背景

概述

此外,认为装置的纳米拓扑图和周围生物结构之间的相互作用可以促进携带生物活性剂的组合物穿过除皮肤屏障以外的其它天然屏障运送以全身递送。具体地,通过利用纳米结构经皮递送装置,通透性不仅在最接近的装置的接触区域中的组织中升高,而且在周围的组织中升高。认为升高的通透性不仅可以在与微针接触的细胞之间发生,而且该作用可以扰乱区域中的其它细胞,包括不同组织类型的细胞。这可以将提高的孔隙度作用转化到附近的结构和组织类型,这可以提高附近血管系统的孔隙度。
认为在装置和接触的组织之间的相互作用导致了皮肤组织的上皮紧密连接的重排,并且这引发了将类似作用传递给局部血管细胞的级联反应,所述细胞例如局部毛细血管的基底膜和内皮两者的细胞。这可以导致毛细血管壁的穿孔,允许生物活性剂直接进入心血管系统。这可以显著地升高受试者的系统对生物活性剂的摄入。
通过使用装置,可以改善包含一种或多种生物活性剂的高粘度组合物的递送。高粘度组合物的粘度可以是例如,大于约5厘泊、大于约10厘泊或大于约25厘泊。在一个实施方案中,组合物的粘度可以为约10厘泊至约50厘泊,例如,约30厘泊至约40厘泊。
组合物的粘度可以根据标准惯例来确定。例如,一种测量粘度的方法需要将活塞插入含有样品流体的密闭容器中,然后测量在容器中旋转活塞所需的力矩。虽然该方法对于测量更大的流体样品的粘度是适当的,但其提供了需要并消耗相对大体积的样品流体的缺点。这样的体积可能不可获得用于分析,特别是在生物分析的情况下。

组合物可以包含一种或多种乳化剂。如本文所使用,术语“乳化剂”一般地是指能够降低非极性和极性相之间的表面张力的药剂,并且包括定义为“自乳化”剂的化合物。适合的乳化剂可以来自任意种类的药学上可接受的乳化剂,包括糖类、蛋白、高分子量醇、润湿剂、蜡和磨碎的固体。当存在时,一种或多种乳化剂可以以按组合物重量计约1%至约15%,约1%至约12%,约1%至约10%,或约1%至约5%的总量存在于组合物中。
组合物可以通过药学、制药学、药物递送、药代动力学、医学或其它相关学科的领域的普通技术人员已知的任意技术制备,其包括将一种或多种赋形剂与治疗剂混合以形成组合物、药物递送系统或其组分。
经皮递送装置可以由多种材料构成,所述材料包括金属、陶瓷、半导体、有机物、聚合物等,以及其复合材料。以举例的方式,可以利用医药级不锈钢、钛、镍、铁、金、锡、铬、铜、这些或其它金属的合金、硅、二氧化硅和聚合物。通常地,装置由能够在表面上携带如本文所述的结构图案的生物相容的材料构成。术语“生物相容的”通常是指基本不会对装置所递送区域中的细胞和组织产生不良影响的材料。其也指不在存活受试者的任何其它区域中产生较大的医学上不良影响的材料。生物相容性材料可以是合成的或天然的。可生物降解的合适的生物相容性材料的一些实例包括羟基酸如乳酸和乙醇酸的聚合物、聚交酯、聚乙交酯、交酯-乙交酯共聚物、具有聚乙二醇的共聚物、聚酐、聚(原酸)酯、聚氨酯、聚(丁酸)、聚(戊酸)、和丙交酯-己内酯共聚物。其它合适的材料可以包括但不限于聚碳酸酯、聚甲基丙烯酸、乙烯醋酸乙烯酯、聚四氟乙烯和聚酯。装置在性质上同样地可以是非多孔或多孔的,在整个装置中关于材料、几何形状、坚固性等可以是均质或异质的,并且可以具有刚性固定的或半固定的形状。
图1说明了一个典型的微针经皮递送装置10。可看出该装置包括一排单个的微针12,将每个微针构造成一定尺寸和形状以便于使这些微针在单个微针不发生损坏的情况下穿透生物屏障。微针可以是图1所示的固体的,多孔的,或者可以包括空心部分。一个微针可以包括空心部分,例如,一个可以贯穿整个针或者部分针延伸的环形的孔,沿着与针平行的方向延伸,或者视情况在针的一侧分支或者开口。例如,图2说明了一个微针阵列14,在每个针的一侧均包含了一个通道16,这些通道用于例如向皮下部位递送药剂。例如,通道16可以至少部分与底部15上的孔对齐以在孔与通道16之间形成一个连接,所述连接可以允许物质通过通道16。
当存在时,可以特别地选择通道16的尺寸以诱导包含生物活性剂的组合物的毛细流动。通常当流体与通道壁之间的粘附力大于液体分子间粘合力时会发生毛细流动。具体而言,毛细管压力与通道16的横截面积成反比,而与液体的表面张力与流体和形成通道的材料接触时的接触角的余弦值的乘积成正比。因此,为了促进贴剂中的毛细流动,可以选择性控制通道16的横截面的尺寸(例如宽度、直径等),更小的尺寸通常会导致更高的毛细管压力。例如,在一些实施方案中,通道的横截面积的尺寸通常为约1微米至约100微米,在一些实施方案中,约5微米至约50微米,在一些实施方案中,约10微米至约30微米。通道16的尺寸可以是恒定的或者其可以根据通道16的长度发生改变。通道的长度也可以发生改变以适应药物化合物的不同体积、流速和滞留时间。例如,通道的长度可以为约10微米至约800微米,在一些实施方案中为约50微米至约500微米,在一些实施方案中为约100微米至约300微米。通道的横截面积也可以发生变化。例如,横截面积可以为约50平方微米至约1000平方微米,在一些实施方案中为约100平方微米至约500平方微米,在一些实施方案中,为约150平方微米至约350平方微米。另外,通道的纵横比(长度/横截面尺寸)可以为约1至约50,在一些实施方案中,为约5至约40,在一些实施方案中为约10至约20。在横截面尺寸(例如宽、直径等)和/或长度根据长度而发生改变的情况下,可以通过平均尺寸确定纵横比。
应当理解,在图中所示微针的数量仅仅是出于阐释的目的。在微针组装中使用的实际微针数量可以例如为约500个至约10,000个,在一些实施方案中,为约2,000至约8,000个,在一些实施方案中,为约4,000到约6,000个。
单个微针可以具有直的或锥形的轴。在一个实施方案中,微针的直径在微针底端是最大的,而在远离底部的一端逐渐变细成一点。也可制造微针使其含有同时包括直(非锥形的)的部分和锥形部分的轴。
