Abstract
本发明公开了一种治疗小儿咳嗽的药物组合物及其制备方法,本发明药物组合物是以柳叶菜、树扁竹、藤黄双黄酮、白矢车菊素为原料药,配比而成,可按常规制剂工艺制成各种剂型,治疗小儿咳嗽疗效显著。

本发明属于中药技术领域,尤其涉及一种治疗小儿咳嗽的药物组合物及其制备方法。
背景技术
小儿咳嗽是儿科临床上常见的疾病之一。一年四季都有可能发病,特别是在冬春季节,由于天气突变或小儿感冒等引起。小儿咳嗽大体上包括现代医学的急性支气管炎、支气管肺炎、哮喘等以咳嗽为主要症状的疾病,任何病因导致的呼吸道慢性、急性炎症都可能引起咳嗽。根据病程的长短可分为三类:急性咳嗽、亚急性咳嗽、慢性咳嗽。小儿形气未充,五脏娇嫩,肌肤柔弱,抵御外部侵袭的功能较弱,容易被燥、热、风、寒等外邪侵袭;小儿脾常不足,容易为乳食、生冷所伤,挑食,饮食不知节制,喜食肥腻,恶食蔬菜,导致体内食积。无论内伤或是外感,都会导致肺气失宣、津液不布、痰阻气道而引发咳嗽。小儿咳嗽注重预防。家长要注意小儿的身体锻炼,提高其抗病能力,避免因外感引发咳嗽。尽量少带小儿到人多的场合,避免与呼吸道传染疾病患者接触。实现小儿咳嗽的防治结合,提高治疗效果。
柳叶菜:柳叶菜科柳叶菜属植物柳叶菜EpilobiumhirsutumL.的全草。全年均可采,晒干。【性味】味苦;淡;性寒。【功能主治】清热解毒;利湿止泻;消食理气;活血接骨。主湿热泻痢;食积;脘腹胀痛;牙痛;月经不调;经闭;带下;跌打骨折;疮肿;烫火伤;疥疮。【化学成份】柳叶菜地上部分含没食子酸(gallicacid),3-甲氧基没食子酸(3-methoxygallicacid),原儿茶酸(Protocatechuicacid)和金丝桃甙(hyperoside)。另外本品还含山奈酚(kaempferol),槲皮素(quercetin),杨梅树皮素(myricetin),槲皮素-3-O-β-D-吡喃葡萄糖甙(quercetin-3-O-β-D-glucopyranoside),杨梅树皮素芸香糖甙(myricetinrubinoside)和异槲斗酸(isovalonicacid)。此外,叶和花还含有棕榈酸(palmiticacid),硬脂酸(stearicacid),亚油酸(linoleicacid),齐墩果酸(oleanolicacid),山楂酸(crategolicacid),委陵菜酸(tormenticacid),阿江榄仁酸(arjunolicacid)和23-羟基委陵菜酸(23-hydroxytormenticacid)。【药理作用】叶,体外对金黄色葡萄球菌有抑制作用,中毒可发生癫痫样惊厥与昏迷。收载于中药大辞典。
树扁竹:本品为兰科鸢尾兰属植物鸢尾兰Oberoniairidifolia(Roxb.)Lindl.[CymbidiumiridifoliumRoxb.]的全草。全年均可采收,开水烫后晒干。【性味】微苦;性凉。【归经】膀胱经。【功能主治】清热利湿;活血通络。主尿路感染;支气管炎;哮喘;肠炎;消化不良;跌打损伤;骨折。【原植物形态】鸢尾兰,多年生附生草本,高12-30cm,簇生多数须根。叶厚,两侧压扁并互相套叠,剑形或长圆状披针形,长4-13cm,宽1-2cm,先端有时稍歪斜,基部有关节。总状花序从植株中心抽出,远长于叶,直立,圆柱形,具多数密集的小花;小苞片薄革质,卵形,长1.5-2mm。花小,直径约2mm,白绿色;萼片与花瓣均反折,花瓣比萼片狭;唇瓣近半圆形,边缘近啮蚀状,先端2裂。蒴果倒卵状椭圆形,长约5mm,柄长1.5mm。花期10-11月,果期11-12月。收载于中药大辞典。
藤黄双黄酮(Morelloflavone):CAS号16851-21-1,分子式C30H20O11,分子量556.48。【药理作用】抗肿瘤。【成分来源】山竹子Garciniamultiflora,藤黄Garciniamorella。
白矢车菊素(Leucocyanidin):CAS号69256-15-1,分子式C15H14O7,分子量306.27。【药理作用】降糖,抑制眼醛糖还原酶。【成分来源】蓼科植物膜萼模根,戟叶酸膜;豆科阿拉伯相思树花,盾柱豆;番荔枝科;桃金娘科等植物。

