标准的细胞传感器或致动器通常是基于流细胞计数法且采用电阻抗感测、光散射测量、和化学或免疫染色随后进行光学感测中的一种或其组合。

发明内容

发明详述

在光学感测领域中,微制造已经允许开发微装置来代替基于玻璃毛细管的流动室,和用于使紧凑的光学器件一体化和提供芯片上的样品转运。
细胞(特别是白细胞)的感测和计数是麻烦的,这部分是由于细胞种群数量。对于基于光学感测的微装置中的白细胞区分,通过硅基片 的各向异性湿法蚀刻制造V型槽微通道并且通过二参数光散射对经过稀释的血液进行3部分的白细胞区分,而没有鞘流动。Altendorf,Proceedings of the Int’l Conference on Solid State Sensors and Actuators,v.1,p.531(1997)。

还经常需要稀释避免堵塞样品室,还为了除去在运行样品之前溶解的红细胞,特别是对于电阻抗或光散射检测来说。这些规程中的一些还需要另外的固定缓冲液。
染料
在本公开中,可以将染料(例如吖啶橙(3,6-二甲基胺吖啶,图1)用于区分靶标,例如细胞、细胞器、颗粒、细胞核、分子(包括双链或单链的核酸,例如DNA、或RNA、染色体、还包括合成的形式)。在一个特定的实施方案中,可以在无需使红细胞溶胞或固定细胞样品的情况下来检测、计数或分选白细胞。某些染料,例如吖啶橙,由于快速扩散到细胞中、容易商购得到、以及与常见光源(即,氩气激光和可见范围中的其它广谱光源)和滤光器的激发和发射波长相容性,也是合乎需要的。Kosenow,Acta Haematol.7,217(1952);Schiffer,Blood,19,200(1962);Jackson,Blood,17,643(1961);Hallermann等人,Verh Deutsch Ges Inn Med.70,217(1964)。
吖啶橙是一种对pH敏感的荧光阳离子染料,其通过静电引力结 合于双链DNA并且将吖啶橙插入在碱基对之间。在结合时,激发最大值变为502nm,发射最大值变为525nm(绿色)。吖啶橙还结合于RNA和单链DNA,激发最大值移位到460nm,发射最大值为650nm(红色)。Adams and Kamentsky,Acta Cytol.15,289(1971);Adams and Kamentsky Acta Cytol.18,389-91(1974);Steinkam等人,Acta Cytol.17,113-17(1973)。吖啶橙在其在中性pH时为疏水性的且可以容易地扩散通过细胞膜和细胞核膜以结合于RNA和DNA方面也是合乎需要的。在活细胞中,吖啶橙在溶酶体的酸性环境中被质子化,这使得其为阳离子型的并防止染料从溶酶体膜泄漏出来。Moriyama等人,J.Biochem.92;1333-36(1982)。在吖啶橙用于白细胞分析时,细胞核被染色为绿色带有轻微混合的红色,是由于双链DNA和单链DNA引起的,而细胞质被染色为红色,是由于RNA和溶酶体。因此,可以通过使用绿色荧光通道的强信号容易地实现白细胞计数。可以通过分析得自红色荧光通道的信号来实现白细胞区分。

装置
本公开的一个实施方案涉及用于细胞计数和/或分化的设备或装置。在特定的实施方案中,所述设备或装置包括由例如硅、玻璃、塑料、金属、或其它材料的材料形成的基板。本公开的一个特定的实施方案是使用软石板印刷术(soft lithography)制造的。Quake,Science290,1536-40(2000)。根据具体的实施方案,也可以使用其它光刻或蚀刻技术。
装置的一个实施方案是使用以10:1比例剧烈混合的两部分PDMS(聚二甲硅氧烷)(Sylgard184,Dow Corning,MI,USA)微制造的(microfabricated)。在真空脱气约30分钟之后,将混合物倾倒在DRIE蚀刻的硅模具上,所述模具已经用HMDS(六甲基二硅氮烷)预先处理以便在烘焙之后容易分离。将模具在80℃烘焙30分钟。将硬化的PDMS从硅模具上分离,并将PDMS片材切割成碎片并在每个碎片上用Luer短柱适配器(Becton Dickinson,NJ,USA)刺扎射流进入孔。将每个PDMS碎片小心地置于经过净化的载玻片上并在80℃烘焙过夜。在一些情况中,对PDMS和载玻片进行氧气等离子体处理(300mTorr,25W,30s),以便改善它们之间的粘着,特别是对于意在重复利用的装置。Bhattacharya等人,J.Microelectromechan.Syst.14,590-97(2005)。
在一个特定的实施方案中,在1cm x1cm PDMS块上模制通道结构,PDMS块的厚度小于3mm。在一个特定的实施方案,通道深度为16μm,以便容纳大的白细胞。