可制造轴的横截面为圆形或非圆形的微针。例如,微针的横截面可以是多边形(例如,星形、正方形、三角形)、椭圆形或者任何其它形状。轴可以具有一个或者多个孔和/或通道。
可根据所需的靶向深度、避免在某特定组织类型中损坏的针的强度要求等对单个针的尺寸进行优化。例如经皮微针的横截面尺寸可以为约10纳米(nm)至1毫米(mm),或者为约1微米(μm)至约200微米,或者为约10微米至约100微米。外径可为约10微米至约100微米,空心针的内径可以为3微米至约80微米。一般尖端的半径小于或者等于1微米。
微针的长度一般取决于其所需的应用。例如,微针的长度可以为约1微米至约1毫米,例如,约500微米或者更短,或者约10微米至约500微米,或者约30微米至约200微米。
一个微针阵列并不需要包括彼此均相同的微针。阵列可以包括长度、外径、内径、横截面形状、纳米结构表面、和/或微针间距不同的微针混合。例如,微针可以按照统一的方式间隔,如按照矩形或者正方形网格或者同心圆的方式。间距取决于许多因素,包括微针的高度和宽度,以及预计穿过微针的任何物质的量和类型。虽然各种微针排列是有用的,但一个特别有用的微针排列是微针间“尖到尖”的间距为约50微米或者更大,在一些实施方案中,为约100到约800微米,在一些实施方案中,为约200到约600微米。
再次参照图1,可以将微针固定在基底20上(即与基底连接或者与基底成为一体)以使其方向与基底垂直或者成一定角度。在一个实施方案中,微针方向可以与基底垂直,每单位面积可提供密度更大的微针。然而,微针阵列可以包括微针方向、高度、材料或者其它参数不同的混合。可以用坚硬的或者柔软的金属、陶瓷、塑料或者其它材料的薄片构建基底20。基底20的厚度可以发生变化以满足装置的需要,例如约1000微米或者更薄,在一些实施方案中,为约1至约500微米,在一些实施方案中为约10至约200微米。
微针表面可以在其上以随机或有序的图案限定纳米拓扑图。图3示意性说明了两种代表性微针22的末端。微针22限定了中央孔24,可以用于通过微针22递送药剂。微针22的表面25限定了纳米拓扑图26。在这个具体的实施方案中,纳米拓扑图26在微针22的表面25上限定了随机的图案。
微针可以包括在表面上形成的多个相同的结构,或者可以包含由不同尺寸、形状及其组合构成的不同结构。结构的预定图案可以包括长度、直径、横截面形状和/或结构间距不同的结构的混合。例如,结构可以以统一的方式间隔,如以距形、正方形网格或同心圆的形式。在一个实施方案中,结构的尺寸和/或形状都可以不同,并且可以形成一个复杂的纳米拓扑图。例如,复杂的纳米拓扑图可以限定分形或者分形样的几何结构。
如本文所使用,术语“分形”通常是指具有在检测的最大和最小等级之间的所有测量等级的碎片形的几何或物理结构,以使所述结构的某些数学或物理特性表现为结构的维数好像大于空间维数。感兴趣的数学或物理特性可以包括,例如曲线的周长或多孔介质中的流速。分形的几何形状可以分割为部分,每个部分都限定了自相似性。此外,分形具有递归定义和在任意小的等级内具有精细结构。
如本文所使用,术语“分形样”通常是指具有一种或多种但不是全部分形特征的几何或物理结构。例如,分形样结构可包括包含自相似部分的几何形状,但是不包括在任意小的等级内的精细结构。在另一个实例中,分形样几何形状或物理结构可以不像分形那样在等级的迭代间可能不按比例减少(或增加),但是在图案几何形状的递归迭代间将增加或减少。分形样图案可比分形更简单。例如,它可以是规则的并且可用传统的欧氏几何语言相对简单地描述,而分形则不行。
限定了复杂纳米拓扑图的微针表面可以包括相同的一般形状的结构(例如柱状物)并且可将柱状物形成为不同的测量等级(例如,纳米级柱状物以及微米级柱状物)。在另一个实施方案中,微针所包括的表面结构可以在尺寸等级和形状上都不相同,或者仅仅是形状不同,但是都制造成相同的纳米级等级。另外,结构由有序排列或者随机分布构成。一般而言,至少一部分结构可以是在纳米级等级制造的纳米结构,例如,限定了横截面尺寸小于约500纳米,例如小于约400纳米,小于约250纳米,或者小于约100纳米。纳米结构的横截面尺寸通常可以大于约5纳米,例如大于约10纳米,或者大于约20纳米。例如,纳米结构的横截面尺寸可以为约5纳米至约500纳米,约20纳米至约400纳米,或约100纳米至约300纳米。在纳米结构的横截面尺寸根据纳米结构的高度变化的情况下,横截面的尺寸可以测定为纳米结构的底部到顶端的平均值,或者测定为结构的最大横截面尺寸,例如圆锥形纳米结构底部的横截面尺寸。
图4说明了一个可在表面上形成复杂的纳米拓扑图的实施方案。该具体的图案包括一个中心大柱状物100和以规则图案提供的尺寸较小的周围柱状物102、104。正如所见,该图案包括迭代的柱状物,每个柱状物由相同的一般形状形成,但是水平尺寸却不相同。该特定的复杂图案是分形样图案的一个实例,所述复杂图案不包括在逐次递归迭代间按比例的相同改变。例如,当柱状物102是第一纳米结构,其限定的水平尺寸是作为微米结构(microstructure)的较大柱状物100的约三分之一,柱状物104是第二纳米结构,其限定的水平尺寸大约是柱状物102的二分之一。
包括了不同尺寸结构的图案可以包括更大的结构,所述更大的结构具有在更大等级下形成的横截面尺寸,例如,横截面尺寸大于约500纳米的微米结构与较小的纳米结构的组合。在一个实施方案中,复杂纳米拓扑图的微米结构的横截面尺寸可以为约500纳米至约10微米,约600纳米至约1.5微米,或者约650纳米至约1.2微米。例如,图4中复杂的纳米拓扑图包括横截面尺寸约为1.2微米的微米级柱状物100。
当图案包含一个或者更多较大的微米结构时,例如,微米结构的横截面尺寸大于约500纳米,所述横截面尺寸是测得的结构的横截面尺寸的平均值或是结构的横截面尺寸的最大值,复杂的纳米拓扑图也将会包括纳米结构,例如,具有不同尺寸和/或形状的第一纳米结构和第二纳米结构等。例如,图4中复杂的纳米拓扑图柱状物102的横截面尺寸为约400纳米,柱状物104的横截面尺寸约200纳米。