本发明的目的是克服背景技术的不足,提供一种有效治疗小儿咳嗽的药物组合物及其制备方法。

柳叶菜140-150重量份树扁竹50-60重量份藤黄双黄酮12.2-13.4重量份白矢车菊素11.2-12.2重量份。
优选的用于治疗小儿咳嗽的药物组合物,是由如下重量份的原料药组成:
柳叶菜145重量份树扁竹55重量份藤黄双黄酮12.8重量份白矢车菊素11.7重量份。
一种治疗小儿咳嗽的药物组合物,其特征在于药物组合物可以采用制剂学的常规方法制备成片剂或胶囊剂或滴丸。

制备方法:按原料药配比取柳叶菜、树扁竹、藤黄双黄酮、白矢车菊素,混匀,用重量百分比浓度62%乙醇作为溶剂,在56℃温浸提取,提取次数为7次,每次提取时间为2.5小时,每次溶剂用量为原料药总重量的25倍,滤过,得药渣A和提取液,提取液回收乙醇,浓缩至相对密度1.16,滤过,药液通过LX-67大孔吸附树脂柱,先用水洗脱,再用重量百分比浓度90%的乙醇溶液洗脱LX-67大孔吸附树脂柱,收集重量百分比浓度90%乙醇洗脱液,回收乙醇,浓缩干燥,即得药物组合物。
实施例4:片剂的制备
取实施例1药物组合物63g,加入淀粉22g,混匀,制粒,干燥,加微晶纤维素14.5g,硬脂酸镁0.5g,混匀,压制成1000片,即得药物组合物片剂。
实施例5:胶囊的制备
取实施例2药物组合物53g,加入淀粉27g,混匀,制粒,干燥,整粒,加入适量硬脂酸镁,混匀,装胶囊1000粒,即得药物组合物胶囊。
实施例6:滴丸的制备
称取聚乙二醇6000132g水浴(80℃)加热煮熔,加入实施例3药物组合物14g,充分搅拌均匀,以液体石蜡为冷却剂,置玻璃管(4*80cm)中,冷却温度为8℃,滴口内外径为7.0/1.5(mm/mm),滴口距液面为1.8cm,滴速以每分44滴为最佳条件,用棉布吸干滴丸表面的冷凝剂,即得药物组合物滴丸。
实施例7:治疗小儿咳嗽的药物组合物
治疗小儿咳嗽的药物组合物的原料药的组成和重量份为:
柳叶菜13重量份藤黄双黄酮1.3重量份白矢车菊素3重量份。