装置的某些实施方案使用聚焦的激光源用于照射,因为非常期望细胞聚集在检测区域240中。然而,本公开中所包括的其它实施方案使用更均匀地散射的光源和缝隙孔径。这种实施方案利用直的通道几何形状而不用细胞集中。在一个实施方案中,检测区域240的通道长度为1000μm。在某些实施方案中,还可以在在样品流动区域220的上游包括过滤器结构230,其将包括红细胞叠连和其它大粒子聚集物在内的污染物过滤掉,以防止检测区域240中的堵塞。在某些实施方案中,过滤器结构230的矩形基柱结构部件的大小为200μmx40μm。在三列基柱中的每一列之间的间距分别为40μm、30μm、和20μm,其甚至允许最大的白细胞通过过滤器区域230。

如图3中所示,在光具座上建立光学系统(透射的激光诱导的荧光检测系统或LIF)。在一个特定的实施方案中,系统设置包括激发或激光光源300、透镜组件340、微流体装置350、可选的另外的透镜组件360、过滤器组件320、330、和图像采集装置390、395。在某些实施方案中,一个或多个发射过滤器包括过滤器组件320、330,在某些实施方案中,图像采集装置395包括电荷耦合器件(CCD)照相机、互补型金属氧化物半导体(CMOS)装置、或光电倍增管(PMT)装置。在特定的实施方案中,图像采集装置395可以偶接为与显示单元或计算装置396例如个人计算机相通讯。本领域技术人员会认识到,有多种和不同的计算机软件程序可用于从系统接受到的数据的积分、汇编、分析、再组合、和其它操作,特别是借助于计算装置396。
在一个特定的示例性实施方案中,将氩气激光(National Laser NLC210BL,488nm,和15-30mW可调,Salt Lake City,UT,USA)用作激发源。将直径50μm的孔口310置于激光输出前面,以促进对准过程和降低照射强度。在某些实施方案中,可选的激光-线带通过滤器(带宽等于约1.9nm,中心波长处于488nm)用于进一步纯化激光光源。在某些其它实施方案中,可选的中性密度过滤器(NDF)用于衰减激光激发。或者,针眼和NDF被两个线性的玻璃偏光器(Edmond Optics TECH SPEC,Barrington,NJ,USA)代替,以便可以容易地调节装置上的照射水平。
在一个特定的实施方案中,使用长工作距离显微镜物镜(USMCO M Plan Apo,10x,0.28NA,Dayton,NV,USA)作为聚光透镜340。使用另一个长工作距离的显微镜物镜(Bausch&Lomb,50x,0.45NA,Rochester,NY,USA)作为物镜360。在相同的实施方案中,在一个特定的试验中使用三个发射过滤器320:488nm长通过滤器(Chroma H1500LP,转换宽度<4.9nm,边沿陡度=2.5nm,Rockingham,VT,USA)、中心波长为525nm且频带宽度50nm的绿色带通过滤器(Chroma D525_50m)、和中心波长650nm且频带宽度50nm的红色带通过滤器(Chroma D650_50m)。使用宽带非偏振混频立方体分束器370(Newport05BC17MB.1,400-700nm,R/T=45%/45%,Irvine,CA,USA)来将光同时引导到光电二极管检测器380和CCD摄像机。
信号经过电学放大并用硅光电二极管接收机组件390(Electro-Optical Systems,UVS-025-H,Phoenixville,PA,USA)或光子倍增管(PMT,Hamamatsu H5784-20,Japan)检测。电压信号被送往深记忆示波器(deep memory oscilloscope,HP54645A,PaloAlto,CA,USA)。在示波器中的缓冲器充满时,数据可以被加载到计算机并用Matlab峰值-检测程序来分析。可以用模拟的CCD摄像机(Hitachi KP-D20B,Japan)以每秒30帧拍取视频,然后转化为数字格式并储存在计算机396中。在本文中所描述的特定的实施方案中也可以使用其它图像采集装置395,例如CMOS、PMT、或其它装置。在某 些示例性试验运行过程中,利用光电二极管检测器和PMT的系统设置比CCD摄像机更灵敏,具有更快的时间响应。在一个示例性的试验运行过程中,首先借助得自CCD摄像机的图像将光学系统粗糙地对准哑设备(dummy device)。使用适当的对准容易地实现检测区域上的10μm直径的照射斑点。