纳米拓扑图可以由任意数量的不同元素构成。例如,元素图案可以包含两个不同的元素,在图4中所示的实例的三个不同的元素,四个不同的元素或者更多。每种不同元素重复的相对比例可以不同。在一个实施方案中,存在的图案的最小元素在数量上要比较大的元素多。例如在图4的图案中,每一个柱状物102都有八个柱状物104,并且中央大柱状物100的周围有八个柱状物102。当元素的尺寸增加的时候,通常在纳米拓扑图中会有更少的元素的重复。通过举例的方式,横截面尺寸约为第二个较大元素的0.5倍,例如约0.3至约0.7倍的第一个元素在拓扑图中是第二个元素的约五倍或者更多。横截面尺寸约为第二个较大元素的0.25倍,或者约0.15到约0.3倍的第一个元素,在拓扑图中是第二个元素的10倍或者更多。
单个元素的间距也可以变化。例如,单个结构的中心间距可以为约50纳米至约1微米,例如约100纳米至约500纳米。例如,结构的中心间距可以是纳米级的。例如,当考虑纳米级结构的间距时,结构的中心间距可以小于约500纳米。然而这不是拓扑图的必要条件,单个结构之间可以离的更远。结构的中心间距可以随着结构的尺寸而变化。例如,两个相邻结构的横截面尺寸的平均值与这两个结构的中心间距的比值可以为约1:1(例如接触)至约1:4,约1:1.5至约1:3.5或者约1:2至约1:3。例如,中心间距可以约为两个相邻结构的横截面尺寸的平均值的2倍。在一个实施方案中,各自的横截面尺寸为约200纳米的两个相邻的结构的中心间距可以为约400纳米。因此,在该情况下,平均直径与中心间距的比值是1:2。
结构间距可以是相同的,即等距离的,或者可以根据图案中的结构变化。例如,图案的最小结构可以间隔第一距离,图案的这些最小结构和较大结构之间的距离或者图案的两个较大结构之间的距离可以与第一距离相同或不同。
例如在图4的图案中,最小的结构104的中心间距为约200纳米。较大的柱状物102和每个周围的柱状物104间的距离较小,为约100纳米。最大柱状物100和每个周围柱状物104之间的距离也小于最小柱状物104之间的中心间距,为约100纳米。当然,这并不是必须的,所有的结构彼此间可以是等距离的或者是任意变化距离的。在一个实施方案中,不同的结构之间可以相互接触,例如,如下面进一步讨论的在彼此之上,或彼此相邻并且彼此接触。
拓扑图的结构可以全部形成为相同的高度,通常为约10纳米至约1微米,但是这不是必须的,图案的单个结构的尺寸可以在一维、二维或者三维内变化。在一个实施方案中,拓扑图的某些或者全部结构的高度可以低于约20微米、低于约10微米或者低于约1微米,例如低于约750纳米、低于约680纳米或者低于约500纳米。例如,结构的高度可以为约50纳米至约20微米或者为约100纳米至约700纳米。例如,纳米结构或者微米结构的高度可以为约20nm至500nm,约30nm至约300nm,约100nm至约200nm,但应懂得结构的横截面尺寸可以是纳米级的,并且具有可以在微米级测量的高度,例如大于约500nm。微米级结构的高度可以与相同图案的纳米级结构的高度相同或者不同。例如,微米级结构的高度可以为约500纳米至约20微米,或者在另一个实施方案中,为约1微米至约10微米。微米级结构的横截面尺寸也可以是微米级的,大于约500nm,并且高度也可以是纳米级的,小于约500nm。
结构的纵横比(结构的高度和结构的横截面尺寸的比值)可以为约0.15到约30,为约0.2到约5,约0.5到约3.5,或者约1到约2.5。例如,纳米结构的纵横比可以落入这些范围内。
如图4所示,装置的表面可以包括单一的图案实例,或可以包括多个相同或不同图案的叠加。例如图5示出了在表面包括图4的图案的多个叠加的表面图案。
在表面上纳米拓扑图的形成可以在不相应增加体积的情况下增加表面积。认为表面积和体积的比值增加可以改善表面与周围生物材料的相互作用。例如,认为表面积和体积的比值增加可以促进纳米拓扑图与周围蛋白质,例如细胞外基质(ECM)蛋白和/或质膜蛋白的机械相互作用。
一般而言,装置的表面积和体积的比值可以大于约10,000cm-1、大于约150,000cm-1或者大于约750,000cm-1。可以根据本领域已知的任意标准方法进行表面积和体积的比值的测定。例如,如本领域所公知以及Brunauer、Emmet和Teller(J.Amer.Chem.Soc,vol.60,Feb.,1938,pp.309-319)所描述,表面的特定表面积可以通过物理气体吸附法(B.E.T法)获得,该方法使用氮气作为吸附气体,将所述参考文献引入本文作为参考。BET的表面积可以小于约5m2/g,在一个实施方案中,例如为约0.1m2/g到约4.5m2/g,或者约0.5m2/g到约3.5m2/g。也可以根据标准的几何运算,通过用于构造表面的模具的几何结构来估算表面积和体积值。例如,在给定区域内,例如单个微针的表面,可以根据每个图案元素的计算体积和图案元素的总数来估算体积。
对于在表面限定了复杂图案的纳米拓扑图的装置,可通过测定图案的分形维数(fractal dimension)来表征所述纳米拓扑图。分形维数为统计学的量,其给出当递归迭代等级持续越来越小时,分形呈现填充空间的完整性的指示。二维结构的分形维数可表示为:

例如,当考虑如图6所示的被称为Sierpenski三角的二维分形时,其中连接等边三角形的三条边的中点并将形成的内部三角形移除,分形维数的计算方法如下:

因此,Sierpenski三角分形显示初始二维等边三角形线长度增加。此外,该线长度的增加并未伴随面积的相应增加。
图4中所示的图案的分形维数约为1.84。在一个实施方案中,装置表面的纳米拓扑图可以显示分形维数大于约1,例如为约1.2至约5,约1.5至约3或者约1.5至约2.5。