树扁竹10重量份藤黄双黄酮4重量份白矢车菊素6重量份。

实验例1:治疗小儿咳嗽作用试验研究
1资料与方法
1.1一般资料
本组病例选自2011年6月至2012年6月在我市医院中医儿科治疗的咳嗽患儿,共40例,其中男20例,女20例;年龄为10个月~10岁,平均年龄为4.2岁;病程为1d~12d,平均病程为6.5d。有9例患儿伴有发热症状,体温为37.4~38.8℃。听诊发现:有18例患儿存在不同程度的肺呼吸音粗糙,5例患儿有气喘声和痰鸣音,8例患儿胸部可闻罗音。
1.2诊断标准
根据小儿咳嗽的诊断标准,诊断要点如下:多在冬春季节发生;肺部听诊发现呼气相延长,肺呼吸音粗糙,有哮鸣音或少量干性罗音;作胸X线片检查,可见肺纹理较粗糙;主要症状为咳嗽,多是在气候突变或感冒之后发生。排除标准:排除有其他呼吸系统合并症的患者。本组40例患者均符合以上标准。
1.3治疗方法
患儿分3组,分别采用药物组合物(实施例1药物组合物批号20110205;实施例2药物组合物批号20110206;实施例3药物组合物批号20110207)治疗,每天2g,可分多次服用。
2结果
2.1疗效评定标准
以治疗后咳嗽患儿的症状表现为疗效评定标准。无效:患者咳嗽症状未减轻,各项检查结果较治疗前无改观;好转:患者咳嗽症状减轻,痰量减少,各项检查结果有改善;治愈:患者咳嗽症状明显消失,体温、发热情况恢复正常,喉间无气喘声和痰鸣声,胸透视检查结果正常。
2.2疗效
本组40例咳嗽患儿中,采用药物组合物总体有效率为92.3%以上。其中总体有效率=(治愈例数+好转例数)/总例数×100%。见表1

2.3典型病例
患者,女,5岁。2012年2月20日医院就诊。因患儿持续发热、咳嗽3天,曾在某医院诊治,使用化痰止咳药物加抗生素治疗,患儿未见好转。患儿咳嗽初期,无痰,有清鼻涕流出,后开始出现发热,体温升高,咳嗽加剧,脸色潮红,纳食不佳,精神不振。舌诊:舌尖红,舌苔淡黄中部厚腻。脉诊:脉象浮数。各项检查:体温为38.9℃,咽红、充血,扁桃体Ⅱ度肿大。听诊结果:双肺呼吸音粗糙,可闻痰鸣音。采用药物组合物(实施例1药物组合物批号20110205)治疗,每天2g,分3次服用,当日热退,纳增,精神好转,但仍有轻微的咳嗽症状,继续服药2天后,痊愈。

制备方法:按原料药配比取柳叶菜、树扁竹、藤黄双黄酮、白矢车菊素,混匀,用重量百分比浓度62%乙醇作为溶剂,在56℃温浸提取,提取次数为7次,每次提取时间为2.5小时,每次溶剂用量为原料药总重量的25倍,滤过,得药渣A和提取液,提取液回收乙醇,浓缩至相对密度1.16,滤过,药液通过LX-67大孔吸附树脂柱,先用水洗脱,再用重量百分比浓度90%的乙醇溶液洗脱LX-67大孔吸附树脂柱,收集重量百分比浓度90%乙醇洗脱液,回收乙醇,浓缩干燥,即得药物组合物。
6.根据权利要求5所述一种治疗小儿咳嗽的药物组合物的制备方法,其特征在于按如下步骤制备:
原料药的组成和重量份为:柳叶菜145重量份树扁竹55重量份藤黄双黄酮12.8重量份白矢车菊素11.7重量份;
制备方法:按原料药配比取柳叶菜、树扁竹、藤黄双黄酮、白矢车菊素,混匀,用重量百分比浓度62%乙醇作为溶剂,在56℃温浸提取,提取次数为7次,每次提取时间为2.5小时,每次溶剂用量为原料药总重量的25倍,滤过,得药渣A和提取液,提取液回收乙醇,浓缩至相对密度1.16,滤过,药液通过LX-67大孔吸附树脂柱,先用水洗脱,再用重量百分比浓度90%的乙醇溶液洗脱LX-67大孔吸附树脂柱,收集重量百分比浓度90%乙醇洗脱液,回收乙醇,浓缩干燥,即得药物组合物。
7.根据权利要求5所述一种治疗小儿咳嗽的药物组合物的制备方法,其特征在于药物组合物可以采用制剂学的常规方法制备成片剂或胶囊剂或滴丸。
8.根据权利要求5所述一种治疗小儿咳嗽的药物组合物的制备方法,其特征在于药物组合物与化学药或中药组成治疗小儿咳嗽药物。