如图12A和图12B中所示,本发明的装置可以被并入到手持的单元中,所述手持单元包括激光光源(例如激光发射二极管或LED120)、至少一个透镜190、至少一个具有可选的分束器的过滤器组件195、在微芯片或其它基板上的本文中所述的微流体装置185、输入/输出端口130、至少一个图像采集装置100、110(其可以是光电倍增管)。在某些实施方案中,手持单元可以用外罩壳或壳体150、180和铆钉或螺栓140、160组装并包装起来。
细胞检测
本公开的一个方面涉及利用微制造的装置从未经稀释的细胞样品(例如人或其它动物的血液)计数和/或区分细胞(特别是白细胞)的方法。在一个示例性的实施方案中,细胞检测利用吖啶橙和新鲜的全人血来进行。
在一个示例性实施方案中,新鲜的人血液得自健康的供体并且在收集的3天内使用。向血液样品中添加EDTA以防止凝固。对于吖啶橙染色,可以加入储备溶液,以得到在Ficoll-Paque Plus中的10μg/mL的最终染料浓度。Ficoll-Paque Plus还用作鞘流动溶液。荧光的聚苯乙烯小珠(5μm绿色荧光小珠)购自Duke Scientific Corporations,Fremont,CA,USA。细胞核染色吖啶橙得自Molecular Probes,Eugene,OR,USA,并溶解于水中,以实现10mg/mL的储备溶液。血液稀释剂Ficoll-Paque Plus购自Amersham Biosciences,Sweden。磷酸盐缓冲盐水(10x PBS)得自Ambion(9625),Austin,TX,USA。
染色结果在具有三频带过滤器模块DAPI-FITC-TRITC的荧光 显微镜(Nikon E800,Japan)下观察,所述过滤器模块具有波长分别为385-400nm、475-490nm、和545-565nm的激发过滤器,波长分别为450-465nm、505-535nm和580-620nm的发射过滤器。在冷却的CCD摄像机(RT-KE color3-shot,Diagnostic Instruments,Sterling Heights,MI,USA)拍取图像。使用血球计(HausserScientific,Horsham,PA,USA)进行白细胞的粗略计数。必要时,血液或荧光小珠用Ficoll-Paque Plus(比重1.077g/mL)稀释,以使溶剂的比重与白细胞匹配。所有的流体使用注射器泵(Harvard Apparatus Pico Plus,Holliston,MA,USA)泵送到装置中。
在这个特定的实施方案中,使用模拟的CCD摄像机以匹配的3nL/min样品流速和30nL/min鞘流速的照相机帧速率来进行视频记录。对于光电二极管检测,使用0.1μL/分钟的样品流速和1μL/分钟的鞘流速。对于光子倍增管器具使用1μL/分钟样品流动和10μL/分钟鞘流动。
为了实现高的信噪比,使用本领域中的常规方法建立用于细胞染色的最大浓度。Adams and Kamentsky,Acta Cytol.15;289(1971)。如图4中所示,使用不同的吖啶橙浓度分析全血样品。白细胞染色的最佳浓度测定为大约1μg/mL。在图4中利用的特定的示例性实施方案中,盖玻片和格栅表面之间的距离为大约100μm。如图4A中可见,在视场下存在有许多的红细胞,而这些细胞不妨碍来自白细胞的荧光信号,如图4B-F中所示。
如图5中可见,在细胞检测中利用的示例性实施方案未经过任何显著的光致漂白。信号拟合为一级指数式衰减,时间常数为6.4+/-0.7秒。另外两个试验证实了对于一个特定的实施方案的光致漂白时间常数为1秒到10秒之间。通过用吖啶橙染色的全血填充装置来表征一个特定的实施方案的光致漂白时间常数。通过激光焦点扫描通道并将照度设置为与用于试验的相同。整个过程用CCD摄像机记录。当观察到发荧光的白细胞具有与背景明显不同的荧光发射时,停止移动激光焦点并且等待,直到白细胞光退色到背景水平。从视频提取图像, 转化为8位灰色标度图像,并用Matlab程序分析。数据拟合为单独的时间常数指数式衰减。
另外,在约2x103/μL的浓度试验绿色荧光小珠,通过CCD摄像机观察,并显示在图6止。样品流速设置为约3nL/min,鞘流动为约30nL/min。在一个示例性试验运行中,如图6A所示的水动力学聚焦的激光束产生如图6B所示的扩大的光环。通常在每个图像中仅出现一个单个的小珠。使用散射的激光照射,如图6C中所示,可以鉴定小珠的踪迹,如图6D中所示。水动力聚焦限制检测区域的横截面积,但不缩小孔道直径,因此可以改善信噪比而不增加堵塞通道的危险。另外,中心流动的横截面的缩减减少了重合效应。最后,用鞘流动围绕中心样品流动使得装置壁中的荧光染料吸收最小化,由此减少背景噪声。如图7中所示,可以容易地鉴定得自光电二极管检测器的小珠信号。