图7A和7B阐述了复杂纳米拓扑图的另一个实例的递增放大图像。图7A和7B的纳米拓扑图包括一系列位于基底上的纤维样柱状物70。在每个单个柱状物的远端,柱状物分裂成多个较小的纤维60。在这些较小纤维60的远端,每条纤维又分裂成多条丝(在图7A和7B中不可见)。在表面上形成的纵横比大于约1的结构可以是柔软的,如同图7A和7B中所示的结构,或者可以是坚硬的。
图7C和7D说明了另一个复杂纳米拓扑图的实例。在这个实施方案中,在基底上形成有多个柱状物72,每个柱状物72各包括一个贯穿其中的环形的空心71。在每个空心柱状物的远端形成多个较小的柱状物62。如同所见的一样,图7C和7D所示的柱状物保持其硬度和直立的方向。另外,与前面的图案相比,该实施方案中较小的柱状物62与较大的柱状物72形状不同。具体而言,较小的柱状物62不是空心的,而是实心的。因此,含有不同等级的结构的纳米拓扑图并不需要所有的结构都具有相同的形状,结构与不同等级的结构的尺寸和形状都可以不同。
图8说明了可以在装置表面上形成的另一个包括纳米级结构的图案。如可以看到的,在该实施方案中,单个图案结构可以具有相同的一般尺寸,但是彼此间的方向和形状却不相同。
除了上述的那些方法或供选择地,可通过其它方法来表征表面,包括但不限于表面粗糙度、弹性模量和表面能。
本领域公知测定表面粗糙度的方法。例如,根据标准操作,原子力显微镜法的接触模式和非接触模式可以用来测定材料的表面粗糙度。可以用来表征微针的表面粗糙度可包括平均粗糙度(RA)、均方根粗糙度、偏斜度和/或峰度。一般而言,具有在其上制造的纳米拓扑图的表面的平均表面粗糙度(即,表面的算术平均高度是在ISO25178系列中定义的粗糙度的参数)可以小于约200纳米,小于约190纳米,小于约100纳米,或者小于约50纳米。例如平均表面粗糙度可以为约10纳米至约200纳米,或者约50纳米至约190纳米。
装置可以用纳米图案化表面的弹性模量进行表征,例如,通过在表面增加纳米拓扑图后弹性模量的改变进行表征。一般而言,由于在表面上增加纳米级结构会导致表面连续性降低和表面积的相关变化,因此在表面上增加多个构成纳米拓扑图的结构可以降低材料的弹性模量。与使用相同材料和相同工艺构成的但表面上无纳米拓扑图图案的相似的表面相比,其上包括纳米拓扑图的装置可以显示弹性模量降低约35%到约99%,例如约50%到约99%,或者约75%到约80%。通过举例的方式,具有纳米图案的表面的有效压缩模量可以小于约50MPa,或者小于约20MPa。在一个实施方案中,有效压缩模量可以为约0.2MPa至约50MPa,约5MPa至约35MPa,或者约10MPa至约20MPa。有效的剪切模量可以小于约320MPa,或者小于约220MPa。例如,在一个实施方案中,有效的剪切模量可以为约4MPa至约320MPa,或者约50MPa至约250MPa。
与不具有在其上限定纳米拓扑图图案的表面的相似的微针相比,在其上包括纳米拓扑图的装置还可以显示表面能升高。例如,与根据相同方法和相同材料制得的但表面上不包括纳米拓扑图图案的相似微针相比,包括在其上形成的纳米拓扑图的微针会显示表面能升高。例如,在其上包括纳米拓扑图的表面的水接触角可以大于约80°,大于约90°,大于约100°或者大于约110°。例如,在一个实施方案中,表面的水接触角可以为约80°至约150°,约90°至约130°,或约100°至约120°。
当在装置表面上形成纳米结构时,结构的堆积密度可以最大化。例如,正方形堆积(图9A)、六边形堆积(图9B),或其一些变化可以用于使元素在基底上形成图案。当在基底上设计横截面A、B和C的各种尺寸的元素彼此邻近的图案时,就可以使用图9C所示的环形堆积。当然,改变堆积密度或者测定表面特征的相关变化在本领域技术人员的能力范围内。
在装置表面上包括制造的纳米拓扑图的装置可以通过一步法构成。供选择地,可以使用多步法进行制造,其中在预形成的表面上制造纳米结构图案。例如,首先形成微针阵列,然后在所形成的微针的表面上制造随机或非随机纳米结构图案。在一步法或两步法中,都可以根据任何合适的纳米拓扑图制造方法将结构制造在表面上或者模具表面上,所述合适的纳米拓扑图制造方法包括但不限于纳米压印,射出成型,石印术(lithography),压花成型等。
一般地,可以根据任何标准的微制造技术形成微针阵列,所述微制造技术包括但不限于石印术;蚀刻技术,如湿化学、干法、和光阻去除;硅热氧化;电镀和化学镀;扩散过程,如硼、磷、砷和锑扩散;离子注入;膜沉积,如蒸发(细丝、电子束、闪光、和遮蔽和阶梯覆盖)、溅射法,化学气相沉积(CVD)、外延附生(epitaxy)(气相,液相,和分子束)、电镀、丝网印刷、层压、立体石印术、激光加工和激光消融(包括投影消融)。
石印术技术,包括光刻法、电子束石印术(e-beam lithography)、X射线石印术(X-ray lithography)等,可用于初级图案的界定以及母模(master die)的形成。然后进行复制以形成含有微针阵列的装置。通常的复制方法包括但不限于溶剂辅助微成型和铸造、压花成型、射出成型等。自组装技术,包括分相嵌段共聚物、聚合物分层和胶体石印术技术,也可以用于形成表面上的纳米拓扑图。
如所知的,可以使用方法的组合。例如,以胶体形成图案的基底可以暴露于反应离子蚀刻中(RIE,也称为干法蚀刻)以便于改进制造的纳米结构的特征,如纳米柱的直径、外形、高度、间距等。对于根据不同工艺例如聚合物分层技术最初形成的制造的纳米结构,也可以采用湿法蚀刻技术来制造供选择的外形。可以通过选择合适的材料和方法控制结构的直径、形状和间距。
