠不安,起卧频繁,呼吸加快,舔前脚,努责;有的频频排尿,下腹部阵阵颤动,后肢开张,取产仔姿势。有的阴门处流有少量血样物,继而流产产仔,给药组也有上述类似反应症状,但与空白对照组比较反应较轻,在较短时间恢复近于正常。
2、试验结果
表1本发明药物制剂对大鼠流产的作用
由上表1可知,本发明实施例1制得的药物胶囊和保胎丸均能降低怀孕大鼠的流产率,其中本发明实施例1的药物胶囊高剂量组的效果更佳,仅有1只大鼠出现流产的表现,以上结果表明,本发明药物制剂对治疗流产具有一定的疗效。
试验例二、本发明药物制剂的临床效果试验
1、试验对象
筛选400名习惯性流产临床志愿患者,年龄25-40岁,平均年龄30岁。将所有志愿患者随机分为治疗A、B、C组和对照组,每组各100例,各组患者在年龄、病情等一般资料无明显差异,符合分组条件。
2、试验方法
治疗A、B、C组:分别服用本发明实施例1-3制备药物胶囊,每天2粒,每天2次,服用7天为一个疗程,共服用4个疗程;对照组:服用保胎灵胶囊(甘肃皇甫谧制药有限责任公司),每次2粒,每天2次,服用7天为一个疗程,共服用4个疗程。记录患者服药后的身体状况,未孕者记录例假情况,治疗过程中受孕者记录患者孕期反应情况、孕妇相关激素检查。
3、试验评价标准
(1)治愈:患者习惯性流产相关症状完全消失,顺利度过习惯性流产期,并生下健康婴儿。
(2)显效:习惯性流产相关症状部分消失,身体状况好转,但仍需进一步治疗。
(3)无效:习惯性流产相关症状没有任何改善,甚至更加严重。
4、试验结果
表2:本发明药物制剂治疗习惯性流产的临床疗效
由上表2可知,临床志愿患者服用本发明药物制剂的治愈率达75%以上,总有效率达90%以上,明显高于对照药的治愈率和总有效率,临床实验结果证明本发明药物制剂治疗习惯性流产效果显著,技术优势明显,故可作为临床治疗习惯性流产的药物使用,临床推广价值高。
由于已经通过以上较佳实施例描述了本发明,在本发明的精神和/或范围内,任何针对本发明的替换/或组合来实施本发明,对于本领域的技术人员来说都是显而易见的,且包含在本发明之中。
Claims (7)
1.一种含艾叶的治疗习惯性流产的药物制剂,其特征在于,所述的药物制剂由以下重量份的原料制备而成:
艾叶10-22份、玉米须10-0份、苎麻根8-20份、竹茹8-16份、黄芩8-14份、山药6-18份、枸杞6-14份、当归8-14份、地骨皮6-14份、砂仁4-14份、枳壳6-12份和甘草4-10份。
2.如权利要求1所述的含艾叶的治疗习惯性流产的药物制剂,其特征在于,所述的药物制剂由以下重量份的原料制备而成:
艾叶10份、玉米须10份、苎麻根8份、竹茹8份、黄芩8份、山药6份、枸杞64份、当归8份、地骨皮6份、砂仁4份、枳壳6份和甘草4份。
3.如权利要求1所述的含艾叶的治疗习惯性流产的药物制剂,其特征在于,所述的药物制剂由以下重量份的原料制备而成:艾叶22份、玉米须20份、苎麻根20份、竹茹16份、黄芩14份、山药18份、枸杞14份、当归14份、地骨皮14份、砂仁14份、枳壳12份和甘草10份。
4.如权利要求1所述的含艾叶的治疗习惯性流产的药物制剂,其特征在于,所述的药物制剂由以下重量份的原料制备而成:
艾叶16份、玉米须14份、苎麻根14份、竹茹12份、黄芩10份、山药12份、枸杞10份、当归10份、地骨皮10份、砂仁9份、枳壳9份和甘草7份。
5.如权利要求1-4任一所述的含艾叶的治疗习惯性流产的药物制剂,其特征在于,所述药物制剂的制备方法包括以下步骤:
