Abstract
本发明公开了一种7-去氢胆固醇的分离纯化方法。首先将胆固醇生物合成所得7-去氢胆固醇发酵物质溶解于溶剂正己烷和水中,充分搅拌后进行过滤,过滤后静置分层,得到水相和有机相;对有机相蒸馏除去正己烷,然后加入甲醇加热搅拌,搅拌后抽滤,得到滤液和滤渣;对滤液降温结晶、抽滤,得到胆固醇;将滤渣中加入正己烷和甲醇的混合物加热搅拌,充分搅拌后进行减压蒸馏,蒸馏后所剩物质降温结晶、抽滤,最后得到高纯度产品7-去氢胆固醇。利用本发明能够有效从产物7-去氢胆固醇发酵物质中分离提纯出高纯度7-去氢胆固醇,并且还可以有效回收未转化的胆固醇,使其重新用于再次投料,从而使其原料得到更大程度的利用,符合目前经济可持续发展的需求。

[0002] 7-去氢胆固醇(7-脱氢胆固醇)是一种固醇类物质。存在于动物皮肤内的皮脂腺及其分泌物中。7-去氢胆固醇为白色结晶粉末或淡黄色结晶粉末,其分子式为C27H44O,亦称维生素DJI。在人体内可由胆固醇转化而成。它储存于皮下,在日光或紫外线照射下可转变为维生素D3,具有调节钙、磷代谢的生物活性。因此,小儿多晒太阳或按计划进行紫外线照射可预防小儿佝偻病。

[0004]目前,7-去氢胆固醇的制备方法通常为:一是7-去氢胆固醇由猪皮中分离提取;二是利用胆固醇经酰化、氧化、腙化、脱腙、皂化得到,7-去氢胆固醇皂化反应合成的一种医药中间体。利用胆固醇生物转化合成7-去氢胆固醇是目前已研制成功的合成方法,但并未进行大规模的工业化生产。

[0008] 本发明提供一种7-去氢胆固醇的分离纯化方法,所述分离纯化方法包括以下步骤:
[0009] a、利用现有胆固醇生物转化合成方法得到的7-去氢胆固醇发酵物质中含有酵母菌培养基、未转化的原料胆固醇和7-去氢胆固醇;首先将所得7-去氢胆固醇发酵物质溶解于溶剂正己烷中,并加入水,所述正己烷和水的加入量均为7-去氢胆固醇发酵物质体积的5〜10倍,加入正己烷和水后在常温下搅拌I〜1.5h ;搅拌后进行过滤,除去不溶于水和正己烷的杂质;所得滤液静置分层,然后进行分液,分出水相和有机相;分液后采用正己烷洗涤所得水相2〜3次,合并有机相;

7-去氢胆固醇,并且还可以有效回收未转化的胆固醇,使其重新用于再次投料,从而使其原料得到更大程度的利用,符合目前经济可持续发展的需求。
[0021] 2、利用本发明技术方案能够得到纯度大于98%的7-去氢胆固醇产品,更好地满足市场对其产品纯度较高的要求。
[0022] 3、目前采用化学法生产7-去氢胆固醇工艺过程中要用到8种有毒有害的有机溶剂,而本发明分离提纯的原料是采用生物转化法合成的,而生物转化法合成过程中仅在产物分离阶段用到少量的(一般是两种)有机溶剂。减少了污染物排放,保护了自然环境,符合世界上生物发酵生产潮流。
[0023] 4、本发明对市场上销售的7-去氢胆固醇标准品和本发明所得产品均作了图谱分析,经分析:标准品纯度为92%,本发明产品纯度为98.24% (详见附图1、2)。

[0030] 本发明7-去氢胆固醇的分离纯化方法,该分离纯化方法的详细步骤如下:
[0031] a、利用现有胆固醇生物转化合成方法得到的7-去氢胆固醇发酵物质中含有酵母菌培养基、未转化的原料胆固醇和7-去氢胆固醇;首先将所得7-去氢胆固醇发酵物质溶解于溶剂正己烷中,并加入水,所述正己烷和水的加入量均为7-去氢胆固醇发酵物质体积的8倍,加入正己烷和水后在常温下搅拌1.2h ;搅拌后进行过滤,除去不溶于水和正己烷的杂质;所得滤液静置分层,然后进行分液,分出水相和有机相;分液后采用正己烷洗涤所得水相3次,合并有机相;
[0032] b、将步骤a得到的有机相进行蒸馏,除去溶剂正己烷,得到的产物中加入50倍体积的甲醇,加热至65°C,在此条件下搅拌1.8小时,搅拌后进行抽滤,抽滤时真空度为0.09MPa、温度为20°C,抽滤后得到滤液和滤渣;
[0033] C、将步骤b得到的滤液降温至室温进行结晶12h,结晶后进行抽滤,抽滤时真空度为0.09MPa、温度为常温,得到胆固醇;
[0034] d、将步骤b得到的滤渣加入正己烷和甲醇的混合物中,所述滤渣与正己烷和甲醇的混合物二者之间的体积比为1:10,所述正己烷和甲醇混合时二者之间的体积比1:1 ;然后升温至45°c,在此温度条件下搅拌I小时;搅拌后所得液体在真空度0.09MPa、15°C条件下进行蒸馏,蒸馏至原液体体积的33% ;然后降温至0°C进行结晶5小时,结晶后进行抽滤,抽滤时真空度为0.09 MPa、温度为(TC,最后得到高纯度产品7-去氢胆固醇(所得产品纯度为98.24%,详见附图2)。