用于形成表面上包括制造的纳米拓扑图的微针的其它方法包括利用超高精度的激光加工技术进行的纳米压印石印术法,对其实例的描述见Hunt等人(第6,995,336号美国专利)和Guo等人(第7,374,864号美国专利),将两者的内容引入本文作为参考。纳米压印石印术是纳米级石印术技术,其中利用可充当纳米压印石印术模具和光刻掩模的混合模具。纳米压印石印术技术的示意图如图10A-10C所示。在制造过程中,通过施加压力将混合模具30压印到基底32中,以在抗蚀层上形成特征(例如,具有纳米拓扑图的微针)(图10A)。一般而言,在与模具30接合之前可将基底32的表面加热到其玻璃化温度(Tg)以上的温度。当混合模具30与基底32接合后,迫使粘性聚合物流进入模具腔内形成特征34(图10B)。随后,模具和基底可以暴露在紫外光下。除了某些被遮蔽的区域外,通常紫外辐射可透过混合模具。因此,紫外辐射穿过可透过部分并且进入抗蚀层。在模具和基底的冷却过程中维持压力。随后,在温度低于基底和聚合物的Tg时(图10C),将混合模具30从冷却的基底32上除去。
如图10C所示,为了促进含有制造特征34的纳米压印的基底32从模具30中释放,以低能量的涂层处理模具30以降低与基底32的粘附是有利的,这是由于模具30的较低的表面能和所产生的在模具30、基底32和聚合物之间的更大的表面能差异可以在材料之间更容易释放。通过举例的方式,可以使用硅模具涂层,如十三-(1,1,2,2-四氢)-辛基三氯硅烷(F13-TCS)涂层。
也可以根据化学添加过程形成结构。例如,膜沉积法、溅射法、化学气相沉积法(CVD);外延附生(气相,液相和分子束)、电镀等可以用于构建表面上的结构。如本领域公知的自组装单层工艺可以用于形成表面上的结构图案。
可以对经皮递送装置的表面进一步进行化学官能化以提高在使用过程中与组织或者单个细胞的相互作用。例如,一个或者多个生物分子,如多核苷酸、多肽、全蛋白、多糖等在使用前可以结合于结构化表面。
在一些实施方案中,包括在其上形成的结构的表面可以已经包含适合的反应性,以使其它所需的官能团可以自发的连接于表面,而没有必要进行表面预处理。然而,在其它实施方案中,在所需化合物连接之前可以对结构化表面进行预处理。例如,通过在表面上增加或者产生氨基、羧酸、羟基、醛、巯基、或酯基基团可以增加结构表面的反应性。在一个代表性的实施方案中,可以通过与含有氨基的化合物如3-氨基丙基三乙氧基硅烷接触对包括在其上形成的纳米结构图案的微针表面进行胺化,以增加表面的氨基官能团,并且可通过添加的氨基官能团在所述表面结合一个或者多个生物分子。
希望结合于图案化装置表面的材料可以包括ECM蛋白如层粘连蛋白、弹性蛋白原或弹性蛋白、原胶原蛋白或胶原蛋白、纤连蛋白等。短多肽片段可以结合于所述图案化装置的表面,如RGD序列,其为结合于许多ECM蛋白的整合素的识别序列的一部分。因此,具有RGD的微针表面的官能化可以促进所述装置与ECM蛋白质的相互作用,并进一步限制在使用过程中针对所述装置的异物反应。
经皮递送装置可以是贴剂的形式,其可以包括多个特征。例如,装置可以包括储层,例如,容器、多孔基质等,所述储层可以储存药剂并提供药剂以进行递送。装置可以包括存在于所述装置自身内部的储层。例如,所述装置可以包含可携带用于递送的一种或者多种药剂的空心或者多个孔。药剂可以通过部分或者整个装置的降解或通过药剂从装置中扩散而从装置释放。
图11A和11B是含有储层的装置的透视图。装置110包含储层112,所述储层112由不透液的背层114和微针阵列116构成。背层和微针阵列116在所述装置的外周处连接在一起,如图118所示。不透液的背层114可以通过粘合剂、热封等连接。装置110也包含多个微针120。在使用所述装置之前可以除去释放衬垫122以暴露微针120。
包含一种或者多种药剂的制剂可以保存在储层112中。适合用作不透液背层114的材料可以包括材料如聚酯、聚乙烯、聚丙烯和其它合成聚合物。材料通常是可热封或者可以其它方式密封到背层,以提供储层的内容物横向流动的屏障。
由不透液背层114和微针阵列116之间的空间和间隙限定的储层112提供了保留待给予的药剂的悬浮液的储存结构。所述储层由各种能够与包含在其中的药物相容的材料构成。通过举例的方式,天然的以及合成的聚合物、金属、陶瓷、半导体材料以及其复合物均可以构成储层。
在一个实施方案中,储层可以连接在微针所处的基底上。根据另一个实施方案,储层可以是独立的,并且可拆卸地与微针阵列连接,或者例如通过合适的管道、路厄锁(leurlocks)等与微针阵列流体连通。
所述装置可以包含一个或者多个储层以贮存被递送的药物。例如,所述装置可以包括储存一种或者多种包含药剂的制剂的单一的储层,或者所述装置可以包括多个储层,其中每个储层都储存向所有或者部分微针阵列递送的一种或者多种药物。多个储层可各自储存不同的可以联合递送的材料。例如,第一储层可以含有药剂,例如药物,第二储层可以包含溶媒,例如盐溶液。不同的药物可以在递送之前混合。可以用任何方式进行混合,包括例如,对分离腔室的壁或膜进行机械破碎(即穿刺、降解、或断裂)、改变孔隙率、或电化学降解。多个储层可包括不同的用于递送的活性药物,可以互相结合递送或顺序递送。
在一个实施方案中,储层可以与经皮装置的一个或者多个微针流体连通,微针可以具有允许将递送的试剂运输至屏障层以下的结构(例如中央孔或者侧孔)。
在供选择的实施方案中,装置可以包含微针组件和储层组件,在使用前两者之间具有止流阀。例如,装置可以包括安置在邻近储层和微针阵列的释放元件。在使用前释放元件可与装置分离以使在使用过程中储层和微针阵列互相流体连通。可以通过部分或者完全拆卸释放元件来实现分离。例如,参照图12-17,显示了释放元件的一个实施方案,将所述释放元件构造成可从经皮贴剂上分离以启动药物化合物的流动。更具体地,图12-17显示了包含药物递送组件370和微针组件380的经皮贴剂300。药物递送组件370包含位于速率控制膜308邻近处的储层306。