如图8中所示,使用红色和绿色发射过滤器,从CCD摄像机拍摄的视频提取的图像都表现出得自用吖啶橙染色的白细胞鉴定的信号,以及得自荧光对照小珠的信号。对于光电二极管检测,对于正常个体的预期的白细胞检测速率为每秒平均为约4-11个细胞。
在一个示例性实施方案中,使用绿色发射过滤器进行用吖啶橙染色的未经稀释的血液样品的超过50秒的时间追踪,实现了每秒最多约1000个白细胞的处理量。通过描绘柱状图研究得自具有525nm发射过滤器的绿色荧光通道的最大信号强度(图9中所示的峰高),如图10中所示。正如所料,较低强度部分可能主要由淋巴细胞提供,而较高强度部分可能主要来自单核细胞,中心区域可能大部分来自粒细胞。Steinkam等人,Acta Cytol.17;113-117(1973)。
在一个示例性实施方案中,使用具有650nm发射过滤器的红色荧光通道进行了用吖啶橙染色的未经稀释的血液样品的超过50秒的时间追踪。如图11中所示,鉴定了两个峰,较低强度主要是淋巴细胞,较高强度峰基本上是单核细胞和粒细胞。细胞染色开始到光电二极管记录之间的时间典型地为大于15分钟。
在两个示例性研究中,利用PMT检测器的一个实施方案的最大的处理量为约每秒1000个白细胞。使用未经稀释的血液,保持了最小的样品体积,增加了处理量。因为试样处理量与体积流率成比例,但是受到最大泵送速率和传感系统的响应时间的限制,在CCD摄像机探测时使用3nL/分钟的中心流动速率。在这个流动速率,典型的白细胞在大约30毫秒内穿过检测区域,这个时间与CCD的帧采集时间大致相等。
对于光电二极管检测,用于各个实施方案的流动速率可以适合地为大约1nL/分钟、大约2nL/分钟、大约3nL/分钟、大约4nL/分钟、大约5nL/分钟、大约6nL/分钟、大约7nL/分钟、大约8nL/分钟、大约9nL/分钟、大约10nL/分钟、大约20nL/分钟、大约30nL/分钟、大约40nL/分钟、大约50nL/分钟、大约60nL/分钟、大约70nL/分钟、大约80nL/分钟、大约90nL/分钟、大约100nL/分钟、大约110nL/分钟、大约120nL/分钟、大约130nL/分钟、大约140nL/分钟、大约150nL/分钟、或其之间的任何数值。同样地,对于PMT检测,用于各个实施方案的流动速率可以适当地为大约200nL/分钟、大约300nL/分钟、大约400nL/分钟、大约500nL/分钟、大约600nL/分钟、大约700nL/分钟、大约800nL/分钟、大约900nL/分钟、大约1μL/分钟、大约2μL/分钟、大约3μL/分钟、大约4μL/分钟、大约5μL/分钟的范围,或其之间的任何数值。
在一个示例性实施方案中,光电二极管接收器模块在低灵敏度设置时的时间响应为0.16毫秒,在高灵敏度设置时为0.6毫秒,而PMT检测器在一个示例性运行中的时间响应为约16微秒。
此外,通过减小横截面积,中心流动的线性流动速度增加(其需要更快的感测),并且通过增加检测区域中细胞之间的平均距离减少了重合效应。
因此,通过利用本文中公开的与微流体装置有关的特定的实施方案,可以在微流动细胞计数器系统中一个接一个地实现白细胞感测、计数、和分选。此外,在本文中描述的某些实施方案中可以使用浓稠 的细胞悬液(例如完整的、未经稀释的血液),这提供了减少样品和废物体积、缩短处理时间、和完全消除芯片上混合和缓冲液储存。在特别的方面中,可以在微流动细胞计数器系统中一个接一个地检测白细胞。
如本文中所述,装置的某些实施方案可以以不同的规格和结构实施,包括但不限于台上装置、手持装置(如图12中所示的)、可植入装置、纳米技术装置、或其它规格或结构。在较小的示例性结构中,高照度LED被用于激发,微型泵用于以抽吸模式操纵样品,而来自绿色和红色通道的荧光信号可以同时检测。

Claims (19)
1.用于白细胞检测的方法,所述方法包括:
a)使包含荧光标记的白细胞和荧光对照小珠的血样流动通过微流体装置;所述微流体装置具有:
入口,逐渐缩窄并包含与第二通道并列的限制性进口的第一通道;包含检测区的第二通道,其中所述第二通道具有40μm-50μm的固定尺寸;和出口;
其中所述入口、第一通道、第二通道和出口流体连通;其中所述荧光对照小珠具有的荧光团不同于荧光标记的细胞的荧光团;所述血样通过微流体装置的流动包括使用泵;
b)使用激发源,激发荧光标记的白细胞和荧光对照小珠;和
c)测定荧光标记的白细胞和荧光对照小珠的荧光。
2.权利要求1的方法,包括使用图像采集装置采集图像,其中所述图像采集装置包括光电倍增管。
3.权利要求1的方法,其中所述血样是全血。
4.权利要求1的方法,其中所述方法进一步包括一个接一个地感测细胞。