[0037] a、利用现有胆固醇生物转化合成方法得到的7-去氢胆固醇发酵物质中含有酵母菌培养基、未转化的原料胆固醇和7-去氢胆固醇;首先将所得7-去氢胆固醇发酵物质溶解于溶剂正己烷中,并加入水,所述正己烷和水的加入量均为7-去氢胆固醇发酵物质体积的10倍,加入正己烷和水后在常温下搅拌Ih ;搅拌后进行过滤,除去不溶于水和正己烷的杂质;所得滤液静置分层,然后进行分液,分出水相和有机相;分液后采用正己烷洗涤所得水相2次,合并有机相;
[0038] b、将步骤a得到的有机相进行蒸馏,除去溶剂正己烷,得到的产物中加入48倍体积的甲醇,加热至64°C,在此条件下搅拌2.0小时,搅拌后进行抽滤,抽滤时真空度为0.09MPa、温度为18°C,抽滤后得到滤液和滤渣;

[0040] d、将步骤b得到的滤渣加入正己烷和甲醇的混合物中,所述滤渣与正己烷和甲醇的混合物二者之间的体积比为1:11,所述正己烷和甲醇混合时二者之间的体积比1:1;然后升温至40°c,在此温度条件下搅拌1.5小时;搅拌后所得液体在真空度0.09MPa、10°C条件下进行蒸馏,蒸馏至原液体体积的30% ;然后降温至0°C进行结晶6小时,结晶后进行抽滤,抽滤时真空度为0.09 MPa、温度为0°C,最后得到高纯度产品7-去氢胆固醇。

[0044] b、将步骤a得到的有机相进行蒸馏,除去溶剂正己烷,得到的产物中加入50倍体积的甲醇,加热至66°C,在此条件下搅拌1.5小时,搅拌后进行抽滤,抽滤时真空度为0.09MPa、温度为15°C,抽滤后得到滤液和滤渣;
[0045] C、将步骤b得到的滤液降温至室温进行结晶14h,结晶后进行抽滤,抽滤时真空度为0.09MPa、温度为常温,得到胆固醇;
[0046] d、将步骤b得到的滤渣加入正己烷和甲醇的混合物中,所述滤渣与正己烷和甲醇的混合物二者之间的体积比为1:12,所述正己烷和甲醇混合时二者之间的体积比1:1 ;然后升温至43°C,在此温度条件下搅拌1.2小时;搅拌后所得液体在真空度0.09MPa、15°C条件下进行蒸馏,蒸馏至原液体体积的35% ;然后降温至0°C进行结晶5小时,结晶后进行抽滤,抽滤时真空度为0.09 MPa、温度为0°C,最后得到高纯度产品7-去氢胆固醇。
Claims (4)
1.一种7-去氢胆固醇的分离纯化方法,其特征在于,所述分离纯化方法包括以下步骤: a、利用现有胆固醇生物转化合成方法得到的7-去氢胆固醇发酵物质中含有酵母菌培养基、未转化的原料胆固醇和7-去氢胆固醇;首先将所得7-去氢胆固醇发酵物质溶解于溶剂正己烷中,并加入水,所述正己烷和水的加入量均为7-去氢胆固醇发酵物质体积的5〜10倍,加入正己烷和水后在常温下搅拌I〜1.5h ;搅拌后进行过滤,除去不溶于水和正己烷的杂质;所得滤液静置分层,然后进行分液,分出水相和有机相;分液后采用正己烷洗涤所得水相2〜3次,合并有机相; b、将步骤a得到的有机相进行蒸馏,除去溶剂正己烷,得到的产物中加入48〜52倍体积的甲醇,加热至64〜66°C,在此条件下搅拌1.5〜2小时,搅拌后进行抽滤,抽滤时真空度为0.09MPa、温度为15〜20°C ;抽滤后得到滤液和滤渣; c、将步骤b得到的滤液降温至室温进行结晶,结晶后进行抽滤,抽滤时真空度为0.09MPa、温度为常温,得到胆固醇; d、将步骤b得到的滤渣加入正己烷和甲醇的混合物中,所述滤渣与正己烷和甲醇的混合物二者之间的体积比为1:10〜12,所述正己烷和甲醇混合时二者之间的体积比1:1 ;然后升温至40°C〜45°C,在此温度条件下搅拌I〜1.5小时;搅拌后所得液体在真空度0.09MPa、10〜15°C条件下进行蒸馏,蒸馏至原液体体积的30〜35% ;然后降温结晶,结晶后进行抽滤,最后得到高纯度产品7-去氢胆固醇; 所述降温结晶为降温至0°C进行结晶5〜6小时;所述结晶后进行抽滤,抽滤时真空度为0.09 MPa、温度为(TC。
2.根据权利要求1所述的7-去氢胆固醇的分离纯化方法,其特征在于:步骤c中所述结晶时的结晶时间为12〜14h。
3.根据权利要求1所述的7-去氢胆固醇的分离纯化方法,其特征在于:步骤d中所述滤渣与正己烷和甲醇的混合物二者之间的体积比为1:10。
4.根据权利要求1所述的7-去氢胆固醇的分离纯化方法,其特征在于:步骤d中所述高纯度产品7-去氢胆固醇的纯度大于98%。