速率控制膜可以有助于降低药物化合物释放后的流速。具体地,经微流体通道从药物储层到达微针组件的流体药物化合物会经历压力下降,导致流速下降。如果该差异过大,就会产生一些反压,从而阻碍化合物流动并且可能抵消在通过微流体通道时流体的毛细管压力。因此,使用速率控制膜可以改善该压力差,并且使药物化合物以更加可控的流速引入微针中。速度控制膜的具体材料、厚度等可以根据多种因素发生变化,如药物化合物的粘度,所需的递送时间等。
速率控制膜可以由本领域已知的通透性的、半通透性的或者微孔材料制造以控制药物化合物的流速,对渗透促进剂的通透性要低于药物储层的对渗透促进剂的通透性。例如,用于形成速率控制膜的材料的平均孔径为约50纳米至约5微米,在一些实施方案中,约100纳米至约2微米,在一些实施方案中,约300纳米至约1微米(例如约600纳米)。合适的膜材料包括,例如,纤维网(例如织造或者非织造)、带孔薄膜、泡沫、海绵等,所述材料由聚合物形成,例如聚乙烯、聚丙烯、聚乙酸乙烯酯、乙烯正丁基乙酸酯和乙烯乙酸乙烯酯共聚物。在第3,797,494号、第4,031,894号、第4,201,211号、第4,379,454号、第4,436,741号、第4,588,580号、第4,615,699号、第4,661,105号、第4,681,584号、第4,698,062号、第4,725,272号、第4,832,953号、第4,908,027号、第5,004,610号、第5,310,559号、第5,342,623号、第5,344,656号、第5,364,630号、和第6,375,978号美国专利中更详细地描述了这种膜材料,出于所有相关目的将其全部内容引入本文作为参考。一种特别合适的膜材料来自LohmannTherapie-Systeme。
参照图12-13,虽然是任选的,但组件370也可以含有与储层306邻近的粘合层304。所述微针组件380同样含有支持物312,从所述支持物中延伸出多个含有通道331的微针330,如上面所描述的那样。如果需要,使用任何已知的粘合技术如粘合剂粘合、热粘合、超声波粘合等将药物递送层组件370和/或微针组件380连接在一起。
无论采用何种特殊的结构,贴剂300还含有位于药物递送组件370和微针组件380之间的释放元件310。虽然释放元件310可任选地与相邻的所述支持物312和/或速率控制膜308粘合,但通常期望其尽可能为轻度粘合,以使释放元件310可以容易地从贴剂300中取出。如果需要,释放组件310也可以含有拉环部分(tab portion)371(图12-13),所述拉环部分向外延伸至至少部分超过贴剂300的周边,以使用户能够容易地抓住该元件并按照所需的方向拉出。如图12-13所示的“不活动”结构中,所述贴剂300的药物递送组件370安全地保留药物化合物307以使其不会在任意明显的程度上流入微针330。通过简单的对释放元件施力使其脱离贴剂,就可以“激活”贴剂。
参照图14-15,显示了激活贴剂300的一个实施方案,其中纵向牵拉释放元件310。如图16-17所示,可以移除整个释放元件310,或者如图14-15所示可简单地对其进行部分剥离。然而,无论在哪种情况下,先前在释放元件310和支持物312上的孔(未示出)之间形成的密封均会被破坏。药物化合物107以这种方式可以开始从药物递送组件170中流出,通过支持物112流入微针130的通道131中。图16-17显示了药物化合物307是如何从储层306流出并进入通道331的。值得注意的是,药物化合物307的流动是被动启动的并不需要任何主动排液机制(例如,泵)。
在图12-17所示的实施方案中,由于已经处理药物递送组件使其和微针组件流体连通,释放元件的脱离可以立刻启动药物化合物向微针流动。然而在某些实施方案中,很希望使用者能更大程度的控制药物化合物释放的时间。这一需求可以通过使用贴剂结构实现,其中所述贴剂结构中的微针组件起初和药物递送组件并不是流体连通的。当需要使用贴剂时,使用者可以物理操作使两个独立的组件流体连通。可以在这种物理操作之前或者之后分离释放元件。
例如,参照图18-23,显示了贴剂200的具体实施方案。图18-19描述了使用前的贴剂200,并显示了由微针组件280构成的的第一部分250和由药物递送组件270构成的第二部分260。药物递送组件270包括置于上述速率控制膜208邻近处的储层206。虽然是任选的,但组件270也包括置于储层206邻近处的粘附层204。另外,微针组件280同样地包括支持物212,从所述支持物向外延伸多个如上所述的具有通道231的微针230。
在这个实施方案中,支持物212和速率控制膜208起初水平放置相互邻近,释放元件210延伸越过支持物212以及速率控制膜208。在该具体的实施方案中,通常期望用粘合剂(例如,压敏粘合剂)将释放元件210可释放地连接于支持物212和速率控制膜208上。如图18-19所示,在其“不活动”结构中,贴剂200的药物递送组件270安全的保存药物化合物207使其不能以任何明显的程度流入微针230。如图20-21所示,当需要“激活”贴剂时,剥离并移除释放元件210以破坏先前在释放元件210和支持物212的孔(未显示)之间形成的密封。之后,沿图22中方向箭头所示的折线“F”折叠第二部分260以使速率控制元件208垂直置于支持物212的邻近处,并与其流体连通。供选择地,也可以折叠第一部分250。无论如何,折叠部分250和/或260均可使药物化合物207开始从药物递送组件270中流出并通过支持物212进入微针230的通道231中(参见图23)。
所述装置可以采用对治疗有益的速度递送药物。根据这个目标,所述经皮装置可以包含具有微电子和其它微机械结构的壳体,以便根据预先设定的时间表或者通过与患者、医疗保健专家主动交流、或者生物传感器来控制递送速率。所述装置可以在表面包括具有预定降解速率的材料,以便控制装置中所包含的药剂的释放。可以通过控制多种因素来控制递送速率,所述因素包括待递送的制剂的特征(例如粘度、电荷、和/或化学组分);每个装置的尺寸(例如,外径和任意开口的体积);经皮贴剂上微针的数量;在承载基质上单个装置的数量;驱动力的施加(例如浓度梯度、电压梯度、压力梯度);阀门的使用等。
可以使用例如阀门、泵、传感器、致动器和微处理器的各种组合控制或者监控药物通过所述装置的运输。可以使用标准的制造或者微制造技术制造这些组件。装置可用的驱动器可包括微泵、微型阀和定位器。例如可以对微处理器预先编程以便控制泵或者阀,从而控制递送速率。
药物在装置内的流动可以通过扩散作用或者毛细管作用发生,或者使用常规的机械泵或者非机械驱动力如电渗或电泳或对流进行诱导。例如,在电渗中,将电极置于生物表面(例如皮肤表面)、微针和/或邻近微针的基底上,以形成朝向或进入递送位点的携带相反电荷的离子种类和/或中性分子的对流流动。
可以通过选择形成微针表面的材料来操控药物的流动。例如,邻近装置微针表面的一个或者多个大凹槽可以被用来引导药物,特别是液态药物的通道。供选择地,可以操控装置的物理表面性质以促进或者抑制材料沿表面的运输,例如通过控制亲水性或者疏水性。
可以使用本领域已知的阀门或者闸门调节药物的流动。阀门可以重复开关,或者可以是一次性使用的阀门。例如,可以将可破坏性屏障或者单通道闸门安装在储层和图案化的表面之间的装置中。在准备使用时,可以破坏屏障或者打开闸门以允许液流通过微针表面。其它用于装置的阀门或者闸门可以通过热激活、电化学激活、机械激活或者磁力激活,以选择性地启动、调控或者阻滞分子通过所述装置流动。在一个实施方案中,通过使用作为“阀门”的限速膜控制流动。
一般而言,任何本领域已知的药物递送控制系统,包括储层、流量控制系统、感测系统等都可以引入装置中。通过实例的方式,第7,250,037号、第7,315,758号、第7,429,258号、第7,582,069号和第7,611,481号美国专利描述了可以引入装置中的储层和控制系统。
通过参照下面提供的实施例可更好地理解本主题。
实施例1
使用与电路设计和制造中所采用的相似的光刻技术制造几个不同的模具。本领域公知并且已经描述了单个处理步骤。
首先,通过使用丙酮、甲醇和异丙醇清洗来制造硅基底,然后根据化学气相沉积法涂覆一层258纳米(nm)的二氧化硅。
利用JEOL JBX-9300FS EBL系统,通过本领域已知的电子束石印术制图工艺在每个基底上形成一个图案。工艺条件如下:
电子束电流=11nA
加速电压=100kV
射距(Shot pitch)=14nm
剂量=260μC/cm2
抗蚀剂=ZEP520A,~330nm厚
显影剂=乙酸正戊酯
显影=浸泡2min,接着用异丙醇漂洗30秒。
然后采用STS先进氧化物蚀刻(Advanced Oxide Etch)(AOE)进行二氧化硅蚀刻。利用55标准立方厘米每分钟(sccm)的He、22sccm的CF4、20sccm的C4F8,在4mTorr,400W线圈功率、200W RIE和404-411V的直流偏压下蚀刻50秒。
随后,使用STS氧化硅蚀刻(SOE)进行硅蚀刻。使用20sccm Cl2和5sccm Ar,在5mTorr,600W线圈功率,50W RIE和96-102V的直流偏压下蚀刻2分钟。硅蚀刻深度为500纳米。
使用缓冲氧化物蚀刻剂(BOE)去除残留氧化物,包括在BOE中浸泡3分钟,接着用去离子水漂洗。
使用Obducat纳米压印机在多种聚合物基底上形成纳米图案。外部水用作冷却剂。UV模块使用波长为200-1000纳米,1.8W/cm2的单脉冲灯。使用250-400纳米的UV滤光器。曝光区为6英寸,最高温度200℃和80巴。所述纳米压印机包括半自动的分离单元和自动控制的脱模。
为了促进纳米压印的膜从模具中释放,使用十三-(1,1,2,2-四氢)辛基三氯硅烷(F13-TCS)处理模具。为了处理模具,首先使用丙酮、甲醇和异丙醇洗液清洗硅模,并用氮气干燥。在氮气气氛中,在热板上放置Petri培养皿并在Petri培养皿中加入1-5ml的F13-TCS。将硅模置于Petri培养皿中并覆盖10-15分钟以便在取出模具之前使F13-TCS蒸汽浸湿硅模。
如下,表1中给出了用于形成各种纳米拓扑图设计的5种不同的聚合物。

形成了几种不同的纳米拓扑图图案,其示意性显示于图24A-24D中。图24E中示出的纳米拓扑图图案是购自NTT Advanced Technology of Tokyo,Japan的平面基底表面。所述图案命名为DN1(图24A)、DN2(图24B)、DN3(图24C)、DN4(图24D)和NTTAT2(图24E)。模具的SEM图像显示于图24A、24B和24C中,膜的图像显示于图24D和24E中。图8示出了使用图24A的模具(DN1)形成的纳米图案膜。在该具体的膜中,通过之前所讨论的温度变化绘制了聚合物的特征。发现图24E的图案的表面粗糙度为34纳米。
图7C和7D中所示的图案也是根据这种纳米压印工艺形成的。如图所示,该图案包括柱状物72和柱状物62。形成的较大的柱状物72的直径为3.5微米(μm),高30μm,中心间距6.8μm。柱状物62高500纳米,直径200纳米,中心间距为250纳米。
在如下表2中提供了聚丙烯膜使用的纳米压印工艺的条件。

实施例2
如以上实施例1所述形成膜,其包含各种不同的图案并由聚苯乙烯(PS)或聚丙烯(PP)形成。下层的基底厚度不同。使用的图案为利用如实施例1中所述的成形工艺的DN2、DN3或DN4。图案模具具有不同的孔深度和特征间距以形成具有指定图案的各种大小不同的特征。通过使用0.6μm微孔聚碳酸酯滤器作为模具形成8号样品(称为BB1)。将25μm聚丙烯膜覆盖于所述滤器顶部,然后加热熔融以使聚丙烯可以流入滤器的孔中。然后冷却模具并用二氯甲烷溶剂溶解聚碳酸酯模具。
图25-33显示了形成膜的SEM,下表3中总结了形成的膜的特征。
对于每种样品,采用AFM对膜进行表征。表征包括形成扫描电子显微镜照片(SEM)、测定表面粗糙度、测定最大检测特征高度、以及测定分形维数。
使用的原子力显微镜(AFM)探针是一系列16硅探针,悬臂可购自μΜasch。悬臂的共振频率为170kHz,弹簧常数为40N/m,长度为230±5μm,宽度为40±3μm,厚度为7.0±0.5μm。探针尖端是n-型的掺磷硅探针,标准探针尖端半径为10纳米,全尖端锥角为40°,总尖端高度为20-25μm,体电阻率为0.01-0.05ohm-cm。
表4中给出的表面粗糙度的值是根据ISO25178系列定义的表面区域粗糙度参数的算术平均高度。
通过分析傅里叶幅值谱计算不同角度的分形维数;对于不同的角度提取傅里叶幅值谱并计算频率和振幅坐标的对数值。然后采用以下公式计算每个方向的分形维数D
D=(6+s)/2,
其中s是双对数曲线的(负)斜率。报告的分形维数是所有方向的平均值。
也可以采用双对数函数由2D傅里叶谱对分形维数进行评估。如果表面是分形的,那么双对数图应当是高度线性的,并具有负斜率(参见,例如,Fractal Surfaces,JohnC.Russ,Springer-Verlag New York,LLC,July,2008)。
实施例3
制造了包括纳米图案表面的微针阵列。首先,通过光刻工艺在硅晶片上制造了如图2所示的微针阵列。每个微针都含有两个相对放置的侧通道,所述通道与微针底部的一个穿透的模孔(在图2中不可见)对齐。
根据典型的微机械过程在硅基晶片上制造微针。根据形成微针阵列的标准方法,晶片在被用抗蚀剂和/或氧化层打底后,接着进行选择性蚀刻(氧化物蚀刻、DRIE蚀刻、各向同性蚀刻(iso etching)),抗蚀剂剥离,氧化物剥离,和石印术技术(例如,各向同性石印术(iso lithography)、孔石印术、狭缝石印术)。
在形成微针阵列后,将如上实施例1中所述的含有在其上形成DN2图案的5μm聚丙烯膜覆盖在微针阵列上,所述聚丙烯膜的特征在表4的样品2中描述过。在升高的温度下(130℃)将晶片/薄膜结构置于加热的真空箱(3英寸H2O真空)上1小时,然后在维持膜表面纳米图案的同时温和地将膜敷在微针的表面。
图34A-34D说明了在增加的放大倍率下的微针阵列顶端上方的膜。
虽然参考具体实施方案对本主题进行了详细描述,但应懂得本领域技术人员在理解前述内容之后可以容易地构思这些实施方案的变化、变体和等价方式。因此,认为本公开的范围为所附权利要求及其任意等价方式。
Claims (28)
1.一种用于穿过皮肤屏障递送生物活性剂的装置,所述装置包括:
微针以及在微针表面上制造的多个纳米结构,所述纳米结构以预定的图案排列,其中至少一部分纳米结构的横截面尺寸小于500纳米且大于5纳米,并且纵横比为0.2至5,其中所述微针还包含通道;和
与所述微针的通道流体连通且包含组合物的储层,所述组合物的粘度大于5厘泊且包含生物活性剂。
2.根据权利要求1所述的装置,其中所述组合物包含的生物活性剂的浓度为大于5mg/mL。
3.根据权利要求2所述的装置,其中所述组合物包含的生物活性剂的浓度为35mg/mL至500mg/mL。
4.根据权利要求1或2所述的装置,其中所述生物活性剂的分子量大于100kDa。
5.根据权利要求1所述的装置,其中所述生物活性剂为蛋白治疗剂。
6.根据权利要求1所述的装置,所述组合物还包含一种或多种药学上可接受的赋形剂。
7.根据权利要求6所述的装置,其中所述药学上可接受的赋形剂选自溶剂、促渗剂、增稠剂、润湿剂、润滑剂和润肤剂。
8.根据权利要求1所述的装置,其中所述图案还包括微米结构,其中所述纳米结构具有比所述微米结构更小的横截面尺寸。
9.根据权利要求8所述的装置,其中所述微米结构的横截面尺寸大于500纳米,所述纳米结构的横截面尺寸小于300纳米。
10.根据权利要求8或9所述的装置,其还包括第二纳米结构,所述第二纳米结构的横截面尺寸小于微米结构的横截面尺寸并且大于第一纳米结构的横截面尺寸。
11.根据权利要求1所述的装置,其中至少一部分纳米结构具有一个或多个以下特征:
a)中心间距为50纳米至1微米;
b)高度为10纳米至1微米;
c)纵横比为0.5至3.5。
12.根据权利要求1所述的装置,其中所述图案具有大于1的分形维数。
13.根据权利要求1所述的装置,其中所述含有多个纳米结构的微针表面具有10纳米至200纳米的平均表面粗糙度和/或4MPa至320MPa的有效剪切模量。
14.根据权利要求1所述的装置,其中至少一部分纳米结构的横截面尺寸为100纳米至300纳米。
15.根据权利要求1所述的装置,其中所述纳米结构具有大致相同的横截面尺寸。
16.根据权利要求1所述的装置,其中两个相邻的纳米结构的横截面尺寸与这两个结构之间的中心间距之比为1:1至1:4。
17.根据权利要求1所述的装置,其中至少一部分纳米结构具有等距离的间距。
18.根据权利要求1所述的装置,其中至少一部分纳米结构是柱状物形式的。
19.根据权利要求1所述的装置,其中所述通道的横截面尺寸是1至100微米。
20.根据权利要求1所述的装置,其中所述通道的长度是10至800微米。
21.根据权利要求1所述的装置,其中所述装置包含含有所述微针的微针阵列。
22.根据权利要求21所述的装置,其中所述阵列包含具有孔的底部,其中所述孔至少部分地与所述微针的通道对齐。
23.根据权利要求22所述的装置,其中所述储层与所述阵列位于其上的基底连接。
24.根据权利要求22所述的装置,其中所述储层与所述阵列可拆卸地连接。
25.根据权利要求8所述的装置,其中至少一部分微米结构的横截面尺寸大于500纳米且小于10微米,并且其中至少一部分纳米结构的横截面尺寸大于5纳米且小于500纳米。
26.根据权利要求8所述的装置,其中至少一部分纳米结构的横截面尺寸为20至400纳米,并且至少一部分微米结构的横截面尺寸为600纳米至1.5微米。
27.根据权利要求8所述的装置,其中至少一部分微米结构的高度为20纳米至1微米。
28.根据权利要求8所述的装置,其中至少一部分微米结构的纵横比为0.2至5。
